Objective : Yukgunja-tang is one of the herbal prescriptions widely used for functional indigestion. In this study, we evaluated the pharmacological effect through the Yukgunja-tang formulation development. Methods : The RAW 264.7 cells were pretreated with Yukgunja-tang tablet (YGJT-T : 50, 100 and 200 ㎍/㎖) and then stimulated with lipopolysaccharide (LPS : 500 ng/㎖). Cell viability, inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2) and inflammatory cytokines (IL-1β, IL-6, and TNF-α) were measured. Also, ICR mice induced acute gastritis by oral administration of 150 mM HCl in 60% ethanol. The YGJT-T (30 mg/kg) was pretreated for 3 days, and 150 mM HCl in 60% ethanol was orally administered 1 hour after the last drug treatment. Mice were sacrificed 1 hour after oral administration of 150 mM HCl in 60% ethanol. The gastric mucosa was observed, and inflammatory cytokines in the gastric tissue were measured. Results : The marker components of YGJT-T were determined by simultaneous analysis using HPLC. In RAW 264.7 cells, pretreatment of YGJT-T was non-toxic and inhibited the production of inflammatory mediators such as NO and PGE2 and suppressed inflammatory cytokines. In addition, pretreatment of YGJT-T improved bleeding and edema due to gastric lesions caused by acute gastritis and suppressed inflammatory cytokines. Conclusion : In summary, our results confirmed that treatment with YGJT-T has anti-inflammatory and anti-gastritis effects in vitro and in vivo. Therefore, in this study, YGJT-T could support a potential strategy for the prevention and treatment of gastritis.
Objectives: Ephedra sinica and Wolbigachul-tang which contains Ephedra sinica are used to treat obesity in Korean medicine. The aim of this study was to analyze the quantities of ephedrine alkaloids by high-performance liquid chromatography. Methods: The analysis was performed using a YMC-Triat C18 column with operating at 25℃, and UV detection at 210 nm. The mobile phase used a gradient flow with 0.1% H3PO4 in water and acetonitrile. Specificity, linearity, precision, accuracy, limit of detection, and limit of quantification were measured for validation anaylsis. This method was applied to analyze the quantities of ephedrine alkaloids in Ephedra sinica and Wolbigachul-tang. Results: The concentration per Ephedra sinica (gram) of ephedrine and pseudoephedrine in Ephedra sinica decoction are 4.74±0.22 mg and 2.19±0.10 mg, respectively and in Wolbigachul-tang decoction are 6.39±0.34 mg and 2.97±0.21 mg, respectively. The retention time of ephedrine was 23.6 min and that of pseudoephedrine was 25.8 min, and norephedrine and methylephedrine were not detected. Conclusions: In conclusion, analyzed the concentration of ephedrine alkaloids in Ephedra sinica and Wolbigachul-tang by the developed validation method.
Purpose: This study was conducted to assess public awareness and policy challenges faced by practicing nurses. Methods: After collecting nurse-related news articles published before and after 'the Thanks to You Challenge' campaign (between December 31, 2019, and July 15, 2020), keywords were extracted via preprocessing. A three-step method keyword analysis, latent Dirichlet allocation topic modeling, and keyword network analysis was used to examine the text and the structure of the selected news articles. Results: Top 30 keywords with similar occurrences were collected before and after the campaign. The five dominant topics before the campaign were: pandemic, infection of medical staff, local transmission, medical resources, and return of overseas Koreans. After the campaign, the topics 'infection of medical staff' and 'return of overseas Koreans' disappeared, but 'the Thanks to You Challenge' emerged as a dominant topic. A keyword network analysis revealed that the word of nurse was linked with keywords like thanks and campaign, through the word of sacrifice. These words formed interrelated domains of 'the Thanks to You Challenge' topic. Conclusion: The findings of this study can provide useful information for understanding various issues and social perspectives on COVID-19 nursing. The major themes of news reports lagged behind the real problems faced by nurses in COVID-19 crisis. While the press tends to focus on heroism and whole society, issues and policies mutually beneficial to public and nursing need to be further explored and enhanced by nurses.
