Effects of Mesangi(Capsosiphon fulvecens) Powder on Lipid Metabolism in High Cholesterol Fed Rats (매생이가 고콜레스테롤 식이 투여 흰쥐의 지질대사에 미치는 영향)

  • This study was conducted to investigate the effects of Mesangi (Capsosiphon fulvescens, CF, a green alga) on lipid metabolism in rats, which was examined by analyzing the lipid composition in serum. Male Sprague-Dawley rats were assigned to one of three groups: the basal diet, high cholesterol, and high cholesterol supplemented with 5% dry Mesangi powder (CF-supplemented group). The body weight gains and food efficiency ratios of the rats fed the CF-supplemented diet were lower than those of the rats fed the basal diet. The levels of total lipid, total cholesterol, and LDL-cholesterol in serum were reduced in the CF-supplemented group as compared to the cholesterol group. However, the level of HDL-cholesterol in blood increased with the addition of CF to the diet. Furthermore, levels of total lipid and cholesterol of liver in experimental group fed CF were significantly lower than the cholesterol group. A decrease in leptin expression levels was observed in the CF-supplemented group as compared to the cholesterol group. These results suggest that the addition of CF in hypercholesterolemic rats has an effect on the improvement of serum and liver levels of cholesterol, which may be related to the regulation of the atherogenic index and lipid metabolism in rats fed CF.

Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats (Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향)

  • Sprague-Dawley male rats were fed the diet of p/s 4.0(soybean oil : I), p/s 0.08(Beef tallow : II) at the level of 15% fat until 8 weeks after weaning. I & II groups were divided into 4 sub-groups by diets with or without 0.3% butylated hydroxytoluene(BHT). 2-AAF was injected at the age of $5_{1/2}$, 6, $5_{1/2}$, 7 weeks. MFO system enzyme(cytochrome p-450, cytochrome p-450 reductase, cytochrome b5) activities and lipid peroxide were determined from isolated liver microsome. 2-AAF injected young rats had growth retardatiion. Lipid peroxide values were not influenced greatly by dietary fat, 2-AAF and BHT. Cytochrome p-450 contents were increased in I-BHT-AAF & II-AAF groups by 2-AAF and its contents were not affected by BHT. But cytochrome p-450 and cytochrome p-450 reductase were not increased in soybean oil diet ybean oil groups. Cytochrome b5 was not influenced by dietary fat, 2-AAF and BHT. Cytochrome p-450 and lipid peroxide, cytochrome p-450 reductase and cytochrome b5, which transfer to MFO system, appeared to have positive correlations(r=0.2474, r=0.2475, p<0.05) each other. This result suggests that MFO system metabolizing 2-AAF was influenced by dietary fats and BHT. 2-AAF induced growth retardation in young rats.

Effects of Capsosiphon fulvescens Extracts on Essential Amino Acids Absorption in Rats (매생이 열수추출물이 흰쥐의 필수아미노산 흡수에 미치는 영향)

  • The aim of this study was to examine the effects of Capsosiphon fulvescens extract (CFE) on levels of essential amino acids in serum and the rate of protein digestion. CFE contains approximately 68% carbohydrate as fiber, 17% crude ash, and 14% crude protein. Experimental rats were divided two groups, one group receiving a normal diet and the other a normal diet containing 5% CFE. To analyze the absorption of essential amino acids into the hepatic portal vein, blood from the hepatic portal vein was collected between 10 a.m. and 5 p.m. for 10 days. In vitro protein digestion was decreased in the group fed 5% CFE; these data indicated that CFE affected protease activity. We measured the absorption of essential amino acids in the serum of the hepatic portal vein, at 30-, 60-, 90-, and 120 minutes after feeding. Although there was no difference in the concentration of total essential amino acids between the two groups, the 5% CFE-fed animals had a decreased rate of absorption. Absorption of Lys and Thr into the hepatic portal vein was lower in the CFE-fed group than in the control group. The rate of absorption of Met was delayed nearly 50% in the CFE-fed group compared to the control group. On the other hand, the rate of absorption of Leu, Ile, and Val was increased; Phe showed the same. Therefore, we suggest that CFE could affect protein metabolism by increasing or decreasing the absorption rate of essential amino acids.

