Transfer Rate of Cross Contamination of Listeria monocytogenes between Pork Meat and Workers' Hands during Pork Meat Processing (포장돈육 가공공정 중 돈육과 작업자 손과의 Listeria monocytogenes의 교차오염 전이율)

  • This study was performed to determine the transfer rates of each foodborne pathogen from pork meat packaging during the processing. We analyzed the transfer rate of Listeria monocytogenes from contaminated pork meat to worker's hands (wearing polyethylene gloves, PEG; cotton gloves, CG; and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 100.00% was higher than that of bare hands 2.513% and PEG 1.511%. In particular, when wearing Co, the transfer rate from the CG to bare hands with CG was 0.08%. Also, the range of transfer rates from contaminated pork meat to cutting board and knife was 0.352-3.791%. In contrast, transfer rates from the workers' hands (with PEG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.001 to 0.141%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that use of PEG could effectively reduce or prevent the cross-contamination compared to CG and provide important information concerning the consecutive transfer of L. monocytogenes during food processing.

  • This study describes cross-contamination events that occur in animal food processing. We analyzed the number of Salmonella spp. contamination transferred from pork meat to workers' hands (wearing polyethylene gloves; PG, cotton gloves; CG, and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 38.80% was higher than that of PG 3.11% and bare hands 1.35%. In particular, when wearing CG, the transfer rate from the CG to bare hands with CG was 0.07%. Also, the range of transfer rates from the contaminated pork meat to cutting board and knife was 0.20-1.99%. In contrast, the transfer rates from the worker's hands (with PG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.0015-0.21%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that the use of PG compared with CG could effectively reduce or prevent the cross-contamination and provide important information concerning the consecutive transfer of Salmonella spp. during food processing.

  • In order to fine some proper bacterial food for Tigriopus japoncus, bacterial flora of the tide pool inhabited by the copepod has been isolated and tested as bacterial food. Food effect and optimum density of the bacteria in terms of survival rate of the copepod was measured in the larval and the adult stages. Among the 264 strains of isolated bacteria, Acinetobacter spp. Moraxella, spp., Flavobacterium spp. and Pseudomonas spp. were certified as effective food for the copepod. According to the experimental results, Acinetobacter spp. AG-3 was the most effective food for all the stages from nauplius to adult, while Moraxella spp. and Flavobacterium spp. were effective for copepodite stage, and Pseudomonas spp. for the adult stage only. The optimum density of bacteria for the food was about $10^6\;cell/ml$, which was the same average density of bacteria in the tide pool.

A Research on the Use and Architectural Changes of Sungnyemun in King Yeongjo's Reign (영조 대 숭례문 문루의 하층 사용과 건축 변화에 대한 연구)

  • This research work is to analyze architectural characteristics of Sungnyemun especially in King Yeongjo's reign in Joseon dynasty. The result of this research is summarized as following: 1. The architectural characteristics of Sungnyemun in King Yeongjo's reign are closely related with Confucian ceremonies such as Jeon-jwa and Heon-goek-rye. To perform these ceremonies, some lower walls of Sungnyemun's wooden pavilion were removed and used as ceremonial space. And after ceremony it was restored. 2. The floor type of center bay of the 1st story of wooden pavilion should have a type of floor using long and narrow fine tree plate, which is same type before the repair work of 1960's dismantlement. 3. The width of east stairway which is reached to east small gate, was changed just before Japanese's rule(1910~1945), should be broaden than present width, which is proven through the recent excavation. 4. The reason of asymmetric characteristic of locations of both east and west narrow-gate, and widths of east and west stairway, are related with order of King's ceremony. * Jeonjwa : a ceremony to see national affairs or receive royalty from officials in main hall or main gate of palace in Joseon dynasty (some times open to public) * Heon-goek-rye : a ceremony after win a war and offering to king enemy's ear or head in Joseon dynasty.

