This study was designed to investigate the effect of cactus (Opuntia ficus-indica var. saboten) extracts on the intestinal bacteria, antioxidative activity and angiotensin -I-converting enzyme(ACE) inhibitory activity. The antimicrobial activities were measured using the 96well-plate method and disc plate method with concentration of 20mg of cactus extract. The stem extract of cactus was inhibitory against Eubacterium limosum, Clostridium perfringens, C. butyricum, C. difficile and Staphylococcus aureus, but was not inhibitory against Bacteroides fragilis, Bifidobacterium bifidum, Lactobacillus acidophilis, Streptococcus thermophilus. The fruit extract of cactus showed no inhibition against Bacteroides fragilis, Bifidobacterium bifidum, Lactobacillus acidophilis, and Streptococcus thermophilis. Their inhibitory activities were not reduced after heat and pH treatment. Antioxidative effects of cactus extracts showed high total polyphenol and flavonoid contents and high activity against free radical DPPH. The stem and fruit extract of cactus showed strong ACE inhibitory activities of 88.8% and 69.2%, respectively. In conclusion cactus (Opuntia ficus-indica var. saboten) extract might be utilized as a functional food material to control intestinal microflora.
A study for recovering of copper and lead from electronic scraps has been carried out using a combination of bioleaching and solvent extraction. It was found that the citric acid generated by Aspergillus niger could be an imporant leaching agent acting in the solubilization of copper, iron, lead and tin from the electronic scrap. Copper could be selectively extracted by 10% LIX84 from the leaching solution and it recoved 99.9% of metallic copper by electrowinning process. Tin and iron were extracted from the remaining solution by 10% Alamine336 and stripped by NaCl solution. Finally, tin could be recovered as a metallic precipitates from the mixed solution of tin and iron by cementation with iron powder.

The study was conducted to investigate the effect of dietary supplementation of Lactobacillus, medicinal plants extract, and essential oil on laying performance, cecal microflora, relative organic weight, leucocyte profiles in laying hens against Salmonella gallinarum. A total of 100 Hy-Line Brown laying hens of 65 weeks of age were assigned into 5 dietary treatments supplemented with no antibiotic (negative control), antibiotic (positive control), Lactobacillus 0.1% (LB), medicinal plants extract 100 ppm (MPE), herb extract 100 ppm (EO). After 7 days of treatment, the 20 hens in each treatment group were divided into challenge group (n=10) and non-challenge group (n=10) for oral administration of Salmonella gallinarums, and were maintained in the same dietary treatments for two additional weeks. Laying hens were selected 20 by each treatment, splitting up into oral Salmonella group and nontreated group. The feeding trial lasted for one weeks and Salmonella gallinarum was orally administered for two weeks. Egg production, egg weight, egg mass, and feed conversion ratio were calculated and cecal microflora, relative organ, and blood were collected at the end of experiment. Compared with the control, laying hens fed Lactobacillus, medical plant extract, and essential oil were not observed to change of laying performance. Salmonella count increased significantly when Salmonella gallinarum was orally administered (P<0.05) but coliform bacteria and Lactobacillus counts were not affected. The relative organs weight was not different in hens with oral administration of Salmonella gallinarum and also in hens fed Lactobacillus, medical plant extract, and essential oil. H/L ratio was not significantly different, but total leucocyte level was somewhat increased in the treatment with oral administration of Salmonella gallinarum and more leucocyte increased in laying hens fed Lactobacillus, medical plant extract, and essential oil than control.
Two experiments were carried out to evaluate the effects of feeding Aspergillus oryzae(AO) ferment on performance, intestinal microflora, serum components, ammonia generation and litter dampness in broiler chicks. In experiment I, three hundred sixty, one day old broiler chicks, Abor Acres, were fed 0 and 0.1% of Aspergiilus oryzae short conidia ferment(AOS) and 0.1% of Aspergillus oryzae long conidia ferment(AOL) for five weeks. In experiment II, three hundred sixty, one day old broiler chicks, Abor Acres were fed 0, 0.1 and 0.2% of Aspergillus oryzae long conidia ferment(AOL) for five weeks. In experiment I, growth rates were not statistically different among dietary treatments. AOS and AOL showed increased tendency in weight gain and feed intake compared to those of control, whereas feed conversion was not different. Litter dampness of AOS and AOL was also tended to decrease compared to that of control, but was not significantly different. Fecal ammonia gas generation was decreased in feeding AOS and AOL, and maintained 1/2 to 3/4 compared to the control. In serum metabolites, AOS and AOL increased glucose and calcium, and decreased total protein, blood urea nitrogen and total cholesterol. In experiment II, body weight of chicks fed 0.1 and 0.2% AOL were heavier than the control(P<0.05). Feed intake of chicks fed 0.1 and 0.2% AOL also were higher than the none, but feed conversion ratio was not different among treatments. Ileal and cecal microflora showed increased tendency in lactic acid bacteria compared to those of the control. Salmonella and E. coli were decreased in ileum of chicks fed 0.1 and 0.2% AOL. In conclusion, feeding AO ferment increased growth performance and improved intestinal microflora of broiler chicks and environments of broiler house.
