A Taxonomic Review of the Korean Atractomorpha Saussure, 1862 (Orthoptera: Caelifera: Pyrgomorphidae) (한국산 섬서구메뚜기속(Atractomorpha)의 분류학적 검토)

  • A taxonomic review of the genus Atractomorpha Saussure in Korea is presented. Two species are identified, Atractomorpha lata (Motschulsky, 1866) and a new record A. sinensis Bolivar, 1905 which is reported from the far southern islands in Korea, Gageo-do and Jeju-do. A key, descriptions, habitus photographs of adults, and distributional information are provided.

Oxya Chinensis Sinuosa Mishchenko Extract: Potent Glycosidase Inhibitor Alleviates Postprandial Hyperglycemia in Diabetic Mice (당뇨 모델을 이용한 벼메뚜기(O. Mistshenk) 추출물의 식후 고혈당 완화 효과)

  • This study was designed to investigate whether extracts from Oxya chinensis sinuosa Mistshenk (an edible insect considered a grasshopper) could inhibit the activity of carbohydrate digestive enzymes and alleviate postprandial hyperglycemia in diabetic mice. Oxya chinensis sinuosa Mistshenk was extracted with 80% ethanol (OEE) or water (OWE) and then concentrated. The carbohydrate digestive enzyme-inhibiting activity of the resulting extracts was evaluated by examining α-glucosidase and α-amylase. The IC50 values of OEE against α-glucosidase and α-amylase were 0.229 mg/ml and 0.106 mg/ml, respectively. This result indicated that OEE has stronger inhibitory effects than OWE and positive control. The blood glucose levels of the diabetic control mice increased after one meal. However, when OEE (300 mg/kg) was added to starch, this increase in postprandial blood glucose levels was significantly suppressed. The area under the curve also significantly decreased following the administration of OEE, which exhibited no cytotoxicity. These results indicate that OEE is more efficacious than OWE and may be used as a carbohydrate digestive enzyme inhibitor, delay carbohydrate digestion and glucose absorption, and thus alleviate postprandial hyperglycemia caused by dietary carbohydrates.

Effects of Grasshopper (Oxya chinensis sinuosa Mistshenko) Powder and Aerobic Exercise on Energy Metabolism in ICR Mice (벼메뚜기(Oxya chinensis sinuosa Mistshenko) 분말 섭취와 유산소성 운동훈련에 의한 마우스의 에너지 대사 변화)

  • The purpose of this study was to analyze the effect of grasshopper (Oxya chinensis sinuosa) powder ingestion with/without aerobic exercise (treadmill running) on energy metabolism. To achieve this purpose, 28 Institute of Cancer Research (ICR) mice were divided into four groups: normal diet control group (CON), a normal diet with exercise control group (COEX), a grasshopper powder-supplemented diet group (GH), and a grasshopper powder-supplemented diet with exercise group (GHEX). Duration of the powder ingestion and aerobic exercise training were 6 weeks. Body weight gain ratio was not significant. Fat mass significantly decreased in GH and GHEX. There were no changes in blood glutamic oxaloacetic transaminase and glutamic pyruvic transaminase levels between groups. Glucose transporter type 2 and glucose transporter type 4 protein levels were not significantly different between groups. Fibronectin type III domain-containing protein 5 level was the highest in GHEX. AMP-activated protein kinase level significantly increased in GHEX compared to the levels in the other groups. Glycogen synthase kinase 3 beta protein level was reduced in GHEX compared to that in CON. These results suggest that grasshopper powder ingestion and endurance exercise training influence energy metabolism.

Lipid Oxidation and Browning during Storage of Dried Grasshopper (벼메뚜기 건제품 저장중의 지질산화와 갈변)

  • Lipid composition of dried grasshopper which the most significant factor of quality deteriorioration for processing and storage were studied. Also brown pigment formation and its effect of lipid autoxidation were studied. Lipid contents of matured female grasshopper were revealed 5.12% and composed of high content of unsaturated fatty acid. The phospholipid comprised 27.35% of the total lipid. On sun ana hot ai' drying, carbonyl values were greatly increased but peroxide values were not determined. On freeze drying, lipid peroxide and carbonyl compounds in grasshopper lipids were nearly not accumulated. Peroxide values were increased during early stage of storage, but carbonyl values were steadly increased for 98 days. Hydrophilic brown pigments which caused by sugar-amino reaction were higher than that of lipophilic. In the water soluble fraction of the browning products has some autioxidative activity, but in liposoluble fraction has not.

