A Technique to Enhance Bacillus thuringiensis Spectrum and Control Efficacy Using Cry Toxin Mixture and Immunosuppressant (Cry 독소단백질 혼합과 면역억제제 첨가를 통한 Bacillus thuringiensis 살충제 적용범위 및 방제력 증진 기술)

  • An entomopathogenic bacterium, Bacillus thuringiensis (Bt), can sporulate along with production of insecticidal Cry toxins. Bt Cry toxins exhibit relatively narrow spectrum to target insects due to their specific interactions with midgut receptors. This study designed several strategies to enhance Bt efficacy in target insect spectrum and insecticidal activity. Four Cry toxins were purified from four different Bt strains and showed relatively narrow target insect spectrum. However, the Cry mixtures significantly expanded their target insect spectra. The additional effect of baculovirus to Cry toxin was tested with recombinant baculoviruses expressing Cry1Ac or Cry1Ca. However, the baculovirus was little effective to expand target insect spectrum. Bacterial culture broth of Xenorhabdus nematophila (Xn) significantly suppressed insect cellular immune response and increased Cry toxicity. The addition of Xn culture broth to Cry mixture significantly enhanced Bt efficacy in target insect spectrum and insecticidal activity.

The Cry mixtures significantly expanded their target insect spectra.   • Screening of Pseudomonas strains that can be used as hosts for expression of crystal protein gene of Bacillus thuringiensis subsp. kurstaki HD-73 was carried out. From rhizosphere soil of 7 kinds or crops as fluorescent Pseudomonas strains were isolated. A hybrie plasmid, pKTC1, composed of the broad host range vector pKT230 and the crystal protein gene was constructed and used for transformation of the 35 Pseudomonas strains. As the result, the crystal protein gene could be introduced into 4 isolates. Several methods including bioassay and immunochemical detection indicated that the crystall protein gene was expressed in the Pseudomonus isolates.

  • 살아있는 유기체에 의해 분비되는 독소는 대부분의 경우 인간에게 유해하다. 가령 여름철 날것이나 오래된 음식에서 쉽게 식중독에 걸릴 수 있는데, 이는 주로 Clorustidium Botulinum이 만들어낸 보툴리눔 독소가 원인이다. 유기체에 의해 생성된 모든 독소는 단백질이며 이는 아미노산 서열로 나타낼 수 있다. 이를 통해 생물정보학 분야의 많은 연구자들이 많은 머신러닝 기술을 통해 단백질의 독성을 예측할 수 있었다. 최근 몇 년 동안 SVM를 사용하는 BTXpred와 CNN을 사용하는 ToxDL과 같은 모델이 각각 박테리아와 동물 독소의 독성을 예측하기 위해 제안되었다. 시대가 변함에 따라 BERT와 같은 성능이 더욱 뛰어난 모델이 시퀀스 분류를 위해 도입되었다. 본 논문은 독성 단백질을 분류를 위해 ProtBERT를 사용할 경우 이의 성능을 보여주고자 한다.

  • A clone pSL 2-1, which is a recombinant plasmid believed to contain the mosquitocidal crystal-line protein gene of the Bacillus sphaericus 1593, was expressed in Escherichia coli JM83 and the product of the clone was purified and identified. The unsolubilized mosquitocidal crystal proteins from the B. sphaericus had formed 43, 58, 64, 100, 113, and 130 Kd bands in the SDS-polyacrylamide gel, but the NaOH-solublized proteins at pH 12 formed 2 protein bands of 43- and 64Kd in the gel because the larger protein (precursor) bands were cleaved. The products of the pSL 2-1 clone was purified by Sephadex G-200 and only the fractions having lethal activity to the 3rd in-star larvae of mosquito Culex pipiens were analyzed by the gel. The only single protein band of 42 Kd toxic to the larvae was formed. The major toxic protein being produced from the B. sphaericus 1593 and the pSL 2-1 clone was found to be the 42 Kd.

