Effects of Banha Extract on the Melanin Biosynthesis and Tyrosinase mRNA Level in Bl6 Mouse Melanoma Cells (반하 추출물이 B-16 마우스 흑색종 세포의 멜라닌 생성과 타이로시네이즈 mRNA 양에 미치는 영향)

  • Melanin pigmentation in human skin is a major defense mechanism against ultraviolet light of the sun. Tyrosinase(EC 1.14.18.1) plays a key role in the biosynthesis of ultraviolet of melanin. This is why much researches have been focused on its regulation in controlling the epidermal melanization. We have found that the water-extract of Banha(Pinelliae ternate B.), an oriental medicinal plant, has no tyrosinase inhibitory activity, but does inhibit the melanin biolsynthesis in B16 mouse melanin cells. We also found that Banha extract lowers the tyrosinase activity in cultured cells. To elucidate the action mechanism of Banha extract we have investigated its effect on the tyrosinase mRNA level using reverse transcription-polymerase chain reaction technique. It was revealed that Banha extract reduced the tyrosinase mRNA level in dose dependent manner; when B16 mouse melanoma cells were cultured with 2mg/ml and 5mg/ml of Banha extract, there were 20% and 44% decrease in tyrosinase mRNA level, respectively. These data suggest that the Banha extract exerts its melanogenic inhibitory effect through the transcriptional regulation of tyrosinase mRNA.

Whitening Effect of Fagopyrum tataricum Extract (쓴메밀 추출물의 미백 개선 효과)

  • This study was performed to DPPH radical scavenging activity, ABTS radical scavenging activity, tyrosinase inhibition activity and intracellular melanin synthesis inhibition to verify the whitening effect of Fagopyrum tataricum (bitter buckwheat) extract as contrasted with Fagopyrum esculentum (sweet buckwheat) extract. F. tataricum extract in consequence showed higher antioxidant activities, tyrosinase inhibition activity and melanin synthesis inhibition compared with F. esculentum extract. We investigated skin bright value during 8 weeks after induction of pigmentation in human skin from UV irradiation. In result, we obtained statistically a significant skin whitening effect from visual and mechanical evaluation. Accordingly, F. tataricum extract is expected to be high availability as functional cosmetic material for skin whitening.

Potential uses of Aristotelia chilensis extracts as novel cosmetic materials (마키베리 추출물의 화장품 신규 원료로서의 가능성)

  • The present study aims to investigate the potential applications of Aristotelia chilensis (A. chilensis) extracts as novel cosmetic materials. The total extracts of A. chilensis were partitioned into chloroform (CHCl3), ethyl acetate (EtOAc), and distilled water (DW) fractions. A. chilensis extracts exhibited no cytotoxicity toward HaCaT human keratinocyte and B16F10 mouse melanoma cell lines. CHCl3, EtOAc, and DW extracts reduced oxidative stress, and EtOAc extract was superior to glutathione, a natural human antioxidant positive control. The extracts of A. chilensis reduced melanin synthesis in cells treated with α-melanocyte-stimulating hormone. The extracts of A. chilensis exhibited antibacterial effects toward Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), and Pseudomonas aeruginosa (P. aeruginosa). In particular, the EtOAc extract was effective in terms of antibacterial activity against S. aureus. In the present study, we identified several potential applications of A. chilensis extracts in terms of novel antioxidant and whitening cosmetic materials as well as antibacterial preservatives.

