Successful Enrichment of Rarely Found Candidatus Anammoxoglobus propionicus from Leachate Sludge

  • Bacteria that mediate the anaerobic oxidation of ammonium (anammox) have been detected in natural ecosystems, as well as various wastewater treatment systems. In this study, sludge from a particular landfill leachate anaerobic treatment system was selected as the incubation seed for anammox microorganism enrichment owing to its possible anammox activity. Transmission electron microscopy observation, denaturing gradient gel electrophoresis analysis, and cloning/sequencing techniques were applied to identify the diversity of anammox microorganisms throughout the incubation. During the early stage of operation, the diversity of anammox microorganisms was similar to the original complex microbes in the seed sludge. However, as incubation time increased, the anammox microorganism diversity within the system that was originally dominated by Candidatus (Ca.) Brocadia sp. was replaced by Ca. Anammoxoglobus propionicus. The domination of Ca. Anammoxoglobus propionicus produced a stable removal of ammonia (70 mg-N/l) and nitrite (90 mg-N/l), and the total nitrogen removal efficiency was maintained at nearly 95%. The fluorescence in situ hybridization results showed that Ca. Anammoxoglobus propionicus was successfully enriched from $1.8{\pm}0.6%$ initially to $65{\pm}5%$ after 481 days of operation. Therefore, the present results demonstrated the feasibility of enriching Ca. Anammoxoglobus propionicus from leachate sludge, even though the original cell count was extremely low. Application of this seldom found anammox organism could offer an alternative to current ammonia-nitrogen treatment.

Diversity and Distribution of Methanogenic Archaea in an Anaerobic Baffled Reactor (ABR) Treating Sugar Refinery Wastewater

  • The diversity and distribution of methanogenic archaea in a four-compartment anaerobic baffled reactor (ABR) treating sugar refinery wastewater were investigated by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). At an organic loading rate of 5.33 kg $COD/m^3{\cdot}day$, the ABR could perform steadily with the mean chemical oxygen demand (COD) removal of 94.8% and the specific $CH_4$ yield of 0.21 l/g $COD_{removed}$. The $CH_4$ content in the biogas was increased along the compartments, whereas the percentage of $H_2$ was decreased, indicating the distribution characteristics of the methanogens occurred longitudinally down the ABR. A high phylogenetic and ecological diversity of methanogens was found in the ABR, and all the detected methanogens were classified into six groups, including Methanomicrobiales, Methanosarcinales, Methanobacteriales, Crenarchaeota, Arc I, and Unidentified. Among the methanogenic population, the acid-tolerant hydrogenotrophic methanogens including Methanoregula and Methanosphaerula dominated the first two compartments. In the last two compartments, the dominant methanogenic population was Methanosaeta, which was the major acetate oxidizer under methanogenic conditions and could promote the formation of granular sludge. The distribution of the hydrogenotrophic (acid-tolerant) and acetotrophic methanogens in sequence along the compartments allowed the ABR to perform more efficiently and steadily.

Response of Syntrophic Propionate Degradation to pH Decrease and Microbial Community Shifts in an UASB Reactor

  • The effect of pH on propionate degradation in an upflow anaerobic sludge blanket (UASB) reactor containing propionate as a sole carbon source was studied. Under influent propionate of 2,000 mg/l and 35℃, propionate removal at pH 7.5-6.8 was above 93.6%. Propionate conversion was significantly inhibited with stepwise pH decrease from pH 6.8 to 6.5, 6.0, 5.5, 5.0, 4.5, and then to 4.0. After long-term operation, the propionate removal at pH 6.5-4.5 maintained an efficiency of 88.5%-70.1%, whereas propionate was hardly decomposed at pH 4.0. Microbial composition analysis showed that propionate-oxidizing bacteria from the genera Pelotomaculum and Smithella likely existed in this system. They were significantly reduced at pH ≤5.5. The methanogens in this UASB reactor belonged to four genera: Methanobacterium, Methanospirillum, Methanofollis, and Methanosaeta. Most detectable hydrogenotrophic methanogens were able to grow at low pH conditions (pH 6.0-4.0), but the acetotrophic methanogens were reduced as pH decreased. These results indicated that propionate-oxidizing bacteria and acetotrophic methanogens were more sensitive to low pH (5.5-4.0) than hydrogenotrophic methanogens.

