Effects of Dietary Soybean Meal in Rainbow Trout (Oncorhynchus mykiss) III. Bioavailability of Phosphors (무지개송어(Oncorhynchus mykiss) 사료원으로써 대두박의 이용 효과 III. 인(P)의 이용성)

  • Feeding experiment was conducted to evaluate effects of dietary soybean meal (SBM) on the phosphorus bioavailability of rainbow trout (Oncorhynchus mykiss). Seven diets, containing 0~70% SBM, were made based on iso-nitrogenous and iso-caloric basis of 44% crude protein. Ajpparent digestibility of phosphorus was measured 60 and 120 days after feeding. The digestibility coefficient increased gradually as the level of SBM increased up to 46~58%, but decreased at 58~70% SBM level. Phosphorus concentrations in feces decreased with an increased in dietary SBM level. Patterns of post-prandial phosphorus excretion were not similar to those of ammonia excretion, but the amount decreased with the increased SBM level in the diets. Total excretion of phosphorus for 24 hours was 19.68mg.kg-1 body wt..day-1 in the control group, but excreted phosphorus by the 58% and 70% SBM groups was 63.4% and 56.1% of the control group, respectively. Phosphorus concentration of whole body decreased with an increase in dietary SBM level for 120 days of feeding. However, there were no significant differences in phosphorus concentrations of liver among the experimental groups, except for 70% SBM diet group.

Optimum Condition for Dyeing Cut Rose 'Denice' by Absorption Method (흡습법에 의한 절화장미 'Denice'의 염색 최적 조건 구명)

  • This research was carried out to produce rainbow-colored roses from white cut rose, Rosa hybrida 'Denice', to meet customers' desire of having various colors of roses. The higher temperature of dyeing solution used, the faster coloring time surveyed. In high temperature, the flowering speed of cut flower accelerated and after treating more than 4 hours, the edges of flowers became too dark or dry. In the condition of concentration of dyeing solution, $11g{\cdot}L^{-1}$ dyed faster than $7.5g{\cdot}L^{-1}$, however, no differences between $15g{\cdot}L^{-1}$ and $11g{\cdot}L^{-1}$ have observed. It looked fast coloration at the stage of early blooming, but all three different petal opening stages need similar time to get even coloration from outer to inner petals. For the consideration of commercial value, flowers with 3-4 petals opened are better quality than flowers with less petals opened. It was possible to make rainbow-colored rose by dipping 'Denice' with 3 primary color combination, Hot Pink, True Blue, and Yellow dyes, at the best result conditions of $20^{\circ}C$ solution temperature, 3-4 petals opening stage, 11g/L concentration of dye solution and 3 hours dipping, respectively.

Effects of Activin on Testosterone-primed Immature Rainbow Trout Gonadotropin Release in vitro (Testosterone 처리한 미성숙 무지개송어 뇌하수체의 세포배양계에서 생식소자극초르몬 분비에 대한 Activin의 효과)

  • The present studies were conducted to evaluate the effects of activin-A on gonadotropins (GTHs) release in testosterone-treated immature rainbow trout Oncorhpynchus mykiss. The administration of testosterone elevated pituitary level of GTH II but not of GTH I. In this study using primary cultures of dispersed pituitary cells in static incubation, dose-dependent increases in GTH II release was observed in the activin-treated group at day 3 of incubation (long-term incubation), but not at day 1 of incubation (short-term incubation). Dopamine, a potent inhibitor of gonadotropin-releasing hormone (GnRH)-stimulated GTH II release in rainbow trout, was only partially effective in decreasing actvin-induced GTH II release. Furthermore, salmon GnRH (sGnRH)-stimulated GTH II release was not potentiated by the pretreatment with activin. However, the control mechanisms of GTH I release by activin and other hormones were not observed in the all tested experiments. The results of these studies support the contention that in contrast with the usual stimulatory effects of activin on GTH release in mammals, activin exerts long-term stimulatory actions on GTH II release in rainbow trout. The control mechanism of GTH I release, however, is a question that remains to be elucidated.

