Mass Culture and Dietary Value of Fabrea salina (Fabrea salina의 대량배양과 먹이효율)

  • Fabrea salina is an euryhaline ciliate living at salt pond. Its size is small as ca. $150{\mu}m$, and its reproductive rate is high, so that this ciliate has been expected to be a new food organism which will be able to replace rotifer, Brachionus plicatilis. However, the dietary value of F. salina on fish larvae has not yet been verified thoroughly. This research was carried out to understand the mass culture technique and dietary value of this ciliate. In this study, six kinds of phytoplankton and three kinds of grain were tested on growth of the ciliate and optimum temperature, salinity and density of food were also examined for its mass culture. Regarding food for the ciliate, Heterosigma sp. and rice showed the highest growth among the phytoplankton and the grain, respectively. In this study, 229 ciliates per mL was cultured with Heterosigma sp., and 220 ciliates per mL with rice. The optimum temperature for F. salina was $33^{\circ}C$, and salinity ranging from 35 to 95 ppt had no significant difference on its growth. The optimum food density and amount for the ciliate were $5\times10^6$ cells/mL in Heterosigma sp. and 2.8 g/L in rice, respectively. With respect to dietary value of F. salina, six-day-old larvae of ayu, Plecoglossus altivelis, fed on the ciliate showed the high mortality over $96\%$ within 3 days, and it was significantly higher than those fed on rotifer. Even though the mass culture of F. salina has been achieved in the study, this ciliate seems to be inadequate as a food organism for fish larvae.

Mg-Al합금의 조성비율에 따른 발화온도특성

  • 최근의 산업활동에서는 신규 원료 개발과 생산 효율성을 높이기 위하여 분체 공정이 증가하고 있는데, 미세 분진의 취급으로 분진운의 형성과 착화가 용이해지므로 분진폭발이나 화재 위험성이 증가하고 있다. 분진을 안전하게 사용하고 저장, 취급하기 위해서는 착화 전의 위험성 지표로서 최저발화온도(MIT ; Minimum Ignition Temperature)를 사전에 파악해 두는 것이 중요하다. 분진농도의 발화온도는 장치 내의 발화위험성이나 분진 취급 공정의 사고예방대책 관리를 위한 실용적 관점에서 중요하게 활용되는 폭발특성값이다. 또한 분진의 발화온도는 분진농도에 의존하며 농도변화에 따른 가장 낮은 온도를 MIT라고 한다. 본 연구에서는 화재폭발사고 빈도가 줄지 않고 있는 Mg 및 Mg-Al합금(60:40 wt%, 50:50 wt%, 40:60 wt%)을 대상으로 조성비율에 따른 최저발화온도를 실험적으로 조사하였다. Mg 및 Mg-Al(60:40 wt%), Mg-Al(50:50 wt%), Mg-Al(40:60 wt%) 시료의 평균입경은 142, 160, 151, $152{\mu}m$이다. MIT실험장치는 IEC 61241-2-1(Methods for Determining the Minimum Ignition Temperatures of Dust, 1994)에 준거하여 제작하여 사용하였다. 실험장치는 가열로, 분진운 시료홀더, 온도조절장치, 압축공기 제어장치 등으로 구성되어 있다. 구체적인 실험방법은 시험분진를 분진홀더에 장착하고 0.5 bar의 압축공기를 0.3 sec 동안 사용하여 일정 온도로 가열된 로의 내부로 분진운을 부유시킬 때에 분진운이 발화하여 가열로 하단부의 개방구에까지 화염이 전파하는지를 디지털비데오카메라로 기록, 평가하여 발화 유무를 판정하였다. Mg합금에 대한 MIT를 측정한 결과 $740^{\circ}C$가 얻어졌으며, Mg-Al(60:40 wt%)의 MIT는 $820^{\circ}C$로 조사되었다. 그러나 Mg-Al(50:50 wt%) 및 Mg-Al(40:60 wt%)에 대해서는 최대 가열로의 설정온도를 $890^{\circ}C$까지로 하여 농도를 변화시키면서 조사하였으나 발화가 일어나지 않았다. 문헌에 따르면 Mg입자 표면의 산화피막은 다공성으로 일정 온도에서 산화반응이 시간에 따라 직선적으로 증가하는데 반하여, Al의 산화피막은 보호 작용을 하여 일정 온도에서 산화반응속도가 표면과 내부의 농도 기울기에 의한 확산속도에 의존한다고 보고하고 있다. 본 연구결과를 토대로 Mg-Al합금의 발화특성을 고찰해 보면, Mg-Al합금에서 자기 전파성이 작은 Al성분의 증가는 착화지연이 증가하여 연소성이 감소하여 최저발화온도의 증가로 이어지는 것으로 추정되었다. 또한 발화온도는 주어진 조건의 온도장에서 분진이 존재하는 시간 길이에 따라 변화하므로, 발화온도를 실험적으로 측정하는 경우에는 측정장치나 방법에 따라 달라지므로 사업장의 현장에 발화온도를 적용하는 경우에는 장치 내의 분진의 존재시간을 고려할 필요가 있다.