Study on the Detection of In vivo and In vitro Hemocytic Pathway in Grasshopper, Euprepicnemis shirakii (메뚜기 체내와 체외에서 혈구생성 경로 추적에 관한 연구)

  • A Differentiating pathway of hemocytes in vitro and in vivo of grasshopper, Euprepocnemis shirakii was described using light and electron microscopes. In the interior of body, the stem cells of the hemopoietic organ differentiated into six types of cells respectively which are prohemoyte, plasmatocyte, granulocyte I, granulocyte II, spherulocyte and oenocytoid. The formation of these hemocytes was derived from the stem cells surrounded by a reticular cell. Hemopoietic tissue cultured in the insect media differentiated different hemocytes, but none of them underwent any mitotic division. Morphological features of the cultured cells in media were essentially the same as those of the hemocytes differentiated from the stem cells in vivo. These results were shown that each stem cell could differentiate into different types of hemocytes. It was confirmed that the stem cells possessed the pluripotent differentiation ability to directly each hemocyte, and that the once formed hemocytes in vivo and in vitro didn t undergo further transformation to other hemocytes. The maintenance of circulating hemocytes in grasshopper had been depended on the widely spreading hemopoietic organ situated in the upper surface of the dorsal alary muscle and located on the first to eighth segments.

Antioxidant activity and antimicrobial activity of the grasshopper, Oxya chinensis sinuosa (벼메뚜기의 항산화 효과와 항균 활성)

  • In this study, to confirm the physiological activity of the grasshopper (Oxya chinensis sinuosa) antioxidant activity, antimicrobial activity, thermal stability of the antimicrobial substance, pH stability, total polyphenol content of the adult grasshopper was measured. Antibacterial activity in accordance with the extraction solvent showed a strong antibacterial activity in a mixed solvent of ethanol for the E. coli strain. Antimicrobial activity in $40^{\circ}C$ hot air drying and a freezedried condition was the highest and there was no difference in the gender. Antibacterial substance was stable to heat and pH. Antioxidant activity of the grasshopper exhibited a high activity in the 50% and 70% ethanol extract. Total polyphenol content was the 12 ~17 mg / 100 g and there was not great difference according to the drying conditions and gender.

Anti-inflammatory and Cellular Protective Effects on Hydrogen Peroxide-induced Cytotoxicity of Grasshopper Extracts (메뚜기류 추출물의 염증 조절작용 및 세포사멸보호 효과)

  • In order to investigate the anti-inflammatory and cellular protective effects of Atractomorpha lata Motschulsky, Oxya japonica japonica Thunberg and Stethophyma magister Rehn, we first examined hydrogen peroxide-induced cytotoxicity in SH-SY5Y cells as well as antioxidant assays including DPPH and FRAP assays. We found that water, ethanol and methanol extracts of Oxya japonica japonica Thunberg and Stethophyma magister Rehn had potentials to anti-oxidant activity and especially water extract of Oxya japonica japonica Thunberg exhibited the most potent protective effects against $H-2O_2$-induced cytotoxicity in SH-SY5Y cells by MTT assay. Taken Together, these findings indicate that water extract of Oxya japonica japonica Thunberg could be useful insect resources for agrobiotechnological or oriental medicinal purposes.

Oxya chinensis sinuosa Mishchenko (Grasshopper) Extract Protects INS-1 Pancreatic β cells against Glucotoxicity-induced Apoptosis and Oxidative Stress (INS-1 췌장 베타 세포에서 벼메뚜기(Oxya chinensis sinuosa Mistshenk) 추출물의 당독성 개선 효과)

  • Type 2 diabetes is a serious chronic metabolic disease, and the goal of diabetes treatment is to keep blood glucose at a normal level and prevent complications from diabetes. Hyperglycemia is a key pathologic feature of type 2 diabetes that mainly results from insulin resistance and pancreatic β-cell dysfunction. Chronic exposure of β-cells to elevated glucose concentrations induces glucotoxicity. In this study, we examined whether an 80% ethanol extract of Oxya chinensis sinuosa Mishchenko (OEE) protected INS-1 pancreatic β-cells against glucotoxicity-induced apoptosis and oxidative stress. Pretreatment with a high concentration of glucose (high glucose = 30 mM) induced glucotoxicity and apoptosis of INS-1 pancreatic β cells. Treatment with OEE significantly increased cell viability. Treatment with 0.01-0.20 mg/ml OEE dose dependently decreased intracellular reactive oxygen species, lipid peroxidation, and nitric oxide levels and increased insulin secretion in high glucose-pretreated INS-1 β cells. OEE also significantly increased the activities of antioxidant enzymes in response to high-glucose-induced oxidative stress. Moreover, OEE treatment significantly reduced the expressions of pro-apoptotic proteins, including Bax, cytochrome C, caspase-3, and caspase-9, and increased anti-apoptotic Bcl-2 expression. Apoptotic cells were identified using Annexin-V/propidium iodide staining, which revealed that treatment with OEE significantly reduced high-glucose-induced apoptosis. These findings implicate OEE as a valuable functional food in protecting pancreatic β-cells against glucotoxicity-induced apoptosis and oxidative stress.