Enterococcus faecium R0026 Combined with Bacillus subtilis R0179 Prevent Obesity-Associated Hyperlipidemia and Modulate Gut Microbiota in C57BL/6 Mice

  • Bacillus subtilis and Enterococcus faecium are commonly used probiotics. This study aimed to identify the effect of live combined Bacillus subtilis R0179 and Enterococcus faecium R0026 (LCBE) on obesity-associated hyperlipidemia and gut microbiota in C57BL/6 mice. Forty male C57BL/6 mice were divided into four groups: normal group (N group), model group (M group), low-dose group (L group), and high-dose group (H group). Mice were gavaged with LCBE at 0.023 g/mice/day (L group) or 0.23 g/mice/day (H group) and fed with a high-fat diet for 8 weeks. In vitro E. faecium R0026 showed an ability to lower the low-concentration of cholesterol by 46%, and the ability to lower the high-concentration of cholesterol by 58%. LCBE significantly reduced the body weight gain, Lee index, brown fat index and body mass index of mice on a high-fat diet. Moreover, LCBE markedly improved serum lipids (including serum triglyceride, total cholesterol, low-density lipoprotein and high-density lipoprotein) while also significantly reducing liver total cholesterol. Serum lipopolysaccharide and total bile acid in L and H groups decreased significantly compared with M group. PCR-DGGE analysis showed that the composition of gut microbiota in the treatment groups was improved. Akkermansia muciniphila was found in H group. The PCA result indicated a similar gut microbiota structure between LCBE treatment groups and normal group while the number of bands and Shannon diversity index increased significantly in the LCBE treatment groups. Finally, qPCR showed Bifidobacterium spp. increased significantly in H group compared with M group, LCBE alleviated liver steatosis and improved brown adipose tissue index.

낙동강에서의 유기성 오염 유무에 따른 질화세균의 변화 (Changes of Nitrifying Bacteria Depending on the Presence and Absence of Organic Pollutant in Nak-Dong River)

  • 유기성 오염원(생활하수 처리시설 방류수) 유무에 따른 질화 세균의 변화를 알아보기 위해 낙동강의 2 조사수역에서 다양한 형태의 질소(T-N, $NH_4$-N, $NO_2$-N, $NO_3$-N) 농도를 측정하였고 조사 수역에 있는 질화세균 종류를 확인한 후, fluorescent in situ hybridization (FISH)법으로 질화세균 수를 정량 평가하였다. 즉 암모니아 산화세균의 종류는 amoA 유전자, 그리고 아질산 산화 세균인 Nitrobacter sp.는 SSU 16S rDNA 특정 기호서열을 목표로 한 PCR-DGGE를 수행한 후 염기서열 분석으로 그들이 각각 Nitrosomonas sp., Nitrobacter sp.임이 확인됨에 따라 그에 상응하는 gene probe, NSO190와 NIT3을 사용해 FISH법을 수행하여 각 수역의 질화세균수를 비교하였다. 아울러 전반적인 세균학적 수질을 모니터링하기 위해 수계에 많은 ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria도 FISH법으로 검출하였다. ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria 분포도와 모든 유형의 질소 측정 결과에 따르면 정점 1 보다 정점 2의 유기물 오염도가 높다고 할 수 있었다. 이에 상응해 NSO160과 NIT3로 검출한 평균 질화세균수도 정점 1 (암모니아산화세균, $7.8{\times}10^5$; 아질산산화세균, $0.8{\times}10^6$ cells/ml)보다 정점 2 (암모니아산화세균, $9.3{\times}10^5$; 아질산산화세균, $1.6{\times}10^6$ cells/ml)에 더 많았고 그들이 총세균수에서 차지하는 평균 비율(%) 역시 정점 1 (NSO190, 18%; NIT3, 23%)에 비해 정점 2 (NSO190, 27%; NIT3, 34%)에서 높았다. 따라서 유기오염도가 낮은 상수원수 취수장 인근 수역인 정점 1 보다 생활하수처리시설 방류수로 인해 유기성 오염원이 상존하는 정점 2에서의 질화작용이 더 활발하다고 결론지을 수 있었다.