Detection of Carbonic Anhydrase in the Gills of Rainbow Trout (Oncorhynchus mykiss) (무지개 송어 rainbow trout, Oncorhynchus mykiss의 아가미에서의 carbonic anhydrase의 존재)

  • Carbonic anhydrase isozymes are a widespread, zinc-containing metalloenzyme family. The enzyme catalyzes the reversible inter-conversion of $CO_2$ and $HCO_3$. This reaction is the main role played by CA enzymes in physiological conditions. This enzyme has been found in virtually all organisms, and at least 16 isozymes have been isolated in mammals. Unlike mammals, there is little information available regarding CA isozymes in the tissues of non-mammalian groups, such as fish. Carbonic anhydrase is very important in the osmotic and acid-base regulation in fish. It is well-known that the gills of fish play the most important role in acid-base relevant ion transfer, the transfer of $H^+$ and/or $HCO_3^-$, for the maintenance of systemic pH. Rainbow trout, Oncorhynchus mykiss, is the most important freshwater fish species in the aquaculture industry of Korea, with annual production increasing each year. In addition, environmental toxicology research has shown that rainbow trout is known to be the species that is most susceptible to environmental toxins. Consequently, carbonic anhydrase was detected in rainbow trout, Oncorhynchus mykiss. The isolated protein showed the specific band with a molecular weight of 30 kDa and pI of 7.0, and it was identified as being carbonic anhydrase. The immunohistochemical result demonstrated that the carbonic anhydrase was located in the epithelial cells of the gills.

Monitoring of the mortalities in the aquaculture farms of South Korea (한국 주요 양식종의 수산동물 폐사피해 모니터링)

  • The practical monitoring was performed to survey the mortalities in aquaculture farms of olive flounder, rockfish, rainbow trout, Japanese eel, white shrimp and abalone in South Korea from May to November, 2011. The aquatic organism disease inspectors, who have the national licenses for the diagnosis and prevention of aquatic disease and have close relationship with the farms, investigated the rates and causes of mortalities according to the standard manual. In 70 flounder farms, the cumulative mortalities rate was 27.9%, and the mortalities were caused by scuticociliatosis, streptococcosis, VHS, non-infectious loss, vibriosis and gliding bacterial disease. The moralities rate of 30 rockfish farms was 13.6%, and those were mainly contributed by gill flukes and streptococcosis. Most of mortalities of rainbow trout were caused by non-infectious loss and protozoan white spot disease. The mortalities rate of Japanese eel was 0.6% by edwardsiellosis, protozoan white spot disease and gill flukes. The loss rate of white shrimp was 71.2%, and most of them was related with non-infectious ones, such as carnivalization, transportation loss, and the rest was caused by viral white spot disease. The mortalities rate in the abalone farms was 10.7% and all of them were related with non-infectious loss.

Analysis of Color Characteristics of Marine Oil Spills Using PlanetScope Images (PlanetScope 영상을 이용한 해양 유출유의 색상 특성 분석)

  • In this letter, we used PlanetScope imagery to conduct experiments on the color characteristics for oil type classification of marine oil spills through Red-Green-Blue (RGB) histogram analysis. The histograms of marine oil spills can be divided into three categories (dark black tones, light silver tones, and light rainbow tones) according to the distribution of pixel values in each band. Thick oil layers with dark black tones can be classified as heavy oil, while thin oil layers with light silver and rainbow tones can be classified as light oil. As more images are analyzed in the future, these oil spill detection and classification methods will become more generalized and reliable.

