Stenotrophomonas maltophilia BW-13 is a potent biocontrol agent to control crisphead lettuce bottom rot caused by Rhizoctonia solani. To define the optimum conditions for the mass production of the S. maltophilia BW-13, we have investigated optimum culture conditions and effects of various carbon sources on the bacterial growth. The optimum initial pH and temperature were determined as pH $6.0\~7.0 and $35^{\circ}C$, respectively. For the selection of effective carbon source for the mass production, we tested the low molecular carbon sources such as sucrose, glucose, lactose, maltose, manose and the high molecular carbon source such as dough conditioner, rice bran, corn starch, sweet potato starch. As the results, the addition of dough conditioner in a basal medium ($1.25\%\;K_{2}HPO_4,\;0.38\%\;KH_{2}PO_4,\;0.01\%\;MgSO_4{\cdot}7H_{2}O,\;0.5\%\;Yeast extract$) was able to achieve higher cell density and the antifungal activity than others. Therefore, the basal medium containing $3\%$ dough conditioner (named as dough conditioner medium) was finally selected the optimized media for the mass production of BW-13 strain.

Botrytis cinerea Pers. was found to be highly virulent to the grapevine plant, especially in greenhouse condition. Pseudomonas species play key roles for the biocontrol of many plant diseases especially in soil. Of the 83 isolates of Pseudomonas spp., a bacterial strain P84, isolated from tomato rhizosphere, was shown to suppress a wide range of phytopathogenic fungi in vitro. The isolate was identified as Pseudomonas putida on the basis of its bacteriological and genetic characteristics. The P. putida P84 strain carry the phlD gene for 2,4-diacetylphloroglucinol biosynthesis and may produce the antibiotics as an antagonistic mechanism involved in biocontrol. The antagonistic activity of the bacterium has a promising implication for its use as a biocontrol agent to control grapevine gray mold.
This study was carried out to clarify the effects of antifungal Streptomyces sp. isolated from the soil grown kiwifruit on the growth inhibition of fruit rot (Botryosphaeria dothidea) infected in kiwi fruit plants in the southwestern districts of Jeonnam. Two hundred and fifty microorganisms were isolated and examined into the antifungal activity against Botryosphaeria dothidea. We screened and isolated six bacterial strains which have a strong inhibition against Botryosphaeria dothidea. And the best antifungal strain designated as the strain #120 showing 96.0% antifungal activity against Botryosphaeria dothidea was finally selected. The strain #120 was identified as Streptomyces sp. #120 based on its morphological, physiological, biochemical and chemotaxonomic characteristics.

There are various crop diseases in green houses that are caused by the cultural environments, especially high temperature and moisture. To solve the forementioned problems, farmers are overusing agricultural chemicals, causing other damages by the chemical residue. In this study, antagonistic bacteria as biological control agents were isolated to produce the environmentally-friendly crops for use in green houses. Eighteen species of antagonistic bacteria were totally isolated from the soil and plants in the Perilla fields, and AK-17 showed the highest activity among the isolates. According to the results of anti-fungal spectrum against several pathogens by AK-17, the antagonism effect of the isolates was remarkable against grey mold rot by Botrytis cinerea, sclerotinia rot by Sclerotnia sclerotiorum, and stem rot by Rhizoctonia solini. To evaluate the biological control effects of the isolates against the major diseases of Perilla, studies were carried out to evaluate the preventive and the curative effects of the diseases throughout the pot experiments. According to the forementioned experiments, the preventive and the curative effects by the isolates against sclerotinia rot were respectively showed as 55% and 92%. For the grey mold rot, those were 40% and 78%, respectively. As to the evaluation of the growth-promoting effect by AK-17, the length and the biomass of the tested plants were increased to 120% and to 164%, respectively. For the leaf numbers and area were respectively increased to 120% and 220%. Furthermore, AK-17 was identified as Burkhoderia sp. according to the results of physiological properties and genetic methods.
In this study, we evaluated environmental-friendly disease control methods using essential oils, antagonistic bacteria, or cooking oil and egg yolk mixture (COY). Among the five tested plant essential oils, Eucalyptus approximans oil shown the highest effect to control of the downy mildew disease (60% disease control value). Interestingly enough, well known antagonistic bacteria against various fungal pathogens were appeared lower control efficiency than plant essential oils. In field conditions, the cooking oil and yolk mixture (COY) treatments shown significantly decreased the downy mildew incidence with 0.3% (disease conttrol value 84%) and 0.5% concentration (disease control value 91%). Our findings suggest that the COY has a great potential as the eco-friendly downy mildew disease control formulation.
