Allelopathic Effect of Volatile Extracts from Eupatorium rugosum (서양등골나물 휘발성 추출물의 알레로파시 효과)

  • This study was carried out to find the allelopathic effect of volatile materials released from Eupatorium rugosum. The GC method was employed for analysis of volatile materials from E. rugosum and 49 chemical substances were identified such as $\beta$-caryophyllene, $\alpha$-terpinenol, chamazulene, bornyl acetate, $\alpha$-pinene, etc. including unidentified three chemicals. Germination test in Phaseolus radiatus was done to find the inhibition effect of volatile materials using some chemicals which were proved to be important component or much amounts ones in E. rugosum. It was strongly inhibited by linalool and terpinen-4-ol. Seedling elongation and radicle growth of that were proportionally inhibited by the concentration of the essential oil, especially $\alpha$-pinene and bornyl acetate. Biomass of receptor plant was slightly decreased more than 58 ${\mu}l$ of the extract in case of $\alpha$-pinene, while it was decreased more than 19 ${\mu}l$ of that in bornyl acetate but it was shown non-significant. From the above results, it was found that volatile materials from E. rugosum showed a allelopathic effect and also $\alpha$-pinene, bornyl acetate, linalool and terpinen-4-ol used in bioassay were some of major allelochemicals in germination inhibition and especially linalool and terpinen-4-ol are prominent effect on growth inhibition of other plant.

Selection of Desirable Varieties for the Year-round Water Culture of Young Welsh Onion (수경재배용 실파 적품종 선발)

  • This experiment was carried out to select the desirable welsh onion (Allium fistulosum L.) varieties for the year-round cultivation in water culture. Ten varieties were tested, of which five were Korean's and the other five were Japanese's. The germination percentage and germinative energy of most varieties we.e more than 90 percent. The fastest germination speed were 'Geumjanguedaepa', 'Gujopa' and 'Fuyune'. The best growth of the varieties were 'Geumjanguedaepa' and 'Tokyokuro' in spring, 'Geumjanguedaepa', 'Gujopa', 'Saesaeksisilpa' and 'Tokyokuro' in summer, 'Geumjanguedaepa', 'Gujopa' and 'Tokyokuro' in fall, and 'Geumjanguedaepa', 'Midorigawa' and 'Tokyokuro' in winter cultivation. Suitable varieties of growth fur all seasons were 'Geurnjanguedaepa' and 'Tokyokuro', though 'Gujopa' was good in summer and fall. In the view of nutritional content, nitrate content was significantly lower in 'Geumjanguedaepa', 'Tokyokuro' and 'Fuyune' than that in'Saesaeksisilpa' and 'Starziam'. Vitamin C content was higher in 'Aqua Green', 'Midorigawa' and 'Seoulbaekpa' than that of the others. Pyruvic acid content was the highest in 'Geumjanguedaepa' and 'Suckchanguedaepa'. Based on the above results, 'Geumjanguedaepa', 'Gujopa' and 'Tokyokuro' were selected as the most desirable young welsh onion varieties for the year-round water culture.

  • To investigate the occurrences of resting cysts of Diplopsalis and Protoperidinium species, sediment trap samples were collected from Gamak Bay, Korea. Based on the germination experiments, nine species of order Peridiniales were identified; cyst of Diplopsalis lebourae, cyst of Protoperidinium excentricum, cyst of Protoperidinium sp. cf. nudum, cyst of Protoperidinium obtusum, cyst of Protoperidinium ventricum, cyst of Protoperidinium sp.1, cyst of Protoperidinium sp.2, Protoperidinium sinuosum (Brigantedinium majusculum) and unidentified Peridiniales.

