Development of Water Soluble Tocopherol Emulsion Using Surfactants (계면활성제를 이용한 수분산 Tocopherol 유화물의 개발)

  • This study was carried out to make water-soluble tocopherol emulsion which can be applicable directly in water. The molecular weight of tocopherol was 340 to 360 and tocopherol emulsion model was decided as O/O/W/W type. In correlation between stability of emulsion and surface tension, the stability in surface tension of emulsion was from 40 to 46 dyne/cm. In the case of lower than 40 dyne/cm of surface tension, the stability of the emulsion was lower. Lipophilic surfactants, especially for a polyglycerine polyricinoleate in 20%, 30% and 40% tocopherol emulsion, was the most effective in emulsion stability. A higher stability of the emulsion among hydrophilic surfactants in the tocopherol emulsion was obtained in the following order; polyglycerine monostearate> polyglycerine monooleate> polyoxyethylene (20) sorbitan monooleate$\geq$ polyoxyethylene (20) sorbitan monolaurate.

  • NAD(P)H quinone oxidoreductase 1 (NQO1) is a flavoprotein that catalyzes the two electron reduction of diverse substrates, including quinones. It uses NADH or NADPH as a cofactor for enzymatic machinery. In the metabolism of quinones, NQO1 has two conflicting functions because of the different stability of converted hydroquinones. The stable form of hydroquinone is excreted from cells by conjugation with glutathione or glucuronic acid. The unstable form of hydroquinone induces cell death by induction of oxidative stress and DNA damage. Certain quinones known as bio-reductive agents have a cytotoxic function following reduction by NQO1. Bio-reductive agents, such as ${\beta}$-lapachone or mitomycin C, induce the depletion of NAD(P)H and the generation of oxidative stress in an NQO1-dependent manner. NQO1 is highly expressed in several cancer tissues. Therefore, NQO1 is a good therapeutic target for cancer treatment with bio-reductive agents.

Isolation and Identification of Bacteria Lysing Anabaena cylindrica (Anabaena cylindrica 분해세균의 분리 및 동정)

  • To isolate the bacteria lysing cyanobacteria, the sediment samples were collected from Dochang and Pal'tang Reservoir and Seokchon Lake. Each sample was smeared on the Anabaena cylindrica lawn and incubated in light chamber for 11 days. Bacteria having cyanobacteria-Iysing activity were isolated from the samples of Seokchon reservoir. Confirmation of cyanobacteria-Iysing activity was carried out to measure chlorophyll a and bacterial cell counting in mixed culture of Anabaena cylindrica and bacteria. Lysis was detected when extracellular meterials was added to the Anabaena cylindrica culture. The isolate was identified by analysis based on 16S rDNA sequence and morphological and physiological properties. The bacterial strain was taxonomically studied by the phylogenetic analysis based on 165 rDNA sequence. This strain was identified as a member of the genus Bacillus and designated as Bacillus sp. CHS1.

Sequences and Phylogenic Analysis of Squid New Kinesin Superfamily Proteins (KIFs) (오징어과의 Kinesin Superfamily Proteins (KIFs)의 유전자분석 및 계통분석)

  • The movement of vesicles from the neuronal cell body to specific destinations requires molecular motors. The squid giant axon represents a powerful model for studies of the axonal transport mechanism because the axoplasm can readily be separated from the sheath by simple extrusion. In a previous study, vesicular movements in the axoplasm of the squid giant axon were inhibited by the kinesin antibody. In the present study, we cloned and sequenced the cDNAs for squid brain KIFs. Amplification of the conserved nucleotide sequences of the motor domain by polymerase chain reaction (PCR) using first-strand cDNAs of the squid optic lobe identified six new KIF proteins. Motif analysis of the motor domains revealed that the squid KIFs are homologous to the consensus sequences of the mouse KIFs. The phylogenetic tree generated by using the maximum parsimony (MP) method, the neighbor-joining (NJ) method, the minimum evolution (ME) method, and the maximum likelihood (ML) method showed that squid KIFs are closest to mouse KIFs. These data prove the phylogenetic relationships between squid KIFs and mouse ones.

Identification of Pseudocercospora bolleana Associated with Angular Leaf Spot on Common Fig in Korea (무화과나무의 모무늬잎마름 증상에 관여하는 Pseudocercospora bolleana 동정)

  • A cercosporoid fungus associated with angular leaf spots on the leaves of common fig (Ficus carica) in Korea is known to be morphologically similar to Passalora, but phylogenetically similar to Pseudocercospora. To clarify the ambiguity, six fig samples with angular leaf spots were collected and examined using a microscope, and two representative isolates were sequenced for multiple genes. The morphological characteristics were consistent with previous descriptions of Passalora bolleana. Molecular phylogenetic analysis based on the internal transcribed spacer and large subunit ribosomal DNA (rDNA) regions showed that the Korean isolates, as well as previously published Korean and Romanian isolates, formed a well-supported group in the clade of Pseudocercospora species. Consequently, the current Korean isolates should be correctly described as Pseudocercospora bolleana. Additionally, Pseudocercospora fici-caricae, a cercosporoid fungus previously described as a leaf pathogen on common fig in Taiwan and Korea, was also compared and discussed.

