Interpretation of Subsurface Fracture Characteristics by Fracture Mapping and Geophysical Loggings (단열조사 및 물리검층을 통한 지표 하 단열특성 해석)

  • As a preliminary study to establish fracture network model in crystalline rocks, detail investigation on fracture characteristics were performed. Five fracture sets were determined on the basis of regional survey of geological structures and fractures on outcrops. Among the fracture sets, S1 set has the highest density and longest trace length of fractures which was identified on surface in the study area. S4 and S5 sets are composed of foliations and foliation parallel shear joints of gneisses, which are very important sets at the aspect of weighting of fracture length. For characterization of subsurface fractures, detail core logging was performed to identify fractures and fracture zones from five boreholes. Acoustic televiewer logging and borehole geophysical loggings produced images, orientations and geophysical properties of fractures which intersect with boreholes. According to the result of the investigations, subsurface fractures can be grouped as three preferred orientations(B1, B2 and B3), which correspond to S1, S2 and S4/S5 of surface fracture sets, respectively. Actually, B1 set is expected to be intensely developed at subsurface. However, it has low frequency of intersection with boreholes due to its parallel or sub-parallel direction to boreholes. According to the inference of conductive fractures, B1 and B3 sets have possibilities of water flow and their intersection lines are also thought to consist of important conduits of groundwater flow. In particular, faults which are parallel to foliations control major groundwater flow in the study area.

Anticariogenic activity of acanthoic acid isolated from Acanthopanax koreanum $N_{AKAI}$ against oral pathogens (섬오가피로부터 분리된 acanthoic acid의 항치아우식효과)

  • This research was aimed to search for natural antimicrobial agents that are sefe for humans and specific for oral pathogens. Acanthoic was isolated from the chloroform fraction of methanol extract of Acanthopanax koreanum $N_{AKAI}$ and its structure were elucidated by 13-NMR, 1H-NNR and ESIMS. Antibacterial activity of acanthoic acid was investigated by the minimum inhibitory (MIC) and minimun bactericidal (MBC) concentration. MIC/MBC of acanthoic acid against Streptococcus mutans $N_{AKAI}$ causing dental caries was determined to be $2/4\;{\mu}g/mL$, which was much lower than these of other natural antimicrobial agents such as $8/16\;{\mu}g/mL$ of sangurinarine and $250/500\;{\mu}/mL$ of green tea extract, $500/600\;{\mu}g/mL$ of thymol and borneol. Acanthoic and significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus $N_{AKAI}$ and Streptococcus sanguis $N_{AKAI}$, and Streptococcus gordonii $N_{AKAI}$ in the MIC range of $4{\sim}32\;{\mu}g/mL$. My finding suggests that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries.

  • The fruiting body of Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2$O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to isolation of four sterols. From the result of spectral data including NMR, MS and IR, the chemical structures of the sterols were determined as ergosta-7,24(28)-dien-3${\beta}$-ol (episterol, 1), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,9(11),22-trien-3${\beta}$-ol (dehydrop-eroxyergosterol, 2), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,22-dien-3${\beta}$-ol (ergoterol peroxide, 3), and $3{\beta}$,$5{\alpha}$-dihydroxy-6${\beta}$-methoxyergosta-7,22-diene (6-O-methylcerevisterol, 4). The ergosterols have been first isolated from this mushroom in this study.

Notes on Rare Species of Mycoparasitic Forming Fungus in Korea (국내에서 발견된 균류에 기생하는 희귀종 버섯)

  • One interesting parasitic forming fungus, being grown on the eggs of Phallus impudicus was collected from Mulchajoreum and on woods of Saryeoni in Jeju island. This taxon identified as a Podostroma solmsii (E. Fisch.) S. Imai f. octospora Yoshim. Doi (Hyporeaceae) based on morphological characteristics. According to the interpretation of Doi(1978), this taxon hsving the stipitate, genus Podostroma, family Hypocreaceae, Hypocreales, Ascomycota was identified as Podostroma solmsi f. octospora and its korean common name designated by authors. This species is characterized in having the fusiforme to ellipsoidal, unicellular and warted ascospores without septate and with blunt apiculus at each end and mycoparasitic habitat. Although there is some mycologists suggest that this taxon having above characters needs to be derived from Hypomyces. But authors are primarily going to record this species as a named "Podostroma solmsii (E. Fisch.) S. Imai f. octospora Yoshim. Doi" for the fungus flora of Korea. In a future, it is need in detail to compare their DNA sequencing data on the type species and some related species. All collections cited here are deposited in the Herbarium Conservation Center of National Academy of Agricultural Sciences.