Sogunjung-tang (SGJT) is traditional herbal prescriptions used to treat abdominal pain. In this study, we evaluated the effect on inflammation and gastritis through SGJT formulation development. SGJT composition herbal medicine was boiled in water at 95~100℃ for 3 hours and then concentrated. Sogunjung-tang mix soft extract (STM) was prepared using pharmaceutical excipients such as purified water, sodium benzoate, β-cyclodextrin and CMC-Na. Anti-inflammatory experiment was conducted using STM and lipopolysaccharide (LPS) in RAW 264.7 cells. Cell survival was measured by MTT method. Nitric oxide (NO) was measured using griess reagents, and pro inflammatory cytokines were measured by enzyme-linked immunosorbent assay and RT-PCR. Also, we verified STM validity in acute gastritis using the ICR mouse. STM was administered oral for three days. And 150 mM HCl in 60% ethanol was oral administered 0.5 mL one hour after the last drug administration. Mice were sacrificed 1 hour after 150 mM HCl in 60% ethanol administration. The gastric mucosa was visually observed. STM were not toxic in RAW 264.7 cells. Treatment of STM inhibited the production of NO and inflammatory cytokine at the protein and mRNA levels. Also, in the acute gastritis model with the mouse, the treatment of STM improved gastric mucosal bleeding and edema. In summary, it was confirmed that the treatment of STM exerts anti-inflammatory and anti-gastritis effects. Therefore, we suggest that STM may provide a preclinical evidence for the prevention and treatment of inflammatory and gastritis diseases.
The aim of this study was to investigate the prevalence of antimicrobial resistance and virulence factors in staphylococci isolated from canine otitis externa. A total 295 causative microorganisms were isolated. The most common isolated species were Staphylococcus (S) pseudintermedius (94 isolates) followed by Pseudomonas aeruginosa (60 isolates), S. schleiferi (25 isolates), Escherichia coli (23 isolates) and Proteus mirabilis (20 isolates). Staphylococci isolates were showed high resistance to penicillin (78.6%), erythromycin (55.9%), tetracycline (52.4%), clindamycin (51.7%) and ciprofloxacin (42.8%). Of the 145 staphylococci isolates, 49 (33.8%) methicillin-resistant staphylococci (MRS) were observed, distributed among S. pseudintermedius (n=34), S. schleiferi (n=6), S. epidermis (n=4), S. hominis (n=2), S. aureus, S. caprae and S. saprophyticus (n=1, respectively). Forty-three (87.8%) of 49 MRS and 10 (10.4%) of 96 methicillin-susceptibility staphylococci harbored mecA gene. About 80% of MRS were multidrug-resistant with resistance to at least one antibiotic in three or more antibiotic classes. Resistance genes blaZ (93/114, 81.5%), ermB (35/81, 43.2%), ermC (3/81, 3.7%), aacA-aphD (50/54, 92.5%), tetM (69/76, 90.7%) and tetK (6/76, 7.8%) were detected among resistant isolates. Virulence factors genes lukF and lukS were found in 100%(145/145) and 43.4%(63/145), respectively. Genes encoding ermA, eta, etb and tsst were not detected. To the best of our knowledge, this is the first study which investigated for the presence of genes encoding antimicrobial resistance and staphylococcal toxins in staphylococci isolated from canine otitis externa. A continuous monitoring and surveillance program to prevent antimicrobial resistance in companion animals is demanded.