Inhibition of Melanin Synthesis by Enhanced Cytosolic Delivery of N-glycosylation Inhibitors Using pH-Sensitive Nano-carrier (pH 감응형 나노입자를 이용한 멜라닌 합성저해 연구)

  • Inhibition of the early N-glycosylation process in the endoplasmic reticulum prevents the activation of tyrosinase, a key enzyme for melanin biosynthesis. This work aims at evaluating the increased activity of N-glycosylation inhibitors in vitro b, employing a nano-sized pH-sensitive liposome as a delivery carrier. Melexsome, a pH-sensitive nano carrier loaded with glycosylation inhibitos, was prepared by the hydration method with phospholipids and cholresterol-based amphiphiles. Inhibitory effects of Melexsome on the N-glycosylation process were evaluated by EndoH & PNGaseF digestion and the western blotting. Melanin synthesis was also monitored after treatment with Melexsome Interestingly, Melexsome effectively increased the efficacy of N-glycosylation inhibitors. Melexsome was also much more efficiently translocated into the cytoplasm as observed in CLSM. These results demonstrated that the amphiphilic lipid-based pH-sensitive nano-carriers could be, used as an efficient delivery system for N-glycosylation inhibitor to enhance the effects of skin whitening cosmetics.

Characterization of Melanin-concentrating Hormone from Olive Flounder (Paralichthys olivaceus) (양식넙치 멜라닌 농축 호르몬의 특성)

  • The melanin-concentrating hormone (MCH), a cyclic hypothalamic peptide composed of 17 amino acids, was initially identified in chum salmon (Oncorhynchus keta) as a regulator of pigmentation. Mammalian MCHs are cyclic hypothalamic peptides composed of 19 amino acids that regulate food intake and energy homeostasis. The present study examined not only MCH expression of different tissues but also the melanohore aggregation and intracellular $Ca^{2+}$ influx of fMCH and the other MCH. Real-time qPCR showed that MCH expressed specially in the brain, gonad, and ovary, and expression of MCH was observed during the developmental stages. In the application of synthetic fMCH and both types of synthetic fMCH, dN-fMCH and dC-fMCH, scale melanophore induced significant changes in aggregation activity with various concentrations of MCH. Also, compared to hMCH and sMCH, fMCH exhibited a 36~99.85% increase in relative potency (%), whereas aggregation of dN-fMCH and dC-fMCH remained in a high concentration. However, dispersion was induced rapidly according to be low concentration of dN-fMCH and dC-fMCH. We show that fMCH and its derivates were bound human MCHR1 and rat MCHR expressed in HEK293T cells with nano-molar affinity and are likely to be ligand-induced to mobilize intracellular $Ca^{2+}$. These results may provide new ligands for binding assay with MCHew ligands, as a structure similar to the mammalian MCH structure was discovered in fish. Once the fMCH receptor system is in place, it can be compared to the MCH system of mammals in terms of MCH function.

Whitening Effects of Angelica dahurica Radix Ethanol Extract (백지 에탄올추출물의 미백효능 연구)

  • To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ${\mu}g/m{\ell}$ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/$cm^2$ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450~500g) and then the test agent of $30{\mu}{\ell}$ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ${\mu}g/m{\ell}$, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.001) than the vehicle control group and this resultsagreed with the results of observation under microscope. From these results, weconcluded that ADEE had positive whitening effect.

Physiological Activities of Cudrania tricuspidata Extracts on the Skin (꾸지뽕나무 추출물의 피부 생리 활성)

  • This paper has shown the experimental results about the physiological activities of water-, ethanol-, ethyl acetate-soluble fractions from ethanolic extracts of leaves, stems and roots of Cudrania tricuspidata on the skin, which has been used for a long time as a traditional herb medicine in Korea and China. The effects of these fractions on the secretion of nitric oxide and cytokines from macrophage(RAW 264.7 cell) exhibited that the ethyl acetate and water fractions from leaves inhibited the release of nitric oxide, all fractions inhibited thoses of inflammatory cytokine $IL-1{\alpha}$, and the ethyl acetate fractions of leaves, stems and roots inhibited thoses of inflammatory cytokine IL-6. The only ethylacetate fraction of leaves demonstrated significantly the reduction of melanin synthesis in melanoma cells. In order to evaluate the efficacy of collagen synthesis, the treatment with extracts on the human normal fibroblast cell(CCD-986sk cell) resulted in finding that the water fractions of leaves, stems and roots and the ethanol fractions of leaves and stems showed the increased synthesis of collagen.

