Effect of Eucommia ulmoides Extracts on Allergic Contact Dermatitis and Oxidative Damage Induced by Repeat Elicitation of DNCB (두충 추출물이 DNCB로 유발된 알레르기성 접촉피부염과 산화적 손상에 미치는 영향)

  • Inhibitory effects of allergic contact dermatitis of hot water extract of Duchung (Eucommia ulmoides Oliver) leaf, bark and stem growing at Sancheong-gun were investigated for female BALB/c mouse induced by repeat elicitation of DNCB (2,4-dinitro-chlorobenzene). Skin reactions, consisting of increased ear thickness and the presence of ear inflammation, were observed in mice treated with DNCB and Duchung. Weight of lymph node, spleen and thymus in mice treated with Duchung extracts were lower than that of mouse treated with DNCB. Ear weight of mouse treated with Duchung extracts was decreased by increasing the concentration of sample as compared to control group and dropped as low as control level at 1,000 mg/kg. Ear thickness became thinner as test time on Duchung extract progressed. MDA (malondialdehyde) contents in liver tissue were not different in sample group with DNCB group, but were different in ear tissue. NO (nitric oxide) contents was decreased in Duchung extract groups at serum and ear tissues as compared to 1% DNCB group. In the present study, the results suggested that Duchung extract inhibits inflammatory response and oxidative damage induced DNCB allergen.

Improvement Effect of the Eucommia ulmoides Extracts on CIA-induced Rheumatoid Arthritis Animal Models (두충 추출물의 류마티스관절염 동물모델에 대한 개선 효과)

  • The present study aimed to evaluate the effect of Eucommia ulmoides extracts on rheumatoid arthritis biomarker in a CIA-induced DBA/1 mice. For evaluation, Eucommia ulmoides extracts was administered orally at dose of 100 mg/kg/day for 4 weeks after production of an animal model of rheumatoid arthritis and we confirmed the treatments' effects based on serum biomarker, radiological, structural parameter analysis. Compared to the negative control group, the Eucommia ulmoides extracts treatments significantly reduced the serum level of inflammation and immunoglobulin markers (i.e., TNF-α, IgG, and hs-CRP), and significantly decreased the monocyte count of white blood cells. Furthermore, the Eucommia ulmoides extracts treatments effectively preserved the joint destruction, and little the joint deformation. Moreover, compared to the negative control group, the Eucommia ulmoides extracts treatments increased the bone volume, and significantly decreased bone inflammation. The results indicate that the Eucommia ulmoides extracts improved rhrumatoid arthritis symptoms. Thus, the Eucommia ulmoides extracts may be a novel therapeutic option for the management of rheumatoid arthritis.

Antioxidative activities of ethanolic extracts of Du-zhong (Eucommia ulmoides Oliver) leaf and bark (두충나무(Eucommia ulmoides Oliver) 잎과 껍질의 에탄올 추출물의 항산화 활성)

  • Antioxidative activities of 50% ethanolic extracts from Du-zhong (Eucommia ulmoides Oliver) leaf and bark were investigated. Yields of the leaf and bark extract were 8.1±0.31 and 17.4±0.89%, respectively. Polyphenol contents of the leaf and bark extract were 64.1±3.35 and 42.4±2.38 ㎍ gallic acid equivalents/mg, respectively. Flavonoid contents of the leaf and bark extract were 24.0±3.15 and 36.7±3.18 ㎍ quercetin equivalents/mg, respectively. As concentration of the leaf and bark extract increased, their antioxidative activities proportionally increased. EC50 values of the leaf and bark extract for cation radical scavenging were 560.6±17.65 and 1,357.4±8.45 ㎍/mL, respectively. EC50 values of the leaf and bark extract for free radical scavenging were 574.2±14.70 and 2,103.1±108.59 ㎍/mL, respectively. EC50 values of the leaf and bark extract for ferric reducing antioxidant power were 319.9±13.42 and 705.9±26.08 ㎍/mL, respectively. EC50 values of the leaf and bark extract for nitrite scavenging were 2,329.2±35.11 and 5,467.6±243.92 ㎍/mL, respectively. In the presence of 74.8 ㎍/mL of the leaf extract and 177.2 ㎍/mL of the bark extract, linoleic acid peroxidation was inhibited by 70.0 and 79.1%, respectively. The Du-zhong leaf extract possessed higher antioxidative activities than its bark extract.