  • In the studies of characterization of crystal prototoxin isolated from Bacillus thuringinensis var. alesti and its bioassay to the silkworm larvae, Bombyx mori L., the following results were obtained. 1. The complete lysis of bacteria grown on the nutrient agar at 30$^{\circ}C$ in an incubator took 30 days, but the period could be reduced by a half in a devised broth media in a fermentation jar. 2. The protein toxin extracted directly from a mixture of crystals and spores without separation of crystals and spores was pure as same as the protein toxin extracted from only crysals separated. 3. By SLS polyacrylamide gel electrophoresis of prototoxin released from crystals in alkali (pH 9.5) three different proteins in molecular weights of 120,000, 87,000, 74,000 were separated. 4. In the bioassay of the toxin to the silkworm larvae, LD$\sub$50/ per gram of the larvae in the 4th instar was 0.080 $\mu\textrm{g}$.

Proteome Profiling of Murine Macrophages Treated with the Anthrax Lethal Toxin (탄저 치사독소 처리에 의한 생쥐 대식세포의 단백질체 발현 양상 분석)

  • Intoxication of murine macrophages (RAW 264.7) with the anthrax lethal toxin (LeTx 100 ng/ml) results in profound alterations in the host cell gene expression. The role of LeTx in mediating these effects is unknown, largely due to the difficulty in identifying and assigning function to individual proteins. In this study, we have used two-dimensional polyacrylamide gel electrophoresis to analyze the protein profile of murine macrophages treated with the LeTx, and have coupled this to protein identification using MALDI-TOF mass spectrometry. Interpretation of the peptide mass fingerprint data has relied primarily on the ProFound database. Among the differentially expressed spots, cleaved mitogen-activated protein kinase kinase (Mek1) and glucose-6-phosphate dehydrogenase were increased in the LeTx treated macrophages. Mek1 acts as a negative element in the signal transduction pathway, and G6PD plays the role for the protection of the cells from the hyper-production of active oxygen. Our results suggest that this proteomic approach is a useful tool to study protein expression in intoxicated macrophages and will contribute to the identification of a putative substrate for LeTx.

  • To describe characteristics of a hemolysin in mosquitocidal Bacillus thuringiensis subsp. gyangiensis strain 21-2, Escherichia coli HB101 was transformed with a gene encoding hemolysin in the strain 21-2. Transformant 47 con-tained 2.5 kb DNA was selected by ELISA, immunoblot and DNA electrophoresis. Transformant 47-5 was recon-structed after digestion of the 2.5 kb DNA with Hind m. Transformant 47-5 contained 1.8 kb DNA and expressed 23 kDa Protein which had mosquitocidal activity to Aedes aegypti. The 23 kDa Protein itself in vitro didn't show hemolytic activity on human erythrocytes, but the protein had the activity after proteinase K treatment.

천연 독소로부터 세포독성물질의 분리 및 정제

  • 코브라과 (Elapidae)에 속하는 뱀에는 극소량으로도 치사시킬수 있는 독을 가지고 있는데 독을 구성하고 있는 물질중 특정한 계열의 단백질이 매우 두드러진 약리학적 활성을 보여줄수 있다. 따라서 본 연구는 독사들의 혈액독소 및 신경독소를 우리나라에서 제일 종류가 많은 위암세포주(SNU-1)에 대해서 세포독성을 검색하고 곤충 및 해양생물의 독소에 대해서도 위암세포주를 이용하여 검색하였다. 검색 결과 king cobra계통인 Ophiophagus hannah의 분획은 SDS-PAGE에서 분자량이 66,000 부근에 나타난 것이 세포독성물질로 추측되며 더욱 정제하여 아미노산 서열을 비교하고 Phospholipase화성을 측정함으로써 세포독성물질의 규명이 가능하리라 생각된다.

  • The prolein components of each crystal toxin of Bacillus thuringiensis kurstaki and alzawai were separated by SDS-PAGE. The major clqistal proteins from both shins were composed of paiypeptides having molecular weights of 130 kd and 64 kd. Digestive mixture of both. toxins with t~ypsin and gut juices shared 62 kd polypeptide which may be major actwe toxh. However, aizawal produced much less amount of 62 kd polypeptide than kurstaki did. On ingestion with Bt &-endotoxin, larvae of Hyphantria cunea developed 45 kd stress protein in the several tissues including fat body, which was induced by heat and cold shock.