Stability of Partial Nitrification and Microbial Population Dynamics in a Bioaugmented Membrane Bioreactor

  • Bioaugmentation of bioreactors focuses on the removal of numerous organics, with little attention typically paid to the maintenance of high and stable nitrite accumulation in partial nitrification. In this study, a bioaugmented membrane bioreactor (MBR) inoculated with enriched ammonia-oxidizing bacteria (AOB) was developed, and the effects of dissolved oxygen (DO) and temperature on the stability of partial nitrification and microbial community structure, in particular on the nitrifying community, were evaluated. The results showed that DO and temperature played the most important roles in the stability of partial nitrification in the bioaugmented MBR. The optimal operation conditions were found at 2-3 mgDO/l and $30^{\circ}C$, achieving 95% ammonia oxidization efficiency and nitrite ratio ($NO_2^-/{NO_x}^-$) of 0.95. High DO (5-6 mg/l) and low temperature ($20^{\circ}C$) had negative impacts on nitrite accumulation, leading to nitrite ratio drop to 0.6. However, the nitrite ratio achieved in the bioaugmented MBR was higher than that in most previous literatures. Denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH) were used to provide an insight into the microbial community. It showed that Nitrosomonas-like species as the only detected AOB remained predominant in the bioaugmented MBR all the time, and coexisted with numerous heterotrophic bacteria. The heterotrophic bacteria responsible for mineralizing soluble microbial products (SMP) produced by nitrifiers belonged to the Cytophaga-Flavobacterium-Bacteroides (CFB) group, and $\alpha$-, $\beta$-, and $\gamma$- Proteobacteria. The fraction of AOB ranging from 77% to 54% was much higher than that of nitrite-oxidizing bacteria (0.4-0.9%), which might be the primary cause for the high and stable nitrite accumulation in the bioaugmented MBR.

Microbial Floral Dynamics of Chinese Traditional Soybean Paste (Doujiang) and Commercial Soybean Paste

  • Traditional soybean paste from Shandong Liangshan and Tianyuan Jiangyuan commercial soybean paste were chosen for analysis and comparison of their bacterial and fungal dynamics using denaturing gel gradient electrophoresis and 16S rRNA gene clone libraries. The bacterial diversity results showed that more than 20 types of bacteria were present in traditional Shandong soybean paste during its fermentation process, whereas only six types of bacteria were present in the commercial soybean paste. The predominant bacteria in the Shandong soybean paste were most closely related to Leuconostoc spp., an uncultured bacterium, Lactococcus lactis, Bacillus licheniformis, Bacillus spp., and Citrobacter freundii. The predominant bacteria in the Tianyuan Jiangyuan soybean paste were most closely related to an uncultured bacterium, Bacillus licheniformis, and an uncultured Leuconostoc spp. The fungal diversity results showed that 10 types of fungi were present in the Shandong soybean paste during the fermentation process, with the predominant fungi being most closely related to Geotrichum spp., an uncultured fungal clone, Aspergillus oryzae, and yeast species. The predominant fungus in the commercial soybean paste was Aspergillus oryzae.

Diversity of Arbuscular Mycorrhizal Fungi Associated with a Sb Accumulator Plant, Ramie (Boehmeria nivea), in an Active Sb Mining

  • Arbuscular mycorrhizal fungi (AMF) have great potential for assisting heavy metal hyperaccumulators in the remediation of contaminated soils. However, little information is available about the symbiosis of AMF associated with an antimony (Sb) accumulator plant under natural conditions. Therefore, the objective of this study was to investigate the colonization and molecular diversity of AMF associated with the Sb accumulator ramie (Boehmeria nivea) growing in Sb-contaminated soils. Four Sb mine spoils and one adjacent reference area were selected from Xikuangshan in southern China. PCR-DGGE was used to analyze the AMF community composition in ramie roots. Morphological identification was also used to analyze the species in the rhizosphere soil of ramie. Results obtained showed that mycorrhizal symbiosis was established successfully even in the most heavily polluted sites. From the unpolluted site Ref to the highest polluted site T4, the spore numbers and AMF diversity increased at first and then decreased. Colonization increased consistently with the increasing Sb concentrations in the soil. A total of 14 species were identified by morphological analysis. From the total number of species, 4 (29%) belonged to Glomus, 2 (14%) belonged to Acaulospora, 2 (14%) belonged to Funneliformis, 1 (7%) belonged to Claroideoglomus, 1 (7%) belonged to Gigaspora, 1 (7%) belonged to Paraglomus, 1 (7%) belonging to Rhizophagus, 1 (7%) belonging to Sclervocystis, and 1 (7%) belonged to Scutellospora. Some AMF sequences were present even in the most polluted site. Morphological identification and phylogenetic analysis both revealed that most species were affiliated with Glomus, suggesting that Glomus was the dominant genus in this AMF community. This study demonstrated that ramie associated with AMF may have great potential for remediation of Sb-contaminated soils.

