Biogenic Synthesis of Metallic Nanoparticles and Their Antibacterial Applications (금속 나노입자의 생체 합성과 항균적 적용)

  • Recent studies on synthesis of metallic nanomaterials such as silver (Ag), gold (Au), platinum (Pt), cerium (Ce), zinc (Zn), and copper (Cu) nanoparticles (NPs) using plants and microbes are attracted researchers for their wide range of applications in the field of biomedical sciences. The plant contains abundant of bioactive contents such as flavonoids, alkaloids, saponins, steroids tannins and nutritionals components. Similarly, microbes produce bioactive metabolites, proteins and secretes valuable chemicals such as color pigments, antibiotics, and acids. Recently reported, biogenic synthesis of NPs in non-hazardous way and are promising candidates for biomedical applications such as antibacterial, antifungal, anti-cell proliferative and anti-plasmodia activity. All those activities are dose dependent, along with their shape and size also matters on potential of NPs. Microbes and plants are great source of metabolites, those useful in biomedical field, such metabolites or chemicals involved in synthesis of NPs in an ecofriendly way. NPs synthesized using microbes or plant materials are reveals more non-toxic, facile, and cost-effective compare to chemically synthesized NPs. In present review we are focusing on NPs synthesis using biological agents such as microbes (bacteria, fungi and algae) and plant, characterization using different techniques and their antibacterial applications on pathogenic Gram-positive and Gram-negative organisms.

Biological Control of White Rot in Apple Using Bacillus spp. (Bacillus spp.를 이용한 사과 겹무늬썩음병의 생물학적 방제)

  • Apple white rot, caused by Botryosphaeria dothidea, is one of the important diseases in Korea. B. dothidea can cause pre- and postharvest decay on apple fruit as well as canker and dieback of apple trees. In this study, we isolated bacteria from the trunk of apple trees and tested their antagonistic activity against B. dothidea. Five bacterial isolates (23-168, 23-169, 23-170, 23-172, and 23-173) were selected that were most effective at inhibiting the mycelial growth of the pathogens. The isolate 23-172 was identified as Bacillus amyloliquefaciens and four isolates 23-168, 23-169, 23-170, and 23-173 were identified as Bacillus velezensis by RNA polymerase beta subunit (rpoB) and DNA gyraseA subunit (gyrA) gene sequencing. All isolates showed strong antagonistic activity against B. dothidiea as well as Colletotrichum fructicola and Diaporthe eres. All isolates exhibited cellulolytic, proteolytic and phosphate solubilizing activities. In particular, two isolates 23-168, 23-169 were shown to significantly reduce the size of white rot lesions in pretreated apple fruits. These results will provide the basis for the development of a fungicide alternative for the control of white rot of apple.

The Effect Estimation of Heavy Metals on the Microbial Activity during Leaf Litter Decomposition (낙엽분해동안 미생물 활성에 미치는 중금속의 영향 추정)

  • This study was to find out influence of heavy metal concentration in plant on microbial activities during decomposition of Artenmisia princeps var. orientalis and Equisetum arvense collected from an abandoned mine and control site in Cheongyang-gun Chungcheongnam-do. Microbial respiration rate showed the highest value at the time of the first collection, and then tended to decline over time. The highest microbial respiration rate appeared in leaf litters with low heavy metal contents, and A. princeps var. orientalis and E. arvense collected and decomposed at the control site showed the fastest decomposition rate. For both A. princeps var. orientalis and E. arvense, litters with low heavy metal content appeared to have higher microbial biomass. There was apparent quantitative correlation between decomposition rate and cumulative respiration rate of leaf litters, and between decomposition rate and microbial biomass of leaf litters. Thus, the study result showed that leaf litter with higher heavy metal content had a negative impacts on the growth and activity of microbial decomposer during decomposition processes.

Control Effects of Micromonospora sp. AW050027 by Media Optimization and Microbial Treatment Against Pine Wood Nematode (Micromonospora sp. AW050027 균주의 배지최적화 및 미생물제제 처리에 의한 소나무재선충 방제효과)

  • Nematicidal activity against pine wood nematode, Bursaphelenchus xylophilus, was tested in the pot and field by the treatment of microbial formulation after media optimization. The optimized media composition was glycerol 10 g, soybean meal 10 g, NaCl 1 g, $CaCO_3$ 2 g, $K_2HPO_4$ 0.125 g per liter and microbial complex formulation was made with liquid and powder type. Most effective antibiotics against symbiotic microorganism with nematode, kanamycin, was added to the formulation. The control effects against pine wood nematodes were checked by pot test and field test. In the result of treatment by trunk injection, five times treatment was more effective than one time and the treatment with the formulation of concentrated culture supernatant was the most effective in the nematicidal activity showing below 10% mortality in pine tree.