Analysis of Tidal Asymmetric Characteristics in the Muan Bay (무안만의 조석비대칭적 특성 분석)

  • Tidal asymmetry would occur owing to shallow water tides at the Western Coast of macro tidal area. Especially, as ebb dominance of Mokpo coastal zone is known as the most prominent in Korea, it had been studied by domestic researchers. The cause of ebb dominance in Mokpo area is considered due to extensive inter-tidal zone in Muan Bay, and this has been studied based on amplification ratio, relative phase and skewness of tide/tidal flow curves in order to analyze qualitative tidal asymmetry. Furthermore, it was possible to figure out tidal characteristics with the difference of tidal amplitude and phase with Mokpo Harbor by observing the tide for 15 days in Muan Bay, which showed 40 minutes shorter ebbing time than at Mokpo Harbor. In addition, tidal flow prediction data in Mokpo North Harbor and Mokpo-Gu were analyzed. Meanwhile, the basis regarding qualitative interpretation of bed sediment and suspended sediment was provided by examining the qualitative changes in tidal asymmetry for spring-neap tidal cycle through the PCA/SWA indices. In addition, by examining long-term changes of ebb dominance in Mokpo Port, tidal characteristics of the past, present and future in this area, which is related to tidal asymmetry, is also provided.

Depositional Sedimentary environments in the Han River Estuary and Around the Kyunggi Bay Posterior to the Han river's developments (한강종합개발 이후 한강하구 및 경기만의 퇴적환경)

  • For the purpose of examining the depositional sedimentary environments in the Han River estuary and around the Kyunggi Bay posterior to the Han river's developments, a hydrological and sedimen-tological survey was carried out. According to the hydrological and sedimentological conditions, the studied area can be divided into 3 depositional sedimentary environments: Fluvial, Estuarine and coastal-Bay. Posterior to the Han river's developments, however, the alterations of hydrodynamic condition in the Han river have caused a substantial change of the sedimentary environments in the lower Han river and its estuary. That is, the contents of total suspended sediment anterior to the Developments decreased from 37mg/l (in the lower Han River) and 500-1750 mg/l (at the Kanghwa Bridge) to 18 mg/l and 208-1142 mg/l posterior to the developments. these changes seem to have caused the siltation near the sin-gok Underwater Dam. Thus the characters of the boundary condition between the fluvial and the estuarine environments have rapidly changed. It is considered that these changes result mainly from the construction of the two underwater dams for the maintenance of the water level of the Han river. As the estuary is a transition zone between land and sea, these changes in the Han River estuary might affect the sedimentary environments around the Kyunggi Bay. In order to verify the effects of these changes, it is necessary that a detailed survey be carried out around the Han River estuary including the Imjin and Yesong River estuaries.

