In the present study, Cordyceps sinensis extract (CSE) was evaluated to determine if it could protect pancreatic ${\beta}$ cells against cytokine-induced cytotoxicity of RINm5F cells. Treatment of cells with cytokines resulted in a decrease of viability, which was caused by increase of nitric oxide (NO) production. CSE protected cytokine-mediated viability reduction in a concentration-dependent manner. Incubation with CSE also induced a significant suppression of cytokine-induced inducible nitric oxide synthase (iNOS) protein and NO production. The molecular mechanism by which CSE inhibited iNOS protein expression appeared to involve the inhibition of $NF-{\kappa}B$ activation. The cytokine-stimulated RIN cells showed increases in $NF-{\kappa}B$ binding activity compared to unstimulated cells. However, pretreatment with CSE inhibited cytokines-induced $NF-{\kappa}B$ activation in RINm5F cells.
The effects of fruiting body of Cordyceps militaris on the growth, the food intake, the food efficiency ratio, the lipid metabolism, the serum protein level ad enzyme activity in male rats were studied. Sprague-Dawley rats were fed four types of diets for five weeks, respectiely: a control diet, a control diet supplemented with 2%, 3% or 4% fruiting body of Cordyceps militaris(CF) powder. In rats fed 2% or 3% CF diets the body weight gain, the food intake, the concentratons of hepatic triglyceride and serum LDL-cholesterol, the atherogenic index, and the total lipid, total cholesterol, triglyceride and phospholipid in serum were similar to those in rats fed the control diet. Whereas, in 4% CF diet these were significantly decrased. But the all CF diets feeding could not decrease the food efficiency ratio, the weights of liver, pancreas, spleen, kidney and heart, and the concentration of serum HDL-cholesterol. Also, it was shown that the concentrations of glucose, total protein, albumin, urea and creatinine, and the GOT, GPT, LDH, ${\gamma}$-GTP and ALP activities were the same levels in serum of rats of fed all the experimental diets.
Cordyceps militaris has shown to have various pharmacological activities including an immune-modulatory effect. Previously, we reported that anti-influenza effect of C. militaris in DBA/2 mice was mediated by increased IL-12 and the activation of NK cells. In this study, we investigated the effect of C. militaris on adaptive immune responses using DBA2 mice immunized with influenza vaccine. To determine the effect of C. militaris on antigen presentation capability, we treated RAW 264.7 cells with various concentrations of ethanol extract of C. militaris, which showed a significant upregulation of CD86 (B7.2), CD284 (TLR4), CD40, H-2k (MHC I) and I-Ad (MHC II). To examine the direct effect of C. militaris on adaptive immune responses, we immunized DBA2 mice with influenza vaccine in presence or absence of C. militaris. After 2 or 4 weeks, influenza-specific T cell proliferation, HAI titers and IFN-${\gamma}$ production were measured in plasma or PBMCs isolated from animals. Influenza-specific T cell proliferation and HAI titers were not considerably increased in immunized mice in presence of C. militaris. However, the production of IFN-${\gamma}$ was much greater in immunized mice with C. militaris as adjuvant than only immunized mice.
Anti-diabetic effect of the exopolysaccharides (EPS) produced from submerged mycelial culture of Cordyceps sinensis (Cs) was studiedin a type II diabetic animal model (C57BL/6J ob/ob). This study was designed to determine whether Cs-EPS improves clinical symptoms of type II diabetes in ob/ob mice. After Cs-EPS treatment at doses of 200 mg/kg body weight, the fasting blood glucose levels decreased by 47% after 7 weeks compared with those of the control mice. According to the oral glucose tolerance test, the glucose levels recovered its baseline after 120 min in Cs-EPS-treated mice, although the blood glucose levels increased significantly after 30 min. On the other hand, the control group (not-treated) did not recovered its initial level of glucose after 120 min. Furthermore, food intake, body weight, total plasma cholesterol and triglyceride concentrations in ob/ob mice treated with Cs-EPS were significantly decreased, compared with those in control ob/ob mice. Cs-EPS treatment increased significantly the plasma insulin level and the expression of leptin mRNA in adipose tissue of Cs-EPS-treated ob/ob mice. From these results, it is demonstrated that Cs-EPS could be effective for regulating normal blood glucose levels by increasing the amounts of plasma insulin and leptin expression in ob/ob mice, indicating that this compound could be a candidate material as a dietary supplement to control hyperglycemia in patients suffering from type II diabetes.
The quality characteristics of breads with different concentrations(2%, 4%) of Paecilomyces japonica (J2, J4) and Cordyceps militaris (M2, M4) powder were compared in terms if sensory and mechanical properties. In the sensory test, J2 and M2 groups gained good scores in taste, texture and odor acceptance as control grourp. Paecilomyces japonica-added group showed higher moisture and bigger volume than that of control group, and Cordyceps militaris group showed lower moisture content and decreasing volume. 'L' values of J and M groups was lower than control, and the longer the storage period, the lower the lightness. The more Paecilomyces japonica was added, the higher the 'a' value was, but Cordyceps militaris showed an opposite trend. The hardness of the bread was the lowest in J2 group and the highest in M4 group, and the longer tile storage period. the higher the hardness was (p<.001). Overall, the addition of 2% Paecilomyces japonica showed a similar tendency with control group, and showed good scores especially in acceptance, moisture and volume. Paecilomyces japonica appeared to be more desirable than Cordyceps militaris.
