The extrusion-cooking condition on Cordyceps pruinosa was designed using twin-screw extruder. Response surface methodology (RSM) was used to investigate extrusion-cooking using a central composition design with varying die temperature $(114-146^{\circ}C)$, feed moisture $(22-38\%)$, feed rate (4-14 ka/h) and screw speed (120-280 rpm). System parameters (die pressure and specific mechanical energy (SME)) and extrudate parameters (density and water solubility index (WSI)) were statically analyzed using RSH. Die pressure was significantly affected by temperature, moisture contents and feed rate. SM was affected by screw speed and feed rate. When die temperature is $130^{\circ}C$ and moisture content $25\%$, the optimum pressure is shown. SME is about 20 Wh/kg, when feed rate is $10\~12kg/min$ and screw speed $200\~250rpm$. WSI was affected by temperature and moisture contents. Density was not affected by any factor. WSI increases by $7\%$ from about $23\%$ to about $30\%$, as temperature is raised from $120^{\circ}C\;to\;140^{\circ}C$. The WSI of Cordyceps pruinosa pulverized after extruding (PE) is about $26.97\%$ higher than that of raw material and $10\%$ higher than that of pulverized after drying (PD). The content of unsaturated fatty acid were not significantly different in PD and PE. Anti-oxidative activity of PE was 1.67-2.2 times higher than that of PD in Cordyceps pruinosa using 1- dipheny1-2-picrylhydrazyl method (DPPH).
Objectives: This study was performed to evaluate effects of Cordyceps militaris(CM) on osteoclast differentiation and its related gene expression. Methods: We used mouse myeloid cells RAW 264.7 stimulated by receptor activator of nuclear factor kappa-B ligand(RANKL) to induce osteoclast differentiation. There are four groups of which RAW 264.7 cells are not stimulated by RANKL (Normal), stimulated by RANKL without CM(Control), stimulated by RANKL with 0.1 ${\mu}g/ml$ of CM(CM 0.1), stimulated by RANKL with 1 ${\mu}g/ml$ of CM(CM 1). Osteoclastogenesis was measured by counting Tartrate-resistant acid phosphatase-positive multinucleated cells [TRAP(+) MNC]. RT-PCR was performed to evaluate the inhibitory effect of CM on gene expression(TRAP, AKT1, JNK1, NFATc1, c-Fos, MITF). Results: 1. CM decreased the number of TRAP(+) osteoclast in RANKL-stimulated RAW 264.7 cell at the concentration of 0.1 ${\mu}g/ml$ and 1 ${\mu}g/ml$. 2. CM decreased the expression of TRAP in osteoclast at the concentration of 1 ${\mu}g/ml$. 3. CM decreased the expression of AKT1, JNK1 in osteoclast at the concentration of 1 ${\mu}g/ml$. 4. CM didn't affect the expression of NFATc1, c-Fos, MITF in osteoclast. Conclusions: Cordyceps militaris has inhibitory effects on osteoclast differentiation and its related gene expression. These results suggest that Cordyceps militaris has a potential as a treatment of osteoporosis.
Objective : This study was undertaken to determine whether Cordyceps sinensis Sacc. (CSS) extract exerts the protective effect against oxidant-induced alterations in membrane transport function in renal tubules. Methods : Membrane transport fucntion was estimated by examining alterations in p-aminohippurate (PAH) uptake in rabbit renal cortical slices. For induction oxidative stress, slices were treated with an organic peroxide cumene hydroperoxide for 60 min at $37^{\circ}C$. Cumene hydroperoxide inhibited PAH uptake in a time dependent manner. Results : CSS at 0.5-5% concentrations prevented cumene hydroperoxide-induced inhibition of PAH uptake. CSS at 1% also attenuated LDH release and lipid peroxidation induced by cumene hydroperoxide. When slices were treated with 0.2 mM mercury chloride, PAH uptake was inhibited and lipid peroxidation was increased. These changes by mercury were significantly prevented by CSS. Conclusion : These results suggest that CSS prevents oxidant-induced alterations in membrane transport function in rabbit renal cortical slices. Such protective effect of CSS may be attributed to inhibition of peroxidation of membrane lipid.

This study was conducted to investigate the effects of the Cordyceps militaris extract on the improvement of the glucide metabolism in serum of streptozotocin (STZ, 50 mg/kg BW, IP injection)-induced diabetic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of creatinine, lipid peroxide and NEFA in sera were significantly higher in the STZ-induced diabetic group (group DMG) and STZ+Cordyceps militaris extract administration group (group DMS) than those in the control group (group BD). However, the concentrations of creatinine, lipid peroxide and NEFA in sera were reduced in the group DMS than those in the group DMG. The activity of creatine phosphokinase (CPK) in serum was lower in the group DMS than in the group DMG. The activity of LCAT in serum was increased in the group DMS (Cordyceps militaris extract administration) than in the group DMG. The results indicate that Cordyceps militaris extract were effective in the improvement of the glucide metabolism in the sera of STZ-induced diabetic rats.

