Virus Diseases Occurred on Watermelon in Jeonnam Province (전남지역의 수박에 발생하는 바이러스 병 발생 실태)

  • Recent occurrence of virus diseases on watermelon plants cultivated in Jeonnam province was investigated from 1998 to 2002. While virus diseases were severely occurred on watermelon cultivated in green house in 1998, those of open field were severer than in green house since 2000. When 128 samples collected from different fields were examined by electron microscopy, 87.8% of the samples contained rod-shaped or filamentous virus particles. RT-PCR analysis of the samples revealed that Cucumber green mottle mosaic virus (CGMMV) was only detected from collected samples at May. Watermelon mosaic virus (WMV) was most frequently found and CGMMV and Zucchini yellow mosaic virus (ZYMV) were slightly at June and July. However Cucumber mosaic virus (CMV) and Papaya ringspot virus (PRSV) have not been detected.

Effect of Chemical Treatment on the Control of Strawberry Anthracnose caused by Colletotrichum sp. (딸기탄저병의 약제방제효과)

  • A total of 55 Colletotrichum isolates from strawberry plants with anthracnose symptoms(crown rot) were inhibited in mycelial growth on potato-dextrose agar(PDA) amended with fungicides in variable degrees depending on the chemicals used, especially showing no growth on PDA with 1 mg/m/tricyclazole. However, in the detached leaf test by treating chemicals before or after inoculation of Colletotrichum sp., tricyclazole was little effective in controlling symptom development; instead azoxystrobin, which had low in vitro inhibition of mycelial growth, inhibited strongly the symptom development. Some chemicals were tested for the control of strawberry crown rot in greenhouse using three methods, sprays soil drenching and plant dipping. No or little control effect were made by chemical spray and soil drenching, but plant dipping in chemical solution, especially azoxystrobin: reduced crown rot development by about 50% in the greenhouse suggesting that the azoxystrobin treatment may be an effective control method of the crown rot of strawberry. No differences in the control efficacy were noted according to the dipping time and chemical concentration of azoxystrobin not less than 10 min and 250 mg/m/, respectively.

Severity of Cowpea mosaic virus and Putkong Disease Monitoring and Purification of Cowpea mosaic virus (석량풋콩재배지에서의 동부모자이크바이러스병의 발생도 및 풋콩병해모니터링과 동부모자이크 바이러스의 순화)

  • One hundred and eighty-six leaves of soybean cv. Seokryangputkong that showed mild mosaic symptoms were collected randomly and ELISA tests were conducted with those leaf samples to screen the presence of Cowpea mosaic virus (CPMV). Ninety-three out of 186 samples reacted positively to CPMV, but those samples did negatively to Soybean mosaic virus (SMV). At least, 55 leaf samples revealed higher values than that of positive control. The results strongly confirmed that CPMV occurred severely in soybean cv. Seokryangputkong. However, a question is raised on the primary reservoir and vector for transmission of this virus. Since the farmer changes seeds every year, seed transmission is excluded. The virus was also purified, the analysis of coat protein conformed the virus of cowpea mosaic virus and UV absorption pattern confirmed that the causal virus of mosaic disease in soybean putkong was cowpea mosaic virus.

Development of a Simple and Effective Bioassay Method to Evaluate Resistance of Watermelon Plants to Fusarium oxysporum f. sp. niveum (수박 덩굴쪼김병에 대한 간편 저항성 검정법 확립)

  • Root-dipping inoculation method has been widely used to determine the resistance of watermelon to Fusarium oxysporum f. sp. niveum causing Fusarium wilt. Although this method leads to the precise results of plant disease responses, more rapid and efficient assay methods have been still required because the root-dipping inoculation method is labor-intensive and time-consuming. In this study, we established a simple and effective bioassay method based on the comparison of various inoculation methods and growth conditions. To develop the system, the occurrence of Fusarium wilt on four resistant and susceptible cultivars was investigated by four different inoculation methods, root-dipping, scalpel, tip and soil-drenching methods. Of these inoculation methods, scalpel method resulted in clear plant disease resistance responses with the simplicity. With the use of scalpel method, we also explored the disease development of the cultivars depending on inoculum concentration, growth stage of seedlings, and incubation temperature after inoculation. Furthermore, we found that the resistance degrees of 23 cultivars derived by scalpel inoculation method were similar to the results by root-dipping method established previously.

  • Root-dipping inoculation method has been used to investigate resistance of radish plants to Fusarium wilt. However, the method requires a lot of labor and time because of complicate procedure. This study was conducted to establish a simple and effective mass-screening method for resistant radish to Fusarium wilt. Radish seedlings of susceptible and resistant cultivars were used to investigate wounding method by scalpel, inoculum concentration, and pathogen-inoculated growth stage of seedlings. We established an efficient mass-screening method based on our results as following: Roots of 14-day-old seedlings of radish are cut with a scalpel at a $90^{\circ}$ angle to a 2 cm-depth at a 1 cm-distance from main stem and then inoculated by pouring with a 10 ml-aliquot of a fungal spore suspension ($1.0{\times}10^7conidia/ml$) on soil. The inoculated plants are cultivated in a growth room at $25^{\circ}C$ for about 4 weeks with 12-hour light a day. The proposed screening method enables to effectively select resistant from mass radish plants cultivars to Fusarium wilt.

