Multi-locus Phylogeny Analysis of Korean Isolates of Phytophthora Species Based on Sequence of Ribosomal and Mitochondrial DNA (핵 및 미토콘드리아 DNA 염기서열을 이용한 국내 Phytophthora 속의 Multi-locus phylogeny 분석)

  • To investigate genetic relationships either interspecies or intraspecies of 14 Korean Phytophthora species, sequence analyses of nuclear DNA (ypt gene and rDNA-IGS region) and mitochondrial DNA (Cox gene, $\beta$-tubuline gene, and EF1A gene) were performed. All of 14 Korean Phytophthora species clearly clustered into foreign isolates of each species. These Korean isolates in Phytophthora species also showed no correlation between molecular classification and morphological classification like as in case of foreigners. P. palmivora KACC 40167 reported previously from genetic groups of Phytophthora species in Korea was not consistent with the classification system, and therefore was required re-examination for the genetic group analysis. Korean isolates of P. drechsleri KACC 40195 showed very close relationship with P. cryptogea KACC 40161 above 94% bootstrap value in P. cryptogea-P. drechsleri complex group. Identification of these isolates is still unclear, because P. cryptogea and P. drechsleri were not differentiated in this study. On the other hand, it was required to unify species for these two species, since P. parasitica and P. nicotianae were clustered into a group on the level of 99 to 100% sequence homology. Comparing to the sequences of foreigners, Korean isolates were newly divided to ten groups in the phylogenic system. These results could be prepared useful informations to understand genetic diversity of Phytophthora species in Korea.

Intraspecific variation and geographic study of Lonicera insularis (Caprifoliaceae) based on chloroplast DNA sequences (엽록체 DNA를 이용한 섬괴불나무(Lonicera insularis Nakai)의 종내변이 및 지리학적 연구)

  • Lonicera insularis Nakai (Caprifoliaceae) is Korean endemic plant that lives along the shore of Ulleungdo and Dokdo. The aim of this study is to construct a phylogenetic relationship within six species (L.maackii, L.chrysantha, L.subsessilis, L. harai, L. morrowii) of genus Lonicera L. and Wigela subsessilis as outgroup and intraspecific variation of L. insularis using chloroplast DNA five regions sequences. Sequence analysis revealed that both L. insularis and L. morrowii showed complete homologies in the intergenic regions of trnL-trnF, trnS-trnG, psbM-trnD and matK coding region. However, sequence in the petN-psbM intergenic region showed a single nucleotide difference between both species, thus we designated them as CP01 and CP02. The plants having CP01 are prevalent in the Ulleungdo and Dokdo, while L. insularis and L. morrowii from Ulleungdo and of Dokdo, and Japan have CP02. This confirmed the existence of two cp DNA lineages with different geographical distributions. We can infer the allopatric speciation by geographical barrier. The result will provide the important basal data to study speciation and specie evolution of ocean islands such as Ulleungdo and Dokdo.

An Ecological Analysis of Lichens Distributed in Rocks of Coast and Field in U-do by Molecular Technique (분자생물학적 기법에 의한 우도해안과 노지암석에 분포하는 지의류의 생태학적 분석)

  • This study was initially performed to obtain fundamental data on lichen distribution depending on environmental condiLichens distributed on coastal rocks atleast 20 m far away from the coast were also investigated. Analyof morphological characteristics and rDNA ITS clones revealed that lichens containing 9 families and 15 genera were distributed in coastal rocks of V-do, while lichens containing 10 families and 14 genera were in field rocks, demonstrating little difference of diversity between both sites. Foliose lichens such as PhaeophysPhyscia, Pyxine belonging to the family Physciaceae were representatives distributed in coastal rocks of V-do, along with Xanthoparmelia belonging to Parmeliaceae, fruticose lichens such as Ramalina to Ramaliand crustose lichen Lecanora to Lecanoraceae. In contrast, foliose lichen Cladonia belonging to the family Cladoniaceae, fruitcose lichen Stereocaulon to Streocaulaceae, and crustose lichen Porpidia to Porpidwere found only on the surface of field rocks. Crustose lichens containing genera Caloplaca, CanDirinaria, Graphis, Rhizocarpon, and Pertusaria were rarely distributed in coastal rocks, and most of them were also found in field rocks.

  • In this study molecular genetic analysis was carried out on 4 human skeletal remains from Osuri, Buyeo. We showed that real-time PCR is the method of the choice to assess the initial number of genuine ancient DNA molecules. Human mitochondrial DNA quantification was accomplished by the real-time PCR for the cytochrome b gene of the mitochondria. Histological results proved to be a good potentiality for biochemical analysis using biomolecule. The level of specimen's preservation state was proved that level of quantitative result was BO-04, BO-01, BO-03, BO-02. Continually, we showed that biochemical and biomolecule results for the level of preservation state were similar. This study will be useful to important material for predicting biochemistry and biology analysis of the ancient bone.

