The aim of this study was to evaluate the effects of three different mulberry cultivars extracts on serum lipid contents in ovariectomized estrogen-deficient rats. Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with $ 80\% $ ethyl alcohol extracts from various kinds of Tajikistan mulberry (OVX-TM), Korea mulberry (OVX-KM) and China mulberry (OVX-CM) at 200mg/kg bw/day, respectively. The mulberry extracts were orally administrated at 1mL per day. The body weights of OVX rats were significantly heavier than the sham-operated rats at all values (p <0.05). The ovariectomy caused an expected increasing in the levels of serum total cholesterol and triglyceride. The serum HDL-cholesterol level on the OVX-TM, OVX-KM and OVX-CM groups were higher than in the OVX-control group.
The ovarian hormone-deficiency induced ovariectomy rat is widely used as an aging model due to its practicality, convenience, and cost effectiveness. The surgically ovariectomized rat induces reactive oxygen species (ROS) generation like aging phenomena. Free oxygen radicals have been proposed as important causative agents of aging. The purpose of this study was to investigate the effect of chondroitin sulfate (CS) to prevent ovariectomy (OVX)-induced oxidative stress. The OVX rats were given intraperitoneally CS at doses of 100 mg/kg and 200 mg/kg daily for fifteen weeks. Malondialdehyde (MDA) levels were determined as well as the activities of superoxide dismutase (SOD), catalase (CAT), reduced-glutathione (GSH), oxidized-glutathione (GSSG), glutathione peroxidase (GPx) in the liver. The liver antioxidative enzyme activity was elevated while MDA concentration decreased in all CS treated animals. The results demonstrated that CS reduced oxidative stress in a dose dependent manner. These results suggest that CS might be a useful candidate for antioxidative reagent.

Menopause is often associated with the incidence of several chronic diseases including osteoporosis, cardiovascular disease, and obesity. The purpose of this study was designed to evaluate the possibility of osteoporosis prevention in postmenopausal women. In this study, we investigated the effects of Ecklonia stolonifera (ES) extracts on bone turnover markers in ovariectomized rats. For this study, the following four groups of 9-week-old Sprague-Dawley rats were evaluated over 6 weeks: normal rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with ES extracts. We measured the osteocalcin and C-telopeptide of collagen cross-links (CTx) content, enzyme ALP activity in serum and collagen content in the cartilage, bone, skin and lungs. We found that the levels of indicators of bone metabolism such as alkaline phosphatase (ALP), osteocalcin and CTx were lower in rats in the ES extract group than the OVX-CON group. In addition, the collagen contents in the bone, cartilage, skin and lungs decreased in response to ovariectomy, but the levels of collagen were greater in the bone of rats that were treated with ES extract than in the bone of rats in the OVX-CON group. These results suggest that the ES may be an effective functional food to prevent osteoporosis in postmenopausal women.

This study was performed to elucidate the effects of dietary calcium-salt, estrogen-treatment, and estrogen/calcium treatment on bone metabolism. Ovariectomized rats were used as an animal model. Female Sprague-Dawley rats with a body weight of $250{\sim}280g$ underwent ovariectomy or sham-operation. The ovariectomized rats were divided into 9 different experimental groups including saline-treated group, estrogen-treated group, high calcium salt-treated group, and estrogen/calcium treated groups, and fed the experimental diet for 6 weeks. The mineral content, weight, length, strength and density in femur and scapula of the animals were determined. The results of the experiment are as follows: there were no difference in weight of wet bone, density, length, strength and content of ash in right femur between ovariectomized rat and sham-operation. When the weight of bones was expressed as bone gram per body weight, it was significantly lower in ovariectomized rat than sham-operation. Estrogen and estrogen/calcium in ovariectomized rats resulted in increased weight of wet bone. Estrogen with its gradual reduction in ovariectomized rats showed the lowest values in strength, content of ash, content of calcium and content of phosphorus among the groups. There were no differences in weight of wet hone, density, content of ash, content of calcium and content of phosphorus in right scapula between ovariectomized rat and sham-operation. In addition skeletal composition was not changed by ovariectomy. Estrogen and estrogen/calcium in ovariectomized rats resulted in decreased skeletal composition however, estrogen with its gradual reduction did not caused the skeletal composition change. Moreover, the gradual reduction of estrogen had a preventive effect on bone loss when the treatment was combined with calcium intensification.
This study was conducted to evaluate suppressive effects of black rice aleurone layer extract (BRE) on body fat, serum lipid, and hormone levels in ovariectomized rats. The rats were divided into five groups (n=8/group): sham operated group (S); ovariectomized control group (OVX); ovariectomized group treated with isoflavone 10 mg/kg B.W. (IF); ovariectomized group treated with BRE 30 or 90mg/kg B.W. (BRE-30 or -90). High fat diet (45% calories from fat) was fed to all rats for 12 weeks. Body weight gain, body fat weight, and adiposity index increased in the OVX group, but they significantly decreased in IF or BRE supplementation. Serum triacylglycerol and leptin levels decreased in BRE groups while serum adiponectin level significantly increased compared to that of the OVX group. Hepatic total lipid, triglyceride, and cholesterol levels increased in the OVX group, but they decreased in BRE supplementation (p>0.05). These data suggest that black rice aleurone layer may be a useful food source to decrease obesity and its related diseases by modulating lipid metabolism in estrogen-deficiency model.
The present experiment was performed to study whether or not ovariectomized female rats can be used as an animal model to investigate the effects of estrogen on body compositions. Six-month-old, female rats were ovariectomized and the changes in body compositions were evaluated 6 months after surgery. As summarized in Fig. 2, body fat accretion was double in ovariectomized rats compared to shamoperated rats. Muscle weights were not d!fferent, while bone weights were lower in ovx rats than in sham rats when the values were expressed as a per\ulcornercentage of body weight. Results of the present study suggest that these ovariectomized rats can be utilized as an animal model to study the body compositions of postmenopausal obese women and related health problems.

