Fluorescent in situ hybridization (FISH) with rRNA-targeted oligonucleotide probes was used to compare the community structures of free-living and aggregated bacteria at thawing period in Lake Baikal. Targeted groups were Eubacteria, $\alpha$-, $\beta$-, $\gamma$- proteobacteria groups, Cytophaga-Flavobacterium group and Planctomycetales. Total bacterial numbers of free-living bacteria were ranged from $0.2{\times}10^6\cells{\cdot}ml^-1$ to $3.2{\times}10^6\cells{\cdot}ml^-1$, which were decreasing with depth, while the aggregated bacterial numbers were dramatically increasing from $0.4{\times}10^4 to 3.3{\times}10^4 \cells{\cdot}ml^-1$ with depth. The ratios of EUB probe binding cells to DAPI counts were ranged from 52.3 to 74.1% in free-living bacteria, and from 39.6 to 66.7% in the aggregated bacteria, respectively. Community structures of the aggregated bacteria were very different from each free-living bacteria at every depth. At 25 m depth, where the chlorophyll a concentration was highest, both structures were quite different from those of surface layers, rendering the fact that the community structures might be affected by phytoplankton. The vertical profile of community structure of aggregated bacteria is particular. The proportion of $\beta$-proteobacteria group was increasing with depth and it was 51.8% at 100 m, but the dominant group was $\gamma$-pro-teobacteria group at 250 m. Taken together, the biodiversity and succession of aggregated bacteria are quite different from free-living bacteria.

Effect of the roasted barley tea in commercial markets on the adherence to the saliva-coated hydroxyapatite(HA) beads and the cell surface hydrophobicity of Streptococcus mutans and Streptococcus sobrinus as cariogenic microorganism was examined in vitro. Adherence activity and hydrophobicity in bacteria tested in all the barley tea samples decreased and the values were different according to the type of tea and the type of treatment. The inhibition of bacterial adsorption to HA beads suggest that barley tea active molecules as catechins and melanoidins may adsorb to a host surface, preventing the tooth receptor from interacting with any bacterial adhesions. The obtained results showed that the barley tea may inhibit bacterial adherence, the first step of the pathogenesis of dental caries in which these microorganism are involved.
To cause invasive disease, microorganism must attach firmly to the tonsillar epithelial cell. Once attached, the microorganisms can proliferated, form colonies and release extracillular toxins which can injure the underlying cells. The purpose of present study was to asertain whether or not there exist in vivo differences in bacterial attachment between patients with acute tonsillitis and healthy individuals as a control. This study was carried out on 20 patients suffering from acute tonsillitis and 20 healthy persons used as control. After scraping of the surface of tonsil, cellular mixture was stained with Acridine orange and the number of attached bacteria was calculated using a fluorescent microscope. The adherence rate was calculated as number of bacteria attached to each of 50 epithelial cells. simultaneously, we peformed conventional bacterial culture. Conclusively, the attachment of more than 10% bacteria to the tonsillar epithelial cell was significantly greater in acute tonsillitis group than control group, and there was a significant correlation between age and the number of the attached bacteria to the epithlium.
For scrutinizing the changes of aggregated bacteria in Lake Paldang, the FISH method was applied by using the rhodamine labeled probes, and total bacteria, chloropyll a concentrations and nutrients concentrations were measured. The aggregates were collected with sediment traps. The T-N, T-P, chlorophyll a concentrations of aggregates were higher 5-15 times, 81-140 and 49-66 times than water samples, respectively. Also, the bacterial numbers of aggregates were 200 times higher than those of water smaples. The ratios of each groups of water sample were 2.1-7.4% for $\alpha$-group, 4.5-8.3% for $\beta$-group, 2.1-7.4% for $\gamma$-group, 2.1-6.1% for Cytophaga-Flavobacterium group and 0.1-2.5% for 'other'group, respectively. While, in aggregates, the ratios of $\alpha$-, $\beta$-, $\gamma$- and Cytophaga-Flavobacterium groups were very small and most abundant group was 'other' bacteria. With these results, the aggregated bacteria in Lake Paldang had a particular group composition of bacteria.
In this study, low molecular weight chitosan (LC) was added to dental cement liquid at 0, 0.5, 1.0, and 2.0 wt% by weight for surface properties and oral bacteria adhesive of dental cement. The evaluated by net setting time and surface properties of surface energy on the surface roughness. The degree of oral bacterial adhesion was assessed using two strains of oral bacteria, S. mutans and E.coli. The results showed that the setting time at the LC0.5 group increased no statistically difference was observed (p<0.05). The surface roughness statically significant LC2.0 group and oral bacteria adhesion experiment results in contrast to the control group LC0, LC-added experimental group showed a somewhat lower adherent surface statistically significant difference was observed (p<0.05). It seems that this LC proved that surface properties and oral bacteria adhesion effect was demonstrated. Therefore, it was suggest that the additional effects of LC and research on a wide range of substances.
