Change of Chemical and Microbial Properties during Fermentation of Cotton Waste for Oyster Mushroom Cultivation (느타리 재배용 폐면 발효 중의 화학성 및 미생물 상의 변화)

  • The changes of microflora and chemical characteristics during fermentation process of cotton waste for oyster mushroom cultivation were investigated with 5 l bench-scale reactors placed in an incubator at different temperatures ($40,\;50\;and\;60^{\circ}C$). Cotton waste was wetted to 70% moisture, and air flow rates to the substrate were 50, 100 and 300 cc/min. In processing of composting, the mesophilic bacterial population decreased sharply but thermophilic bacterial population increased. In case of fungi, both mesophilic and thermophilic population decreased. The daily $CO_2$ evolution showed little difference in all treatments, while $NH_3$ dropped sharply after 3 days. The desirable composting temperature and air flow based on the mycelial growth of oyster mushroom were $50^{\circ}C$ and 100 cc/min, respectively.

Isolation of Wild Yeasts from Soils of Reed Fields in Seocheon-gun County, Chungcheongnam-do, South Korea, and Characterization of Unrecorded Yeasts (충남 서천군 갈대밭 주변 토양에서 야생효모의 분리 및 미기록종 특성)

  • The goal of this study was to isolate wild yeasts from reed fields in the Seocheon-gun county in Chungcheongnam-do, South Korea. Molecular analysis of the D1/D2 domain of the 26S rDNA of the yeasts isolated from soil samples was performed using Basic Local Alignment Search Tool (BLAST). In total, 20 yeast strains from 11 species were isolated from 20 different soil samples. Candida species were observed to be the most common yeasts. Of these 20 strains, Candida subhashii (6 strains), Candida tropicalis (3 strains), Candida boleticola MS008, and Lachancea thermotolerans (2 strains) were identified as alcohol-fermenting yeasts. Further, Bullera japonica YJ10-1, Candida subhashii J7-1, Kluyveromyces yarrowii YJ11-1, and Ustilago shanxiensis Y10-1 were newly recorded yeast strains in Korea, and therefore, their microbiological characteristics were investigated further. All of these unrecorded yeast strains had oval cells; only Candida subhashii J7-1 formed ascospores and pseudomycelia. Kluyveromyces yarrowii YJ11-1 grew in vitamin-free medium, and all of these strains, except Candida subhashii J7-1, grew in 5% NaCl-containing YPD broth.

Utilization of various substrates for the cultivation of oak mushrooms (Lentinula edodes) (다양한 기질에 대한 표고버섯 톱밥재배 가능성)

  • We investigated the effect of different media components on the characteristics and productivity of Lentinula edodes cultivated in sawdust. Ten substrates were used for this study. Soybean and cotton seed meal were unsuitable substrates as fungal mycelia did not grow to maturity during the incubation period. Lentinula edodes (NIFoS culture number 2462) was grown on the ten substrates, and soybean hull, mixed medium, cotton seed hull and corn grain showed greater effect on the productivity than wheat bran. However, wheat bran induced higher productivity in NIFoS 2778 than the other substrates. Sawdust medium with soybean meal produced smaller fruiting bodies than the other substrates. In contrast, corn meal media produced larger mushrooms than the other substrates. The external characteristics of the mushrooms varied based on the substrates upon which they were grown. This is not surprising given that the substrates differed with respect to carbohydrate and protein content, e.g., cotton seed and soybean meal contained a higher crude protein and crude ash than the other substrates tested.

Characteristics of fruiting bodies color mutants in Pleurotus ostreatus (원형느타리버섯 백색돌연변체의 특성)