이동통신 주파수 대역 전자파에 대한 생체 반응

  • 우리 나라 인구의 절반 정도에 해당하는 수의 이동통신 전화가 사용되고 있다. 이동 전화 사용자의 수는 앞으로 더욱 증가할 것이며, 또한 이를 이용한 각종 정보의 교환이 늘어남으로써 개인당 사용량도 늘어날 것으로 생각된다. 사용빈도의 증가와 더불어 이동전화에 의한 뇌종양, 유방암 및 기형 등 각종 인체 질환 발생 가능성에 대한 우려가 늘어나고 있다. 이러한 이동통신 주파수 대역 전자파는 크게 이미 잘 알려진 전자기장이나 자외선, 방사선 등과 인체에 대한 효과가 비슷할 것이라고 생각할 수 있다. 물론 이들은 주파수 대역이 전혀 다르며 이에 따라 서로 다른 물리적 성질을 가지고 있다. 단순히 보유하고 있는 에너지의 크기만으로 비교할 수 있는 것이 아닌 것이다. 그럼에도 불구하고 자외선이나 방사선처럼 전자파도 인체 질환을 유도할 것으로 생각하고 있다. 전자통신제품 사용의 증가에 따라 전자파에 대한 생체 영향에 대한 연구결과들이 발표되고 있다. 특히 이동통신 사용에 따라 암발생이 증가하고 기형이 발생할 수 있다는 역학 및 생물학 연구 결과들이 있었다. 하지만 이 보다 더 많은 수의 논문들이 전자파와 종양 발생이 관련 없다고 얘기하고 있다. 이에 대한 논란은 더 많은 연구와 토론이 필요할 것이다. 정확한 이동전화 주파수대역 전자파가 생체에서 어떻게 작용하는지 알기 위해서는 전자파 자체의 생물리학적 작용기전에 대한 이해가 필요하다. 전자파에 대한 인체 영향을 정확히 분석하기 위해서는 DNA나 단백질 같은 분자수준에서 세포나 실험동물에서 어떤 변화가 발생하는지 분석하는 것이 필요하다. 세포 수준의 연구는 전자파와 한 종류의 세포의 관계를 직접 분석할 수 있다. 세포내에서 DNA나 단백질의 변화는 바로 조직 및 장기내에서의 변화를 의미한다. 하지만 생체내에서는 하나의 세포가 외부 자극에 반응하는 것보다 면역계, 신경계 및 순환계 등 타 장기나 세포의 영향이 존재하는 상황이다. 따라서 세포 수준에서 전자파만의 효과를 장기나 개체내에서 분석하는 전자파에 의한 종양발생 관계를 더 정확히 알 수 있을 것이다. 전자파 중에서 이동전화 주파수 대역에 대한 생체 연구는 많지 않으나 이들로부터 이 주파수 대역의 전자파에 대한 생체 영향 평가 자료를 많이 모을 수 있다. 이러한 자료를 이용하면 핸드폰과 전자파에 대한 위해성 평가를 할 수 있다. 앞으로 다양한 수준에서 생체 영향을 분석함으로써 전자파 노출에 대한 효과를 알 수 있을 것이다.

Detection of Carbonic Anhydrase in the Gills of Rainbow Trout (Oncorhynchus mykiss) (무지개 송어 rainbow trout, Oncorhynchus mykiss의 아가미에서의 carbonic anhydrase의 존재)

  • Carbonic anhydrase isozymes are a widespread, zinc-containing metalloenzyme family. The enzyme catalyzes the reversible inter-conversion of $CO_2$ and $HCO_3$. This reaction is the main role played by CA enzymes in physiological conditions. This enzyme has been found in virtually all organisms, and at least 16 isozymes have been isolated in mammals. Unlike mammals, there is little information available regarding CA isozymes in the tissues of non-mammalian groups, such as fish. Carbonic anhydrase is very important in the osmotic and acid-base regulation in fish. It is well-known that the gills of fish play the most important role in acid-base relevant ion transfer, the transfer of $H^+$ and/or $HCO_3^-$, for the maintenance of systemic pH. Rainbow trout, Oncorhynchus mykiss, is the most important freshwater fish species in the aquaculture industry of Korea, with annual production increasing each year. In addition, environmental toxicology research has shown that rainbow trout is known to be the species that is most susceptible to environmental toxins. Consequently, carbonic anhydrase was detected in rainbow trout, Oncorhynchus mykiss. The isolated protein showed the specific band with a molecular weight of 30 kDa and pI of 7.0, and it was identified as being carbonic anhydrase. The immunohistochemical result demonstrated that the carbonic anhydrase was located in the epithelial cells of the gills.