Isolation and characterization of 6 unrecorded Pseudomonas spp. from Korean soil (토양에서 분리한 국내 미기록종 Pseudomonas 속 6종의 생화학적 특성과 계통 분류)

  • In 2017, as a study to discover indigenous prokaryotic species in Korea, a total of 6 bacterial strains assigned to the genus Pseudomonas were isolated from soil. From the high 16S rRNA gene sequence similarity (${\geq}99.5%$) and phylogenetic analysis with closely related species, the isolated strains were identified as independent Pseudomonas species which were unrecorded in Korea. The six Pseudomonas species were Pseudomonas mandelii, P. canadensis, P. thivervalensis, P. jessenii, P. lurida, and P. brenneri. Gram reaction, culture conditions, colony and cell morphology, basic physiological and biochemical characteristics are described in the species description section.

A Taxonomic Reconsideration of the Genus Lemna L. (Lemnaceae) in Korea (한국산 좀개구리밥속(개구리밥과)의 분류학적 실체에 대한 재고)

  • Duckweed family (Lemnaceae Martinov), including the genus Lemna L., is a typical floating aquatic perennial plant, and about five genera and 40 species in the family are in wide distribution around the world except the polar regions. The genus Lemna is the smallest and the simplest plant among the angiosperms. It has a characteristic of doubling every three days with fast vegetative propagation, which helps the organisms to increase in rapid growth. As such, the plant is ideal for environmental pollution assessment and toxicity test. Although taxonomists and scholars have used different scientific names for the species, many of them have agreed that there is only one member of species of the genus Lemna in Korea. Paying attention to the external morphological variation observed in the Korean genus Lemna, we conducted a molecular phylogenetic analysis to identify the entity of the Korean Lemna species and to investigate the possibility of two or more members of the species existing in Korea. We determined and aligned the DNA sequences of the atpF-H region of the chloroplast DNA in 37 populations of the nationally distributed Lemna species. The results showed that the sequence length of the cp DNA atpF-H region was 463-483 bp, the length of the aligned sequences was 488 bp, and the number of variation site in nucleotide sequences was 47. There were two types of aligned sequences of the cp DNA atpF-H region from 37 populations of Lemna species in Korea. The maximum parsimony analysis revealed that the Korean Lemna consists of two clades, and one of them had two subclades. The results suggest that, contrary to the general understanding, at least two taxa (L.aequinoctialis, L.minor) exist in Korea.

Cloning of Acetate Kinase Gene from the Copepod Paracyclopina nana and its Expression in Escherichia coli (요각류 Paracyclopina nana Acetate Kinase의 클로닝 및 대장균에서의 발현)

  • The acetate kinase gene from the copepod Paracyclopina nana was cloned. The open reading frame (ORF) was 1,200 bp, and poly(A) signal sequence was located in the end of the ORF. After the molecular phylogenetic analysis of P nana acetate kinase gene, it was revealed that it formed the same branch with that of Aspergillus. Also P. nana acetate kinase showed the difference with those of other prokaryotic microorganisms but showed the same clade with those of fungi. We also confirmed that the recombinant protein of P. nana acetate kinase made approximately 50 kDa after expression of recombinant gene construct in E. coli. This may be useful to compare this protein to those of other organisms in biochemical characteristics.

Development of specific SNP molecular marker from Thistle using DNA sequences of ITS region (엉겅퀴의 ITS 영역 염기서열 분석을 통한 특이적 SNP 분자마커의 개발)

  • Thistle is a perennial plant that is widely used for medicinal purposes. Information on the genetic diversity of thistle populations are great important for their conservation and germ plasmic utilization. Although thistle is an important medicinal plant species registered in South Korea, no molecular markers are currently available to distinguish them from other similar species from different countries. In this study, we developed single nucleotide polymorphism (SNP) markers derived from the nuclear ribosomal DNA internal transcribed spacer (ITS) regions of genomic sequences to identify distinct Korean-specific thistle species via an amplification refractory mutation system (ARMS)-PCR and high resolution melting (HRM) curve analyses. We performed molecular authentication of four different kinds of thistle species from different regions using DNA sequences in the ITS intergenic region. We also developed a quantitative PCR assay using species-specific ITS primers, which allowed us to estimate the ratio of Korean-specific thistle species using varying ratios of mixed genomic DNA templates from the two species. The SNP markers developed in this study are useful for rapidly identifying specific thistle species from different countries.

Studies on Mild Mutants of Tobacco Mosaic Virus II. Biochemical Properties of Ribonucleic Acid and Coat Protein (약독 담배모자이크바이러스 II. RNA 및 외피단백질의 특성)

  • The biochemical properties of ribonucleic acid (RNA) and coat protein of the mild tobacco mosaic virus (TMV) mutant, Tw 333 are described. The molecular weight of the RNA calculated from polyacrylamide gel electrophoresis was $2.03\times10^6$ daltons. The molar ratio of the bases of the RNA was 25.4 guanine, 29.2 adenine, 17.5 cytosine and 27.9 uracil in moles. The hyperchromicity on Tw 333-RNA by thermal denaturation was $25.1\%$, indicating Tm value of $47^{\circ}C$. The virus coat protein migrated as a single component in SDS-polyacrylamide gel electrophoresis and had a molecular weight of 17,500 daltons. A total of 158 amino acid residues are present in the protein. Separation of the tryptic peptides by electrophoresis and chromatography yielded ninhydrin-positive compounds. The biochemical properties of RNA and coat protein of the mild mutant we very similar to those of wild type of TMV-OM strain, but some difference between the strains were observe in the base composition, hyperchromicity, amino acid composition and tryptic peptide map.
