Isolation of Sorbic Acid from the Mountain Ash Berries (I) (국산(國産) 건조(乾燥)마가목 열매로 부터 Sorbic Acid의 분리(分離)에 관한 연구(硏究) (I))

  • This investigation was primarily carried out to establish the optimum conditions for the separation of sorbic acid, as an enzyme inhibitor, from dry mountain ash berries. The berries were crushed to $35{\sim}40\;mesh$, and leached with water to produce a reddish juice, which was concentrated to syrup after evaporation. Parasorbic acid was separated from the syrup by steam distillation in the presence of acid. In this study, the optimum experimental results for separation of parasorbic acid were obtained as follows : (1) the most applicable leaching time of the dry ash berries was about 6 hours at room temperature and less than 4 hours at $40^{\circ}C$ and (2) in steam distillation, after removal of malic acid, addition of 30 ml of sulfuric acid per 50 ml of syrup extracted from 100 g of the ash berries was very adequate. The purity of sorbic acid obtained experimentally through the isomerization technique was qualitatively examined by spectrophotometeric, paper chromatographic, and melting point measurements.

  • $5{\sim}6$에 향기 짙은 순백색 꽃이 소박하고 탐스럽게 피었다가 10월에 붉은색으로 단장하는 열매는 마치 나무의 정수를 탄생시킨 양 매우 열정적이고, 고귀한 아름다움을 간직하고 있다. 또한 겨울에 하얀 설경을 배경으로 달려있는 열매는 관상적 가치는 물론 생명의 강인함을 넘어 우리 인간들에게 무엇이라 꾸짖는 듯한 외침이 서려있는 도전적 자태에 우리의 마음을 빼앗기는 아름다움으로 세파에 찌들인 도시인들에게 조경수 및 관상수 인기가 급상승하고 있는 수종이다. 특히 마가목은 한자명으로 馬牙木(마아목)이라고 불리는데 이것은 봄에 생겨나는 새순이 말의 이빨처럼 힘차게 돋아난다고 해서 동적인 감정과 함께 용기와 생동감을 주는 나무이다. 특히 공해에 강한 수종으로 도시에서 쉽게 접할 수 있는 장점을 가지고 있다. 일반적으로 병해충에 강하다고 알려져 있으나 몇몇 해충은 관리를 소홀히 하면 피해를 볼 수 있고 심하면 고사하므로 각별한 주의가 요망된다.

Variation of Fruit and Seed Morphology of 6 Natural Populations of Sorbus commixta Hedl. in Korea (마가목 6개 천연집단의 열매와 종자 형질 변이)

  • This study was conducted to investigate the variation of fruit and seed morphology among populations and among individuals within population of Sorbus commixta Hedl. distributed in Korea. Fruits collected from 42 trees in six natural populations and their six fruit and four seed characteristics were analyzed. In all characteristics, there were significant differences among populations and among individuals within populations. Especially, number of fruits per fruit-bearing branch and number of seeds per fruit showed higher values among populations in total variance component. Coefficients of variation in the number of fruits per fruit-bearing branch and seed weight are relatively high (42.0~75.3%) compared to other traits (11.9~32.1%). As a result of simple correlation analysis, the number of fruits per fruit-bearing branch showed a significant positive correlation with latitude but showed a negative correlation with longitude. According to cluster analysis, geographically close populations showed the tendency of clustering into the same group. Three principal components (PC) were deduced from principal component analysis, which explain the 87% of total variance of fruit and seed characteristics. The highest contribution was seed length and seed weight in PC1, fruit width and seed index in PC2 and fruit length and number of fruits per fruit-bearing branch in PC3.

Anticancer Effect of Sorbus commixta Hedl Extracts (마가목의 항암활성탐색)

  • We prepared extracts from bark, leaf and fruit of Sorbus commixta Hedl using by water and ethanol. To test for mutagenicity and antimutagenicity on extracts, we used Rec assay and Ames test. The result of Rec assay, the extracts of the Sorbus commixta Hedl did not show a DNA-damage activity. The results of Ames test were provided that extracts of Sorbus commixta Hedl showed $43{\sim}73\;%$ antimutagenic activities. In the inhibition ratio of the growth of several human cancer cells (A549, HepG2, MCF7), 91% of MCF7 cell growth, 94% of A549 cell growth and 91% HepG2 cell growth were inhibited by adding 1mg/ml of fruit ethanol extracts, bark ethanol extracts and fruit ethanol extracts, respectively. There was a little cytotoxicity on human normal hephatocyte (WRL68), extracts(1mg/ml) showed over the 70% survival.

  • This study was carried out to investigate the effect of Sorbus commixta Hedl. fruit extracts on the lipid metabolism in rats. Experimental groups were divided into 3 groups; Sprague-dawley rats were fed either diets containing 1% of water extracts(WE group), 1% methanol extracts(MeOH group) and diets without extracts(Control group) during 4 weeks. Food intake, weight gain and liver weight were not shown in meaning full difference among the groups. The concentration of serum triglyceride and total cholesterol were significantly decreased in WE and MeOH group compared to control group. Serum HDL-cholesterol, phospholipids and glucose contents were slightly lower in WE and MeOH group than control group. The levels of liver triglyceride was significantly decreased in MeOH group compared to control group. The levels of total cholesterol and phospholipids in liver were significantly lower in WE and MeOH group than control group.