A powerful antagonistic bacterium against Phytophthora capsici causing phytophthora blight of red pepper was isolated from the cultivated soil in Kyongju Korea, The bilogical control mechanisms of the isolated strain were caused by strong antifungal antibiotic, siderophore and cellulase. The strain was identified as Chryseomonas luteola by the cultural morphological and physiological characteristics. The opti- mal culture medium for the antibiotic production was determined as follows : 0.15%D(+) cellobiose, 0.55% $NH_4$CI, 0.01% KCI 0.7% $K_2$$HPO_4$ 0.2% $KH_2$PO$_4$ and 0.5% sodium citrate at pH 7.0 The optimal incubation time was 84 hours at $30^{\circ}C$ In pot bioassay, the treatment of C luteola 5042 protected red pepper plant against the blight of Phytophthora capsici.
This study was carried out to identity the effects of antifungal bacteria isolated from the soil grown kiwi fruit plants on the growth inhibition of Botrytis cinerea causing gray mold in kiwi fruit plants in the southern districts of Jeonnam. Antagonistic microorganism CHO 163 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. CHO 163 showed 86.9% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. CHO 163 showed almost all of antagonistic activity against Botrytis cinerea. And we also confirmed that in vitro the treatment of Bacillus sp. CHO 163 cultured by SD+B+P broth efficiently controled the growth of Botrytis cinerea causing gray mold in kiwi fruit plants.
In oder to select the powerful rhizophere-dorminatable biocontrol agent, we had isolated an indigenous antagonistic bacterium which produced antibiotic and siderophore from a disease suppressive local field soil of Gyungsan, Korea. And we could select the Pseudomosp. 4059 which can strongly antagonize against Fusarium oxysporum and Phytophthora capsici by two kinds of antifungal mechanism that can be caused by the antibiotic of Phenazin, a siderophore and a auxin like subThe selected strain was identified as Pseudomonas fluorescens (biotype A) 4059 by biochemical tests, API $\textregistered$ test, MicroLog TM system and 16S rDNA analysis. The selected antagonistic microorganism, Pseudomosp. 4059 had an antifungal mechanism of antifungal antibiotic and sidrophore. And we were confirmed the antagonistic activity of P fluorescens 4059 with in vitro antifungal test against Phytophthora capsici and in vivo by red-pepper.
Sclerotium rolfsii is a well known broad host range soil borne plant pathogenic fungus and caused serious damage to various vegetable crops. To develop an effective biological control agent for S. rolfsii, an isolate which showed strong inhibitory effect on the mycelial growth of S. rolfsii was selected among the antagonistic bacterial isolates collected from vinyl-house soil. The bacterial isolate was identified as Bacillus amyloliquefaciens KBC1009 based on the morphological, physiological characteristics and by 16S rRNA sequence analysis. The growth conditions for B. amyloliquefaciens KBC1009 were optimized in LB media(pH7) by culturing at 30℃ for 72 hrs. Glucose and yeast extract were confirmed as the best carbon and nitrogen sources, respectively. In order to test the inhibitory effect of B. amyloliquefaciens KBC1009 to stem rot of pepper, green house experiment was conducted. Drench of 1/500 diluted bacterial suspension of B. amyloliquefaciens KBC1009(5×108 cfu/ml) to each pepper plant 3 times with 10 days interval showed 66.7% control effectiveness. These results suggest that B. amyloliquefaciens KBC1009 is one of promising biocontrol agent to control stem rot caused by Sclerotium rolfsii.
Pseudomanas fluorescens was selected from mushroom and studied in both batch and continuous culture in order to find out optimum conditions for cultivation. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. Cells of P. fluorescens were grown well on medium containing 30g/L of glucose, whereas the growth was inhibited with the glucose concentration at higher than 30g/L. The highest value of specific growth rate and productivity were obtained when using 10g/L of yeast extract. Optimum concentrations of $NH_4Cl$ and $(NH_4)_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfur source was 1.0g/L. It was also found that the cell concentrations were at the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P.fluorescens at high initial dissolved oxygen (D.O) value led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 68 for the initial D.O value.
