Yangkyuksanhwa-Tang (YKSH), consisting of nine different herbs, is commonly used in Soyangin-type individuals with stroke, based on the Sasang Constitution Theory in Korea. However, no evidence has yet confirmed a beneficial effect of YKSH in ischemic stroke treatment. In this study, we investigated the effects of YKSH on ischemic brain injury in a mouse model of cerebral ischemia. Focal cerebral ischemia in mice was induced by photothrombosis, and behavioral recovery was evaluated. Infarct volume, inflammation, and newly generated cells were evaluated by histology and immunochemistry. YKSH treatment resulted in a significant recovery from the motor impairments induced by focal cerebral ischemia, as determined with wire grip and rotarod tests. YKSH treatment also decreased the infarct volume and the number of cells positive for tumor necrosis factor-${\alpha}$ and myeloperoxidase when compared with a vehicle-treated control group. By contrast, YKSH treatment considerably increased the number of cells positive for glial fibrillary acidic protein and ionized calcium-binding adapter molecule 1, as well as the number of cells doubly positive for Ki67/doublecortin when compared with the vehicle-treated group. These results suggest that YKSH treatment attenuated the infarct size by anti-inflammatory action, astrocyte and microglia activation, and neuronal proliferation, thereby facilitating neurofunctional recovery from a cerebral ischemic assault. YKSH could therefore be a potential treatment for neurofunctional restoration of the injured brains of patients with stroke.

Recently, adipose mesenchymal stem cells (AdMSC) that are similar to bone marrow MSC and blood derived MSC are thought to be another source for stem cell therapy. However, the diseases that can be applied for stem cells therapy are age-dependent degenerative diseases. Accordingly, the present study investigated the growth and differentiation potential to neural cells of human AdMSC (hAdMSC) obtained from aged thirty, forty and fifty. The growth of cells and cell viability were measured by passage and neural differentiation of hAdMSC was induced in neural differentiation condition for 10 days. Our results demonstrated that cell number, viability and morphology were not different from hAdMSC by age and passage. Immunofluorescence analysis of neural cell marker (TuJ1, NSE, Sox2, GFAP or MAP2) demonstrated no significant differences in neural cell differentiation by age and passage. As the number of passage was increased, the mRNA level of MAP2 and Sox2 was decreased in hAdMSC from age of 50 compared to hAdMSC from age of 30. In conclusion, the present study demonstrated that ability of neural cell differentiation of hAdMSC was maintained with ages, suggesting that autologous stem cells from aged people can be applied for stem cell therapy with age-dependent neural disease with the same stem cell quality and ability as stem cell derived from young age.
Eimeria tenella, an intracellular protozoan parasite infecting the epithelial cells of the ceca of chickens, causes severe diarrhea and bleeding that can lead its host to death. It is of interest that 2. tenezla first penetrate into the mucosal intraepithelial Iymphocytes (IEL) before they parasitize crypt or villous epithelial cells. This in vitro study was undertaken to know whether the penetration of E. tenella into such a lymphoid cell is a beneficial step for the parasite survival and development. Three sequential experiments were performed. First, the in vitro established bovine kidney cell line, MDBK cells, were evaluated for use as host cells for E. tenella, through morphological observation. Second, the degree of parasite development and multiplication in MDBK cells was quantitatively assayed using radioisotope labelled uracil ($^3H-uracil$) . Third, the E. tenella sporozoites viability was assayed after preincubation of them with thicken spleen cells. E. tenella oocysts obtained from the ceca of the infected chickens were used for the source of the sporozoites. Spleen cells (I) obtained from normal chickens (FP strain) were preincubated with the sporozoites (T) at the E:T ratio of 100:1, 50:1 or 25:1 for 4 or 12 hours, and then the mixture was inoculated into the MDBK cell monolayer. Morphologically the infected MDBK cells revealed active schisogonic cycle of E. tenella in 3~4 days, which was characterized by the appearance of trophozoites, and immature and mature schizonts containing merogoites. The 3H-uracil uptake by E. tenella increased gradually in the MDBK cells, which made a plateau after 48~60 hours, and decreased thereafter. The uptake amount of $^3H-uracil$ depended not only upon the inoculum sixte of the sporozoites but also on the degree of time delay (preincubation; sporozoites only) from excystation to inoculation into MDBK cells. The 3H-uracil uptake became lower as the preincubation time was prolonged. In comparison, after preincubation of sporozoites with spleen cells for 4 or 12 hours, the 3H-uracil uptake was significantly increased compared with that of control group. From the results, it was inferred that, although the penetration of E. tenella sporozoites into the lymphoid cells such as IEL is not an essential step, it should be at least a beneficial one for the survival and development of sporozoites in the chicken intestine.