In order to investigate the effects of supplemental ionic surfactants in in vitro ruminal fermentation, N-Lauroylsarcosine sodium salt(N-LSS) and sodium dodecyl sulfate(SDS) for negative(-) ionic surfactant, and hexadecylpyridinium chloride monohydrate(HPCM) and hexadecyltrimethyl ammonium bromide(HTAB) for positive (+) ionic surfactant were supplemented by 0.05% and 0.1% into the Dehority’s artificial medium containing rice straw(1mm) as a substrate. In vitro DM digestibility, the growth of rumen mixed microbes, pH, cumulative gas production and SEM(Scanning Electron Microscopy) observation of microbial attachment on rice straw particle were investigated through the experiment composing 9 treatments (two supplemental levels of two positive ionic(+) surfactant, two supplemental levels of two negative(-) ionic surfactant) including the control. The sample collection was at 6, 12, 24, 48 and 72 h post fermentation with 3 replications per treatments. DM digestibility in treatments supplemented (+) or (-) surfactants almost stopped afterward 12 h fermentation, in vitro DM digestibility at 72 h post fermentation in the ionic surfactants was at half level of that of the control(P<0.05). Accumulative gas production in in vitro was less(P<0.05) with addition of ionic surfactants compared to the control. The amount of rumen mixed microbes recovered from in vitro incubation fluid pleateaued at 12 h post fermentation for the positive (+) ionic surfactants, but steadily increased as fermentation time elapsed for the control. Rumen microbial growth rate was significantly(P<0.05) low in the negative(-) ionic surfactant compared to the control. pH of the incubation fluid was ranged from 6.02 to 7.20, and was the highest in the negative(-) ionic surfactants, and was the lowest in the control(P<0.05). In SEM observation, rumen microbial population attached on rice straw particle was less with addition of ionic surfactants than the control. In conclusion we could not found any positive effects of negative- and positive- charged surfactants on rumunal fermentation characteristics and rumen microbial growth rates.
This study was performed to investigate the antimicrobial effect of the Fraxinus rhynchophylla extracts against food-borne pathogens. First, the Fraxinus rhynchophylla was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Fraxinus rhynchophylla was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Fraxinus rhynchophylla extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Fraxinus rhynchophylla showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. The synergistic effect has been found in combined extracts of Fraxinus rhynchophylla and Portulaca aleracea as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Fraxinus rhynchophylla against Staphylococcus aureus and Shigella dysenteriae. The ethyl acetate extract of Fraxinus rhynchophylla showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Fraxinus rhynchophylla retarded the growth of S. aureus more than 24 hours and Shigella dysenteriae up to 36 hours. The ethyl acetate extracts of Fraxinus rhynchophylla has been shown the antimicrobial effect against Staphylococcus aureus and Shigella dysenteriae.
This experiment was carried out to investigate the effect of red ginseng extract on the growth of Lactobacillus sp. (L acidophilus, L casei, L salivarius), Escherichia coli and Listeria monocytogenes in pH controled medium by $\beta-Glycerol\;PO_4$ buffer. The growth of Lactobacillus sp. was show a similar pattern in control and MRS broth with red ginseng extract $1.0\%$ but was remarkably show inhibiting in MRS broth with over $2.0\%$ red ginseng extracts. The growth of E coli was inhibited in Trypticase soy broth with $1.0\%$ red ginseng extracts. Also the growth of L monocytogenes was inhibited in Trypticase soy broth with $5.0\%$ red ginseng extract The growth of L acidophilus KCTC3150, L casei KCTC3189, L salivarius ssp. salivarius CNU27, and E coli KCTC1039, L monocytogenes KCTC3443 were remarkably inhibited in pH non-control medium and pH control medium with $10\%$ red ginseng extract These results was suggested to effect of inhibition of microorganisms growth not pH decrease by organic acid but another components in red ginseng extract.