The author examined the origins, the course and the period of introduction of 94 types of vegetables and fruits mainly used in Korea. Then, based on it, the author looks into the relationship between food culture in Korea and those in the Silk Road. Among the vegetables and fruits, 57 types are not originated from Korea 17 types of stem and leaf vegetables, 9 types of root and fruit vegetables. 7 types of fruits, 6 types of seed, 6 types of pomes, 2 types of berries and grapes, and 1 type of nut. Their origins are spread in Europe, Southwest and South Asia but interestingly, they are located near or along the Silk Road. Therefore, it can be presumed that the vegetables and fruits were introduced into Korea from its neighboring countries by way of the Silk Road even before the Three Koreas Period and they were eaten widely in the Joseon Dynasty. Thus, the Silk Road helped some of the vegetables and fruits used in Korea to be introduced into Korea and eventually, contributed to diversifying Korean food culture. The cultural exchange is not one-way but two-way communication and the cultural exchange through the Silk Road is no exception. It is certain that by way of the Silk Road, foods and recipes were introduced into Korea from other countries but at the same time, Korean foods and recipes were propagated to other countries. In the future, more researches and studies should be conducted to find out how the foods and recipes are exchanged among the countries by the way of the Silk Road.
본 연구의 목적은 장애인보호작업장 종사자들이 직업에 대한 만족감을 느끼고 서비스의 효과성을 증진시킬 수 있도록 하는데 기초자료로 제공하고자 하였다. 이를 위해 전국 장애인보호작업장 종사자를 대상으로 설문조사를 실시하였다. 본 연구의 결과 첫째, 장애인보호작업장 종사자의 직무스트레스는 낮은 수준으로 나타나고 있으며 직무만족은 상대적으로 스트레스에 비해 높고 직무스트레스는 역할갈등이나 역할과다 요인이 심각한 수준이고 직무만족은 승진이나 보수요인에 낮게 나타났다. 둘째, 장애인보호작업장 종사자의 일반적 특성에 따른 직무스트레스는 경력요인이 많은 영향을 미치고 있다. 셋째, 장애인 보호작업장 종사자의 일반적 특성에 따른 직무만족은 연령의 경우에서만 차이가 있는 것으로 나타났다. 넷째, 직무스트레스와 직무만족도간의 상관관계는 모든 요인 간에 상관이 있는 것으로 나타나 직무스트레스가 높을수록 직무만족은 낮아지는 것으로 분석되었다. 따라서 직무만족을 높이기 위해 승진이나 보수수준에 대한 정책적인 개선과 종사자의 직무스트레스를 낮출 수 있도록 개인의 노력과 더불어 지속적인 연구가 필요하다.
STZ 투여로 제 1형 당뇨를 유발시킨 다음 B. sarmienti 에탄올 추출물을 8주간 급여 후 혈당 및 혈중지질 변화를 알아보았다. 시험기간 동안 당뇨대조군 및 당뇨실험군의 체중증가는 정상군과 비교하여 유의적으로 낮게 나타났으며, B. sarmienti 에탄올 추출물 급여가 당뇨실험군에서의 체중변화에는 영향을 미치지 않는 것으로 조사되었다. 또한 수분섭취량, 사료섭취량, 혈당 모두 정상군과 비교하여 당뇨대조군 및 당뇨실험군에서 8주 동안 유의적으로 높게 지속되었다. 혈중 총 콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤 변화를 측정한 결과 정상군과 비교할 때 당뇨대조군에서는 상대적으로 증가하였으나, B. sarmienti 에탄올 추출물 투여로 유의적인 감소현상과 함께 정상군과 유사한 수준을 유지 하였다. 특히, B. sarmienti 에탄올 추출물 투여로 인한 혈중 총 콜레스테롤 변화는 BE0 29.1%, BE1 26.1%, BE2 23.6%로 평균 26.2%의 감소효과를 나타내었다. 또한 혈중 중성지방 변화는 BE0 55.6%, BE1 64.1%, BE2 68.4%로 평균 62.7%의 감소현상을 나타내었다. 혈중 콜레스테롤 및 중성지방은 B. sarmienti 에탄올 추출물 투여의 농도 의존적 감소현상은 관찰할 수 없었으나, 혈중지질 개선에 많은 영향을 미치는 것으로 조사되었다. 본 연구에는 제 1형 당뇨모델에서 8주 동안 B. sarmienti 에탄올 추출물을 투여하면서 혈당조절 효과는 관찰할 수 없었으나, 혈중지질 성분인 triglycerides, 총 콜레스테롤 및 LDL 콜레스테롤의 수준을 떨어뜨리는 효과가 있는 것으로 조사되었다. 결론적으로 B. sarmienti 에탄올 추출물 투여가 고혈당을 중심으로한 지질대사 이상과 함께 많은 합병증 유발이 문제시 되고 있는 당뇨병의 체내 지질대사 개선에 효과가 있을 것으로 사료된다.