Changes in the Qualify Properties of Doenjang Prepared using the Powder or extracts of Paecilomyces japonica were investigated during fermentation at 20$^{\circ}C$. The moisture content was increased during fermentation but was not significantly different in all treatments. The pH of Doenjang with p. japonica was lower than the control group and decreased with the fermentation time. The amino nitrogen content increased gradually for up to 60 days and decreased slightly at 90 days. The L, a and b value decreased in proportion to the fermentation period and those of Doenjang with P. japonica powder were the lowest. From the results of the sensory evaluation, the color of the control group was similar to "yellow" but that of the Doenjang made from powder of P. japonica was close to "dark brown" and those of the Doenjang made from the P. japonica extract were darker than that of the control group. The texture was "glossy and smooth" in all cases and preference about the texture was high. The Doenjang with added P. japonica Powder had a saltier taste and the Doenjang with P. japonica Powder had the least sweet taste. In the flavor and overall Preference, the Doenjang with P. japonica powder was the lowest.
Objectives : This study was performed to investigate the effects of Cordyceps Sinensis (CS) and Cordyceps Militaris (CM) on anti oxidation in the livers of ${CCl_4}-treated$ rats. Methods : Hepatotoxicity in rats was induced by carbon tetracWoride. $CCl_4-induced$ rats were administered with the extract of CS and CM. Results : In vitro, CS and CM didn't affect levels of lipid peroxide and the activities and type conversion ratio of xanthine oxidase. However, hydroxyl radicals and DPPHL radicals were decreased. In vivo, in the ${CCl_4}-treated$ rats, lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were increased but superoxide dismutase was decreased. After CS and CM were administered to ${CCl_4}-treated$ rats, levels of lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were decreased but superoxide dismutase was increased. Conclusions : These results suggest that CS and CM decrease the activities of free-radical-generating enzymes which form lipid peroxide and increase the activities of oxygen free radical scavenging enzymes.

This study was carried to investigate the antidiabetic metabolism effect of water extract $Cordyceps$$Militalis$(C.M.) in Streptozotocin(STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg,b.w. dissolved in citrate buffer. The water extract of C.M. was orally administrated once a day for 7 days at a dose of 500mg/kg,b.w(body weight). or 1,000mg/kg.b.w. The content serum glucose was significantly decreased in C.M. treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in C.M. treated group compared to the those of STZ-control group. These results indicated that water extract of C.M. would have antidiabetic metabolism effect in STZ-induced diabetic rats.
Cordyceps militaris (CM) occurring from a fruiting body by a host insect is a kind of mushroom, which is composed of animal host and plant fruit body. CM contains large amounts of useful ingredients including polysaccharides, vitamins, amino acids, minerals, etc. The essential amino acids from CM including cystine, lysine, and methionine can be expected to improve damaged hair treatment as effective ingredients. In this study, the improvement effect of the CM extracts on chemical and physical properties for damaged-hair treatment was investigated. The cysteic acid and cystine monooxide produced by oxidation of cystine were analyzed their chemical structure by FT-IR spectroscopy. It was confirmed that the vibration absorption peak ($1,041cm^{-1}$) of cysteic acid was reduced and increased sulfur content considerably which means meaningful improvement effect on damaged-hair treatment. Apparently, the cuticle morphology of the damaged-hair was improved significantly by treatment with CM extracts. Especially, confocal laser scanning microscope images of the damaged-hair treated with the extract showed highly increased fluorescence intensity which means promising effect in hair treatment. The tensile strength of the damaged hair treated also increased by 168% compared with the damaged hair.

Paecilomyces sinclairii was administered ad libitum feeding at percentage levels of 0, 1.25, 2.5, 5 and 10 percentage (calculated about 8 g/kg)/feeder for a period of 3 months. There was no observed clinical signs or deaths related to treatment in all groups tested. Therefore, the approximate lethal dose of P. sinclairii was considered to be higher than 8 g/kg in rats. Mild decreases in body weight gain were observed dose-dependently in P. sinclairii treated groups in dose response manner after 2 weeks. Interestingly, the weight of abdominal adipose tissues surrounding epididymides were greatly reduced by this Dongchunghacho, in parallel with the mild increase in body weight gain. However, the absolute weight change of other organs was not observed. There were not significantly different from the control group in urinalysis, ocular examination, hematological, serum biochemical value and histopathological examination. From these results, it is concluded that the no-observed-adverse-effect level (NOAEL) of P. sinclairii is less than 1.25% (1 g/kg) in rats in the present study.
