Thirty three entomopathogenic species belonging to twelve genus were collected throughout 16 collecting sites from 1990 to 1996. Among those collected species, 14 unrecorded species such as Cordyceps bifusispora, C. martialis, C. oxycephala, C. paludosa, C. pentatomi, C. rosea, C. ryogamiensis, Shimizuomyces paradoxa, Akanthomyces aculeatus, Polycephalomyces ramosus, Tilachlidiopsis nigra were added to Korean entomopathogenic species through this study. In nature, occurrence of Cordyceps nutans, C. sphecocephala and Paecilomyces tenuipes were from early June to late September. On the other hand, C. militaris, C. kyushuensis and C. pruinosa were mainly found from mid July to mid August when relative humidity are increased. Nine species of the genus Cordyceps including C. bifusispora and four deuteromycetous species were isolated. As a result of cultural test using six Cordyceps species, anamorph of C. militaris, C. kyushuensis were proved as Verticillium sp. C. pruinosa as Acremonium sp., C. sphecocephala as Hymenostilbe sp. and C. scarabaeicola as Beauveria sp., respectively.

The effects of mycelium of Cordyceps militaris on the growth, the lipid metabolism, the serum protein levels and the enzyme activities in male rats were studied. Sprague-Dawley rats were given four different types of diets for a succeeding period of five weeks: either a control diet, a control diet supplemented with 2%, 3% or 4% mycelium of Cordyceps militaris (CM) powder. The body weight gain, hepatic weight, feed efficiency ratio and the feed intake of the rats given diets with 2%, 3% or 4% CM were similar to those in rats given the control diet. The concentrations of hepatic total lipid and triglyceride of rats fed the 3% or 4% CM diets were significantly lower than those of rats fed the control diet. But the concentrations of hepatic total cholesterol and phospholipid of rats fed the all CM diets were similar to those of rats fed the control diet. The concentrations of total lipid, total cholesterol, triglyceride phospholipid, and the atherogenic index, and the activities of glutamic oxaloacetic transaminase and lactic dehydrogenase in serum of rats fed the all CM diets were significantly lower than those of rats fed the control diet. No differences were noted in the concentrations of HDL-cholesterol, LDL-cholesterol, total protein, albumin and creatinine, and the activities of glutamic pyruvic transaminase, ${\gamma}$ -glutamyltranspeptidase and alkaline phosphatase in the serum among the rats with on all the experimental diets. These results showed that the all CM diets feeding decreased the total cholesterol, the triglyceride, the phopholipid, and the atherogenic index in serum of rats.
To classify fungal species employed for pharmacological effects, mycelial proteins of six isolates of Ganoderma lucidum, five isolates of Schizophyllum commune and five isolates of Cordyceps spp. were separated on polyacrylamide gel to compare them by esterase, acid phosphatase, leucine aminopeptidase and peroxidase patterns. Similarity of isozyme patterns among the isolates of G. lucidum IY003, IY004, IY005 and IY008 was indicated over 70%, but that among the isolates of G. lucidum IY009, IY010 and others was indicated from 48% to 9%. Highest similarity of isozymes of S. commune was observed to be between IY803 and IY805, and similarity between these two isolates was 57%. Similarity among other isolates was shown to be from 40% to 56%. Isozyme patterns of Cordyceps spp. were comparatively different, even though they were originated from the same kind of insect as their isolate. Similarity between Cordyceps spp. IY901 and IY904, which was isolated from moths, was 67% and that of IY905 and IY909, which was originated from the larvae, was 42%. Similarity among other isolates was shown to be from 12% to 67%.
Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) materials were extracted from Paecilomyces sinclairii. In vitro cytotoxicity tests. Fr. HW was not cytotoxic against MH3T3, Sarcoma 180 and HepG2 cancer cell lines at the concentration of $0{\sim}2,000\;{\mu}g/ml$, while HT-29 cell line was cytotoxic at the concentration of $100\;{\mu}g/ml$. Fr. NaCl and Fr. MeOH were slightly cytotoxic to the cell lines. Intraperitoneal injection with Fr. NaCl and Fr. MeOH exhibited antitumor activity with life prolongation effect of 32.3% in mice inoculated with Sarcoma 180. Fr. NaCl improved proliferation of spleen cells and the immunopotentiation activity of B lymphocyte by increasing spleen cell numbers and the alkaline phosphatase activity by $2.4{\sim}2.6\;and\;2.7{\sim}3.9$ folds, respectively. Fr. NaCl generated $89\;{\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7, a mouse macrophage cell line, at the concentration of $50\;{\mu}g/ml$, while iipopolysaccharide, a positive control, produced $79\;{\mu}M$. The Fr. NaCl showed the hightest antitumor effect and activation of B lymphocytes and macrophage. From these results, antitumor effect of P. sinclaitii was likely due to immunopotentiation activity.
Cordyceps militaris(CM) has been used as a traditional folk medicine to strengthen the immune system and regain energy for hundreds of years in Far East Asian countries. In the present studies, we determined that the stimulatory effect of Cordyceps militaris on testosterone production in male mouse rats. Results illustrated that changes of the body weight, food and water intake of the rats were not observed in this study but the concentration of testosterone in the serum of the rats was significantly increased by CM(p < 0.05). Therefore fruiting bodies of CM grown on the drone bee medium may be an integrative medicine for the treatment of reproductive problems caused by insufficient testosterone levels in human males.
This study was carried out to identify the genetic characteristics among isolates of Paecilomyces spp.and Cordyceps spp. by URP-PCR analysis. Twenty URP (universal rice primer) primers of 20 mer which were designed from repetitive sequence of rice, were used for producing PCR DNA fingerprints of the mushrooms. Of them, 5 URP primers, URP2F, URP2R, URP9F, URP4R, and URP17R amplified genomic DNA of the mushrooms with polymorphic PCR patterns. On isolates of Cordyceps militaris, primers URP1F, URP2R, URP6R and URP17R produced PCR polymorphic bands of 4 types. Isolates of Cordypces sp. that are isolated from different area of Korea were identical to isolate of C. militaris, while other species of Cordypces were different to the PCR profiles. However, the URP primers did not identify the polymorphism of PCR profile on isolates of P. japonica.
A infectious fungus (J131) recently isolated from artificial cultivation farms of Paecilomyces tenuipes is known to cause a great yield loss and deterioration of products. In this study, its occurrence and characterization were made. The isolate (J131) was identified as Paecilomyces farinosus (Holm ex S. F. Gray) Brown & Smith. Colonies of the isolate on PDA grew moderately fast, attaining a diameter of 19mm within 14 days at 24${\pm}$1$^{\circ}C$. Vegetative hyphae are smooth-walled and hyaline. Conidiophores mainly arose from the submerged mycelium, usually measuring 1.3-1.5$\mu\textrm{m}$ wide and consisted of verticillate branches with whorls of 2 to 4 phialides. Phialides of 3.8-5.8${\times}$1.5-1.8 $\mu\textrm{m}$, with a swollen basal portion tapered into a distinct neck of about 0.6-0.8 $\mu\textrm{m}$ wide. Conidia arisen from the phialides, were ellipsoidal to fusiform, smooth-walled, hyaline, 1.8-2.4${\times}$1.1-1.4$\mu\textrm{m}$. .When silkworm larvae were injected with concentration of 1${\times}$10$\^$8/ conidia/$m\ell$ of infectious fungus J131, infection rate was 41.3% and synnemata formation rate was 23.6%. Black spots were appeared on the surface of silkworm larvae in 5-9 days after injection. The surface of larvae was covered with hyphae which were gradually turned into yellow from white color during the incubation period of 3-4 days. 1 to several 4-6mm long synnemata of yellow or orange color were formed on the surface. The matured synnemata were ramified from its end and produced much conidia of white color.

