Experiments were conducted to measure rooting and growth of rose cuttings as affected by medium composition and volume. Rosa hybrida 'Red Sandra' and 'Little Marble' were used as test plants. For medium composition test, rockwool, phenolic foam, perlite, vermiculite, peatmoss, coir and a commercial medium (Tosilee medium) were applied in single or mixed forms on the propagation bed with fog (200 sec./5 min.) installed to control the humidification time. 'Red Sandra' and 'Little Marble' cuttings were planted in 16-cell plastic pots and 72-cell plug trays, respectively. For the medium volume test, 72-cell plug trays and 16-cell or 9-cell plastic pots were used. Cuttings with an uniform node with a five-leaflet leaf was soaked in a $500mg{\cdot}L^{-1}$ IBA solution for 30 sec. A fogging system installed on the propagation bed was used for (100 sec./5 min.). Rockwool 1 : peatmoss 1 (v/v) was the most effective on growth and rooting for both cultivars and high volume of Tosilee medium and rockwool were most effective media far both cultivars.

In order to promote in vitro rooting in SOD2-transgenic rose plantlets, which were not well rooted in a rooting medium (MS medium with NAA $0.03mg{\cdot}L^{-1}$), we dipped the plantlets into liquid $Kelpak^{(R)}$ before placing them in the rooting medium. After 4 weeks, $Kelpak^{(R)}$ significantly promoted in vitro rooting in the plantlets. Therefore, $Kelpak^{(R)}$ can be used successfully to aid in the in vitro rooting of rose plantlets with roots that are not well-generated.

Cuttings based on water media are relatively low-cost and easy to manage compared to those based on soil media. Nevertheless, species that root out in water media are very few. Water-medium rooted cuttings are not widely used. In water media, the survival rate of cuttings is high, but in many cases, they only form callus while not rooting. If cuttings with callus formed in a water medium were to root and survive after being transplanted in soil, water-medium cuttings could be used as an important means of propagating landscape tree species. In this study, cuttings of Ternstroemia japonica having various reactions to water media were soaked in different types of water media. Their callus formation and rooting conditions were analyzed. The water-medium cutting swere then transplanted in soil, and conditions for their survival were examined. The study concluded: 1. Cuttings that only formed callus in water media were able to root and survive after being transplanted. 2. Cuttings with satisfactory callus formation showed more satisfactory survival conditions than those with unsatisfactory callus formation. 3. Cuttings rooted in water media showed a high survival rate of 96.6% after being transplanted. 4. Water-medium cutting of Ternstroemia japonica proved to be more effective in Late July or August than in late June. 5. Cuttings pre-treated in 50ppm-IBA for 24 hours showed better survival conditions than those that were not pre-treated inIBA. 6. Cuttings pre-treated in 50ppm-IBA for 24 hours and soaked in water media with 0.02% atonic, 0.02% polyoxin, and 0.01% polyoxin showed a rooting and survival rate of 80 to 85%. Regardless their low rooting rate, water-medium cuttings can show a high survival rate when these factors are considered: proper cutting time, plant hormone treatment, and types of water media. Accordingly, water-medium cuttings will emerge as major means of propagating landscape tree species that require small volumes of multi-species.
The shoot tips of Delphinium cv. Princess Caroline were cultured on the MS medium supplemented with cytokinin and auxin alone or in combination. Among cytokinins, BA was most effective in shoot multiplication, adequqte concentrations being 1.0-5.0 mg/L. Shoot multiplication was very favorable on the media with 1.0-3.0 mg/L BA and 0.1-0.5 mg/L IAA. Additions of BA and IAA did not stimulate shoot multiplication, but increased a little fresh weight. Shoots were scarcely rooted on the media with IBA or NAA, and were not done utterly on the media containing activated charcoal. Therefore, shoots were treated by Rootone and planted in the cultural media for in vivo rooting. The highest rate of rooting was 68% in the mixed cultural medium composed of Perlite 1 and Vermiculite 1.
This experiments were carrid out to examine the effects of liquid medium addition in same vessels on shoot elongation and rooting, and soil survival of plantlets after the shoot cluster sections of Spathiphyllum floribundum 'Cupid' were pre-cultured. The shoot clusters with 3 to 4 small shoots were proliferated on LS medium supplemented with 2.0 mg/L BA for 8 weeks, and then 15 mL of various kinds of liquid medium was added in the same vessels. The addition of 15 mL liquid medium containing l/2 MS macro elements, 50 g/L sucrose and 5.0∼10.0 g/L activated charcoal was significantly stimulated the elongation and rooting of proliferated shoots. The medium addition was resulted in the enhanced soil survival of plantlets.