Morphogenetic Identification of Eel's Larva (Leptocephalus) Collected by Set net in Namhae, Korea (남해 정치망에서 채집한 엽상자어(Leptocephalus)의 형태 및 유전학적 특성)

  • The present study was tried to identify whether the eel's larva was close to a conger (Conger myriaster), a pipe conger (Muraenesox cinereus) or four species of Anguilla. Experimental fishes were collected by set net in the gulf of enggang, Namhae, Korea from May to June. Their morphological characteristics were compared with adult fishes of a conger, a pipe conger and four species of Anguilla. For genetic classification, DNA was isolated and amplified by using 12S rRNA and 16S rRNA primer set. The PCR products were direct sequencing in both directions. The nucleotide sequences were analyzed using softwares. As results of morphological measurement on eel's larva, the percentages of head length and preanal length against total length were similar with a conger. Based on the nucleotide sequences, the phylogenetic tree also revealed a close relationship to a conger. Therefore, eel's larva, caught in Namhae from May to June, was identified into a conger's larva.

Occurrence of Powdery Mildew Caused by New Race 2F of Podosphaera xanthii on Cucumber in Korea (Podosphaera xanthii의 새로운 Race 2F에 의한 오이 흰가루병 국내 발병 보고)

  • Powdery mildew is a common and serious disease of the Cucurbitaceae including cucumber (Cucumis sativus) in most areas of the world. To identify causal agents of the powdery mildew and their physiological race(s), we collected cucumber leaves displaying typical symptoms of powdery mildew from different locations in Korea. Based on morphological and molecular characteristics, all powdery mildew isolates were identified as an obligate biotrophic fungal pathogen, Podosphaera xanthii. After inoculation at melon (Cucumis melo) differentials to identify the fungal race(s), P. xanthii isolate MI180427 and IC190611 were identified as race 1 which has been repeatedly reported as dominant race in Korea. However, another isolate SE180328 produced different disease response in the tested differentials, being identified as race 2 which has not been reported in Korea. To confirm the race of SE180328, we inoculated additional melon differentials and determined the isolate as race 2F that is the prevalent race of powdery mildew in Beijing, China. Report of this new race 2F in Korea will be helpful for future breeding programs to develop resistant varieties to this race.

Induction of Apoptosis by Ethanol Extract of Scutellaria baicalensis in Renal ell Carcinoma Caki-1 Cells (황금(黃芩) 에탄올 추출물에 의한 인체 신세포암 Caki-1 세포의 자가세포사멸 유도)

  • Scutellaria baicalensis, belonging to the family Labiatae, is widely distributed in Korea, China, Mongolia, and eastern Siberia. It has been used in traditional medicine for various diseases, such as dysentery, pyrexia, jaundice, and carbuncles. In addition, S. baicalensis is reported to possess various beneficial pharmacological activities, including anti-inflammatory, antidiabetic, antiviral, antihypertension, antioxidant, and anticancer effects. However, the molecular mechanisms of its anticancer activity have not been clearly elucidated. In the present study, we investigated the proapoptotic effects of ethanol extract of S. baicalensis (EESB) on human renal cell carcinoma Caki-1 cells. The anti-proliferative activity of EESB was associated with apoptosis induction, which was associated with the up-regulation of death receptor 4, the Fas ligand, and Bax and the down-regulation of Bid, XIAP, and cIAP-1 proteins. EESB treatment also induced mitochondrial dysfunction, proteolytic activation of caspase-3, -8, and -9 and degradation of caspase-3 substrate proteins, such as poly (ADP-ribose) polymerase, ${\beta}$-catenin, and phospholipase C-${\gamma}1$. However, pretreatment of a pan-caspase inhibitor, z-VAD-fmk, significantly attenuated the EESB-induced apoptosis. Taken together, these findings suggest that EESB may be a potential chemotherapeutic agent. Further studies will be needed to identify the active compounds that confer the anticancer activity of S. baicalensis.

Bioinformatic Analysis of the Canine Genes Related to Phenotypes for the Working Dogs (특수 목적견으로서의 품성 및 능력 관련 유전자들에 관한 생물정보학적 분석)

  • Working dogs, such as rescue dogs, military watch dogs, guide dogs, and search dogs, are selected by in-training examination of desired traits, including concentration, possessiveness, and boldness. In recent years, genetic information has been considered to be an important factor for the outstanding abilities of working dogs. To characterize the molecular features of the canine genes related to phenotypes for working dogs, we investigated the 24 previously reported genes (AR, BDNF, DAT, DBH, DGCR2, DRD4, MAOA, MAOB, SLC6A4, TH, TPH2, IFT88, KCNA3, TBR2, TRKB, ACE, GNB1, MSTN, PLCL1, SLC25A22, WFIKKN2, APOE, GRIN2B, and PIK3CG) that were categorized to personality, olfactory sense, and athletic/learning ability. We analyzed the chromosomal location, gene-gene interactions, Gene Ontology, and expression patterns of these genes using bioinformatic tools. In addition, variable numbers of tandem repeat (VNTR) or microsatellite (MS) polymorphism in the AR, MAOA, MAOB, TH, DAT, DBH, and DRD4 genes were reviewed. Taken together, we suggest that the genetic background of the canine genes associated with various working dog behaviors and skill performance attributes could be used for proper selection of superior working dogs.
