The purpose of this study is to discuss and analyze the change of GLUT-4 and muscle fiber type of streptozotocin(STZ)-diabetic rats over a period of 6 weeks. We divided into four groups; I(aquatic exercise and feeding of Cordyceps militaris; n=6), test group II(feeding of Cordyceps miliaris; n=6), test group III(aquatic exercise; n=6), control group IV(non-treatment; n=6). After experimenting we measured the blood glucose, body weight, muscle fiber type and GLUT-4 protein content. The change of glucose levels decreased greater in group I than the other group. The body weight gain was lower in the all groups. The change femoris muscle fiber type, the size of muscle fiber TypeII lessened more than the one of Type I in group IV. Decrease of muscle fiber size more diminishment in group I than the other group. GLUT-4 protein quantity decrease in group IV compared to normal group. It was significantly increased in group. I, III compared to group IV. But there was more increase in group I (p<.001). These results suggest that GLUT-4 and muscle fiber type II decrease in STZ-diabetic rats and that when we apply aquatic exercise and feeding of Cordyceps militaris in diabetic rats over a period 6 weeks, it is increase GLUT-4 and the increase of insulin sensitivity of peripheral tissue. So it is considered to be helpful in improvement of glucose homeostasis and in prevent from muscle atrophy resulted from complication.
In this study, we studied the effect of dissolved cement powder on Carassius auratus by analysis of the morphophysiological changes. The gill exposed to dissolved cement powder showed the thickened primary lamellae and the activity of chloride cells and mucous cells was also significantly increased and the proliferation, separation and clubbing of gill filament was observed in the secondary lamellae. In the kidney tissue, the space in Bowman's capsule was widen and the arrangement of dermis was irregular due to the thinned epidermis in the integument tissue. The activities of antioxidant enzymes and LDH tended to increase with the duration of cement exposure. It was confirmed that the up-regulated proteins were identified as involved in glycolysis and energy metabolism and down-regulated proteins were myofibrillar proteins which were involved in muscle contraction by the cement exposure to the integument. With these results, dissolved cement powder was thought to be a big threat to the survival of the fish because it causes the morphological changes and weakens the physiological activity in C. auratus tissues.
Myostatin is a transforming growth and differentiation factor-${\beta}$ family member that acts as a negative regulator of muscle growth. Previously, mutations in the myostatin gene were known to be related to double muscling phenotypes in cattle. Because myostatin gene is highly related to muscle mass, also meat quality, in cattle, we sequenced whole myostatin mRNA and investigated the SNPs (Single Nucleotide Polymorphisms) in Korean cattle (Hanwoo). The results indicated that Hanwoo had an SNP in nt2385 and this mutation can be a useful marker with further verifications. We also investigated expression patterns of the myostatin gene from various muscle tissues and organs. Northern blotting results indicated that myostatin expression was restricted in muscles with variable expression levels. The results presented here can be used as a valuable information for meat quality related traits and muscle mass in cattle.

This case report describes capture myopathy in an 8-month-old female red-necked wallaby (Macropus rufogriseus) that died after a 14-day history of depression, lameness and astasia following a bathing protocol for dermatitis. In a blood test performed the day after the wallaby fell down, serum chemistry showed a normal rangeof LDH but elevated CK, ALT and AST. Upon histopathologic examination after the animal's death, we found degeneration and necrosis of myofibers in skeletal muscle and myocardium. According to history that the wallaby was pressed by other individuals, suspected vitamin E insufficiency in the diet, physical stress during bathing as well as results of a blood test and microscopic examination, we diagnosed this case as capture myopathy. Although capture myopathy is a common problem in marsupials, this is the first reported case in Korea and understanding this case will help to manage future cases of capture myopathy in captive settings in Korea.
Histological study of eel was carried out to determine the effect of drying on muscular tissue and fat movement. The results are as follows: (1) Changes of muscular tissue during drying may he divided into two stages early and late. In the early stage there was apparent dehydration of connective tissue while in the late stage muscle fibre dehydration appears to occur. (2) Movement of fat during drying depends upon division and contraction of the myocommata caused by inner structural changes. This phenomenon appeared to be more apparent in the late rather than the early drying stage.(3) After freeze-drying traces of ice crystals were detected around the muscle cells. These ice crystals remained even after rehydration. (4) When muscular tissue was heated the connective tissue melted while muscle fiber dehydrated and solidified.
