A Study on the Possibility of Produced by Supercritical Fluid Extraction from Lycii Fructus Seed for Cosmetic Ingredients (구기자 종자 초임계유체 추출물의 화장품소재로서의 가능성 평가)

  • The purpose of this study was to investigate the possibility of applying cosmetic material about extracted from Lycii Fructus seed supercritical fluid. This study was conducted to evaluate the anti-melanogenesis effect and antimicrobial activity about the extracts from Lycii Fructus seed. These researches studied for emulsion physical stability of pH, viscosity, particle size from emulsion containing Lycii Fructus seed extract. As a result, the supercritical fluid extract from Lycii Fructus seed significantly inhibited melanin synthesis by 81.86 % at the concentration $750{\mu}g/mL$. Antimicrobial effects of extract was determined against Staphylococcus aureus, Candida albicans, Escherichia coli. except by Staphylococcus epidermidis. The physical stability of viscosity and pH on the emulsion containing Lycii Fructus seed extract were stable for 28 days. Emulsion containing Lycii Fructus seed extract did not change particles at observation into optical microscope. These results suggest that extracts from Lycii Fructus seed may have value as the potential cosmetic formulation.

A convergence study of cytotoxicity evaluation of adult dentifrices (성인용 치약의 세포독성 평가의 융합적 연구)

  • This study was conducted to effect the cell activity and cytotoxicity of dentifrice. For the study, 6 kinds of general dentifrice, 3 kind of whitening dentifrice, 2 kinds of natural dentifrice and SLS(sodium lauryl sulfate) of positive control group. Immortalized human gingiva fibroblast cell was used for the study, WST test for cell activity and Agar diffusion test for cytotoxicity. Agar diffusion test showed high cytotoxicity in general dentifrice test group and whitening dentifrice test group, but low cytotoxicity in natural dentifrice test group. As a result of cell nucleus staining, cell shape and nuclear activity showed that the highest activity in natural dentifrice group, followed by whitening dentifrice group and general dentifrice group. As a result of this study, the cytotoxicity of different ingredient and according to the use to dentifrice. As a result of this study, we confirm cytotoxicity of kind and components according to the purpose of using dentifrice. Therefore, it is necessary to indicate the detailed ingredients of dentifrice for the smart choice of consumers.

Antioxidative Activities and Whitening Effects of Ethyl Acetate Fractions from The Immature Seeds of Abeliophyllum distichum (미선나무 미성숙 종자의 항산화 및 미백 활성)

  • Abeliophyllum distichum Nakai is deciduous shrubs of flowering plant in Oleaceae. It is important plant resources and consists of one species in the world. Also the endemic plant of A. distichum has been protected and designed endangered plant in Korea. For this reason, study on the immature seeds of A. distichum (ADS) hasn't progressed. In the present study, we evaluated the antioxidant activity and inhibitory effects on proteins and mRNA levels were related in the whitening effect in B16F10 cells. ADS was effective for reaction oxygen species (ROS). ROS causes various diseases such as aging, inflammation, cancer, and etc. Antioxidant properties were evaluated DPPH, ABTS radical scavenging activity and Reducing power. Plants were known that contained phenolic compounds were related in antioxidant activity. Phenolic compounds were phytochemicals commonly named natural polyphenols. These are secondary metabolites of plants involved in the defense against different types of stresses. In results, ADS suppressed the expression and transcription of Tyrosinase, TRP-1, TRP-2, and Microphthalmia-associated transcription factor (MITF). Tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 1 (TRP-2) are known to play an important role in melanin biosynthesis. MITF regulated the expression and transcription of Tyrosinase, TRP-1, and TRP-2. In conclusion, ADS was effective in both antioxidant activities and whitening effects. Also, they were associated with the content of phenolic compounds. We suggested that ADS can be use antioxidants and skin-whitening functional cosmetics material derived from natural plant resources.