Influence of Land Use Change in the Forest Catchment on Sediment Accumulation at the Outlets of Rivers: Results of a Study in Kushiro Mire, Northern Japan (산림유역 개발이 하천 출구의 토사 퇴적에 미치는 영향: 일본 쿠시로습지의 연구결과)

  • The purpose of this study was to examine the influence of land use change in the forest catchment on sedimentation rate at the outlets of rivers in Kushiro Mire that have been impacted by forest clearing, agricultural activity and river regulation. We analysed Caesium-137(Cs-137) concentration in sediment cores, and we estimated sedimentation rates and Cs-137 inventories over the last 50 years. Cs-137 from atomic bomb testing first entered the environment in 1954 which provides easily identifiable chronological markers in the sediment. Because Cs-137 is strongly absorbed into sediment particles, its redistribution occurs in association with sedimentary particles. Since the 1950s, the forest catchment areas draining into the mire have been developed intensively from forest areas to agricultural lands. The sediment accumulations at the outlets of rivers after 1954 ranged from 36 to 148 cm. The Cs-137 inventory is significantly greater than the reference sites which reflected natural accumulation conditions because sediment containing Cs-137 was carried from catchments into the outlets of the rivers. In addition, the Cs-137 inventory was correlated with the sedimentation rate. However, the Cs-137 inventories in Kuchoro and Kushiro river profiles were slowly increased with the sedimentation rates. This is because the sediment originating from scoured areas such as streambeds and banks contains a low level of Cs-137 concentration.

Physico-chemical properties and biological activity of controlled-release granular formulations for the herbicide dicamba (방출조절형 dicamba 입제의 물리화학성 및 생물효과)

  • Dicamba (3,6-dichloro-o-anisic acid) granular formulations for controlled release (DGFCRs) were prepared with biodegradable polymers, corn starch and pregelatinized starch, to minimize harmful side effects, extend weed control performance, and control the releasing rate of the active ingredient. Physico-chemical properties and biological activity of DGFCRs were studied. Six different granules were formulated by applying two processes, granulation and extrusion. Formulation efficiencies of active ingredient (A.I.) in the granules prepared by granulating and extruding were $90.0{\sim}96.3%$. Incorporation ratios of A.I. in the granules prepared by granulating and extruding showed $89.5{\sim}94.5%$ and $46.7{\sim}82.0%$, respectively. The highest swellability was DG-2 formulation prepared with corn starch. Whereas, the lowest floatability in water was DG-2 formulation, while the highest one was DG-1 formulation prepared with pregelatinized starch, Miragel 463. The degradation rates of dicamba in the granules under the elevated temperature of $50^{\circ}C$ were less than 5% for DG-1 and DG-2 formulations even after 90 days, meanwhile, those of DE-1 formulations prepared with pregelatinized starch, Mirasperse, were more than 5%. The release rates of A.I. from the granules into water under a static condition were about 100% after 2 weeks. Weeding effects of the granules on broad leaf weeds tested in greenhouse were more than 90% after 30 days.

High-Resolution Seismic Characteristics of the Holocene Mud Deposits in the Southeast Innershelf, Korea (한국 남동 내대륙붕해역의 홀로세 뻘질 퇴적층의 고해상도 탄성파 특성)

  • The Holocene mud deposits in the southeastern innershelf of the Korean Peninsula were studied using the shallow seismic reflection profiles coupled with sediment sampling. The Holocene mud deposits are developed as a belt in the innershelf area from the mouth of the Nakdong River to off Pohang along the coastline. The surficial sediments in the study area gradually become finer and well sorted from south to north. The seismic facies in the Holocene sequence change northward from parallel reflectors in the mouth of the Nakdong River and northeastward prograding reflectors in the southern part off Ulsan to transparent layer in the nearshore off Ulsan to Pohang. The regional difference of seismic facies indicates that the Holocene sediment characters are varied with localities. By combining the surficial sediments properties with seismic facies patterns, the suspended sediments mostly supplied from the Nakdong River may be transported northward along the shore by the north-flowing Tsushima warm current. The Holocene mud sequence overlying the ravinement surface which is produced by erosion of underlying sediments during a rapid transgression can be interpreted as the highstand system tracts probably formed during the highstand of sea level similar to the present-day sea level.