Dried fruits of Schizandra chinensis Baillon, Fructus Schisandrae, have been widely used for many years to prevent and treat various diseases in Asian countries including Korea and Russia. It has recently been reported that extracts of Fructus Schisandrae are effective for controlling muscle and skeletal diseases. In this study, we investigated the efficacy of ethanol extract of Fructus Schisandrae (EEFS) on apoptosis and inflammatory response in gastrocnemius muscle of dexamethasone-induced catabolic muscle atrophy mice as part of natural substance discovery and functional analysis for improving muscle function. According to the results of this study, EEFS supplementation attenuated body weight gains and suppressed calf thickness loss in dexamethasone-induced muscle atrophic mice. Gastrocnemius muscle immunohistochemistry showed that expression of caspase-3 and poly(ADP-ribose) polymerase, which are representative apoptotic markers, was markedly increased in dexamethasone control mice; however, their expression was effectively reduced in the EEFS-fed mice. EEFS supplementation also prevented dexamethasone-induced increases in immunoreactivity of muscle fibers for myostatin, an important negative regulator of skeletal muscle mass. In addition, EEFS significantly normalized the increased numbers of nitrotyrosine, 4-hydroxynonenal and inducible nitric oxide synthase-positive muscle fibers compared to that found in dexamethasone control mice. These results suggest that EEFS protects dexamethasone-induced muscular atrophy by decreasing apoptosis and inflammatory responses, and EEFS is more likely to be developed as a muscle strengthening agent.
Skeletal deformity in hatchery-reared river puffer (Takifugu obscurus) fingerlings occurred. The vertebral abnormality was recognized with naked eyes 80 days after hatching. Affected fish had a good appetite but no clinical signs were found except the vertebral abnormality. As more than 90% of the hatchery-reared fish exhibited vertebral abnormality, the fingerlings could not be used for commercial seeds any more. Morphological changes in the vertebrate and the swim bladder were observed with a soft X-ray. Histopathological changes on the caudal muscle around the deformed vertebrae, gill, and spleen were also examined to clarify the cause of the deformity. Spinal curvature occurred between vertebrae 10 and 12, but any malfunction of the swim bladder was not found. The diameters of the muscle fibers around the deformed vertebrae were much smaller and more irregular than those in normal fish, and the gabs between the fiber bundles were greatly enlarged. No evidence of inflammation response was found in the muscle layer. In the hatchery, feed was putting at the position of water inflow, which might attract the fingerings to move toward the feed in spite of incomplete development of their caudal muscle fibers. From these results, it is suggested that the high speed of water current in rearing aquaria might be associated with the development of vertebral abnormality.
In order to investigate the general characteristics of ATPase and ATPase thermostability between porcine white muscle and red muscle, myofibrillar proteins were prepared and compared their physicochemical characteristics. SDS-polyacrylamide gel electrophoretic analyses showed that a protein band of 30,000 daltons was detected noticeably in myofibril from red muscle, but negligibly in myofibril from white muscle. The noticeable differences were found between porcine white muscle and red muscle for the activities of EDTA-ATPase, Ca-ATPase and Mg-ATPase. Myofibrillar proteins from white muscle showed higher thermostability than those from red muscle. Thermodynamic parameters, enthalpy $({\Delta}H^#)$, entropy $({\Delta}S^#)$, etc., showed characteristic variations between porcine white muscle and red muscle.
Congenital myotonia is a hereditary disorder of the skeletal muscle. The most characteristic features of the disease are myotonia and variable muscular hypertrophy. Molecular biologic investigations have revealed that mutations in the gene of the human skeletal muscle chloride ion channel protein are a cause of the disease. The Becker's type congenial myotonia is clinically similar to the autosomal dominantly inherited congenital myotonia (Thomsen's disease). Both disorders are characterized electrophysiologically by increased excitability of muscle fibers. reflected in clinical myotonia. In general, Becker's type congenital myotonia is more severe than Thomsen's disease in muscular hypertrophy and weakness. The authors recently experienced a 25-year-old female patient who has no family-related disease history and who has conspicuous muscular hypertrophy and the stiffness with muscles which occurred from the age of 3 or 4. Clinically she showed the authors a percussion myotonia. On electrophysiological study, exercise and repetitive stimulation of the abductor digiti quinti muscle disclosed a decline in the compound muscle action potential. Biopsy of biceps muscle revealed enlargement of muscle fibers with marked nuclear internalization. After the oral taking the Mexiletine, the patient showed a favorable turn a little with her stiffness of muscles. So we authors are reporting one case of Becker's type congenital myotonia with review of literatures.
Purpose: The purpose of this study was to examine the effects of daily exercise before steroid treatment on mass, the type I and II fiber cross-sectional area, and myofibrillar protein content of hindlimb muscles in a rat model. Method: Adult male Sprague-Dawley rats were randomly assigned to one of three groups: a control group(n=10) that had a normal saline injection for 7days, a steroid group(n=10) that had a steroid injection for 7days, and an exercise-steroid group(n=10) that ran on the treadmill for 7days before a steroid treatment. Body weight and food intake were measured every day. At 15 days all rats were anesthetized and the soleus, plantaris and gastrocnemius muscles were dissected. Result: The exercise-steroid group showed significant increases as compared with the steroid group in body weight, muscle weight of the soleus and gastrocnemius, type II muscle fiber cross-sectional area of plantaris, and myofibrillar protein content of the soleus, plantaris, and gastrocnemius. As compared with the control group, the steroid group showed significant decreases in body weight and diet intake, muscle weight, the type II fiber cross-sectional area and myofibrillar protein content of the soleus, plantaris, and gastrocnemius muscles. Conclusion: Daily exercise before steroid treatment attenuates hindlimb muscle atrophy, with type II muscle changes more apparent than type I muscle changes.
