Analysis of n-Hexanal in Headspace Vapor over Cooked Brown Rice by Direct Vapor Injection Gas Chromatography (현미취반시 생성되는 n-hexanal의 가스크로마토그래피에 의한 분리및 정량)

  • n-Hexanal in headspace over the cooked brown rice stored at $5^{\circ}C$ and $35^{\circ}C$ for 0. 4, 8 and 12 months was determined by a modified direct vapor injection gas chromatographic method. The retention time of n-hexanal was 3.5 min and n-hexanal could be rapidly separated from other compounds at the operational conditions of gas chromatography. n-Hexanal contents of cooked brown rice also showed a standard deviation of less than 10% of the average.

  • Lysozyme is an important antibacterial material, as effective food preservative. A number of lysozymes are found in nature such as egg white, where exists about 3.5% of egg proteins. In this study, carboxymethyl cation exchange chromatography has been used for separation of lysozyme. A simulation study by ASPEN was also performed for saving time and cost in chromatography purification experiments. Important parameters in experimental chromatography were sample loading amount, NaCl concentration, and pH of eluent. Simulation results were successfully fitted with chromatograms from experiments with change of parameters mentioned above.

  • Enantiomer separation of N-fluorenylmethoxycarbonyl (FMOC) $\alpha$-amino acid was performed on covalently immobilized chiral column (Chiralpak IB) based on polysaccharide derivative as a chiral selector by reversed phase liquid chromatography. The effect of the reversed mobile phase on the chromatographic parameters of the enantioselectivities, resolution factors and retention times using covalently immobilized Chiralpak IB was shown. Also the enantiomer separation of N-FMOC $\alpha$-amino acid in the reversed and normal phase was compared and the results obtained in the former mobile phase were generally lower than those in the latter mobile phase.

  • 스트레스로 인한 인체의 반응은 중추신경계, 시상하부, 변연계 및 기타 표적기관으로부터 시작되는데, 자율신경계 반응, 내분비계 반응, 면역계 반응 등을 통하여 복합적인 신체 증상으로 발현되며, 스트레스 상황에서는 시상하부-뇌하수체-부신 축 (HPA axis)과 교감신경계의 작용으로 여러 신경전달물질 방출에 변화가 생기며 이러한 변화는 면역기전에 중요한 역할을 하고 일부는 면역세포의 활성에 직접적으로 영향을 미쳐 신체 각종 질병의 원인이 될 것으로 추정된다. 한의학에서는 천인상응(天人相應)의 관점에서 육기(六氣)를 생체자극의 외적 요인으로 간주하고, 생체내적 현상인 정신이 외적 자극을 통하여 나타나는 생체반응을 칠정(七情)으로 보았으며, 이러한 관점에서 스트레스는 신체에 오장(五臟)의 허실(虛實), 혈허(血虛), 정손(精損), 기역(氣逆), 기(氣)의 순환장애, 담연(痰涎), 화(火) 등의 병적인 요인을 만들어 준다. 본 연구에서 재료로 사용된 호초 (Piper nigrum Linne)는 후추나무의 과실을 말린 것으로서 세계적으로 널리 사용되는 향신료이며, 한의학에서는 온중제한하기(溫中除寒下氣), 쾌격소담(快膈消痰), 해독(解毒)등의 효능으로 한담식적(寒痰食積) 완복냉통(脘腹冷通) 곽란(癨亂) 토사(吐瀉)등의 치료에 활용되어 왔다. 특히 쾌격소담(快膈消痰)하는 작용은 정신적 스트레스에 유효할 것으로 생각되므로 본 연구에 이용하게 되었다. 실험동물은 ICR계 생쥐를 이용하였으며, 심리적 스트레스는 옆쪽 cage에서 다른 마우스의 신체에 가해지는 전기 충격을 하루 1시간 동안 지켜보게 하는 것으로 유발하였으며, 이 상태에서 약물을 투여한 그룹을 실험군, 그렇지 않은 그룹을 대조군으로 하였다. 정상군은 아무런 자극 없이 하루 1시간 동안 일정 공간에 가두어 두는 것으로 하였다. 실험 결과, 호초(胡椒) 추출물을 100mg/kg/day 용량으로 5일간 투여한 실험군은 아무런 처치를 하지 않은 대조군에 비해 혈장 중 corticosterone 함량이 유의하게 감소되었고, 뇌에서의 noradrenalin 분비량이 유의하게 증가되었으며, plus maze test에서의 머무름 시간이 연장되는 것으로 나타나 호초(胡椒)가 심리적 스트레스를 효과적으로 억제하고 진정작용이 있는 것으로 사료되나 구체적인 작용기전 및 인체에서의 효과에 대해서는 향후의 보다 자세한 연구가 필요할 것으로 생각된다.