Quality Characteristics of Fruit Spirits from a Copper Distillation Apparatus (동증류기를 이용한 과실증류주의 품질 특성)

  • The objective of this study was to investigate the quality characteristics of spirits derived from fruit using copper distillation equipment. First, optimal yeast strains were chosen through a fermentation test on raw materials (apples, mandarins and rowanberries). The normal fermentation condition for rowanberries observed at a rowanberry concentration of 8% during mashing included chaptalization with sugar to increase the alcohol content. During the mashing, fruits were fermented and distilled through one of three different types of distillation apparatuses (pot distiller, vacuum distiller, or multistage distiller made of copper). The results revealed that the type of apparatus used affects the content of alcohol and flavor components. The distilled spirits prepared through a copper multistage distiller had a much higher tendency to retain components of fruit aromas such as ethyl acetate and provided higher yields than spirits prepared with a pot distiller or vacuum distiller. Thus, the copper multistage distiller apparatus can be applied to positively influence the taste and flavor of fruit distilled spirits by enhancing fruit aromas, removing impurities such as sulfur components and enhancing yields.

Effect of Cryptochlorogenic Acid Extracted from Fruits of Sorbus commixta on Osteoblast Differentiation (마가목 열매에서 추출한 Cryptochlorogenic Acid 처리에 의한 조골세포 분화 촉진 효능)

  • Chlorogenic acid, a well-known polyphenol, and its derivatives, ester of caffeic acid on quinic acid moiety, are abundant in coffee, tea, fruits, and various vegetables. This study examined the effects of cryptochlorogenic acid (CCA) on osteoblast differentiation. CCA-induced mRNA expression levels of osteogenic genes in MC3T3E1 and C3H10T1/2 cells were determined by RT-PCR and qPCR. CCA regulated expression of key osteogenic genes in the early stage of differentiation, including distal-less homeobox 5 (Dlx5), DNA-binding protein inhibitor (Id1), and runt-related transcription factor 2 (Runx2). These results suggest that CCA may enhance osteoblast differentiation through expression of osteogenic genes such as Id1, Dlx5, and Runx2, especially in the early stage.

A Study on Perception of Alcoholic Beverage among Women and Development of Anti-Oxidant Beer Natural Ingredients (국내 성인 여성의 음주 인식과 형태 및 천연 소재를 활용한 항산화 맥주 개발에 관한 연구)

  • For the study, 366 copies of questionnaire were distributed among and collected from Korean women in their 20s to 60s between April and May 2016. To investigate the utility of anti-oxidant beers that contain extracts of natural substance respectively, the anti-oxidantive activity were measured and preference for anti-oxidant craft beers was surveyed among the subjects. The result suggested that the subjects had positive perception toward drinking, Korean adult women highly prefer beer among different types of alcoholic beverage, and the form of drinking changed according to the stamina, mood, situation, place, and company. Also, natural substance extracts showed high levels of antioxidant effects and, in the survey on preference for craft beers, the aronia group scored higher than other groups. The findings in this study suggest, for Korean adult women, beer with anti-oxidant ingredients can be beneficial for skin and overall health. Also, the findings can be used as basic data related to drinking among adult women and development of anti-oxidant beer. The Journal of Digital Policy & Management. Chlorogenic Acid Isomers from Sorbus commixta of Ulleung Island Origin and Their Inhibitory Effects against Advanced Glycation End Product (AGE) Formation and Radical Scavenging Activity (울릉 마가목의 클로로겐산 이성체의 최종당화산물의 생성 저해 및 라디칼 소거 활성)

  • Advanced glycation end product (AGE) formation and reactive oxygen species are potential therapeutic targets for the prevention of diabetic nephropathy and other pathogenic complications. Activity-guided isolation of an ethylacetate-soluble portion of 80% methanolic extract from fruits of Sorbus commixta of the Ulleung Island origin using AGE formation inhibition assay led to the isolation and identification of three caffeoylquinic acid derivatives of a previously known structure, 3-O-caffeoylquinic acid (neochlorogenic acid; 1), 4-O-caffeoylquinic acid (cryptochlorogenic acid; 2), and 5-O-caffeoylquinic acid (chlorogenic acid; 3). The structures of these compounds were confirmed by interpretation of nuclear magnetic resonance and mass spectrometry data. Among the isolates, the major metabolite, neochlorogenic acid (1) showed the most potent inhibitory effect against AGE formation with an $IC_{50}$ value of $167.5{\pm}3.5{\mu}M$. Furthermore, all isolated chlorogenic acid isomers were evaluated for their radical scavenging activity against peroxynitrite, and structurally related isomers 1, 2, and 3 exhibited potent inhibitory effects in this radical scavenging assay. This result suggests that the monocaffeoyl quinic acid derivatives isolated from S. commixta might be beneficial for the regulation of diabetic complications and related diseases.

Anti-coagulation and Anti-platelet Aggregation Activity of the Mature Fruit of Sorbus commixta (성숙 마가자의 혈액 응고저해 및 혈소판 응집저해 활성)

  • The mature fruit of Sorbus commixta is known as magaja in Korea and is consumed in the form of tea and wine. In addition, it has been used to treat hypertension and articular neuralgia in folk medicine. In this study, the ethanol extract of magaja and its subsequent organic solvent fractions were prepared, and their in vitro anti-coagulation, and platelet aggregation inhibitory activities were evaluated. Among the fractions, the ethylacetate fraction demonstrated significant inhibition against thrombin, prothrombin, blood coagulation factors and platelet aggregation, without hemolysis activity up to 0.5 mg/ml. Our results suggest that the ethylacetate fraction of magaja has the potential to be used as a new anti-coagulation agent.