산천어의 생식선자극호르몬 유전자발현에 미치는 에스트로젠의 영향과 에스트로젠수용체 cDNA cloning

  • 어류의 번식활동은 뇌-뇌하수체-생식소로 이어지는 내분비기관이 주로 담당하고 있으며, 그 중에서 뇌하수체에서 만들어지는 생식선자극호르몬 (GTH)은 생식소 발달에 매우 중요한 역할을 한다. 한편, GTH는 생식소에서 생성되는 스테로이드호르몬에 의해 유전자의 발현이 조절된다. 본 연구에서는 산천어 (Oncorhynchus masou)의 GTH 유전자발현에 미치는 estradiol-17$\beta$ (E2)의 영향을 생체내에서 먼저 조사하고, 유전자발현조절의 메카니즘을 구명하기 위하여 에스트로젠 수용체 (ER)의 cDNA를 cloning하였다. 또한 내분비교란물질로 알려져 있으며 에스트로젠 수용체와의 유사결합작용이 알려진 노닐페놀 (NP)의 영향도 아울러 조사하였다. 미성숙 산천어를 E2 및 NP (각각 5 mg/kg 및 10 mg/kg 체중)로 처리하면 72 시간째에 뇌하수체내의 $\beta$-actin mRNA에 비해 GTH (FSH$\beta$, LH$\beta$) mRNA가 상대적으로 증가하는 경향을 나타내었다. 뇌하수체에서 분리한 ER의 기본적인 구조는 DNA결합도메인 (Zf-C4), 호르몬결합도메인 (hormone_rec), 기타 전사활성화도메인으로 구성되어 있었으며 (그림 1), 기존에 보고된 에스트로젠 수용체중에서 무지개송어 및 대서양연어의 ER과 각각 98%, 96%의 높은 상동성이 관찰되었다. 향후, 준비된 ER을 재료로 하여 GTH 유전자발현의 호르몬의존성을 조사할 예정이다.

  • Infectious pancreatic necrosis(IPN) virus was known as a causative agent of newly recognized viral disease of young rainbow trout characterized by highly contagious, high mortality and necrosis of pancreas. Several strains of IPN viruses were recovered from young rainbow trout that have been shown a typical cinical sign of infectious pancreatic necrosis disease. The field isolate produced cytopathic effect, and multiplied up to $10^{6.0}$ to $10^{6.5}$ $TCID_{50}/0.1ml$ in BT cell culture. In the indirect immunofluorescent assay with trout anti-IPN virus IgG and goat anti-trout IgG FITC conjugate, these isolates were proved to be a IPN virus that were closely related with VR277 strain of IPN virus antigenically.

  • Effects of muscle fluid concentrates from Styela clava on fish diets were investigated in rainbow trout(Oncorhynchus mykiss). Fish were fed on one of the isonitrogenous(48%) and isolipic(20%) feed containing 5 to 20% of muscle fluid concentrates for 8 weeks. Hematological parameters such as hemoglobin, hematocrit, albumin, glucose, total bililubin, triglyceride and glutamic pyruvate transaminase of the rainbow trout which were fed on the diets varied, but no specific trend became apparent. However, glutamic oxaloacetic transaminase value was significantly higher than that of normal fishes. Therefore, concentrated ascidian fluid which is normally discarded, can be combined with commercial diets for the normal growth of rainbow trout.

Composition in Carotenoids of Rainbow Trout Fed with CLA and Ascidian Tunic Extracts (CLA와 멍게 껍질 추출물 급이에 따른 무지개송어 색소조성)

  • Astaxanthin is a valuable pigment source for many aquacultured species, including salmonoids, shrimp, sea bream, and ornamental species. Conjugated linoleic acid (CLA) and ascidian tunic extracts were mixed with the basal diet of rainbow trout to investigate their pigmentation effects. Synthetic Carophyll Pink and natural carotenoids that came from the tunic extracts were incorporated into muscle and skin tissues. The main carotenoids found in muscle after 8 weeks were canthaxanthin in CP12 (13.4%), and CP52 (17.2%), and astaxanthin in CP12 (58.5%), and CP52 (59.2%) in the Carophyll Pink group, while those in skin were canthaxanthin in CP14 (34.5%), and CP54 (29.2%), and astaxanthin in CP14 (32.0%), and CP54 (36.5%) in the ascidian tunic extract group. The total carotenoid content in skin (53.0-69.3 mg/kg) was greater than that in muscle (9.5-13.8 mg/kg).
