Reproductive Ecology and Genetic Variations in the Sea Urchins Anthocidaris crassispina and Hemicentrotus pulcherrimus in the Cheju Coast (제주연안에 서식하는 보라성게 Anthocidaris crassispina와 말똥성게 Hemicentrotus pulcherrimus의 지역별 번식생태학적 특성과 유전적 변이의 비교)

  • Genetic variabilities of local populations of Anthocidaris crassispina and Hemicentrotus pulcherrimus were analysed by random amplified polymorphic DNA (RAPD) technique with phenotypic variabilities in timing of gonadal maturation, reproductive output and size of individuals in Cheju. H, pulcherrimus, collected from 4 locations during March 1997, indicated that Daepo individuals were significantly smaller than that at Hamdok, Wimi site A and Wimi site B (ANOVA, p>0.001). Gonodal-somatic index (GSI) of the Wimi site B population was significantly higher than that of three other locations (ANOVA, P>0.0001). An ANOVA test conducted on test size of A. crassispina, harvested from six different locations of Cheju during June 1997, indicated that the size of individuals from Pophwan was significantly smaller (P>0.0001) than that from five other locations. GSIs of urchin m Wimi and Hanlim were significantly higher than that from Pohwan and Oedo (ANOVA, p>0.0001). Genetic similarity, calculated from k13 primer analysis of total DNA, among the six different populations of A. crassispina varied from 0.67 to 0.92, and the values for H. pulcherrimus from 0.60 to 0.73; thus there was no genetic variation among different populations of the same species. Therefore, the populations are genetically homologous and the observed phenotypic variabilities were possibly associated with water temperature and food.

  • The 34 potently pig pheromonal odorants (1-32, 5755 & 7113) through structure-based virtual screening and ligand-based virtual screening method were selected and their ADMET and pharmacokinetics characters were evaluated and discussed quantitatively. The pheromonal odorants were projected on the following pre-calculated models, Caco-2 cell permeability, blood-brain barrier permeation, hERG inhibition and volume-distribution. From the results of in silico study, it is found that an optimal compound (31) either penetrating or have a little ($P_{caco2}$=-8.143) for Caco-2 cell permeability, moderate penetrating ability ($P_{BBB}$=0.082) for blood-brain barrier permeation, the low QT prolongation ($P_{hERG}$=1.137) for the hERG $K^+$ channel inhibition, and low distribution into tissues ($P_{VD}$=-5.468) for volume-distribution. Therefore, it is predicted that the compound (31) a topical application may be preferable from these based foundings.

  • 외래종이라 함은 도입종, 귀화종, 침입종을 통칭하는 용어로 특정한 인위적 목적 여부에 관계없이, 본래의 자생지가 아닌 외부에서 들어와 다른 생물의 서식지를 점유하여 번식하는 생물종을 지칭하는 것으로, 침입외래종은 생태계에 도입되고 확산되면서 생물다양성과 연계된 생태계서비스를 위협하거나 또는 악영향을 미치는 외래종을 의미하며, 위해우려종이란 국내에 유입될 경우 생태계 등에 위해를 미칠 우려가 있어 환경부장관이 지정 고시하는 생물종 (살아있는 것으로서 개체의 일부 알 종자 등을 포함)을 지칭한다. 본 연구는 환경부고시에 따른 한반도 미유입 환경위해우려종43종에 대하여 농업환경위해성 평가를 실시하고, 분류동정 기법을 개발하는 등 기초 자료를 제공하고자 수행되었다. 이를 위해, 해외식물표본 확인과 미국 현장조사를 실시하였으며, 우선적으로 8종의 미유입 식물에 대하여 보고하고자 한다. - Carduus acanthoides L.   • This experiment was carried out to develop a sensitive, rapid, solid-phase microtitre plate assay of progesterone using the monoclonal antibody to this hormone. Monoclonal antibody to progesterone was much higher titre and binding affinity about 10 times than conventional polyclonal antibody to progesterone. Dot-blot analysis of monoclonal antibody revealed a single precipitation band when reacted with anti-mouse IgM and anti-mouse K. A competitive reaction was used with a reaction time of 2 hours. The standard dose-response curve was linear through 1,000pg/well. This ELISA system approach is applicable to evaluation for the rapid assessment of luteal function and reproductive status in both clinical and research in a wide variety of species.