Antioxidant activity of extracts from the submerged-liquid culture of mushrooms was measured using two systems : linoleic acid and mouse liver microsomes induced by various free radical sources. Mushrooms of Pleurotus ostreatus (Neutari), Phellinus linteus (Sanghwang), Paecilomyces japonicus (Dongchunghacho), Hericicum erinacium (Norugungdengyee) and Agaricus blazei (Shinryeong) in 1% mulberry tree powder-supplemented medium were incubated in a shaking incubator (200 rpm, $25^{\circ}C$) for 3 days. Hot water extracts of mycelial cultures were freeze-dried, followed by fractioning with hexane, chloroform, ethylacetate and butanol in the order. Antioxidant activity of each sample was examined in free radical-induced linoleic acid oxidation in phosphate-buffered saline (PBS ) solution by measuring the amount of malonaldehyde (MA), and mouse liver microsomal systems by measuring the amount of thiobarbituric acid reactive substances (TBARS). In linoleic acid oxidation system, hot water extracts from the cultures of Pleurotus ostreatus, Phellinus linteus, and Paecilomyces japonicus exhibited stronger antioxidant activity than aqueous or butanol fraction and the combined fraction of hexane, chloroform and ethylacetate, but the hot water extract from Pleurotus ostreatus culture was the strongest activity. The antioxidant activity of the hot water extract from Pleurotus ostreatus culture was stronger than any other fractions in mouse microsomal system. These results suggest that hot water extract of Pleurotus ostreatus culture, and the cultures of Phellinus linteus and Paecilomyces japonicus could be useful for functional materials to reduce the oxidation of lipids in food systems induced by free radicals.
The fallowing are the results of the experimental studies of Cordyceps militaris (CM) on the penile nitric oxide synthase (NOS) activity and the level of blood testosterone in hydrocortisone acetate-treated rats. CM was tested for the effects on activity of xanthine oxidase and lipid peroxidation in penis of hydrocortisone acetate-treated rats. In vitro, CM didn't effect the levels of lipid peroxide and the activity of NOS. In the penis of hydrocortisone acetate-treated rats, lipid peroxide, the activities and ratio of type conversion of xanthine oxidase were increased but activity of NOS and content of nitrite were decreased. In vivo, after administration of CM to hydrocortisone acetate-treated rats, levels of lipid peroxide in penis was decreased. Also, the activities and ratio of type conversion of xanthine oxidase were decreased, too. The body weight and concentration of testosterone in the blood were increased. The effects of Cordyceps militalis Broth did better than the effects of Cordyceps militalls Mycelia, These results suggest that CM decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide and increase the penile NOS activity and the level of blood testosterone in hydrocortisone acetate-treated rats. Conclusively, CM is capable of improving of sexual ability in hydrocortisone acetate-treated rats.
Effect of single intraperitoneal administration of Cordyceps sinensis (Cs) extract at 24 hour before whole-body ${\gamma}$ - irradiation on the survival ratio, body weight, organ weight changes and serum metabolites in the irradiated mice were investigated. The single pre-administration of Cs extract increased the 40-day survival ratio of irradiated mice from 66.7% to 83.4%. The administration of Cs extract completely prevented weight reductions of spleen and thymus produced by ${\gamma}$ - irradiation (P<0.05, P<0.01). Similar but somewhat less radioprotective effect was also found in the testis of the Cs treated mice. The administration of Cs inhibited the serum hyperglycemia produced by irradiation on the day 7th(P<0.01). However, it did not influence the serum cholesterol and protein levels on the days examined. The present study is the first report regarding Cs which was tested and found to be radioprotective.
One hundred and six Cordyceps cultures including five cultures of Paecilomyces tenuipes were used for production of artificial fruiting body. In the test of artificial fruiting body formation, no fruiting bodies were induced on media containing PDA and ground silkworm pupae with liquid nitrogen. The best fruiting body formation was showed on media which mixed at the ratio of 1 unsticky rice to 3.5 water. But fruiting bodies formed on media mixed at the ratio 1 unpolished rice to 2.5 water. Optimal temperature in inducing artificial fruiting body was at $20^{\circ}C$. Twenty seven isolates were selected as good cultures for production of artificial fruiting body. Maturation of fruiting bodies incubated on rice grain media was completed for about 50 to 65 days.
This experiment was carried out to study formation of fruitbody with Cordyceps scarabaeicola (EFCC C-252) isolate. This isolate was the one of best fruitbody formation on brown rice 60 g plus 30 g silkworm pupa media among EFCC C-251, EFCC C-252, EFCC C-1092 from EFCC (Entomopathogenic Fungal Culture Collection) of Kangwon National University. Fruiting body was formed only isolate EFCC C-252 among tested isolates on the medium of brown rice (60 g) and silkworm pupae (30 g). The optimal temperature and light for fruiting body formation were $25^{\circ}C$ and fluorescent light (300 lux). The maximal fruiting body formation was observed at 70 g of brown rice and 80 g of silkworm pupa medium which was treated separately. Fruiting body was formed maximally by 2 days interval of irrigation.
The isolates of Cordyceps militaris preserved in EFCC, Kangwon National University were investigated to form the fruiting bodies under artificial conditions. The fruiting bodies were observed to be better in the 1l polyethylene bottle containing $60{\sim}80\;gm$ of brown rice and $100{\sim}110\;ml$ of water. Addition of $10{\sim}20\;gm$ of pupae per bottle showed higher fruiting. Similarly, addition of sucrose, peptone or hemoglobin also had favorable effect on fruiting. $25^{\circ}C\;and\;20^{\circ}C$ were favorable for mycelial growth and fruiting respectively. Light intensity of 500 lux and 12 h of light/dark period produced highest amount of fruit bodies.