Antiproliferation Effects of Ethanol and Water Extracts from Germinated Rough Rice (발아 벼 열수 및 에탄올 추출물의 암세포주 증식억제 효과)

  • This study was conducted to investigate the effect of 70% ethanol and water extract of Korean rough rice ('Ilpum', 'Goami2', 'Keunnun', 'Sulgaeng', 'Baegjinju', and 'Heugkwang') before and after germination on proliferation of human cancer cell lines (HepG2, PC-3, and MCF-7). Antiproliferation effect was higher in ethanol extract than water extract, and was higher in after germination. 'Ilpum' ethanol extract after germination showed higher anti-proliferation effect on HepG2 and PC-3 cell lines than before germination. The cell viability on HepG2 and PC-3 cell lines of 'Ilpum' ethanol extract after germination was 27.23% and 5.05% at 1.0 mg/mL, respectively. The anti-proliferation effect on MCF-7 was the highest in 'Ilpum' and 'Heugkwang' 70% ethanol extract after germination and those cell viabilities were 7.27% and 17.00% at 0.5 mg/mL, respectively. These results suggest that germinated rough rice might have a potentially preventive effect on human cancer cells.

Characterization of Protein Function and Differential Protein Expression in Soybean under Soaking Condition (Proteomics를 이용한 콩의 발아 전 침종처리에 따른 단백질 발현 양상 비교 분석)

  • Soybean is very useful crop to supply vegetable protein for human. However, cultivation arear of this economically important crop is gradually diminished in upland field. Hence, cultivation area of soybean is increased in paddy field. During the growth duration of soybean, excessive moisture injury is serious problem for sustainable production and supply. We investigated protein expression according to different period of seed soaking and germination after seed soaking. For comparison on expression of protein according to different condition, we performed two-dimensional electrophoresis. After electrophoresis analysis, we selected differentially expressed protein spots according to different condition such as soaking period and germination after soaking to identify protein function by using MALDI-TOF. Results revealed that pattern of expression of protein according to soaking period and germination after soaking were generally not different in major spots. However, degree of expression of protein in some protein spots was increased in accordance with decrease of soaking period. Especially, in Hwangkeum-Kong, Danyeop-Kon, and Pecking, the degree of expression of protein was remarkably increased for 4 days after soaking. But, according to germination after soaking, degree of expression of protein in germinated seeds of all cultivars was higher than un-germinated seeds. In results of MALDI-TOF analysis, specific proteins were identified by different soaking period such as Allergen Gly m Bd 28K, P24 oleosin isoform B. Also, in accordance with germination, degree of protein expression of the related protein, Gibberellin was increased in un-germinated seeds of Iksan-Kong. In ungerminated seeds of Sinpaldal-kong, proteins were identified as down-regulated by soaking such as ATP binding and Inhibitor II', proteinase.

N-phenyl Substitutent Effect on the Herbicidal Activity of 2-(4-(6-chloro-2-benzoxazolyloxy)phenoxy)-N-phenylpropionamide Derivatives against Rice Plant with Pre- and Post-emergence (발아 전 후 벼의 약해에 미치는 2-(4-(6-chloro-2-benzoxazolyloxy)phenoxy)-N-phenylpropionamide 유도체들 중 N-phenyl치환기의 효과)

  • The influence of the 2-(4-(6-chloro-2-benzoxazolyloxy)phenoxy)-N-phenyl- propionamide derivatives on the herbicide activities against rice plant with pre-emergence and post-emergence in down land were examined and the structure activity relationship (SAR) were analyzed by Free-Wilson and Hansch method. In pre-emergence, the SAR approach is shown that the optimal, $({\pi})_{opt}=0.91$, hydrophobicity with electron donating effect of the ortho substituted mono substituents and 2,3,4-substituted three substituents were found to be contribute the herbicidal activity. Whereas, in post-emergence, the optimal, ({\pi})_{opt}=0.50$, hydrophobicity with electron withdrawing effect of meta substituted mono subsituents and 2,3-substituted two substituents were found to be contribute the herbicide activity. The herbicide activities with post-emergence more increase than that of pre-emergence. It is assumed from the SAR equations that the 2-methyl-3-methoxy-4-cyano group substituent is selected as the most lowest herbicide activity against rice plant with post-emergence in green house. The hydrolysis reaction was proceeded through nucleophilic addition-elimination (Ad_{Nu-E})$ with the orbital control between LUMO of substrate and HOMO of water molecule. And molecular electrostatic potential (MEP) of none (H) substituent was discussed.