Effect of Medicinal Herb Prepared through Traditional Antidiabetic Prescription on α-Glucosidase Activity and Evaluation Method for Anti-Melanogenesis Agents Using α-Glucosidase Activity (당뇨 처방에 근거한 생약재의 α-Glucosidase 활성 저해 효과 및 이를 활용한 미백 소재 평가법)

  • For the purpose of investigating the in vitro antidiabetic activity of a medicinal herb and herb mixture extracts prepared through traditional antidiabetic prescription, this study examined ${\alpha}$-glucosidase inhibitory activity. Tyrosinase, a type I membrane glycoprotein, is synthesized and glycosylated in the endoplasmic reticulum (ER) and Golgi. The enzyme is subsequently transported to melanosomes, where it participates in melanogenesis. Previous studies showed that disruption of early ER N-glycan processing by an ${\alpha}$-glucosidase inhibitor suppresses tyrosinase enzymatic activity and melanogenesis. According to the results, most oriental medicinal herbal extracts were stronger than acarbose and N-butyldeoxynojirimycin, known as an ${\alpha}$-glucosidase inhibitor. Interestingly, ethyl acetate layer of enzyme hydrolyzed Cheongsimyeonjaeum had an inhibitory effect on melanin synthesis in B16F1 cells, although it did not inhibit tyrosinase activity directly. Together, ${\alpha}$-glucosidase inhibition activity could be used to evaluate anti-melanogenesis, although cross-checking with melanin inhibitory assay is recommended.

A Study on Antioxidant, Skin-Whitening and UV Absorption Effect of Beta vulagaris Using Lactiplantibacillus plantarum SM4 (Lactiplantibacillus plantarum SM4 를 이용한 레드비트 생물전환물의 항산화, 피부 미백과 자외선 흡수 효능 연구)

  • The purpose of this study was to confirm the skin whitening and UV absorption effects of bioconverted Beta vulagaris (BBE) using Lactiplantibacillus plantarum SM4, and the effects were evaluated by measuring total polyphenol content (TPC), total flavonoid content (TFC), radical elimination activity (RSA), and tyrosinase activity inhibition (TAI). TPC and TFC of BBE were 25.0 mg GAE/g DM and 8.05 mg QE/g DM, which were 1.3 and 1.1 times higher than hot-water extract (HWE) respectively. RSA, an indicator of antioxidant activity, and TAI, an indicator of skin-whitening effect, were 36.8%, and 68.6%, respectively, 1.1 and 1.2 times higher than that of HWE. UVA and UVB absorption of BBE were 21.4 and 87.6%, which was 1.4 and 1.7 times higher than that of HWE respectively. When main substances of BBE were analyzed using LC-MS/MS, kaempferol and isorhamnetin, a type of polyphenols known to have high antioxidant and skin-whitening effects, were identified. As a results, BBE is expected to be used as a functional cosmetic material as it has excellent antioxidant, skin whitening, and UV absorption properties.

Comparison of Melanogenesis-Inhibiting Activity by Extracts of Prunus persica Flower and Calyx (도화 및 도화악 추출물의 미백활성 비교)

  • The antioxidant activity and whitening effect of the distilled water (DW) and ethanol extracts of the Prunus persica flower and calyx were studied. In the oxygen radical absorption capacity (ORAC) assay for antioxidant activity measurement, it was confirmed that the flower extract was stronger than the calyx extract, and that the ethanol extract was relatively stronger than the DW extract. To define the whitening effect, an experiment was conducted involving tyrosinase inhibitory assay and measurement of the melanin content of B16F10. As a result of the use of tyrosinase, the DW extract of calyx showed 53% inhibition as the highest activity. The melanin content inhibitory rates were defined as 57% for the ethanol flower extract and 63% of the ethanol calyx extract, based on a $10{\mu}g/mL$ concentration. Based on these results, mixture with the whitening effect in the extract of P. persica and another compounds should be researched for development as a cosmetic ingredient.