Imitation of Phosphoenolpyruvate to Oxaloacetate Pathway Regulation of Rumen Bacteria in Enteric Escherichia coli and Effect on C4 Metabolism (반추위 미생물이 가진 Phosphoenolpyruvate에서 Oxaloacetate 경로 조절기작의 대장균에서의 모사와 C4대사의 영향)

  • One of the fermentative metabolism of enteric Escherichia coli was imitated after rumen bacteria, which have high C4 metabolism. E. coli expresses phosphenolpyruvate carboxylase (PPC) for the pathway between phosphoenolpyruvate (PEP) and oxaloacetate (OAA) during glycolytic condition while expresses phosphoenolpyruvate carboxykinase (PCK) during gluconeogenic condition. In contrast to enteric E. coli, rumen bacteria express the PEP-OAA pathway only by PCK. To verify the effect of the regulation imitation on the C4 metabolism of E. coli, PPC-deficient E. coli strain with PCK expression in glycolytic condition was constructed. The PEP-OAA regulation modified E. coli strain increased 2.5-folds higher C4 metabolite than the wild type strain. The potential use of C4 metabolism by regulation control is discussed.

Antibacterial Properties Associated with Microorganisms Isolated from Arctic Lichens (북극 지의류 유래 미생물의 항균성)

  • A total of 5 different polar microorganisms were isolated from Arctic lichens and their bioactive compounds were extracted from cell culture using different solvents including acetone, water, chloroform, diethylether, ethanol, ethyl acetate, methanol, and petroleum ether. The antibacterial properties of the extracts were evaluated by disk diffusion tests and minimal inhibitory concentration tests against 6 bacterial pathogens; Staphylococcus aureus, Bacillus subtilis, Micrococcus luteus, Enterobacter cloacae, Pseudomonas aeruginosa and Escherichia coli. Among the extraction samples, ethyl acetate extracts of Burkholderia sordidicola S5-$B^T$ (KOPRI 26644) showed the highest activity (inhibition zone, 7-10 mm; MIC value, 57.5-1000 ug/ml) against targeted bacteria. Among the various solvents used for extraction, chloroform extract exhibited the weakest, but still obvious, activity.

Selection of Antagonistic Soil Actinomycetes Against Both Colletotrichum orbiculare and Botrytis cinerea in Cucumber Plants (오이 탄저병원균과 잿빛곰팡이병원균에 억제 효과가 있는 방선균 선발)

  • The purpose of this study is to select actinomycetes with antifungal activity against Botrytis cinerea and Colletotrichum orbiculare, which are airborne pathogens in cucumber plants. In 2020, 560 species of actinomycetes were isolated from rhizome soils of various vegetables in Korea; 7 strains showing simultaneous antifungal activity against two pathogens were selected. Finally, strain S20-465 was selected through dual culture and plant assay. This strain was identified as Streptomyces sp. based on 16S rRNA analysis. The culture filtrate of strain S20-465 inhibited mycelial growth of both pathogens by more than 60%. In addition, when cucumber plants were treated with 20-fold and 40-fold diluted culture filtrates of S20-465, lesions caused by B. cinerea and C. orbiculare on cucumber leaves were significantly reduced compared to the control. This results suggest that strain S20-465 produces specific secondary metabolites with antifungal activity against both pathogens.

Modulation of Human Macrophage Phagocytic Activity by C-reactive Protein (C 반응성 단백질이 사람 Macrophage 탐식 기능에 미치는 영향)

  • The effects of CRP purified from human ascites fluid on phagocytic activity of the human macrophage were investigated. CRP was purified using affinity chromatography including absorption on p-diazonium phosphocholine or C-polysaccharide coupled sepharose 4B and gel filtration on hydroxylapatite column chromatography. Macrophage was separated ficoll hypaque gradient density and absorption method, and then was confirmed phagocytic uptake test using latex method. CRP was able either to inhibit or to enhance phagocytic activity of human macrophage against bacteria in vitro. The effects of CRP on phagocytic activity of human macrophage were in time and dose-dependent manners. The additional sequence of reaction mixture against bacteria in vitro shows a threshold stimulus on the activation of phagocytic response upon the CRP.

Isolation of Polyene Antifungal Antibiotics Against Gummy Stem Light Caused by Didymella bryoniae (Streptomyces sp. 유래 Polyene 계 항만고병 항생물질의 분리)

  • Antifungal agents, flavofungin and fungichromin were isolated from the fermentation culture broth of a Streptomyces sp. SKM338. Biological evaluation of these antibiotics indicated that the compounds possesses broad spectrum antifungal activity against various pathogens. Especially, these compounds inhibited throughly growth of Didymella bryoniae, caused Gummy stem blight of melons, occurs in the southeastern Korea. Inhibition of this pathogen may be prevented from directly reducing both pre- and post-harvest yields.

Purification and Characterization of Cholesterol Oxidase Produced by Soil Microorganism HSL613 (토양 미생물 HSL613이 생산하는 Cholesterol Oxidase의 정제 및 특성)

  • The extracellular cholesterol oxidase produced from a soil microorganism HSL613 was purified and partially characterized. Through a series of purification procedures including concentration with CH2 concentrator, DEAE-cellulose column chromatography and gel filtration on Superose12, the purified enzyme was shown to have a specific activity of 108 units/mg protein giving 30.8-fold purification and final yield of 66%. The molecular weight of the enzyme was estimated to be 59,500 daltons by SDS-PAGE. The optimum temperature and pH for this enzyme were $50^{\circ}C$ and 6.0, respectively. The activity of the purified cholesterol oxidase was inhibited by $Ag^{2+}$, $Hg^{2+}$ and SDS.