  • The white-colored and the dark gray-colored mutants were frequently happened in cultivated areas of Pleurotus ostreatus (Wonhyeong-neutari). These caused conflicts between farmers and spawn companies. Our studies were conducted to elucidate the mechanism of mutagenesis. The results from the studies would provide valuable informations that could be used to prevent the color-related mutation, and also will be applied in breeding programs of P. ostreatus. Oyster mushroom variety, Wonhyeong-neutari, is somatic hybrid of Pleurotus and has genetic makers for arginine, ornithine, proline, riboflavine. Genetic markers analysis of monospore isolates derived from color mutants show identical tendency with that of Wonhyeong-neutari, these results indicate that color mutants were derived from Wonhyeongneutari. Twenty-one and four homokaryons were selected from the white-colored mutant MGL 2205 and gray-colored ASI 2029. All 34 F1 hybrids derived from the white-colored mutant MGL 2205 produce white-color fruiting bodies, indicating that the white color trait is heritable. In the first generation hybrids between the white-colored MGL 2205 and the gray-colored ASI 2029, all 16 hybrids produced pigmented fruiting bodies. Homokaryons isolated from the hybrid MGL 2205 X ASI 2029 were mated with homokaryon tester strains derived from MGL 2205. By these result, we could assumed that white color trait is a heritable character which is controlled by more than one recessive gene.

Antimicrobial Activity of Nano Materials against Acidovorax citrulli and Other Plant Pathogens (나노 화합물을 이용한 Acidovorax citrulli 및 식물병원성 미생물의 항균활성 효과 검정)

  • Antimicrobial activities of nano-materials were tested against several plant pathogens. Twelve different nano-materials were used to observe the antagonistic activity against three kinds of mold and sixteen different kinds of watermelon fruit rot pathogens (Acidovorax citrulli). According to the results, no antagonism have been found against the pathogen, Cylindrocarpon destructans. However in the case of Pythium ultimum, combination of Brass/Glucose 1,000 ppm confirmed the mycelial growth reduction by 94%. In addition, little effect was found against Rhizoctonia solani by Ag/Glucose 3,000 ppm. The remaining other nano-materials have different antimicrobial effect depending on the strains of A. citrulli. But in the case of lime (Cu/Salt 1,000 ppm) highest antimicrobial activity was observed with 97%. Moreover growth of five different strains of A. citrulli was checked by 99% with the combination of Ag/Glucose 1,000 ppm. 92% reduction of A. citrulli growth was observed with $Brass/CaCO_3$ 3,000 ppm. Tested nano-materials against different plant pathogens in this study showed the antimicrobial activity at the range of 24-70%.

Mating type and metalaxyl sensitivity of Phytophthora infestans isolated from Kangwon area in Korea (강원 지역에서 분리한 감자역병균(Phytophthora infestans)의 교배형(mating type) 및 metalaxyl 저항성)

  • Isolates of Phytophthora infestans obtained from several locations of Kangwon area in 1998 and 1999 were examined on mating types and sensitivity to metalaxyl. Both A1 and A2 mating type isolates were isolated in 1998 and 1999. The majority of the P. infestans isolates were A1 mating type. About 64.3% of the isolates collected in 1998 and 99.1% in 1999 were determined as A1 mating type. Sensitivity of the P. infestans to metalaxyl was examined by measuring mycelial growth on V8 juice agar amended with $10{\mu}g/mL$ matalaxyl. About 44.6% of the isolates examined in 1988 were resistant to metalaxyl, 55.4% of the isolates were intermediate resistant, but sensitive isolate was not isolated. However, 10.5% of the isolates examined in 1999 were sensitive, 88.6% of the isolates were intermediate resistant, and 0.9% of the isolates were resistant to metalaxyl. This studies indicate that A1 mating type is displacing A2 mating type and metalaxyl sensitivity of the P. infestans isolates of Kangwon area is increasing. This result is quite different from trends of early in 1990s.

Effect of Sodium Hypochlorite(NaOCl) Treatment On Bacterial Yellow Blotch in Oyster Mushroom, Pleurotus ostreatus (Sodium Hypochlorite(NaOCl) 처리가 느타리버섯의 세균성 갈반병에 미치는 효과)

  • Studies were conducted to determine the potential of sodium hypochlorite(SHC) on the control of bacterial yellow blotch in cultivated oyster mushroom, Pleurotus ostreatus. SHC at the concentration of 80 ppm was effective on the control of Pseudomonas agarici causing yellow blotch in oyster mushroom except number 916 isolate. In vitro the mycelial growth was slightly inhibited at the concentration higher than 100 ppm of sodium hypochlorite, but retardation of the mycelial growth was soon recovered. Spray of SHC solution at the concentration of 40-50 ppm per day significantly reduced the incidence of the yellow blotch without impairing the growth of oyster mushroom in field culture. However, the higher concentration of SHC(67 ppm) induced yellow brown or dark gray in color and deformed cap and elongated stripe in morphology of fruiting body. Results indicate that periodical spray of sodium hypochlorite seems to be the recommendable method for protection against bacterial yellow blotch disease in oyster mushroom without reducing food quality.

