Effect of microbial product on microorganisms in soil and growth of cabbage and tomato (미생물제재 처리에 의한 토양 미생물상의 변화 및 배추와 토마토의 생장에 미치는 영향)

  • Effect of the microbial product, which consisted of Bacillus coagulans DL-1 and rice bran, on the microorganisms in soil and growth of cabbage and tomato was investigated. Bacillus congulans DL-1 was isolated form the soil and identified in this study. Total number of microorganisms in the soil treated with the microbial product was higher than the untreated soil. The growth of cabbage and tomato on the soil treated with microbial product was faster than that on the untreated soil. The treatment of microbial product in the soil resulted in the increase of useful microorganisms, which seemed to enhance the growth of cabbage and tomato. It seemed that microbial product can increase the number of certain microorganisms and change the ratio of different species of microorganisms.

  • The term mycoherbicide started in 1970, but its interest heightened due to increase costs of chemical herbicides. A classical biocontrol agent is expected to become a permanent part of its new environment and do no harm to it. Contemporary biological control agent(BCA) must be produced by artificial culture and could be applied like chemical herbicides over weeds. BCA is different from the classical approach in that it released through natural spread. To date 26 species of fungi are used as classical BCA against 26 species of weeds in seven countries. There are a number of examples of pathogens attacking non-target plants. But through risk assessments which include understanding the taxonomy, biology and ecology, the target and non-target species, it will be safe to introduce of exotic pathogens to control weeds. But pathogens have not been successfully used in practice. Many mycoherbicides show potential in laboratories, but are ineffective in the field and not consistent from year to year or field to field. There is also a lack of understanding humidity, dew formation and temperature and their effects on suppression of weeds by plant pathogens. Potential pathogen must be selected as a BCA. Previous studies suggest that these pathogens must (1) produce abundant and durable inoculum in artifical culture, (2) be genetically stable and weed specific and (3) kill weeds in control. A granular preparation of mycoherbicide into sodium alginate is lighter than liquids and less bulky than organic matter. Gel forms have also been used.

  • Effect of the microbial product made of Bacillus stearothermophilus DL-3, which was isolated from the soil and identified in this study, and rice bran on microorganisms in soil and growth of lettuce (Red skirt lettuce) and Chinese cabbage (Ga rack new No.1 Chinese cabbage) was investigated. Total numbers of microorganisms in the pot with untreated soil, treated with standard amount of microbial product and treated with double amounts of microbial product for growth of lettuce after 6 weeks were 2.78${\times}$10$^{7}$ CFU/g, 2.72${\times}$10$^{8}$ CFU/g and 3.63${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the soil without treatment of microbial product and treated with standard amount of microbial product were 2.06${\times}$10$^{8}$ CFU/g and 5.49${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the pot with untreated soil, treated with standard amount of microbial product and treated with double amounts of microbial product for growth of Chinse cabbage after 6 weeks were 1.43${\times}$10$^{7}$ CFU/g, 3.42${\times}$10$^{8}$ CFU/g and 7.22${\times}$10$^{8}$ CFU/g. Total numbers of microorganisms in the soil without treatment of microbial product and treated with standard amount of microbial product were 5.75${\times}$10$^{8}$ CFU/g and 7.96${\times}$10$^{8}$ CFU/g. On basis of leaf length, leaf width, leaf number, wet weight and dry weight, the growth of lettuce and Chinese cabbage on the soil treated with microbial product was faster than that on the untreated soil. The treatment of microbial product in the soil resulted in the increase of useful microorganisms, which seemed to enhance the growth of lettuce and Chinese cabbage.

Environmental Bioresources Laboratory, Korea Research Institute of Biosicence and Biotechnology, Taejon, Korea, 1 Department of Agricultural Chemistry, Kyungpool< National University, Taegu, Korea - The biodegradation of chlorinated phenols in an artificial wastewater was investigated using a mixed culture. The mixed culture was composed of 8 microorganisms isolated from the soil contaminated with various chlorinated phenols. Pseudomonas sp. BM as a main constituent of a mixed culture was Gram-negative, catalase- and oxidase-positive, and rod-shaped, and did not grow at 41°C. It degraded 99% of initial 500 mg!1 of pentachlorophenol (PCP) in the minimal salts medium as a sole source of carbon and energy within 3 days. The degradation efficiency of Pseu.domon.as sp. BM was not affected by the other organic carbon and nitrogen compounds. Pseudomonas sp. BM was able to grow in a broad range of pH 5 - 8, and degrade 2,000 mg/1 PCP. In the experiment with an artificial wastewater containing chlorinated phenols, the degradation efficiency of the mixed culture was the range of 73% (2,4-dichlorophenol) -96% (2-chlorophenol) during an incubation of 7 days. In a continuous culture experiment, the degradation efficiency of mixed culture plus activated sludge was about 2 times higher than that of the control containing only activated sludge. These results indicate that it is possible to apply the mixed culture to other wastewaters containing chlorinated phenols. Effect on Digestion Efficiency by Adding Microbial Agent in Mesophilic Two-stage Anaerobic Digester (중온2단혐기성소화조에 미생물제재 주입시 소화효율에 미치는 영향)