Antioxidant and Tyrosinase Inhibitory Effects of Brassica oleracea L. Fractions (적채 분획물의 항산화 및 Tyrosinase 억제 효과)

  • This research was designed to investigate the antioxidant activity of red cabbage (Brassica oleracea L., BO) fractions on the liposomes consisted of L-$\alpha$-dilinoleoylphosphatidylcholine (DLPC-liposome) and the tyrosinase activity of BO fractions. The methanol extract of BO (BOM) was fractionated into five different partition layers: hexane (BOMH), ethylether (BOMEE), ethylacetate (BOMEA), butanol (BOMB) and aqueous (BOMA) layers. The antioxidant activities of BOM fractions toward oxidized DLPC-liposome were examined by spectrophotometry measuring oxidized conjugated dines. The antioxidant activities of BOMEE and BOMEA fractions toward oxidized DLPC-liposomes were similar to the antioxidant activities of $\alpha$-tocopherol and weaker than that of BHT. The synergy effects of antioxidation of BOMEE and BOMEA fractions added with vitamin C and $\alpha$-tocopherol had even stronger antioxidant activities than the fractions without vitamin C and $\alpha$-tocopherol. These results showed that the fractions of BOMEE and BOMEA could be developed as a potent antioxidant. Out of five different partition layers of BOM fractions, BOMEA exhibited the strongest tyrosinase activity of 94% at a concentration of 40 $\mu\textrm{g}$/mL. This result suggests BOMEA fractions inhibit the foramation of melanin and therefore can be used as the inhibitor of melanin synthesis. Results of antioxidant activity and tyrosinase inhibition indicate that useful bioactive substances exist in BOMEE and BOMEA fractions. Both fractions from BO (red cabbage) have the potential of being developed into health related products.

  • Hair follicles develop as a result of epithelial-mesenchymal interactions between epidermal keratinocytes and dermal cells. Moreover hair follicles constitute multiple cells that influence hair follicle development and cyclic activity. We isolated some cells using explantation and enzymatic digestion method from human scalp hair follicles. So we could culture some follicular cells, such as outer root sheath (ORS) cells, dermal papilla (DP) cells, dermal sheath (DS) cells, matrix cells and melanocyte.

Effect of Halophilic Bacterium, Haloarcula vallismortis, Extract on UV-induced Skin Change (호염 미생물(Haloarcula vallismortis) 용해물의 자외선유발 피부변화에 대한 효과)

  • Skin carrys out protective role against harmful outer environment assaults including ultraviolet radiation, heavy metals and oxides. Especially, ultraviolet-B (UVB) light causes inflammatory reactions in skin such as sun burn and erythma and stimulates melanin pigmentation. Furthermore, the influx of UVB into skin cells causes DNA damage in keratinocytes and dermal fibroblasts, inhibition of extracellular matrix (ECM) synthesis which leads to a decrease in elasticity of skin and wrinkle formation. It also damages dermal connective tissue and disrupts the skin barrier function. Prolonged exposure of human skin to UVB light is well known to trigger severe skin lesions such as cell death and carcinogenesis. Haloarcula vallismortis is a halophilic microorganism isolated from the Dead Sea, Its growth characteristics have not been studied in detail yet. It generally grows at salinity more than 10%, but the actual growth salinity usually ranges between 20 to 25%. Because H. vallismortis is found mainly in saltern or salt lakes, there could exist defense mechanisms against strong sunlight. One of them is generation of additional ATP using halorhodopsin which absorbs photons and produces energy by potential difference formed by opening the chloride ion channel. It often shows a color of pink or red because of their high content of carotenoid pigments and it is considered to act as a defense mechanism against intense UV irradiation. In this study, the anti-inflammatory effect of the halophilic microorganism, H. vallismortis, extract was investigated. It was found that H. vallismortis extract had protective effect on DNA damage induced by UV irradiation. These results suggest that the extract of halophilic bacterium, H. vallismortis could be used as a bio-sunscreen or natural sunscreen which ameliorate the harmful effects of UV light with its anti-inflammatory and DNA protective properties.