The Aroma Components of Duchung Tea and Persimmon Leaf Tea (두충차와 감잎차의 향기성분)

  • The aroma components of duchung tea and persimmon leaf tea were collected simultaneous distillation and extraction method. Those were analyzed and identified by GC-MS using a fused sillica capillary column. Seventy seven components, including 17 alcohols, 3 hydrocarbons, 15 ketones, 16 aldehydes, 8 esters,8 acids, 8 heterocyclic compounds and 2 phenols were confirmed in duchung tea. Seventy one components, including 17 alcohols, 10 hydrocabrons, 11 ketones, 13 aldehydes, 8 esters, 6 acids, 5 heterocyclic compounds and 1 phenol were confirmed in persimmon leaf tea. The most abundant components of duchung tea were aldehydes(23.31%) including 2-pentenal and heterocyclic compounds(16.71%) including $epoxy-{\beta}-ionone$. The most abundant components of persimmon leaf tea were alcohols(25.57%) including linalool and aldehydes(19.45%) including hexanal and hydrocarbons(10.40%) including ${\alpha}-copaene$.

Effects of Eucommia ulmoides olivon Ethanol Extract on Lipid Metabolism and Antioxidant Enzyme Activities of Rats Fed High Fat Diet (고지방 식이를 투여한 흰쥐의 지질대사와 항산화 효소 활성에 미치는 두충 에탄올 추출물의 영향)

  • To investigate the effects of Ewomia ulmoides olivon extract on the serum and hepatic lipid status and anti-oxidant enzyme activities, male Sprague Dawley (SD) rats were given high fat diets and Eucomia ulmoides olivon ethanol extracts for 6 weeks. Rats were divided into (our groups consisting the control (C), Eucomia ulmoides oliuon extract fed group (CE), high fat diet group (CL) and the extract and high fat fed group (CLE). Eucomia ulmoides oliuon ethanol extract lowered total cholesterol, but increased HDL-cholesterol and HDL-cholesterol/total cholesterol of the serum compared to the control. On the other hand, the ratio of HDL-cholesterol/total cho-lcsterol (HTR) was increased by 55% (p<0.05). High fat diet significantly increased serum cholesterol contents, but decreased HDL-cholesterol level and HTR (p<0.05). Atherogenic index (AI) of CL group was increased by almost four times of the control (p<0.05). Triglycerides (TG) and phospholipids were also increased by high fat diet. Eucomia ulmoides oliuon ethanol extract decreased the levels of TG (p<0.05) and phospholipids (p<0.05). Although liver antioxidant engyme activities including glutathione sulfur transferase (GST), superoxide dismutase (SOD) and catalase were decreased by high fat diet, those were stimulated by the administration of Eucommia ulmoides olivon ethanol extract.

Effects of Feeding Eucommia ulmoides Leaves Substituted for Rice Straw on Growth Performance, Carcass Characteristics and Fatty Acid Composition of Muscle Tissues of Hanwoo Steers (볏짚을 두충잎으로 대체급여 시 거세한우의 생산성, 도체특성 및 육의 지방산 조성에 미치는 영향)

  • This study was conducted to examine the growth performance, carcass characteristics, physico-chemical properties and fatty acid composition of muscle tissues of Hanwoo steers when they were fed diets containing four levels of E. ulmoides leaves. Steers were allotted to one of four dietary treatments, which were designed to progressively substitute Eucommia ulmoides for 0, 3, 5 and 10% of the rice straw in the basal diet. Seventy two Hanwoo steers (321±13kg) were used. Average daily gain (0.81-0.86kg) and feed conversion (10.05-10.59) were not changed by feeding E. ulmoides leaves. Emission of fecal ammonia gas was decreased by increasing substitution levels of E. ulmoides leaves. Emission of ammonia gas in feces of steers were significantly (p<0.05) decreased in the 5% and 10% treatments at 40℃ compared with in control (30.6ppm) and 3% treatment (29.8ppm), respectively 20.3 and 21.6ppm. Back fat thickness was higher (p<0.05) in steers fed control diet (15.0mm) than 5 and 10% E. ulmoides diets (10.2 and 10.5mm respectively). The grade ‘A’ appearances of meat yield of steers were increased up to 20% by the 3% substitution of E. ulmoides leaves, and the grades ‘1+’ and ‘1’ appearances of meat quality were significantly improved (p<0.05) by the 5 and 10% Eucommia ulmoides feeding. There was no statistical difference in meat color of loin and top round among all treatments. It appeared that the 5% and 10% E. ulmoides leaves feeding affected (p<0.05) a increase in oleic acid concentration in loin and top round muscles. The concentration of serum cholesterol was lower (p<0.05) in steers fed 10% E. ulmoides (53.3mg/100g) than that of the other treatments (55.7-57.0mg/100g). The diarrhea incidence was about 20.26% in control, while it was lower (p<0.05) in the 5 and 10% E. ulmoides leaves treatments (10.4-12.5%) than control. There was no statistical difference in the occurrences of respiratory disease among all treatments. It is concluded that E. ulmoides leaves should be a prospective feed additive because it contains various functional substances. It is likely that air-dried E. ulmoides leaves can reduce the back fat thickness of Hanwoo steers and the occurrences of diarrhea. Therefore, the 5 and 10% substitution of E. ulmoides for roughage are highly recommended to be used in practice.