Effects of kanamycin concentration on regeneration and rooting of transgenic 'McIntosh Wijcik' were investigated to establish the efficient Agrobacterium-mediated transformation system. Relatively high regeneration frequency of explants appeared even at the high concentration of $150mg{\cdot}L^{-1}$ kanamycin, but the regeneration frequency and the number of normal shoots decreased significantly at a concentration of higher than $100mg{\cdot}L^{-1}$ kanamycin in the gelrite-gellifying medium. Rooting response varied with the transgenic lines in the agar-solidifying medium supplemented with the different concentrations of kanamycin and they were grouped with the inhibition level at $30mg{\cdot}L^{-1}$ concentration. No correlation between copy number and root response was observed. The optimum concentrations of kanamycin for the regeneration of 'McIntosh Wijcik' apple in the medium gellified with gelrite and for indirect-selection of putative transformants in the rooting medium solidified with agar were found to be $100mg{\cdot}L^{-1}$ and $30mg{\cdot}L^{-1}$ respectively.
In order to develop an efficient micropropagation system for a rare tree species, Empetrum nigrum var. japonicum K. Koch, the effect of medium salt, cytokinins and auxin at different concentration were evaluated. Shoot induction from axillary bud was better on WPM medium than on MS medium. Although there was no significant differences observed in shoot induction among the salt strengths of WPM medium, whereas healthy shoots were developed on basal WPM medium. In comparison of the cytokinins affecting shoot proliferation, zeatin was better than BA, whereas BA exhibited more effectiveness on shoot elongation. In vitro root formation was better on WPM medium than on 1/2MS medium and achieved the highest rooting rate when 5.0 mg/L IBA treatment. 93% of rooted plantlets were survived on artificial soil mixture after 4 weeks of acclimatization. Above results suggest that a rare tree species, E. nigrum var. japonicum can be micropropagated via axillary bud cultures.
This experiment was carried out to know the effect of media and agar concentrations, aeration and growth regulators on rooting and acclimatization of the shoots harvested from bioreactor culture in Rehmannia glutinosa. Half MS media with 1.2% agar improved rooting and acclimatization of shoots. Shoots were effectively acclimatized and rooted well in case of aeration by using membrane filtered vessels. Shoots acclimatized in vessel with membrane Inter were healthier and had higher ex vitro survival rate than those without membrane Inter on plug tray. Addition of paclobutrazol 0.3-0.4 mg/L, to acclimatization media enhanced shoots growth and root development.
This study was conducted to determine the optimum medium and humidification period for the strawberry (Fragaria × ananassa Duch. cv. Maehyang) cutting seedling production. The cuttings were planted in coir (CO), rockwool (RW), phenolic foam (PF), and organic foam medium (OFM) with fogging treatment in 0, 3, 6, 9, or 12 days on February, 2019. And, as the field verification examination research, cuttings were planted in CO, RW, PF, and OFM, with misting treatment in 0, 6, 9, 12, or 15 days on June, 2019. In fogging treatments, rooting and survival rates of seedlings tended to increase with longer fogging periods, and rooting and survival rates were showed significantly higher in CO medium with more than 9 days of fogging periods. In misting treatments, rooting rate was significantly higher in CO and RW medium with more than 9 days of misting periods, and survival rate was significantly higher with more than 12 days of misting periods. There was no significantly difference by growth medium and humidification period in shoot and root growth. Therefore, when considering the rooting and survival rate the CO medium with 9 days of humidification period could be beneficial for the production of the 'Maehyang' strawberry cutting seedlings.
Present study describes a method on the application of efficient tissue culture systems for the micro-propagation of juvenile and mature sawtooth oak(Quercus acutissima). Nodal segments with axillary buds were used as initial explant sources. WPM(Woody Plant Medium) was the best in growth and proliferation of shoot among the media tested. Although the single effect of zeatin revealed on two dorminant shoot elongation with normal growth until the elevation of levels up to 3.0mg/l, BAP($N^6$-benzyl amino purine) usually showed better response than zeatin on shoot multiplication and/or elongation. In addition, the incorporation of BAP and zeatin onto the culture media represents more effectiveness in shoot proliferation and its growth. Optimum concentrations of BAP and zeatin were 0.5 and 0.05~1.0mg/l, respectively. Ninety percent of the proliferated shoots was rooted on half-strength GD (Gresshoff and Doy, 1972) medium containing 0.5mg/l IBA(indole butyric acid) in 4 weeks after culture. More than 70% of the rooted plantlets survived after 5 months of transplanting into artificial soil mix containing equal amount of peatmoss and perlite. Among 27 plus tree clones which were grafted twice onto the juvenile rootstocks, only 4 clones revealed the possibility for shoot multiplication through tissue culture system. The capacity for the micropropagation using mature explant sources was highly depended on clonal differences compared with those of octet age. More than 90% of rooting ratio was obtained from the best responding clone. Among the 7 rooting media tested, GD medium was the best far rooting. The most effective rooting was obtained on half-strength GD medium containing 0.2 to 2.0mg/l IBA. More than 60% of rooted plantlets survived after 5 months of transplanting into the artificial soil mix.