표고버섯으로부터 유래한 eritadenine (EA)은 혈중 low density lipoprotein (LDL), triglyceride (TG), phospholipid를 감소시키는 물질이다. 최근 저희 연구팀은 in vitro 연구에서 EA가 angiotensin converting enzyme (ACE)의 활성을 저해함을 밝혔다. 본 연구에서는 EA와 EA 함유한 표고버섯균사체 고체배양 효소추출물(EA-LEMCEE)이 본태성 고혈압 쥐 SHR의 혈압을 낮추었음을 보고한다. EA 및 EA-LEMSCEE는 인체 탯줄 혈관내피세포 EA.hy926 세포의 ACE 활성을 억제하였다. EA-LEMSCEE는 SHR의 수축기 및 이완기 혈압을 Control 처리구에 비해 유의성 있게 낮추어 처리 5주에는 Positive control인 captopril (CP) 처리와 비슷한 혈압감소 효과를 나타내었다. EA-LEMCE는 LEMSCEE에 비해 수축기 및 이완기 혈압을 감소시켰지만 유의성은 없었다. EA- LEMSCEE는 renin과 ACE 활성을 Control 처리에 비해 유의성 있게 억제하였고, angiotensin II (Ang II) 함량을 감소시켜, 처리 5주 후에 이러한 CP의 효과와 유사하였다. 또한 EA-LEMCEE는 LEMCEE에 비해 유의성 있게 renin 및 ACE 활성을 억제하였고 및 Ang II 함량을 감소시켰다. 이와 같은 결과는 EA 및 EA-LEMCEE는 본태성 고혈압 SHR의 renin과 ACE 활성을 억제하여 혈압을 낮추었고, EA 및 EA-LEMSCEE는 인체 고혈압을 낮출 수 있음을 의미한다.
본 연구의 대상은 제주도내에 거주하고 있는 만 20세 이상에서부터 만 60세 미만의 성인장애인 총 57명을 대상으로 설문조사를 실시하였으며, 장애인식개선교육강사 역할을 수행하고 있는 남성장애인 1명을 2015년 5월에 3회에 걸쳐 면접조사하였다. 연구결과 해석에 대한 보충자료로 사용하였다. 이러한 연구를 통해 얻어진 결론은 다음과 같다. 우선 장애인의 경제활동이 자기효능감에 미치는 관계를 알아본 결과, 장애인의 경제활동은 자기효능감 중 자신감에 유의미한 관계를 나타냈고, 자기조절효능감과 과제난이도 선호에는 유의미한 관계를 나타내지 않았다. 둘째, 장애인의 경제활동영역이 자기효능감의 하위요인에 미치는 차이를 알아본 결과에 대해서는 장애인의 경제활동여부와 마찬가지로 자신감에 유의미한 차이를 나타냈고, 자기조절효능감과 과제난이도 선호에는 유의미한 차이를 나타내지 않는 것으로 나타났다. 셋째, 장애인의 경제활동영역에 따른 자기효능감의 차이를 $Scheff{\acute{e}}$ 검증한 결과, 업체취업(M=23.48, p<.04)과 강사활동(M=22.06, p<.04)은 미취업(M=18.93, p<.04)보다 높지만 업체취업과 강사활동의 차이는 밝혀지지 않았다.