A Study on the Classified Jang(Fermented Soybean) in Goryeo and Chosun Dynasty Period (고려시대 및 조선시대 장류)

  • On the basis of the cookbooks and Data Base of the Korean Classics(http://db.itkc.or.kr/itkcdb/mainIndexIframe.jsp), this paper analyzed the fermented soybean listed in the general documents of the Chosun Dynasty(1392~1897) and the Goryeo Dynasty(918~1392). In the Goryeo Dynasty, there are 15 kinds of Jang(soybean paste or solution), among which are Jang (soybean paste fermented by mold)(6 documents), Yeomgjang, Yeomshi(2), and Gaejang(1). However, the cookbook at that time is defunct. The Goryeo Court relieved the famine-stricken people by proving them with Jang. In the Chosun Dynasty, 111 kinds of Jang were listed in the general documents, and 153 kinds in cookbooks. There were 55 kinds of general Jang, such as Jang(204), Yeomjang(63), Chojang, Goojang(7), and Gaejang(6), are listed in the general documents, and in the cookbooks, there are 55 kinds of Jang, such as Sookwhangjang(9 cookbooks), Daemaekjang(8), Myeonjang(8), Saengwhangjang (8), and Yooinjang(8), and among them, 13 kinds belong to the Chinese origin. A total of 9 Kinds of Ganjang(soybean solution fermented by mold), such as Soojang(30), Cheongjang(23), Gamjang(8), and Ganjang(3) are found in the general documents. In the cookbooks, 12 kinds of Jang, as Cheongjang(10), Cheonrijang(4), Ganjang(3), and etc., are listed. There were 9 kinds of Gochoojang(red pepper-soybean paste), such as Chojang(12), Gochojang(3), and etc., are listed in the general documents, and 9 kinds as Gochojang(7), Manchojang(7), rapid Manchojang(4), and etc., are in the cookbooks. In addition, 16 Kinds of Yookjang(fermented soybean-meat paste) as Haejang(15), Hyejang(11), Yookjang(11), and etc., are found in the documents, and 22 kinds as Nanjang(9), Gejang(6), Yookjang(5), Shoigogijang(4), and etc., are in the cookbooks. Eighteen Kinds of Shi(soybean paste fermented by bacteria) as Yeomshi(40), Shi(35), Shijang(6), and etc., are recorded in the documents, and 19 kinds as Jeonkookjang(6), Shi(4), Sooshijang(4), and etc., are in the cookbooks, and among them 11 kinds belong to the Chinese origin. Six kinds of Jipjang(aqueous soybean paste) as Jipjang(7), Uoopjang(4), Pojang (2), Jangzoop(2) are recorded in the documents, and 15 kinds as Jipjang(9), Zoopjeo(7), and Hajeoljipjang(5) are in the cookbooks. Soybean paste, or solution for relieving hunger is not recorded in the documents. However, the Chosun court, for the purpose of relieving famine-stricken people, used general Jang. Such 21 Jang to relieve the famine-stricken people as Pojang(7), rapid Jang(6), and Sasamgilgyeongjang(4) are listed in the cookbook. Geonjang(dried soybean paste), Nanjang (egg-soybean paste), Doojang(soybean paste), Maljang(random soybean paste), Myeonjang(wheate-soybean paste), Sodoojang (red bean-soybean paste), Yookjang(soybean-meat paste) and Jang(soybean paste) are recorded in the documents, as well as in the cookbooks. Chinese-original Jang and Shi are recorded in the cookbooks, with no list in the general documents. Therefore, it seems that it didn't pass down to the general public.

Optimum Reinforcement Conditions of Large Diameter Reinforcement for Steep Slope of Conventional Railway Embankment under Train Loading (기존선 성토사면 급구배화를 위한 열차 하중 하 대구경 봉상보강재의 최적 보강조건)

  • A reinforcement is required to ensure the structural safety in case of railway embankment excavation under railway load. A large diameter soil nailing with concrete wall is applied as the reinforcement method instead of the conventional soil nailing system. In this study, a series of 3 dimensional numerical analyses are performed to investigate the optimum reinforcement considering 15 different conditions based on the length, lateral spacing, diameter, and inclination of the reinforcement. The interface between soil nail and perimetric grout is considered by means of cohesion, stiffness and perimeter of the grout. 0.3 m of reinforcement diameter is assessed as the most appropriate based on the economical viewpoint though ground displacement decreases with the increase of diameter, however the difference of displacement is negligible between 0.4 m and 0.3 m of diameter. Surface settlement, lateral displacement of wall, and stress of reinforcement are calculated and economic viewpoint to reinforce embankment considered. Consequently, the optimum reinforcement conditions considering those factors are evaluated as 3 m in length, 0.3 m in diameter, 1.5 m in lateral spacing, and 10 degree of inclination angle in the case of 3 m of excavation depth. Additionally, inclined potential failure surface occurs with approximately 60 degrees from the end of nails and the surface settlement and wall lateral displacement are restrained successfully by the large diameter soil nailing, based on the result of shear strain rate.