Indigenous bacteria isolated from contaminated sites play important roles to remediate contaminated groundwater. Chromium has the most stable oxidation states. Cr(VI) is toxic, carcinogenic, and mobile, but Cr(III) is less toxic and immobile. In this study, indigenous microorganism (MMPH-0) was enriched from Cr(VI) contaminated groundwater, and identified by 16S rRNA gene analysis. Using MMPH-0, the effect of stimulating with e-donors (glucose, lactate, acetate, and no e-donor control), respiration conditions, biomass, tolerance, and geochemical changes on Cr(VI) reduction were investigated in batch experiments for 4 weeks. The changes of Cr(VI) concentration and geochemical conditions were monitored using UV-vis-spectrophotometer and Eh-pH meter. And the morphological and chemical characteristics of MMPH-0 and precipitates in the effluents were characterized by TEM-EDS and SEM-EDS analyses. MMPH-0 (Enterobacter aerogenes) was able to tolerate up to 2000 mg/L Cr(VI) and reduce Cr(VI) under aerobic and anaerobic conditions. MMPH-0 performed faster and higher efficiency of Cr(VI) reduction with electron donors (over 70% after 1 week with e-donor, 10-20% after 4 weeks without e-donor). The changes of Eh-pH in effluents showing the tendency from oxidizing to reducing condition and a bit of acidic change in pH due to microbial oxidation of organic matters donating electrons and protons suggested the roles of MMPH-0 on Cr(VI) in the contaminated water catalyzing to transit geochemical stable zone for more stable $Cr(OH)_3$ or Cr(III) precipitates. TEM/SEM-EDS analyses of MMPH-0 and precipitates indicate direct and indirect Cr(VI) reduction: extracellular polymers capturing Cr component outside cells. These results suggested diverse indigenous bacteria and their biogeochemical reactions might enhance more effective and feasible remediation technology of redox sensitive heavy metals in metal-contaminated in groundwater.
Aquifer recharge and recovery is a technology used to ensure a stable supply of clean water. During the process, river water is injected into a soil aquifer and stored. The stored water is then recovered and used to produce drinking water. It is important to understand quality improvement of the injected water while it is stored in the aquifer. In the present study, a lab-scale column reactor containing saturated-zone soil was employed to mimic an aquifer. The reactor was used to investigate microbial removal of nitrate that is a major inorganic contaminant detected in the Nakdong River. The reactor was introduced with river water that contained nitrate at concentrations (5.07, 6.81, 8.27, and 11.07 mg $NO_3{^-}/l$) detected downstream of the Nakdong River in the past 2 years. The nitrate concentrations decreased during the introduced water is retained in the reactor. Effluent from the reactor contained 1.49 mg $NO_3{^-}/l$ or less and had an average pH of 7.98 regardless of the nitrate concentrations of the influent. However abiotic control reactor showed similar nitrate-concentrations in its influent and effluent. Considering the result of abiotic control, the decreased nitrate concentration observed in the test column suggested that microorganisms in saturated-zone soil removed nitrate in the river water introduced into the reactor. Results of this study will be used to better understand microbial improvement of water quality in aquifer recharge and recovery technology.

In order to help the preservation of the unshiu orange and pear, antimicrobial paper incorporating grapefruit seed extract and zeolite was applied to pack fruits. Unshiu orange was packed in a box (24${\times}$24${\times}$22 cm) attached with antimicrobial paper and then stored respectively at l$0^{\circ}C$. Pears were wrapped individually before storage at l$0^{\circ}C$. During the storage, weight loss, pH, total acidity, soluble solid content, microbial load and decay were measured as quality indices. Steady pH increase in unshiu orange was observed to slightly decrease total acidity during the storage with little difference between the packaging treatments. The microbial loads of total aerobic bacteria, and yeast/mold counts were suppressed during storage by the antimicrobial paper packaging, which also contributed to reducing the decayed unshiu orange. Limited reduction of total aerobic bacteria and yeast/mold counts was observed only for initial storage period for the pears wrapped with 9 and 12% antimicrobial agent-added papers. Antimicrobial paper was useful for the reduction of microbial load in unshiu orange and pear without other quality deterioration.