The aim of this study was to evaluate the effects of Sedum sarmentosum Bunge (SS) on the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with SS at 50 mg/kg BW/day (OVX-SS50), 100 mg/kg bw/day (OVX-SS100) and 200 mg/kg bw/day (OVX-SS200), and an ethyl ether fraction of SS at 10 mg/kg bw/day (OVX-EE) and an ethyl acetate fraction of SS at 10 mg/kg bw/day (OVX-EA). The SS ethanol extracts and SS fractions were orally administrated 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the SS extracts tended to gain weight less than OVX-control. Supplementation with the SS extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of SS. Therefore, it may be used to possibly improve the quality of life in menopausal women.

The purpose of this study was to investigate the effects of Porphyra tenera (PT) extracts on formation of collagen cross-link in ovariectomized rats. From day 3 until 42 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with PT at 50 mg/kg bw/day (OVX-PT5O), 200 mg/kgbw/day (OVX-PT200). The PT ethanol extracts were orally administrated 1 mL per day. Body weight gain, food intake and food efficiency ratio were significantly different among the groups. The change of collagen content was studied in lung, bone, cartilage and skin of ovariectomized rats. The effects of PT extracts on the amount of collagen were examined by measuring the hydroxyproline, which is a specific amino acid existing in collagen. Pyridinoline is pyridinium cross-link formed in the mature form of collagen from lysine and hydroxylysine residues. Pyridinoline content was analyzed by HPLC. Pyridinoline content in bone collagen was decreased by ovariectomy but supplementation with the PT extracts was similarly increased to Sham. These results suggest that the PT supplementation could decrease bone loss in postmenopausal women.

In this study, the effects of Colpomenia sinuosa (CS) extract on serum lipid level and bone formation in an ovariectomized animal model were investigated. Twenty four nine-week old female Sprague-Dawley rats were randomly assigned to four groups: sham-operated rats (SHAM), ovariectomized control rats (OVX-CON), and ovariectomized rats supplemented with CS extract at 50 mg/kg bw (OVX-CS50) or 200 mg/kg bw (OVX-CS200). Three OVX groups were surgically ovariectomized while the SHAM group was sham-operated. CS extract was orally administrated at 1 mL per day. Analysis of serum lipid contents found that the total cholesterol, triglyceride, and low density lipoprotein cholesterol levels in the OVX-CON group were higher than those in the SHAM group. Notably, upon administration of CS extract after ovariectomy, triglyceride levels tended to significantly decrease. In addition, platelet aggregation ability decreased in groups treated with CS extract compared to the OVX-CON group. Serum alkaline phosphatase activity as an indicator of bone formation was lower in the CS extract group compared to the OVX-CON group. Collagen contents in bone and cartilage were reduced by ovariectomy, whereas the CS extract-supplemented groups exhibited higher concentrations in bone. According to these results, CS extract improved serum lipid parameters and osteogenesis in ovariectomized rats.