A Study on the Landscape Characteristics and Propagation Methods of Korean native Lindera obtusiloba Blume (자생 생강나무의 조경적 특성 및 번식방법에 관한 연구)

  • This study was carried out to investigate the growth characteristics and propagation methods of L. obtusiloba as a woody landscape plant. The results are obtained as follows: L. obtusiloba was dioecious shrub and shaped with bush type. Leaves were ovate, obtuse, cordate, 7.05 cm long, 7.20 cm wide, and petiole 2.0 cm long. Flowers of L. obtusiloba were diclinous. Soil acidity ranged from pH 4.06 to 5.53 with the lowest at the Mt. Soo-Ri. Mt. Soo-Ri located near factory district, which was considered to damaged by environmental deteoration. While soil organic matter was highest at Mt. Soo-Ri, inorganic nutrients were low. L. obtusiloba grows in the area with low soil acidity and low content of inorganic. Therefore it seemed to be tolerant to air pollution. L. Obtusiloba was high seed germination rate in the plug box and its shoots were longer than seeding box and softwood cutting of L. obtusiloba showed the rooting rate of 50% at 5,000ppm on June 23. To develop a mass propagation method of Korean native L. obtusiloba through an axillary bud culture as a woody landscape plant, about 2∼3 cm shoots induced from explant were subcultured to new media contained different growth regulators. Shoots multiplied most effectively on a WPM containing 1.0 mg/l BA, producing 5.5 shoots with a shoot length of 2.5 cm per shoot explant.

Effects of Seed Tuber Processing and Cultural methods on Tuber yield of Amorphophallus Konjac. K. (구약감자의 종서처리 및 재배방법이 수량에 미치는 영향)

  • This experiment was conducted to determine tuber yield increase of Konjac by sowing time, seed tuber split method and cultural methods. Tuber yield per unit area was generally increased in early planting than conventional planting time. Tuber yield of polyethylene film mulching culture among storage methods was high, while the emergence rate of konjac for seedling plus PE mulching, and tunnel culture were prompted by 20 days, and especially seedling was to be greatly controlled due to temperature difference of the day and night at emergence. All of the cutting methods(two and four split method) were higher than conventional method because of increasing number of bulblet, accordingly, that method using seed tuber was profitable for mass propagation. Both botanical characteristics and tuber yield tend to be increased at 30 to 50 percent shading than natural condition.

Biochemical characterization and PFGE pattern of Brucella canis isolated from kennels in Gyoengbuk province (경북지역 애견 번식장에서 분리한 Brucella canis의 생화학적특성 및 PFGE 양상)

  • A biochemical characterization and antimicrobial drugs susceptibility study was conducted in four breeding kennel which was canine abortion caused by Brucella canis in Gyeongbuk province in 2003-2006. Total of 267 dogs domesticated in the four kennel were examination. Among them, 143 (53.6%) dogs were sero-positive and 25 of blood samples were isolated to Brucella canis. At amplification of 35KDa-BCSP gene using PCR, 711 bp DNA fragment was same visible in 25 isolates and B canis RM6/66. Biochemical characterization of B canis isolated was non-hemolytic, no production of $H_2S$, no fermentation of carbohydrates, catalase-positive, oxidase-positive, indol-negative, hydrolyzation of urea, reduction of nitrate and development of thionin dye medium. Using disk-diffusion method, all of 25 strains tested were found to be highly susceptible to tetracycline, aminoglycoside, quinolone, macrolide antibiotics, rifampin and ampicillin in vitro. Using PFGE with restriction enzyme Smi I, 25 isolates tested were typed to 2 pattern, S1 and S2.

New Cultivar 'Yellow Tiara' of Leaf Variegated Hosta (비비추의 잎변이 품종 'Yellow Tiara' 육성)

  • Hosta 'Yellow Tiara' was induced using methyle methane sulfonate (MMS) to obtain mutagens. Among the induced leaf-variegated hosta plants, some with yellow-green leaves were selected. They were cultivated via vegetative propagation. Assessment of the botanical characteristics was conducted for three years since 2004. The major characteristic of these cultivars is that they maintain their yellow-green leaves uniformly all year round. These cultivars are useful as materials for pot plants or as ground cover.

  • In mammals, male and female germline stem cells are derived from primodial germ cells. Despite many efforts to identify stem cells from gonads, there has been little successe to identify germline stem cells yet. In this study, we isolate and characterized porcine germline stem cells using only stem cell markers that are prevalently expressed in various tissues. Gonadal cells derived from both male and female formed colonies and showed AP activities and different lectin binding properties. Pluripotency of germline stem cells was also identified by positive signals against putative stem cells markers such as SSEA-1 and SSEA-3. In addition, nestin was also found in primary gonad cells that have a similar morphology to the AP-positive cells. The nestin expression suggests that the germline stem cells may have similar expression of the prevalent stem cell markers found in other tissues. The demonstration of nestin expression together with pluripotent cell markers calls further investigation of the possible differentiation of nestin-positive cells into neurons.

Development of A New Phalaenopsis Cultivar 'Little Dew' with White Miniature Type Flowers (백색 미니 다화성계 호접란 신품종 '리틀 듀' 육성)

  • A new Phalaenopsis cultivar 'Little Dew' has been developed by a cross breeding and a line selection at the Flower Breeding Research Institute, Gyeongnam ARES from 2000 to 2007. Characteristics trials for this cultivar was carried out three times from 2004 to 2006. 'Little Dew' developed from a cross between Phal. 'Timothy Christopher' and Phal. amabilis showed white miniature type flowers and a multiflora shape. It has 3~4 flower stalks and many flowers, and long life span of flowers. This new cultivar was registered for commercialization in 2007 and would be cultured well in greenhouse conditions in Korea.