Whitening and Antioxidant Activities of Solvent Extracts from Hot-Air Dried Allium hookeri (열풍건조 삼채의 분획별 미백 및 항산화 활성에 관한 연구)

  • The study analyzed the antioxidant and whitening activities of fractions from hot-air-dried Allium hookeri (HADAH). The total polyphenol contents in fractions from HADAH extract showed the highest value ($171.07{\pm}7.55mg/g$ GAE) in ethyl acetate fraction of roots. Antioxidant activities of leaf and root parts were significantly higher in the ethyl acetate fraction. HADAH showed higher whitening activity (n-hexane fraction of leaf; 52.22% melanin production) than arbutin (65.82% melanin production), which is known as a whitening ingredient. These results suggest that HADAH extracts can be used as antioxidant and whitening materials. Thus, study of HADAH extracts showing effective whitening and physiological activities can provide efficient data for industrial use of Allium hookeri.

Antioxidant and Whitening Activities of Various Cultivars of Korean Unripe Peaches (Prunus persica L. Batsch) (국내산 복숭아 유과의 품종별 항산화 활성 및 미백활성 평가)

  • This study was carried out to investigate the biological activities, including the total polyphenolic contents, antioxidant activities, and tyrosinase inhibition activities, of various cultivars of unripe peaches (Prunus persica L. Batsch). The results indicate that the total polyphenolic content (gallic acid equivalent mg/g) was the highest in Yumefuji among six-cultivars (Takinosawa Gold, Kawanakawase Hakuto, Madoka, Yumefuji, Nagasawa Hakuho, and Hong Bak) of unripe peaches, whereas it was the lowest in Madoka. Antioxidant effects of unripe peaches were determined in three different in vitro bioassays measuring DPPH radical scavenging, ABTS radical scavenging, and reducing power activity. In the results, antioxidant activity was also higher in Yumefuji than other cultivars. Tyrosinase inhibition activity ranged from 4.65% to 8.58%, and Hong Bak displayed the highest tyrosinase inhibition activity, although the difference was not significant. This study will provide information on antioxidant and tyrosinase inhibition activities for the development of natural compounds as functional cosmetics.

Inhibitory Effect of Angelica keiskei Extracts on Melanogenesis (신선초 추출물의 멜라닌 생성 억제활성)

  • Angelica keiskei is a perennial herb belonging to the Umbelliferae family. In this study, the whitening effect of A. keiskei extracts was examined through melanogenesis and tyrosinase inhibitory assays. The ethanol extract (50%) significantly inhibited tyrosinase in a concentration-dependent manner. RT-PCR revealed that the extract exhibited decreased expression of tyrosinase, tyrosinase-related protein-1, tyrosinase-related protein-2, and melanocyte-inducing transcription factor. These results suggest that the extract can be used as an ingredient for the development of cosmeceuticals.

Anti-Oxidant, Anti-Melanogenic, and Anti-Inflammatory Activities of Zanthoxylum schinifolium Extract and its Solvent Fractions (산초 추출물 및 분획물의 항산화, 미백 및 항염증 활성)

  • This study was designed to explore new nutraceutical and cosmetic resources possessing biological activities from the plant kingdom. To fulfill this purpose, we analyzed the anti-oxidative, anti-melanogenic, and anti-inflammatory activities of Zanthoxylum schinifolium extract (ZSE) and its solvent fractions using in vitro assays and cell culture model systems. Three kinds of ZSE treated with methanol, ethanol, and water exhibited potent anti-oxidative activities through DPPH radical scavenging capacity, and inhibited in vitro DOPA oxidation. Furthermore, Z. schinifolium methanol extract (ZSME) inhibited the ${\alpha}$-melanocyte stimulating hormone, which induces melanin contents in B16F10 cells. Its anti-melanogenic activity originates from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Moreover, lipopolysaccharide induced nitric oxide production in the RAW 264.7 cell line was also ameliorated by ZSME treatment in a dose dependent manner. Among the four solvent fractions of ZSME treated with dichloromethane, ethyl acetate, n-butanol, and water, three fractions, except water, showed significant anti-melanogenic and anti-inflammatory activities. Taken together, these results provide important new insights into Z. schinifolium, indicating that it possesses numerous biological activities such as anti-oxidative, anti-melanogenic, and anti-inflammatory activities. Therefore, it may well serve as a promising material in the field of nutraceuticals and cosmetics.