A Laboratory-Scale Study of the Applicability of a Halophilic Sediment Bioelectrochemical System for in situ Reclamation of Water and Sediment in Brackish Aquaculture Ponds: Establishment, Bacterial Community and Performance Evaluation

  • In this study, we investigated the potential of using sediment bioelectrochemical systems (SBESs) for in situ treatment of the water and sediment in brackish aquaculture ponds polluted with uneaten feed. An SBES integrated into a laboratory-scale tank simulating a brackish aquaculture pond was established. This test tank and the control (not containing the SBES) were fed with shrimp feed in a scheme that mimics a situation where 50% of feed is uneaten. After the SBES was inoculated with microbial sources from actual shrimp pond sediments, electricity generation was well observed from the first experimental week, indicating successful enrichment of electrochemically active bacteria in the test tank sediment. The electricity generation became steady after 3 weeks of operation, with an average current density of $2.3mA/m^2$ anode surface and an average power density of $0.05mW/m^2$ anode surface. The SBES removed 20-30% more COD of the tank water, compared to the control. After 1 year, the SBES also reduced the amount of sediment in the tank by 40% and thus could remove approximately 40% more COD and approximately 52% more nitrogen from the sediment, compared to the control. Insignificant amounts of nitrite and nitrate were detected, suggesting complete removal of nitrogen by the system. PCR-DGGE-based analyses revealed the dominant presence of Methylophilus rhizosphaerae, Desulfatitalea tepidiphila and Thiothrix eikelboomii, which have not been found in bioelectrochemical systems before, in the bacterial community in the sediment of the SBES-containing tank. The results of this research demonstrate the potential application of SBESs in helping to reduce water pollution threats, fish and shrimp disease risks, and thus farmers' losses.

펜톤산화 및 생물학적 연속처리를 통한 유류오염토양의 효율적 처리 (Efficient Remediation of Petroleum Hydrocarbon-Contaminated Soils through Sequential Fenton Oxidation and Biological Treatment Processes)

  • 유류의 유출로 인한 총석유계탄화수소(total petroleum hydrocarbons: TPH)는 종종 토양과 지하수의 오염을 초래하고 있다. TPH는 환경에 노출이 될 경우 물리화학적 과정을 거쳐 분해가 되나 그 반응은 상대적으로 느리다. 본 연구에서는 TPH로 오염된 토양의 환경친화적인 처리기법을 궁극적으로 개발하기 위해서 화학적 및 생물학적 통합기술을 도입하고자 시도하였다. 여기서 펜톤유사반응을 전처리단계로 도입하고 이후 디젤분해 혼합균을 처리하여(생물증강법) 오염유류를 처리하고자 하였다. 계면활성제 OP-10S (0.05%)과 산화제($FeSO_4$ 4%, 및 $H_2O_2$ 5%)를 사용할 경우 토양으로부터 효율적으로 TPH를 처리, 제거할 수 있는 것으로 나타났다. 디젤분해 혼합균을 토양슬러리에 접종할 경우 100배 이상 분해균의 밀도상승이 관찰되었는데 이는 접종된 분해균이 오염된 토양에서 성공적으로 존재할 수 있음을 의미한다($10^8-10^9$ CFU/g slurry). Fenton으로 처리된 토양에서의 TPH 제거 효율은 분해균으로 생물증강을 실시할 경우 최소한 57% 정도 상승되는 것으로 나타났다. 그러나 화학적, 생물학적 연속처리를 실시할 경우 대조구(무처리; 재거효율 95%)에 비해 상대적으로 낮은 처리효율(79-83%)을 나타내었는데, 이는 화학처리 중에 발생하는 자유기(free radicals) 함유 산화물질이 분해를 억제한 것에 기인하는 것으로 보인다. 본 연구에서의 얻어진 결과는 환경에 있어서 TPH로 오염된 토양과 저질을 효율적으로 정화하고 토양생태계의 신속한 회복에 활용할 수 있을 것으로 판단된다.