The Seasonal and Regional Distribution of Phytoplankton Communities in the Fisheries Resources Protection Area of Korea in 2016 (2016년 한국 수산자원보호해역에서 식물플랑크톤 군집의 계절 및 해역별 분포)

  • This study was conducted to understand the characteristics of the seasonal and regional distribution of phytoplankton communities in the Fisheries Resources Protection Area of Korea (FRPA). We investigated the phytoplankton composition, abundance and dominant species collected from five different regions (Cheonsu, Tongyeong-I, Tongyeong-II, Hansan, and Jindong) in 2016. According to the results, most environmental parameters, such as temperature, salinity and nutrients, showed statistically significant seasonal differences. Suspended particulate material (SPM) only showed a statistically significant regional difference. The mean abundance of phytoplankton ranged from 13 to $4,062cells{\cdot}ml^{-1}$, with large spatio-temporal fluctuations. In particular, the bloom of phytoplankton (>$10^3cells{\cdot}ml^{-1}$) in Cheonsu Bay occurred in April and October with Skeletonema spp. and Chaetoceros socialis being the dominant species during these two seasons, respectively. The dominant species in the FRPA were diatoms (Pseudo-nitzschia spp., Skeletonema spp., and Chaetoceros pseudocriniuts) and dinoflagellates (Scrippsiella trochoidea and Tripos furca). The seasonal distribution of phytoplankton communities showed typical characteristics of coastal waters, i.e., that diatoms usually dominated in winter and autumn, while dinoflagellates tended to dominate in spring and summer. Meanwhile, the dominance rate of diatoms in the phytoplankton community in Cheonsu Bay, which is located in a high-turbidity region, was 9~27 % greater than that of diatoms in the phytoplankton community found in the south coastal waters, which is a low turbidity region.

  • Bartonella henselae is the causative agent of cat scratch disease. Although cats are the main zoonotic reservoirs of Bartonella spp., unusual cases of cat scratch disease caused by a domestic dog scratch have been recently reported. For the in vivo B. henselae infection, eight dogs were inoculated intradermally with $2{\times}10^8CFU$ of B. henselae Houston-1 suspended in 1 ml of phosphate buffered saline on day 0 and subsequent injections of the same amount given intradermally on days 21, 28, 36, 58 and 64. After in vivo canine B. henselae infection was confirmed by nested PCR, the IFN-$\gamma$ levels of the culture supernatant of PBMC stimulated with B. henselae was significantly higher in the B. henselae-PCR positive group than the B. henselae-PCR negative group. Our results showed that the canine immune responses against B. henselae were different from those of cats. Th1 activation by B. henselae stimulation was characterized in dog peripheral blood mononuclear cells, whereas Th2 activation was reported in B. henselae-infected cats.

Optimal Dimension of PE Film Bag According to Fruit Size in MAP Storage of 'Fuyu' Persimmon Fruit ('부유' 단감 MAP 저장에서 과실 크기에 따른 PE 필름 봉지의 적정 규격)

  • Model for optimally dimensioned PE film bag was developed according to fruit size to maintain best quality of 'Fuyu' persimmon in modified atmosphere packaging (MAP) storage based on relationship between quality and oxygen and carbon dioxide in PE film bag. Harvested persimmons were graded into five sizes, and average weights were LL:261, L:217, M:188, S:168, and SS:154 g. Five fruit units of each grade were optimized in five PE film bag sizes of $150{\times}376,\;140{\times}357,\;130{\times}344,\;130{\times}333,\;and\;120{\times}3l8\;mm$. To minimize quality deterioration such as softening and discoloration, optimal oxygen and carbon dioxide concentrations in PE film bag were 0.5-1.0 and 6.0-8.0%, respectively, and optimal thickness of PE film bag according to fruit size were LL:45, L:50, M:55, S:60, and $SS:65\;{\mu}m$. For all fruit sizes, model for PE film bag area $(mm^2)$ was good quadratic simple equation by fruit weight (g): $Y=-4055.707+627.993X_1-0.701{X_1}^2$. Model far optimal oxygen and carbon dioxide (Y) concentration in PE film bag was suited to linear multiple equation by fruit weight $(X_1,\;g)$ and PE film thickness $(X_2,\;{\mu}m)$. Equations for oxygen and carbon dioxide concentrations (%) were $Y=5.798-0.0109X_l-0.0491X_2\;and\;Y=-2.427+0.01927X_l +0.09646X_2$, respectively.