Selection and Characterization of Horticultural Traits of Tomato leaf curl virus (TYLCV)-resistant Tomato Cultivars (토마토 황화잎말림바이러스(TYLCV) 저항성 품종 선발 및 원예특성 분석)

  • This study was conducted to evaluate imported tomato $F_1$ cultivars as breeding materials for the resistance to Tomato yellow leaf curl virus (TYLCV) by molecular markers and bioassay. From marker genotyping and disease evaluation of 40 $F_1$ cultivars, most of the cultivars declared as TYLCV-resistance carried heterozygous marker genotype for the TYLCV resistance genes Ty-1, Ty-3, or Ty-3a, and showed low disease rates. Whereas, 4 of 5 $F_1$ cultivars declared as intermediate resistance showed marker genotype for susceptibility and disease rates ranged 18.1-33.3%. However, the xx cultivars showed inconsistency in marker genotype and disease rate. Characterization of horticultural traits of the $F_1$ cultivars with TYLCV-resistance indicated that large-size fruit cultivars were higher in yield and similar in sugar contents and solid-acid ratio compared to a control cultivar preferred in the domestic market, although hardness remained to be a problem. On the other hand, cherry tomato cultivars showed lower yield and brix, but longer internode compared to a control cultivar, indicating that breeding for TYLCV-resistance using these cultivars will require more efforts and time compared to large-sized.

The effects of speakers' age on temporal features of speech among healthy young, middle-aged, and older adults (연령세대에 따른 말 산출의 시간적 특성: 말속도와 쉼을 중심으로)

  • The purpose of the this study is to observe the effects of healthy adults' age on temporal features of speech and identify which could differentiate older and young adults. We examined speech rates(i.e., overall speaking rate, articulation rate), occurrence of pause, and duration of pause per utterance by utilizing the National Institute of Korean Language's open corpus. We selected a total of 30 healthy adults (10 young, 10 middle-aged, and 10 older adults) in this study. There were significant differences among the groups in the overall speaking rate, articulation rate, total occurrence of pause, the occurrence of pause between syntactic words, total duration of pause, and duration of pause between syntactic words. The older and middle-aged adults showed slower speech rates and longer and more frequent pause than young adults. But there were no significant differences among the three groups in terms of pause within syntactic word. The overall speaking rate significantly differentiated older adults from young adults. These findings suggested that the effect of speakers' age was reflected in gradual changes in the temporal features of their speech.

Identification of Alfalfa Mosaic Virus from Soybean (대두에서 발생한 알파파 모자이크 바이러스의 분류동정에 관한 연구)

  • A yellow stripe and bud benting disease of soybean was commonly observed on the field at Suweon area. The causal agent was identified as alfalfa mosaic virus (AMV) by indicator plant reactions, physical properties, serological test and electron microscopy. AMV produced vein clearing, top necrosis, top bent and mottling on the parts of soybean plants. Local lesions were produced on the inoculated leaves of Vigna sesquipedialis, Vicia faba and Tetragonia expansa, while Chenopodium am, anticolor, C. quinoa, Pisum satvium, Petunia hybrida and Nicotiana tabacum 'Bright yellow' were systemically infected. The thermal inactivation point was $60^{\circ}C$, dilution end point was $10^{-3}$, and longevity in vitro was 2 days at room temperature. AMV from soybean was reacted with AMV - antiserum in agar gel diffusion test. Electron microscopy of AMV from soybean exhibited bacilliform particles of 60nm in length.

Detection of Cucumber green mottle mosaic virus in Bottle Gourd Seeds by RT-PCR (RT-PCR에 의한 박 종자의 오이녹반모자이크바이러스 검정)

  • Cucumber green mottle mosaic virus (CGMMV) was a major pathogen of watermelon and had affected seriously to watermelon production in Korea. Rapid and sensitive detection method of CGMMV associated with bottle gourd (Lagenafia siceraria) seeds was developed by using RT-PCR in this study. A pair of primeri Wmfl and Wmrl, specific for CGMMV was designed from coat protein gene sequences of CGMMV-W and used for amplifying 420 bp product in RT-PCR. To simplify the virus extraction procedure and reduce an inhibitor from the extract for the RT-PCR, some methods using ethanol precipitation, double filtration, polyethylene glycol precipitation and phenol/chloroform/isoamyl alcohol extraction procedure were compared and the phenol/chloroform/isoamyl alcohol extraction procedure was selected by its enhanced sensitivity. This detection method using the selected extraction step and the primers for RT-PCR could reliably detect an infected level of one CGMMV-infested seed in 1,000 seeds. This rapid and sensitive RT-PCR assay provides auseful tool for the specific detection of CGMMV in bottle gourd seed samples containing high levels of back-ground inhibitors.

Incidence of Viral Infection of Main Garlic Growing Areas of Gangwon Province in Korea (강원도 마늘 주산지 재배마늘의 바이러스 감염 실태)

  • This research was conducted to investigate the incidence of viral infection in Samcheok-shi and Jeongseon-gun, main garlic growing areas of Gangwon province in Korea. Four primers sets (GLV, LYSV, GCLV, OYDV) were used for RT-PCR to test of 100 samples collected from each location. Infection rates of GLV, LYSV, GCLV and OYDV of garlic samples from Samcheok-shi and Jeongseon-gun were 95, 95, 92 and 33%, respectively. All garlic samples tested in this research were infected at least one virus. Coinfection of 3 kinds of virus (GLV, LYSV and GCLV) and 4 kinds of virus (GLV, LYSV, GCLV and OYDV) were 60 and 25%, respectively. While infections of GLV, LYSV and GCLV were evenly found in both two regions, OYDV infection of garlic samples collected from Jeongseon-gun was higher than Samcheok-shi. Viral infections at garlic fields of whole country and Gangwon province are very serious now. Therefore, it is very necessary to develop technic for virus-free bulb and renew seed bulb of garlic.