  • These experiments were conducted to learn the antibiosis of microfloral organisms in the casing soil to Mycogone perniciosa, and interactions between M. perniciosa and Agaricus bisporus. The results obtained were as follows: 1. In vitro tests, the development of M. perniciosa was suppressed from the unidentified microfloral organisms in the casing soil. All the infected sporophores of A. bisporus occurred within an area applied with spore suspension of M. perniciosa as spot treatment and no infected ones in the area around the spot treated under cropping conditions. 2. In vitro tests, although the antagonistic relationship between M. perniciosa and A. bisporus was somewhat different in varying the kind of media, A. bisporus ultimately overgrew the colony of M. perniciosa. When spore inoculation of M. perniciosa was applied on the surface of grain spawn of A. bisporus and the mid layer of casing soil under cropping conditions, no infected sporophores were produced, whereas the infected sporophores were only produced on casing soil inoculated on the surface of casing soil with spore suspension.

Investigation of Active Antifungal Compounds of Essential Oil from Chamaecyparis obtusa Against Dermatophytes, Microsporum canis and Trichophyton Mentagrophytes (피부사상균 Microsporum canis 및 Trichophyton mentagrophytes에 대한 편백정유의 항진균활성물질 탐색)

  • The present study was conducted to evaluate the application of Chamaecyparis obtusa and to investigate potential utilization of essential oil from C. obtusa as plant-based medicine. The antifungal activity of essential oil from leaves and twigs of C. obtusa (Sieb. Et Zucc) was determined and the major components of active fractions against Microsporum canis (KCTC6591) and Trichophyton mentagrophytes (KCTC6077) were identified by GC/MS analysis. In treatment of essential oil from C. obtusa, the strain M. canis was more resistant than the other, T. mentagrophytes. In the agar diffusion assay, essential oil from C. obtusa inhibited hyphal growth of M. canis and T. mentagrophytes at the concentration of more than 5,000 ppm. The zones named B and C in the TLC assay of essential oil from C. obtusa showed antifungal activities. Among four sub-fractions of n-hexane extract from B and C zones, named as B-1, B-2, C-1 and C-2, the C-2 showed the highest antifungal activity. Instrumental GC/MS analysis for sub-fractions showed that a major component of C-1 was ${\alpha}$-terpineol as terpene alcohol, while C-2 contained sesquiterpenes such as elemol, cedrol and eudesmol.

Studies on Biological Activity of Wood Extractives(VII) - Antimicrobial and Antioxidation Activities of Extractives from the Heartwood of Prunus sargentii - (수목추출물의 생리활성에 관한 연구(VII) - 산벚나무 심재 추출성분의 항균 및 항산화활성 -)

  • Antimicrobial and antioxidative activities on heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. Six flavanones including pinostrobin, eriodictyol, naringenin, pinocembrin, taxifolin and verecundin were isolated from Prunus sargentii which has been selected due to its high antimicrobial and antioxidative activities among the tested species. According to the results of antifungal test, pinocembrin was evaluated as the highest antifungal compound among the test compounds, which showed 80% of hyphal growth inhibition rate. Antifungal activity of pinocembrin was similar to hinokitiol(${\beta}$-thujaplicin), strong antimicrobial compound isolated from Thujopsis dolabrata. Naringenin followed pinocembrin in its antifungal activity. However, verecundin did not show any antifungal activity. No compound was effective in antibacterial activities. As a result of the measurement of free radical scavenging activity, antioxidative activities of taxifolin and eriodictyol were 2 times that of ${\alpha}$-tocopherol, and antioxidative index of these compounds were even superior to that of ${\alpha}$-tocopherol. In this regard, it could inferred that high antifungal and antioxidative activities of extractives of P. sargentii were derived from pinocembrin, taxifolin and eriodictyol, respectively.