This research was carried out to determine the effect of Cudrania tricuspidata fruit (CTF) on the performance of laying hens, egg quality, yolk fatty acid composition, Immunoglobulin G (IgG) and blood composition. A total 288 Brown Nick laying hens of 48 weeks old replaced into 4 groups with 4 replicates of 18 birds in each. Treatments were added with 0, 0.25, 0.5, 1.0% CTF containing dried seed and pulp. Metabolizable energy (ME) and crude protein (CP) were 2,800 kcal/kg and 16.0% in basal diets, respectively. Egg production and daily eggmass were significantly increased in 1.0% CTF addition treatment compared to those of other treatments (P<0.05). Egg weight and feed intake were not different among treatments. Feed conversion ratio showed the lower result in 1.0% CTF treatment than other treatments (P<0.05). Eggshell color was not influenced by the addition of CTF. Albumin height, Haugh unit and eggshell breaking strength was not statistically different, but seemed to be higher as dietary CTF addition increased. Yolk color was increased as dietary CTF increased and was the highest in 1.0% CTF group (P<0.05). Fatty acids were not shown the consistency in yolk. Blood cholesterol and triglyceride (TG) of birds fed CTF were significantly low compared to those of control. IgG was also statistically higher in CTF treatments than control (P<0.05). As the results of this experiment, the addition of 1% CTF would be the most effective on performance and egg quality, whereas the 0.25% CTF addition in basal diets seemed to be optimum to improve the blood cholesterol, TG,high density lipoprotein cholesterol (HDL-CHOL), IgG in laying hens.
Effects of various combinations of corn oil (CO) and perilla oil (PO) as respective dietary sources of ${\omega}$-6 and ${\omega}$-3 polyunsaturated fatty acids on fatty acid profiles of immune organs were studied in young chicks. Seventy-five 1-day-old male (ISA Brown) chicks were assigned to five treatments with three replications. Semi-purified-type diets containing glucose and soybean meal as major ingredients were added with 8% CO, 6% CO+2% PO, 4% CO+4% PO, 2% CO+6% PO and 8% PO and fed for 7 weeks. There were no significant differences in body weight gain, feed intake and relative weights of liver and immune organs (g/100g weight) among dietary groups. Dietary fatty acid patterns were generally reflected in the fatty acid compositions of all immune organs such as spleen, thymus and bursa of Fabricius. The levels of a-linolenic acid(LNA), eicosapentaenoic acid (EPA) and docosahexaenoic acid in various immune organs increased with increasing levels of perilla oil in the diets, whilet the levels of linoleic acid (LA) and arachidonic acid (AA) decreased. Thymus appeared to have capacity to retain remarkably higher (P<0.05) levels of LA and LNA up to 37 and 22%, respectively, compared to the other organs. Thymic tissue contained ${\omega}$-3 fatty acid and ${\omega}$-6 fatty acid 10~36 times and 3~5 times higher than the other organs, respectively. Spleen tissue was specifically higher (P<0.05) in the levels of AA and EPA and the ratios of AA/LA and EPA/LNA, compared to the other organs, suggesting that the tissue might have high desaturase activity to convert LA or LNA to AA or EPA, respectively. BSA antibody production tended to increase by 18 ~ 32% with higher levels of perilla oil in diet, although the increase was not statistically significant. In conclusion, fatty acid compositions of immune organs very depending on the lipid composition of the diets and each organ appears to respond differently for its fatty acid profile to dietary lipids. Considering AA and EPA are precursors of many important eicosanoids, further studies are required to clarify the responses of the immune organs to the dietary fatty acids.
Two different Eelwardsiella tarda isolates, KFE and Edk-2, were obtained from Korea and Japan respectively. On the base of the previous results showing higher pathogenicity of E. tarda KFE compared to that of E. tarda Edk-2 isolate, we tried to determine the differences of antigenicities of these two isolates in loach which is one of the important species in freshwater aquaculture in Korea. Concentration of specific antibody in the serum appeared to be much higher in loach immunized with FKC of E. tarda Edk-2 than those found in loach immunized with FKC of E. tarda KFE. Cross-reaction analysis using agglutination test with normal and antigen-absorbed antisera implied the differences epitopes in the antigens of these E. tarda isolates. For the comparison of bactericidal activity of the produced antibody with different antigens, absorption analysis was performed and confirmed the presence of critical epitopes in the FKC of E. tarda KFE strain. The prophylactic agent, polysaccharide-bound protein (PS-K) injected 1 week before the artificial infection with E. tarda KFE isolates decreased the cumulative mortality in loach and would be on effective method to prevent the occurrence of bacterial infection including E. tarda.

