  • The purpose of this study was to investigate anti-melanogenesis effects of mulberry seed extracts (MSE). MSE inhibited melanogenesis in melan-a cells at $10{\mu}g/mL$ without cytotoxicity. Also, MSE decreased tyrosinase, tyrosinase-related protein-1 (TRP-1) protein expression in the melan-a cells. To identify the signaling pathway of MSE, the ability of MSE to influence extracellular signal-regulated protein kinase (ERK) activation was investigated. MSE induced ERK protein expression in a dose-dependent manner. In addition, MSE presented inhibition of the body pigmentation in vivo zebrafish model. These results suggest that MSE may be an effective anti-melanogenesis agent regulating the expression of ERK protein and melanogenic enzymes.

  • Melanogenesis is involved in the pigmentation of the hair, eyes, and skin in living organisms. Various signaling pathways stimulated by ${\alpha}-MSH$, SCF/c-Kit, $Wnt/{\beta}-catenin$, nitric oxide and ultraviolet activate melanocyte, leading to melanin production by tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 expressed via the microphthalmia-associated transcription factor (MITF). However, the abnormal regulation of melanogenesis causes dermatological issues such as graying hair and vitiligo. Therefore, the activators that promote melanogenesis are crucial for the prevention of graying hair and the treatment of hypopigmentary disorders. Many melanogenesis stimulators have been studied for the development of novel drugs derived from synthesized compounds and natural products. Here, in addition to providing a description of a common signaling pathway in the melanogenesis of graying hair and the vitiligo process for the development of novel anti-hair graying agents, this article reviews natural herbs and the active ingredients that promote melanin synthesis as a pharmaceutical agent for the treatment of vitiligo. In particular, compounds such as Imatinib and Sugen with a stimulating effect on melanogenesis as a side effect of the drugs, are also introduced. Recent advances in research on natural plant extracts such as Polygonum multiflorum, Rhynchosia Nulubilis, Black oryzasativa, and Orysa sartiva, widely known as traditional and medicinal extracts, are also reviewed.

  • Gray hair is a representative sign of aging. Intrinsic aging, stress, and the external environment cause hair graying. Stress is known to be a major factor in the early onset of hair graying. We previously found that Pueraia lobata root extract (PLRE) can prevent hair graying by promoting melanin formation. However, it remains unknown whether PLRE can prevent hair graying induced by conditions of stress. In this study, we confirmed the effect of PLRE on stress-induced hair graying. A reporter cell line was newly constructed to confirm the expression of microphthalamia-associated transcription factor (MITF), the main transcription factor for melanin production. MITF expression and melanin pigmentation were reduced in human hair follicle tissue treated with the stress hormone cortisol or H2O2 to induce oxidative stress. PLRE treatment restored MITF expression and increased the amount of melanin pigment in the hair follicle. The expression of Tyrosinase related proteins-2 (TRP-2), a melanin synthesis enzyme in the hair follicle, also increased. In conclusion, PLRE can effectively prevent the inhibition of melanin synthesis by stress hormones and oxidative stress.

  • Purpose: Gingival whitening is one of dental treatment purposes which is close to treating aesthetic disorders. Initial gingival whitening treatment was done by dermabrasion using a high power Diode Laser. However, this treatment method cannot be free from any infection or pain after the treatment. Therefore, we have decided to progress gingival whitening treatment using a low power LED laser. Materials and Methods: The laser was irradiated on pork meat then the safety of output power, temperature change and skin denaturalization was measured. Bison 365 nm LED laser was irradiated on oral mucosal pigment of a 15 - 20 kg beagle for 15 min for 1 - 2 weeks, one or two times each. Any pigment loss was checked through Hematoxyline-Eosin staining. Results: The melanin pigments at the area of 365 nm LED Laser irradiation were decreased. Conclusion: The 365 nm LED Laser proposed in this study is considered to compensate the bleaching effect achieved by either using Diode laser or surgical methods.

  • This study investigated whether the whitening effects of fullerene serum occurred through improvement of melanin production and skin tone and evaluated its potential for use as a cosmetic material for adult women aged 30-55 with dull skin color and pigmentation. Subjects were divided into a control group with fullerene-less serum (B) and an experimental group with serum containing 0.1% fullerene (A) and evaluated over 8 weeks. Visual assessment and measurement of skin conditions (melanin index, skin brightness) were conducted before use of and 4 and 8 weeks after use of the product, and a questionnaire survey and safety assessment were conducted at the end of the experiment. The results revealed that the highest decrease was 0.37% in the experimental group (A) after eight weeks of use of the product, but that this difference was not statistically significant. Moreover, no significant differences were observed upon inter-group comparison. Evaluation of changes in the melanin index and inter-group comparison revealed a significant decline in the experimental group (A) at all time points. Additionally, the $L^*$ value (skin brightness) of the experimental group (A) showed a significant increase (improvement) after eight weeks, while in inter-group comparison revealed a significant increase (improvement) at all time points (4 and 8 weeks later) (p<0.05). Based on a questionnaire survey of efficacy, positive response rates such as improvement of the target parts on week 8 were high in terms of 'improvement of skin color.' Moreover, the safety assessment revealed no adverse reactions. In conclusion, serum containing 0.1% fullerene revealed improved melanin production and skin tone (brightness) and confirmed its potential for use as a cosmetic material with brightening efficacy. Taken together, the results of this study indicate systematic investigation of the effective application and cosmetics formulations of this product are warranted.

  • Soybean (Glycine max), as one of the foods with high plant proteins, contains a large amount of bioactive compounds and known to be effective in cardiovascular disease and obesity as well as in improving skin condition. The purpose of this study was to investigate the anti-photoaging effects of soybean fermented with Lactobacillus Rhamnosus ($Bio-Peptone^{(R)}$) by assessment of cytotoxicity against UVB, collagen synthesis after UVB-irradiation, tyrosinase activity, and melanin synthesis. Results showed that $Bio-Peptone^{(R)}$ protected skin fibroblasts against UVB-induced cytotoxicity and increased type I collagen synthesis. Furthermore, $Bio-Peptone^{(R)}$ significantly inhibited tyrosinase activity and reduced melanin contents. This study suggests that $Bio-Peptone^{(R)}$ has protective effects against UVB-induced skin damage. Thus, it is concluded that $Bio-Peptone^{(R)}$ is able to prevent skin damage against UVB and thus acts as anti-photoaging materials by protecting skin forming wrinkles and skin pigments.

  • In this study, liquid culture of Tricholoma matsutake mycelia was performed via biomass production, and its inhibitory effects on melanin biosynthesis were evaluated. The Tricholoma matsutake mycelia extract inhibited 38.6% of tyrosinase activity at 100 ppm, which is higher than that of extracelluar medium at same dose. In addition, when 100 ppm of Tricholoma matsutake mycelia extract was treated to melan-a cells for 3 days, 19% of melanin production was reduced without cell toxicity. These results suggested that cultured Tricholoma matsutake mycelia might be useful as a skin depigmenting material.

  • When fruit bodies of Coprinellus congregatus were matured, they were autolysed to form black ink. During the developmental changes, cell walls of basidia were degraded. Laccase formed melanin which was the typical black pigment of fungi, and chitinase hydrolyzed the chitin which was a component of fungal cell wall. When laccase and chitinase genes were used as the probe for the Northern analysis to confirm their expression during the fruit body development, both gene expressions were increased as the mushroom was getting matured.

  • This study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 36 ~ 86%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as skin whitening agents.