  • In the near future, the capacity of conventional anaerobic digester is thought to be insufficient because of the increase of the total solids from expansion of intercepting sewer, sewage quantity and direct input of night soil from near apartment districts. The objectives of this study was to investigate the improvement of digestion efficiency using microbial agent(Bio-dh). The system was a pilot-scale, two-staged, anaerobic sludge digestion system. The first-stage digester was heated and mixed. The agitation velocity of the first-stage digester was 120rpm. The second-stage digester was neither heated nor mixed. The Digestion temperature was kept at $35{\pm}1^{\circ}C$ The detention time of digester was 19 days. The dosage of sewage sludge and microbial agent were $0.65m^3/day$ and $0.5{\ell}/day$, respectively. The experiments was run for 25days. Three times a week, $COD_{Mn}$ and SS of effluent, TS, VS, and biogas production rate were measured. Temperature, pH, and alkalinity were measured daily. The results were as follows ; Without microbial agent, digestion efficiencies ranged 46.0%~50.9%(mean=48.6%), with microbial agent(Bio-dh), digestion efficiencies ranged 52.8%~57.3%(mean=54.2%). Consequently, microbial agent(Bio-dh) increased the sludge digestion efficiency about 12%. Also, Without microbial agent, the mean concentration of $COD_{Mn}$ and SS of second-stage digester effluent were 1,639mg/L, 4,888mg/L respectively. With microbial agent, the mean concentration of $COD_{Mn}$ and SS of second-stage digester effluent were 859mg/L, 2,405mg/L respectively. Consequently, microbial agent(Bio-dh) increased the removal efficiency of $COD_{Mn}$ and SS about 47.6% and 50.8%, respectively.

  • Effect of microbial products made of Bacillus stearothermophilus DL-3 on growth of chickens and pigs was investigated. Two types of microbial product were made in this study. One is the microbial product made of culture broth of B. stearothemophilus DL-3 and rice bran which named as the microbial product A. The other is the microbial product made of culture broth of B. stearothermophilus DL-3, apple pomace, soybean pomace and rice bran which named as the microbial product B. Chickens were divided into three groups and each group was fed with $100\%$ general feed, $90\%$ general feed supplemented with $10\%$ microbial product A or $90\%$ general feed supplemented with $10\%$ microbial product B. The average chicken weight of each group was $41.1{\pm}2.5g,\;41.6{\pm}3.2g\;and\;42.3{\pm}2.9g$ and those after 28 days was $547.7{\pm}91.7g,\;560.1{\pm}17.2g\;and\;562.2{\pm}32.5g$, respectively. The average weight gain for each group was 506.6 g, 518.6 g and 519.9 g, respectively, and weight increases of groups fed with $90\%$ general feeder and $10\%$ microbial product A and B were $2.4\%\;and\;2.6\%$ higher than the group fed with $100\%$ general feed. Pigs were also divided into three groups and each group was fed like chickens. The average weight of each group was $9.3{\pm}1.0kg,\;9.4{\pm}1.1kg\;and\;9.6{\pm}1.0kg$ and those after 37 days was $19.3{\pm}4.1kg,\;20.2{\pm}3.9kg\;and\;20.8{\pm}4.2kg$, respectively. The average Weight gain for each group was 10.65 kg, 10.82 kg and 11.20 kg, respectively, and weight increases of groups fed with $90\%$ general feeder and $10\%$ microbial product A and B were $1.6\%$ and $5.2\%$ higher than the group fed with $100\%$ general feed.

  • Organic farming is necessary to sustainable agriculture, preserve biodiversity and continued growth the sector in agriculture. In organic farming, reduced usage of chemical agents that adversely affect human health and environment, employing amino acids and oil cake fertilizer, plant extracts, and microbial agents are used to provide safe agricultural products to consumers. To investigation microbiome structure, we proceeded on the pepper plant with difference fertilizers and treatments in organic agriculture for three years. The microbial communities were analyzed by the next generation sequencing approach. Difference soil microbiota communities were discovered base on organic fertilizer agents. Occurrences of virus and anthracnose diseases had a low incidence in conventional farming, whereas bacteria wilt disease had a low incidence in microbial agents treated plots. Microbe agents, which applied in soil, were detected in the microbial community and the funding suggested the applied microbes successfully colonized in the organic farming environment.

  • Recently, several studies have been conducted on biogas and organic compost production using food waste in an anaerobic digester. In this study, basic experiments were conducted to produce biogas and compost by fermenting food wastes with microbial agents. First, a microbial agent was developed by combining various microorganisms. Then, the amount of generated biogas was identified through a food waste batch experiment. Further, we could maximize and demonstrate biogas production in an anaerobic digester by examining biogas production and composting in a pilot plant.

Combined Effects of Sanitizer Mixture and Antimicrobial Ice for Improving Microbial Quality of Salted Chinese Cabbage during Low Temperature Storage (저온 저장 중 절임배추의 미생물학적 품질 향상을 위한 혼합 살균제재와 항균성 얼음 병합처리 효과)

  • The combined effects of a sanitizer mixture solution and antimicrobial ice on the quality of salted Chinese cabbages were examined. Salted Chinese cabbages were treated with a sanitizer mixture (comprised 50 ppm aqueous $ClO_2$ and 0.5% citric acid), packed in 2% brine and antimicrobial ice, and stored for 12 days at 4 and $10^{\circ}C$. Microbiological data on the salted Chinese cabbages after washing with the sanitizer mixture indicated that the populations of total aerobic bacteria, and yeast and molds decreased by 2.20 and 1.28 log CFU/g after treatment with the sanitizer mixture. In addition, coliforms population of salted Chinese cabbage after 12 days storage at $4^{\circ}C$ in the combined mixture of the sanitizer and antimicrobial ice was 3.22 log CFU/g, which was a significantly different from that of control (5.46 log CFU/g). The combined treatment of sanitizer mixture, antimicrobial ice, and low temperature at $4^{\circ}C$ suppressed reduction of pH and elevation of titratable acidity, resulting in delaying the growth of lactic acid bacteria. Differences in salinity, hardness, and Hunter's $L^*$, $a^*$, and $b^*$ values among treatments were negligible during storage at $4^{\circ}C$. Therefore, this study suggests that a combination of sanitizer mixture, antimicrobial ice treatment, and low temperature storage could improve the microbial safety and quality of salted Chinese cabbages during storage.