Effects of Feeding Frequency on Oxygen Consumption of Nile Tilapia, Oreochromis niloticus, in a Recirculating Aquaculture System (순환여과식 사육 시설에서 사료 공급 횟수에 따른 나일틸라피아, Oreochromis niloticus의 산소 소비량)

  • This study was conducted to test actual oxygen consumption rates of Nile tilapia (Oreochromis niloticus) at a commercial scale high density culture tank in the recirculating culture system, with a special emphasis on the oxygen consumption rate of this species with different daily feeding frequencies. Nile tilapia, an average of 400 g and a total wright of 390 kg, were stocked in a circular concrete tank of 2.9 m in diameter and 0.8 m in depth, in a recirculating culture system. The fish were fed commercial diet 1, 2, 5, and 9 times per day between 09:00 h and 19:00 h. feed was evenly supplied where the fish were fed more than once a day. Dissolved oxygen of influent and effluent water was monitored every 50 seconds by a computer with electrodes for 24 hours. The standard metabolic rate of tilapia was $39.31{\pm}4.4mg\;O_2/kg$ fish/hr in the present study. The maximum oxygen consumption was reached between 30 to 50 minutes after feeding. The maximum oxygen consumption levels of 1, 2, 5, and 9 feeding times per day were 161.2, 123.4, 111.1 and 111.1 mg $O_2/kg$ fish/hr, respectively. While the mean oxygen consumption levels of 1, 2, 5, and 9 feeding times per day were $79.9{\pm}21.5,\;81.3{\pm}14.8,\;84.2{\pm}9,9$ and $98,9{\pm}11.0mg\;O_2/kg$ fish/hr, respectively. These results show that oxygen consumption rates are not much different between the feeding frequencies. These results could be used to design to provide desirable oxygen supply system in the recirculating high-density tilapia culture system.

Effects of Dietary Bee Venom on Serum Characteristic, Antioxidant Activity and Liver Fatty Acid Composition in Broiler Chickens (정제 봉독의 급여가 육계의 혈청성상, 항산화능 그리고 간 내 지방산 조성에 미치는 효과)

  • This study was conducted to investigate the effect of dietary bee venom on serum characteristics, antioxidant activity, and hepatic fatty acid composition in broiler chickens. A group of 875 one-day-old feather-sexed male broiler chicks were randomly allocated to five treatments with seven replicates (25 birds/replicate) for three weeks. A corn-soybean meal-based diet was used as the basal diet. Five dietary treatments were compared: 1) basal diet, 2) basal diet containing $10{\mu}g/kg$ of bee venom powder, 3) basal diet containing $50{\mu}g/kg$ of bee venom powder, 4) basal diet containing $100{\mu}g/kg$ of bee venom powder, and 5) basal diet containing $500{\mu}g/kg$ of bee venom powder. At 21 days, one bird per pen was slaughtered by asphyxiation in $CO_2$ gas, and blood was collected to measure serum characteristics and antioxidant activity. In addition, the liver was excised to measure the concentration of malondialdehyde and determine fatty acid composition. Increasing dietary bee venom in the diet failed to affect most serum parameters except for triglyceride and non-esterified fatty acids. Dietary bee venom inclusion quadratically increased the concentration of stearic acid (P<0.05), but decreased palmitoleic acid, oleic acid, linoleic acid, mono-unsaturated fatty acids, and poly-unsaturated fatty acids. Finally, dietary bee venom tended to lower hepatic malondialdehyde contents quadratically (P=0.054). In conclusion, our study revealed that dietary bee venom improved antioxidant capacity and affected fatty acid metabolism in broiler chickens.

Dry Matter Distribution during Seedling Development in Soybean (콩의 유묘기간 중 건물중의 분배)

  • To obtain the basic information about the translocation of seed reserves of soybean[Glycine max(L.) Merrill] cultivars, 'Hill', 'Paldalkong' and 'Jangyeobkong' , the dry matter changes of emerging organs during the germination and seedling development were observed at the research farm of College of Natural Resources, Korea University, on May 26, June 5 and June 14. Mean fresh weight, dry weight and moisture content of three soybean cultivars were increased until the V2 stage in all seedling parts except cotyledons. Cotyledon dry weight was continuously decreased and the decreased amount at the V2 stage was about 80% of the cotyledon dry weight at the germination stage. Structural component of cotyledons was 22.2% of the cotyledon dry weight at the germination stage. Metabolic components of cotyledons were markedly decreased until V2 stage, and about 91% of cotyledon metabolic components at germination stage was utilized. However, those of whole seedling were increased after the V1 stage. Therefore, it appeared that role of cotyledons as nutrient supplier for germination and seedling growth was important until the V1 stage especially.

Effects of Radiation Mutant Perilla frutescens var. crispa in Preventing and Alleviating Symptoms in a Monosodium Iodoacetate-Induced Osteoarthritis Rat Model (MIA로 퇴행성관절염을 유도한 랫드에 방사선 형질전환 차조기가 증상 예방 및 완화에 미치는 효과)