This study was carried out to identify bacterial strains adhered to human scalp hair and to investigate the antibiotic susceptibility of them. A total of 39 isolates were obtained from patients in intensive care units and healthy persons. The most common species isolated was Staphylococcus epidermidis (19 isolates), followed by S. aureus (14 isolates), S. waneri (5 isolates), and S. pasteuri (1 isolate). The susceptibility of isolates to amikacin, ampicillin, bacitracin, carbenicillin, cefazolin, cefoperazone, chloramphenicol, erythromycin, gentamicin, methicillin, nalidixic acid, neomycin, oxacillin, penicillin, streptomycin, tetracycline and vancomycin was determined by the disk diffusion method. All of the antibiotic resistant isolates were obtained from patient scalp hair. To examine the effect of conventional shampoo and detergent SDS on removing of bacteria from hair, we treated hair with culture solution of S. aureus. The bacteria attached to hair were not removed even by repeated washing with detergents. These results suggested that hair could be a source of bacterial contamination in hospital.

The use of biological-activated carbon (BAC) processes in water treatment involves biofiltration, which maximizes the bacteria's capabilities to remove organic matter. In this study, the distribution of the bacterial community was assessed in response to different types of BAC processes applied downstream in the Nakdong River. The bacterial biomass and activity were $1.20{\sim}34.0{\times}10^7$ CFU/g and 0.61~1.10 mg-C/$m^3{\cdot}hr$ in coal-based BAC, respectively. The attachment of the bacterial biomass and the removal efficiency of the organic carbon were greatest with the coal-based activated carbon. The bacteria attached to each activated carbon material were detected in the order of Pseudomonas genus, Chryseomonas genus, Flavobacterium genus, Alcaligenes genus, Acinetobacter genus, and Spingomona genus. Pseudomonas cepacia was the dominant species in the coal-based materials, and Chryseomonas luteola was the dominant species in the wood-based material.
The purpose of the present investigation was to improve several procedures being used in the adherence assay of Streptococcus sanguis cells to hydroxyapatite (HA) beads and to study the effect of the beads on the counting of radioactivity. The standard adhere assay involved the adherence of radiolabeled bacteria to 40mg of HA beads. The beads were mixed with ['H]thymidine-labeled bacterial cells and incubated for 60 minutes at room temperature. Unadsorbed cells were removed, the beads with adsorbed cells were dried, and the radioactivity was monitered in a scintillation spectrometer. The 30 seconds sonication of cells in a form of long chains appeared to be adequate for obtaining mostly singlet or doublet cells. Unlike the counting of S. sanguis cell suspension, bacterial cells adhered to HA or saliva-coated HA(SHA) required smaller volume (2.5ml) of scintillaton fluid for better counting. Eighteen percent quenching of counts could be attributed to the beads. Among 3 procedures commonly used to equilibrate the beads for adherence assay, no differences were found in their effectiveness. The HA beads on which the bacteria remained attached in scintillant during the counting were found to be the source of sample self-absorption representing 34.5% of the total radioactivity counts resulting from the beads dissolved in HCl solution.
A Gram (-), rod-shaped bacterium in size of 1.3∼$1.8{\times}0.35{\mu}m$ inhibiting the growth of cyanobacterium (Ana-baena cylindrica) was isolated and designated NG-2 in this manuscript. This isolate showed positive reactions for catalase and oxidase, and optimal growth conditions of 35∼TEX>$40<^{\circ}C$ and pH 9.0. In a mixed-culture of A. cylindrica and the isolate, each microorganism grew inverse-proportionally, and the cyanobacterial vegetative cells almost completely disappeared within 24 hours. NG-2 lysed A. cylindrica only under light, which means that lytic activity of NG-2 was dependent on the photosynthetic activity of host. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas het-erocysts have not been lysed. When cyanobacterial cell walls have been lysed partly, NG-2 attatched around A. cylindrica filament and formed colony, then encouraged complete lysis of cyanobacterial cells. The isolate showed similar lytic activity in natural water as in an artificial medium. And lytic activity of NG-2 was enhanced when attached on expandable polystyrene bead.
In this study BGn-incorporated non-fluoride release of pit and fissure sealant $Concise^{TM}$ was developed to improve the mechanical properties and promote antibacterial effect of fit and fissure sealant with the original material. The mechanical properties and antibacterial activity of BGn incorporating vary-ing amounts bioactive glass nano particles(BGn) (0,0.5,1.0 and 2.0 wt% in sealant) were characterized composition of the resulting were investigated. The solubility to aid absorption was calculated by weighing specimens with a diameter of 10 mm and a thickness of 2 mm according to ISO 4049 (2009). The antimicrobial effect was evaluated using three strains of S. mutans, S. aureus and E. coli. The absorbance of the test results was as high as the addition of BGn increased, and the lower the solubility as the solubility was added(p<0.05). Adhesion experiment results S. mutans in contrast to the control group $Concise^{TM}$, BGn-added experimental group showed a somewhat lower adherent surface but no statistically significant difference was observed (p<0.05). However S. aureus and E. coli statistical analysis indicated a significant difference for antibacterial agents between control and BGn containing(p<0.05). It seems that this BGn proved that even a antibacterial effect was demonstrated. Therefore, it was suggest that the additional effects of BGn and research on a wide range of substances.