The effects of Sparassis crispa extracts fermented with isolated strain from S. crispa on antioxidant and immunological activities were determined. S. crispa extracts fermented with Meyerozyma guilliermondii FM showed significantly higher total phenol contents and DPPH radical scavenging activities compared to those fermented with lactic acid bacteria. In methotrexate-induced immunosuppressed rats, reduced levels of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-2, and immunoglobulin E (IgE) and increased levels of IL-10 were detected in S. crispa extract injected groups regardless of fermentation. We confirmed that rats treated with S. crispa fermented with M. guilliermondii FM showed higher blood leukocyte contents compared to other treatments. These results suggest that M. guilliermondii FM has high potential as a starter culture for fermentation of S. crispa extracts with increased antioxidant and immunological activities.

This study was carried out to investigate the functional activities of microorganisms isolated from kochujang, such as fibrinolytic, immunostimulating, and cytotoxical activities, and to apply these microorganisms to kochujang products. Ninety-one microbial strains with proteolytic activity were selected from 294 strains isolated from traditional and commercial kochujang. Three strains (TPP 0014, TPP 6013, and TPP 6015) with high fibrinolytic activity were tested for their immunostimulating and cytotoxical activities. For the assessment of macrophage activation, cytokines such as tumor necrosis factor, $interleukin-1{\alpha}$ and nitrogen oxide were measured with the murine macrophage cell line RAW 264.7. In addition, the cytotoxical activities of the three strains were examined by MTT assay on the colon cancer cell line SNU-C4 and normal cell line CHO-K1. Using an API identifying kit, two of the microbial strains (TPP 0014 and TPP 6015) were identified as Bacillus stearothermophilus and the other strain (TPP 6013) was identified as B. amyloliquefacience.
Platycodon grandiflorum (Korean name, Doraji) has been widely used in traditional herbal medicine as an expectorant for pulmonary disease and a remedy for respiratory disorders in Asia. Here, we investigated the effects of BtOH extract saponin fraction of P. gradiflourm (PGS) on phagocytosis and anti-tumor activity with related cytokine productions in RAW264.7 macrophage cells. The results showed that PGS increased phagocytosis, anti-tumor activity, TNF-${\alpha}$ and nitric oxide (NO) production without direct tumor cell cytotoxicity. To further investigate whether NO is involved in anti-tumor and phagocytic activities of PGS, cells were co-treated with specific iNOS inhibitors, L-NIL (N6-(1-iminoethyl)-L-lysine, dihydrochloride), to block NO production. PGS decreased anti-tumor activity in L-NIL-treated cells, whereas phagocytic activity was not inhibited under the same conditions, indicating that the anti-tumor activity by PGS appears to be conducted by NO. These findings suggest that P. grandiflorum could be used a potential nutrition therapeutic agent for cancer patients.
A total of fourteen, 1-wk-old male Holstein calves were allotted into two groups consisted of control (CON) and IGY which was orally administrated with immunoglobulin yolk (IgY) for 1wk. Calves in both groups were provided with milk replacer according to feeding program and had ad libitum access to timothy hay for the entire experimental period (7wks). At 0, 7 and 49 day of experiment, blood samples were collected from the jugular vein of calves to investigate blood biochemical profiles and the differential count (%) of white blood cell (WBC). We also monitored growth performance and colony forming unit (CFU) of fecal microbial population in calves. The adminstration of IgY in calves did not affect body weight and weight gain during 49 days feeding trial compared with control group. The CFU of E. coli and Lactobacilli in the feces of calves were not significantly affected by IgY treatment, whereas the score of the calf scours during day 43 to 49 in IgY group showed a significant (P<0.05) solid type. There were no differences in plasma biochemical components including total protein, albumin, immunoglobulin and the other indicators. As for WBC differential count (%), there was no statistical difference in the percentages of neutrophil, lymphocyte, monocyte, eosinophil and basophil at 0, 7 and 49 days after the oral supplementation of IgY. In conclusion, the oral supplementation of IgY as an immunostimulant did not affect growth performance, fecal microbial population, blood biochemical profile and WBC differential count in Holstein calves.