  • The present study aimed to evaluate the effect of radiation mutant Perilla frutescens var. crispa on bone metabolism and the inflammatory response in a monosodium iodoacetateinduced rat model of osteoarthritis. radiation mutant Perilla frutescens var. crispa was administered orally at doses of 25, 50 or 100 mg/kg/day for 2 weeks before direct injection of monosodium iodoacetate (3 mg/50 μl of 0.9% saline) into the intra-articular space of the rats' right knees. The rats subsequently received the same doses of oral radiation mutant Perilla frutescens var. crispa for another 4 weeks. It was evaluated that the treatment effects based on serum biomarkers, and morphological and histopathological analysis of the knee joints. Compared with those in control rats, the radiation mutant Perilla frutescens var. crispa treatments significantly reduced the serum levels of inflammation, bone metabolism markers (i.e., COX-2, LTB4, MMP-3, and COMP), and the amount of fibrous tissue. Otherwise, it was significantly increased the concentration of TIMP-1 and calcitonin. In addition, the radiation mutant Perilla frutescens var. crispa treatments effectively preserved the knee cartilage and synovial membrane. As a result, it indicates that radiation mutant Perilla frutescens var. crispa prevented and alleviated osteoarthritis symptoms. Thus, radiation mutant Perilla frutescens var. crispa can be used in food and drug material for the management of osteoarthritis.

Hanja Information in the Entries of Korean Unabridged Dictionary (국어대사전의 표제어에 나타나는 한자 정보)

  • For language information processing that includes both Hangul and Hanja, an electronic dictionary supporting Hangul and Hanja simultaneously is necessary. This paper examined statistical information on Hanja entries of Korean Unabridged Dictionary such as the number of entries that include Hanja based on the KSC-5601 character set, the frequency of the pronunciation and meaning of each character of Hanja included in the entries, the frequency per part of speech of Hanja in entries and the average number of Hanja characters per entry. At least one or more of Hanja characters appear in 303,951 entries out of 440,594, accounting for 68.99% of the total. 858,595 characters of Hanja are included in the 440,594 entries, which is 1.95 Hanja characters per entry. As the average syllable length of the entries is 3.56 and the average count of the Hanja characters per entry is 1.96, it can be said that 54.7% of all the characters of the entries are in Hanja. Among 4,888 Hanja character codes, 4,660 are used once or more, whereas 228 Hanja codes never appear in any entry. There were 5 characters which appear more than 4,000 times. A total of 858,595 Hanja characters used in all the entries correspond to 471 Hangeul codes.

Metagenomics analysis of methane metabolisms in manure fertilized paddy soil (메타게놈 분석을 이용한 돈분뇨 처리에 의한 논토양에서 메탄대사에 미치는 영향 조사)

  • Under flooded rice fields, methanogens produce methane which comes out through rice stalks, thus rice fields are known as one of the anthropogenic sources of atmospheric methane. Studies have shown that use of manure increases amount of methane emission from rice. To investigate mechanisms by which manure boosts methane emission, comparative soil metagenomics between inorganically (NPK) and pig manure fertilized paddy soils (PIG) were conducted. Results from taxonomy analysis showed that more abundant methanogens, methanotrophs, methylotrophs, and acetogens were found in PIG than in NPK. In addition, BLAST results indicated more abundant carbohydrate mabolisetm functional genes in PIG. Among the methane metabolism related genes, PIG sample showed higher abundance of methyl-coenzyme M reductase (mcrB/mcrD/mcrG) and trimethylamine-corrinoid protein Co-methyltransferase (mttB) genes. In contrast, genes that down regulate methane emission, such as trimethylamine monooxygenase (tmm) and phosphoserine/homoserine phosphotransferase (thrH), were observed more in NPK sample. In addition, more methanotrophic genes (pmoB/amoB/mxaJ), were found more abundant in PIG sample. Identifying key genes related to methane emission and methane oxidation may provide fundamental information regarding to mechanisms by which use of manure boosts methane emission from rice. The study presented here characterized molecular variation in rice paddy, introduced by the use of pig manure.

Analysis and risk assessment of formaldehyde in water from water purification plant in korea (국내 정수장 먹는 물 중 폼알데히드 함유실태 조사 및 위해성 평가 연구)

  • Formaldehyde is used in lether manufacture, a dry plate and an explosive. It is by-product of ozonizing process in filtration plant. The effects of exposure are eye pruritus, tickle, runing nose, blocking nasal passages and headache. It also makes a dried throat and causes inflammation. It is classified as B1 group for inhalation by US. EPA, which can cause cancer in human. For analysis of formaldehyde, formaldehydes-DNPH derivative was extracted with solid cartridge and was analyzed by High Performance Liquid Chromatography/Diode Array Detector (HPLC/DAD). The detection limit was $3{\mu}g/L$ and the recoveries were 72.3~109.1% (RSD 2.9~11.5%). Water samples were collected in four Korean rivers, four times per year seasonally for 10 years from 1998 to 2007. The monitoring results were 48.8% (630/1291), $5.15{\sim}101.9{\mu}g/L$ in purified water. Because of non-carcinogen in drinking water, hazard index is calculated with RfD. Results of excess cnacer risk was below 1 and was considered as safe value.