To develop technique for the production of P. tenuipes stromata on a large scale, the infection of P. tenuipes and the growth of stroma were investigated by silkworm (Bombyx mori) variety. Also, studied about biological activites of fruiting body formed on silkworm. Infection rate of the 5th instar larvae of the silkworm with P. tenuipes was the highest in Yangwonjam, followed by Hachojam, Baegokjam and Chilbojam in that order. Also, as the inoculation times was increased, infection rate tended to be raised. The rate of fruiting body formation of the silkworm pupae infected with P. tenuipes was the highest in Baegokjam, followed by Yangwonjam and Chilbojam in the order. But, actually the fruiting body formation of the 5th instar larvae of the silkworm tested was good in Chilbojam, followed by Yangwonjam and Baegokjam in that order in 3 times spraying inoculation. The fruiting bodies of Yangwonjam and Chilbojam infected with P. tenuipes had high amount of Mannitol, but Baegokjam and Hachojam had high concentration of Glucose on a dry weight basis. The mean content of total amino acid in the fruiting bodies of P. tenuipes was 1.03 ${\mu}mole/g$. The distribution rate of amino acid components decreased in the order of Arginine (12.2%)>Glycine (10.5%)> Proline (9.6)>Tyrosine (8.9%)>Serine>Leucine>Threonine. The most abundant amino acid in the fruiting bodies of the Baegokjam, Chilbojam and Hachojam infected with P. tenuipes was arginine, while Yangwonjam was Glycine. The most abundant fatty acid in P.tenuipes was Oleic acid on a dry weight basis. The unsaturated fatty acids such as Oleic acid, Linoleic acid and Linolenic acid accounted for more than 78% of the total fatty acids.
Entomopathogenic fungus Cordyceps militaris is famous for its medicinal efficacies. It has been reported to have various pharmacological activities such as anti-tumour, insecticidal, antibacterial, immunomodulatory and antioxidant. In this study, we investigated the effect of the extract of C. militaris (MPUN8501), which was identified by the analysis of the nucleotide sequences of 5.8S ribosomal RNA, on the function of liver. C. militaris powder was extracted using hot water extracts method as time, volume and temperature and using method as differential polarity of organic solvent. Each fraction was tested for the improvement of hepatic enzyme alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. The BuOH extracts (CME) had highest activity which was used for the test of toxicity and efficacy of C. militaris. The enhancing effect of CME on the activity of ADH and ALDH was much more than medicine, drink, natural tea etc. Thus CME promoted the resolution of alcohol and acetaldehyde in rats, inducing recovery to normal condition rapidly. Furthermore, oral administration of CME effectively protected the carbon tetrachloride-induced acute hepatic injury as revealed by the hematological parameters (levels of sGOT and sGPT) and histological observation. CME was ascertained to be safe by regulatory toxicity studies of single dose toxicity and genotoxicity. These results suggest that CME would be useful for the maintaining normal hepatic activity as a functional health food.
The growth characteristics of Paecilomyces tenuipes according to the passage in the two kind of liquid media were investigated by comparing the mycelium and conidium formation degrees. The potato dextrose broth medium and the silkworm larvae medium containing the silkworm powder were used as the liquid media, and the potato dextrose agar medium and the brown rice medium containing the powder of silkworm pupa were used as the solid media. The conidium formation degree in liquid media differed by the passages but that in solid media was not. This suggested that the passage in liquid media did not affect significantly the conidium formation in solid media. When the brown rice media were inoculated with the concentration of $1{\times}10^{10}$ conidia/ml, $1{\times}10^8$ conidia/ml and $1{\times}10^6$ conidia/ml, respectively, the conidium formation degrees were similar. This indicated that the optimal inoculation concentration of conidium to the brown rice media is $1{\times}10^6$ conidia/ml.