Fermentation of Germinated- and Nongerminated-Yellow Soybean Chungkookjang Using Bacillus subtilis (Bacillus subtilis에 의한 발아 및 미발아 황태 청국장 발효)

  • In order to investigate changes in quality and enzyme activity during Chungkookjang fermentation, germinated- and nongerminated yellow soybeans were fermented by Bacillus subtilis and traditional methods. When the soybean was soaked for 6 h and then watered for 4 days with 2 h-interval at $25^{\circ}C$, the highest germination rate was obtained. The germinated soybeans had a higher total isoflavone ($988.4{\mu}g/g$) than that of the nongerminated soybeans ($859.5{\mu}g/g$). Amino type nitrogen contents, protease and amylase activities were higher in germinated soybean Chungkookjang, which was fermented with B. subtilis, than nongerminated soybean Chungkookjang, which was fermented with B. subtilis and traditional methods. Reducing sugar and amino type nitrogen contents, the number of viable cells and protease and amylase activities, were higher for Chungkookjang fermented with B. subtilis, than Chungkookjang fermented by traditional methods. ALP and SOD activities in the Chungkookjang diet group were considerably higher than in the control group. AST activity in the germinated soybean Chungkookjang diet group was higher than in the nongerminated soybean Chungkookjang diet group. In conclusion, it is suggested that Chungkookjang prepared with germinated soybeans using B. subtilis D7 could be practically used as a functional product.

  • Natural infection of purple stain seemed to be higher in early maturing lines comparing to late maturing group. It was positively correlated with length of the period from flowering to maturity and significantly associated with weather conditions during the maturing period.