  • 현재 널리 상용되어 있는PDP는 3전극 변방전형이다. 3전극 면장전형 PDP는 주방전이 유전체 아래에 서로 평행하게 위치하고 있는 ITO투명전극 사이에서 발생한다. 따라서 방전시의 전기장은 MgO 보호막 위에서 아치형태로 형성되게 된다. 플라스마 방전 시 전자에 의해 이온화된 이온 입자들은 전기장에 의해 그 방전경로가 정해지게 된다. 물론 전기장은 표면에서 수직이지만 전기장에 의해 가속되어진 이온입자들은 MgO 보호막에 기울어져서 입사하게 된다. 따라서 플라스마 방전시의 이온들의 MgO 보호막으로의 입사각은 매우 다양하다. $\gamma$-FIB (Focused ion beam) 시스템은 이온입사에 의한 물질의 이차전자방출계수 측정에 효과적인 장비이다. 본 실험은 이러한 $\gamma$-FIB 시스템을 이용하여 다양한 각도로 입사하는 이온빔에 의한 MgO 보호막의 이차전자방출계수를 측정하였다. 또한 이온화 에너지가 다른 여러 종류의 불활성 기체를 사용하여 이온의 입사하는 각도에 따른 MgO 보호막의 일함수를 측정하였다. 이온빔의 입사각은 각각 $0^{\circ}$, $10^{\circ}$, $20^{\circ}$, $30^{\circ}$로 변화시키면서 이차전자방출계수 및 일함수를 측정하였다. 이러한 실험을 통해 입사각이 클수록 이차전자방출계수는 증가하고 일수는 감소하는 것을 확인 할 수 있었다.

Evaluation of Biological Critical Control Points Using Escherichia coli Genotyping (Escherichia coli Genotype을 이용한 생물학적 Critical Control Point의 적합성 평가)

  • This study was performed to evaluate the effectiveness of biological critical control points using the genetic profile of Escherichia coli isolates from pork cutting plants. Samples were collected from carcasses, equipment (knife, table, glove, transport belt, boning and skinning machine), the environment (wall and floor), and meat cuts during the cutting process from two plants. Pulsed-field gel electrophoresis (PFGE) was used to characterize the E. coli isolates. An identical genotype was detected from the carcasses, equipment, environment, and final meat cuts, and showed that the incoming carcasses, which were contaminated during transportation from slaughterhouses, were a major source of E. coli that was spread throughout processing. Also, consistent cross-contamination due to improper cleaning and disinfection procedures was another possibility. As a result, incoming carcasses and cleaning procedures should be considered critical control points in pork cutting plants, since a heating step is not used to inactivate microorganisms. Furthermore, the high rate (59.6%) of E. coli isolation indicates E. coli can be a good indicator in livestock processing plants even though it has genetic diversity.

A Numerical Study on the Estimation Method of the Results of Static Pile Load Test Using the Results of Bi-directional Pile Load Test of Barrette Piles (바렛말뚝의 양방향재하시험을 이용한 정적압축재하시험 결과 추정방법에 관한 수치해석적 연구)

  • Bi-directional pile load test (briefly called 'BDH PLT') cannot be performed at loading levels where ultimate bearing capacity could be assessed in field, it is not possible to precisely determine both ultimate load and yield load and under loading. Since the load is transmitted separately to the skin and the end unlike the static pile load test (briefly called 'SPLT') and the direction of loading on the skin is opposite, such methods could have a result different from actual movements of shafts. In this study, three-dimensional finite element method (briefly called '3D FEM') analysis was conducted from results of the BDH PLT, made with barret piles, which were large-diameter cast-in-place concrete piles, and the calculated design constants were applied to the 3D FEM analysis of the SPLT to interpret them numerically and then, actual behaviors of cast-in-place concrete piles were estimated. First, using the results of the BDH PLT with cast-in-place concrete piles, behaviors of the piles made by loading upwards and downwards were analyzed to calculate load-displacement. Second, the design constants, calculated by the 3D FEM analysis and the back analysis, were applied on the 3D FEM analysis for the SPLT, and from these results, behaviors of the SPLT through the BDH PLT was analyzed. Last, the results of the 3D FEM analysis of the SPLT through the BDH PLT was expressed in relationships as {A ratio of bearing capacity of the SPLT and of the BDH PLT (y)} ~ {A ratio of reference displacement and pile circumference (x)}, and they were all classified by reference displacement at 10.0 mm, 15.0 mm, and 25.4 mm.