Analysis of Organic Acids in Urine by Gas Chromatography/Mass Spectrometry (기체 크로마토그래피/질량분석법을 이용한 요 중에서의 유기산 분석)

  • GC/MS was used to find an optimum experimental condition for the screening of organic acids in urine. Urinary organic acids were isolated through the liquid-liquid extraction method (LLE) to examine the influence of pH and the effect of including the back extraction and oximation processes respectively on the extraction. When pH was adjusted to 0.5 during the extraction without oximation process, relatively higher recovery rate and the smallest relative standard deviations (0.3-12.4%) were obtained. By removing the interference, the addition of back extraction made possible surer identification of organic acids with retention time of 15-16 minutes. Under this condition, we obtained the content distribution of urinary organic acids in healthy Korean children (n=16) by establishing the calibration curves for 51 standard organic acids.

  • The adsorption and separation behaviors of transition metal ions using a merrifield resin bound 1,12-diaza-3,4:9,10-dibenzo-5,8-dioxacyclopentadecane (NTOE) and 1,12,15-triaza-3,4:9,10-dibenzo-5,8-dioxacycloheptadecane(NDOE) were investigated in aqueous solution. The orders of adsorption degree(E) and distribution ratio(D) of transition metal ions were Cu(II)$t_R$) of metal ions were affected by adsorption degree(E) and distribution ratio(D). This results showed good separation efficiency of Ag(I) from mixed metal solution.

  • The adsorption and separation behaviors of alkali, alkaline earth and transition metal ions using a poly-dibenzo-18-crown-6 were investigated in aqueous solution. The adsorption degree(E) and distribution ratio(D) of alkali, alkaline earth metal ions were Li(I)$t_R$) of metal ions were affected by the adsorption degree(E) and distribution ratio(D). This results showed good separation efficiency of K(I), Sr(II), Ag(I) and Pb(II) from the mixed metal solution.

  • The retention behaviors of polystyrenes influenced by mixed solvents are examined in thermal field-flow fractionation(ThFFF). Experimental data are obtained with polystyrene samples of molecular weights of 35,000, 110,000, 200,000 and 470,000 dissolved in organic solvents. The pure and mixed solvents are tetrahydrofuran(THF), chloroform(CHL), cyclohexane(CH), and benzene(BZ), respectively. The values of retention ratio(R) and thermal diffusion coefficient($D_T$) are measured with change of molecular weight and composition of mixed solvents. Atempts are then made to correlate the measured values with various physicochemical parameters of polymers and solvents. Studies suggest that R is significantly increased with the density of solvent and a good correlation is found between them. $D_T$ values decreases in the mixed solvent having has a higher concentration of poor solvent.

  • Analytical method using the reversed phase ion-pair chromatography (RP-IPC) for the determination of kasugamycin(5-amino-2-methyl-6-(2,3,4,5,6-pentahydroxy cyclohexyloxy)tetrahydropyran-3-yl-amino-${\alpha}$-imino acetic acid), pesticide as fungicide bactercide has been established. The retention behavior of kasugamycin in the RP-IPC was examined with respect to the effect of concentrations of organic modifiers, pH of eluent and types and concentrations of the counter ions as ion-pair reagent. This method developed by the optimum factors, can be used for the application of the quality control in the crude product and its formulation.

  • Dihexyloctanamide(DHOA) was used as an extractant or phase modifier with the diamide extractants in a solvent extraction process for a radioactive liquid waste treatment. The degradation compounds of the DHOA extractant, irradiated with $^{60}Co$ gamma ray, were octanoic acid and dihexylamine which are identified by a Fourier transform infrared(FT-IR) and gas chromatograph/mass spectrometer(GC/MS) analysis, and determined by the GC/MS with selected ion monitoring(SIM) mode. Retention behavior of octanoic acid, tridecane (internal standard) and dihexylamine in total ion chromatogram (TIC) were 8.65 min., 9.79 min., and 10.27 min., respectively. With increasing the absorbed dose of the $\gamma$-ray irradiated DHOA, the concentration of octanoic acid was decreased and that of dihexylamine was increased.