Studies on the Germination Characteristics of Sesame (Sesamum indicum L.) (참깨의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This study was carried out to define the effects of external factors including temperature, moisture, oxygen and light quality on the germination of sesame seeds and to investigate the change of major chemical constituents of seeds during germination. The results obtained are summarized as follows: 1. The average germination ratio was from 95.8% to 97.2% when it was tested every $5^{\circ}C$ intervals from $20^{\circ}C$ to $35^{\circ}C$ and no significant difference in germination ratio was found within $20^{\circ}C$ to $35^{\circ}C$. But the germination ratio dropped rapidly to 32.2% when seeds were germinated at $15^{\circ}C$ and the coefficient of variation become greater(77%) 2. The days required for germination ranged from 1.16 to 1. 64 at the temperatures of $35^{\circ}C$ to $25^{\circ}C$ and they were 3.07 and 10.4 at the temperatures of $20^{\circ}C$ and $15^{\circ}C$, respectively. 3. Considering the germination ratio and days needed, $15^{\circ}C$ was assumed to be the minimum temperature for germination practically and this temperature is recommended for testing low temperature tolerance of seed germination of sesame cultivars. 4. The varieties shown the highest low temperature tolerance were Shirogoma and Turkey. The next varieties shown some degree of low temperature germination were Suweon #29, Naebok and IS 58. The varieties with 70 to 80% of germination ratio were Maepo, Suweon #14, Kimpo, Moondeok, and Haenam. Among the 90 varieties tested, the varieties with comparatively high degree of low temperature tolerance were about 10%, and 70% of the low temperature tolerant varieties were domestic varieties. 5. At $12^{\circ}C$ the Shirogoma was the only variety which showed over 50% of germination ratio, 71.4% of the varieties showed less than 20% of germination ratio. When the temperature was raised to $27^{\circ}C$ 18 days after placement at $12^{\circ}C$ all the varieties showed over 90% of germination ratio within 2days. 6. The amounts of water imbibition needed for seed germination were 0.48 to 0.62 times of the seed dry weight at $25^{\circ}C$ and were significantly different among sesame cultivars. About 63% of water required for germination was imbibed in 2 hours after placement of seeds under the germination condition. 7. Under saturated moisture condition the average germination ratio was 0.42%. In the soil of which water potential was -0.4bar 64.8% of the seeds germinated and the most adequate soil water potential for sesame seed germination was about -0.4 to -5.5 bar. The germination ratio decreased as the soil water potential declined below -5.5 bar. 8. Six out of 10 varieties were not influenced by 5% of oxygen in air germination chamber, while varieties such as Yecheon, PI 158073, IS 103 and Euisangcheon showed 64 to 91% of germination under the 5% oxygen content. Under anaerobic condition, cotyledones were not emerged but only hypocotyl was emerged and elongated. The germination ratio of IS 103 decreased significantly under anaerobic condition. 9. When the seeds were dried for 24 hours after 12 hours imbibition of water, the seeds of Cheongsong did not lose their germination ability and 27.5% was germinated but Suweon #9 and Early Russian failed to germinate. However, the germination ratio of IS 103 decreased when the seed were dried 24 hours after 4 hours imbibition of water and the germination ability of IS 103 was maintained even though the seeds were dried for 24 hours after 24 hours imbibition of water. 10. During germination, sugar content of sesame seed increased rapidly and activity of ${\alpha}$-amylase increased gradually while starch content decreased significantly. The rates of increase in sugar content and enzyme activity and decrease in starch content were significantly lower at $15^{\circ}C$ compared with those at $25^{\circ}C$. 11. During germination of sesame seeds, lipid content in the seeds dropped rapidly and the activity of alkaline lipase increased significantly at early stage of germination. The rate of decrease in lipid content and increase in emzyme activity was lower at $15^{\circ}C$ than at $25^{\circ}C$. 12. Four out of 6 varieties were not affected in germination by light wave length. But Suweon #8 was inhibited in germination by 600-650nm. and IS 103 by 600 to 650nm and 500 to 550nm of light wave length. Suweon #8 showed high germination ratio under 650 to 760 nm and 500 to 560nm, and IS 103 under 400 to 470nm and complete darkness. 13. The germination ratios increased significantly in the seeds of which 1000 grain weight is heavier. When the seeds were placed at soil 4cm deep, Cheongsong and Early Russian failed to emerge their cotyledones, but Suweon #9 and IS 103 showed 32.5 and 50% cotyledone emergence, respectively. The extracts from sesame plant and soil where the sesame was cultivated previously did not affect in the-germination of sesame seeds. 14. The covering by black or transparent polyethylene films increased germination ratio compared with uncovered seeds. The covering was effective in shortening the days needed for germination and in improving the early seedling growth, number of capsules per plant and grain yield. Difference was not so seizable between the two polyethylene films but the transparent film appeared somewhat more effective than the black one. 15. Simcheon, Cheongsong. Suweon #9. PI 158073 and IS 103 showed lower rate of water absorbtion by seed during germination and Suweon #8, Suweon #26, Orotall and Euisangcheon showed high increase in seed weight after water absorbtion by seed.

Investigation on Disease Incidence and Yield of Rice Cultivars for Use in Processing of Eco-friendly-grown Germinated Brown Rice (친환경 발아현미 생산에 적합한 병 저항성 및 생산성이 우수한 벼 품종선발)

  • In order to select the optimum rice cultivars for the use in processing of eco-friendly-grown germinated brown rice (GBR), disease incidences and yields in rice cultivars bred in National Institute of Crop Science were investigated in eco-friendly paddy fields (Gokseong, Jeonnam Province, Korea) during the years 2009-2011. The incidences of rice sheath blight and blast on the cultivar Samgwang were higher than those on one reference cultivar Chucheong, but much lower than those on the other reference cultivar Gosihikkali. The cultivars Keunnun and Haiami selected as special rices showed disease incidences and yields similar to the reference rice cultivars Chucheong and Gosihikkali. These results indicated that the cultivars Samkwang, Keunnun, and Haiami can be selected as rice cultivars for the use of in processing of eco-friendly grown GBR because their disease incidences and yields in Gokseong may be similar to the reference cultivars.