Characterization and Transdermal Delivery of Ethosomes Loaded with Eucommia ulmoides Extract (두충 추출물을 담지한 에토좀의 특성 및 경피 전달)

  • In this study, ethosomes loaded with Eucommia ulmoides extracts that have anti-inflammatory and anti-aging activities on the skin were prepared as cosmetic materials. The particle size, physical property, and entrapment efficiency of ethosomes to identify their stabilities during 4 weeks were measured. Ethosomes containing 0.01~0.05% E. ulmoides extracts did not show either precipitation or separation. The mean particle size of these ethosomes was 136.4 nm, and monodisperse. The particle size of ethosome containing 0.05% E. ulmoides extracts was 151.15 nm and the entrapment efficiency was 81.79%. Therefore, in vitro skin permeation experiment was performed with ethosome containing 0.05% E. ulmoides extracts that showed the high loading efficiency and stability during 4 weeks. The order of skin permeation of E. ulmoides extracts was ethosome (77.16%) > liposome (62.80%) > ethanolic solutoin (41.59%). These results suggest that ethosomes containing 0.05% E. ulmoides extracts are effective for the skin permeation and may be used as a cosmetic formulation containing plant extracts.

Effects of the Dietary Mixture of Eucommia ulmoides Oliver on Osteoporosis (두충[Eucommia ulmoides Oliver] 조성물이 골다공증에 미치는 효과)

  • Inhibitory effects of Eucommia ulmoides Oliver against osteoporosis were studied with ovaryectomized rats. The diet mixture A of calcium, vitamin D$_3$, and vitamin K$_1$and the additional mixture B with extract of Eucommia ulmoides Oliver were fed for 7 months to ovaryectomized rats. The urine content of pyridinoline and deoxypyridinoline was used as bone biochemical markers. Concentrations of deoxypyridinoline of ovaryectomized rats fed with the mixture A and B decreased 9.0% and 26.9%, respectively, when they were compared to the ovaryectomized control. The trabecular bone areas of proximal tibias of ovaryectomized rats were measured and compared to the ovaryectomized control. The areas fed with the mixture A and B increased 9.9% and 42.0%, respectively, that proved the inhibitory effect of Eucommia ulmoides Oliver against osteoporosis.

Antioxidative Properties and Whitening Effects of the Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma (두충, 단삼, 진피 및 천궁의 항산화 활성 및 미백 효과)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Eucommiae Cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma were carried out. Comparison of the four ethanol extracts revealed that, Aurantii nobilis pericarpium had the highest electron-donating ability(79.0%),; however, Salviae miltiorrhizae radix had the highest SOD-like ability(21.9%). The xanthine oxidase experiment exhibited a hindrance effect of 79.3% in Salviae miltiorrhizae radix, 57.5% in Eucommiae cortex and 71.9% in Aurantii nobilis pericarpium. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 12.4% in the Eucommiae cortex, 22.8% in the Salviae miltiorrhizae radix, 27.5% in the Aurantii nobilis pericarpium and 59.5% in the Cnidii rhizoma. Based on these results, we suggest that the ethanol extracts of Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma. can be used as food and cosmetic ingredients.

Sensory Evaluation and Electronic Nose Analysis for the Development of Mixed Eucommia ulmoides Leaf Tea (두충혼합차 개발을 위한 관능검사 및 전자코 분석)

  • The leaves of Duchung(Eucommia ulmoides), an oriental medicinal plant, have a peculiar aroma of Chinese medicine and astringent taste, which make the consumer be reluctant to Duchung leaf tea. Therefore, the purpose of this study was to develop a mild flavored Duchung leaf tea by mixing with other plants. The flavor patterns of developed tea were analyzed using an electronic nose. Polygonatum odoratum and Elsholtzia splendens were used for improving the flavor of Duchung leaf tea. The addition of 20, 30 and 40% of Polygonatum odoratum improved the overall acceptance in hedonic sensory evaluation. The flavor pattern of the tea was described by principal component analysis(PCA) and the resistance ratio(R/cub gas/R$\_$air/) of sensors. The PCA plot was also used to explain the mild flavor of the tea, which was extended from the right side(positive value of the first principal component) to the left side(negative value). Analysis by using an electronic nose with metal oxide sensors could be applied to detect whether mixed Duchung leaf tea was acceptable or not.