Organic acid production and phosphate solubilization by Enterobacter intermedium 60-2G (Enterobacter intermedium 60- 2G의 유기산 생성과 불용성인의 가용화)

  • A phosphate solubilizing bacterium. strain 60-2G, possessing a strong ability to solubilize insoluble phosphate was isolated from the rhizosphere of grass. On the basis of GC-FAME profile, carbon utilization pattern, and the DNA sequence of a conserved partial 16S rRNA gene, the 60-2G was identified as Enterobacter intermedium. The analysis by HPLC revealed that the strain 60-2G produced mainly gluconic and 2-ketogluconic acids with small amounts of lactic acid in broth culture medium containing hydroxyapatite. During the incubation period of the strain 60-2G in broth culture, pH of the medium decreased upto 3.8 while the soluble phosphate concentration increased. The reversed correlation between pH and soluble phosphate concentration indicated that the solubility of P was due to the produced organic acids. The sequence homology of the deduced amino acids suggested that E. intermedium 60-2G synthesized PQQ which is essential for the oxidation of glucose by glucose dehydrogenase.

Phylogenetic Diversity and Community Analysis of Marine Bacteria Associated with Ulva pertusa (구멍갈파래(Ulva pertusa)에 서식하는 해양세균의 계통학적 다양성 및 군집구조 분석)

  • The present study was done to assess the diversity of the bacterial community associated with Ulva pertusa collected from Jeju Island using Restriction Fragment Length Polymorphism (RFLP) marker analysis. For RFLP analysis, a total of 145 bacterial strains associated with Ulva pertusa were screened and cultivated using Marine agar and R2A agar. The PCR amplicons of the 16S rRNA gene from all the isolated strains were digested with HaeIII and RsaI restriction enzymes and then classified into different groups according to their restriction patterns. Strains selected based on the RFLP patterns showed more than 91% 16S rRNA gene sequence similarity when compared with known bacterial species, which include 4 phyla - proteobacteria (alpha-proteobacteria, beta-proteobacteria, gamma-proteobacteria - 63%), firmicutes (11%), actinobacteria (4%), bacteroidetes (22%)–as well as 7 classes (actinobacteria, flavobacteriia, cytophagia, bacilli, α-proteobacteria, γ-proteobacteria, β-proteobacteria), 13 orders, 18 families, and 27 genera. These results confirmed a wide diversity of bacterial communities as contrasted with other regions. The newly isolated 10 strains, which show 16S rRNA sequence similarity of <97% compared to previously identified bacteria, could be noble species. Further experiments, such as morphological, physiological, and biochemical classification, are necessary to confirm the novelty of the newly isolated 10 strains.

Phylogenetic Study of Genus Sorbus in Korea by Internal Transcribed Spacer Sequence (ITS) (ITS에 의한 한국내 마가목 속 분류군의 유전적 계통분류학적 연구)

  • Genus Sorbus is a long lived woody species that is primarily distributed throughout Asia and Europe. This species is regarded as very important herbal medicines in Korea and China. Sorbus commixta is primarily distributed throughout Europe. We evaluated a representative sample of the four taxa with nuclear ribosomal DNA internal transcribed spacer sequences (ITS) to estimate genetic relationships within genus. Aligned nucleotide sequences of the length of ITS1 were nearly constant within genus Sorbus varying from 219 in S. aucuparia to 218 in the rest species. Especially, the 5.8S subunit of all taxa of Sorbus was found to constant of 165 bp nucleotides. However, aligned nucleotide sequences of the length of ITS2 vary from 240 in S. sambucifolia var. pseudogrcilisto 245 in S. aucuparia. Total alignment length is 629 positions, of which 35 are parsimony-informative, 32 variable but parsimony-uninformative, and 552 constant characters. The base furtherance showed the difference to the by a total taxon: an average A and T are 17.7% and G and C are 30.4%, 34.2%, respectively. All the four taxa beginning with conserved base paired triplets emerging from single strand regions (domain I). Noteworthy, in the RNA secondary structure proposed for the three Korean Sorbus taxa RNA transcript ITS2, which shows a remarkedly well-conserved folding (domain II). When compared to the European Sorbus (S. aucuparia) of ITS2. ITS analysis may be useful in germ-plasm classification several taxa of genus Sorbus.

Seasonal Variations and Species Composition of Planktonic Ciliates in the Southern Coastal Waters of Jeju Island, Korea (제주도 남부해역의 부유성 섬모충류의 종 조성과 계절 변동)

  • As part of study on the microbial food web in the southern Jeju Island of Korea, Planktonic tintinnids and aloricate oligotrichs were investigated from July 1998 to June 2000 to understand seasonal variation and water mass indication with environmental factors. 39 species of tintinnids were identified representing 1 order, 11 families, 20 genera. Tintinnid abundance ranged from 100 to 5,400 cells.1$^{-1}$ (mean 314 cells.1$^{-1}$ ), and oceanic species were mainly dominant in fall and winter season, while neritic species were a little pre- dominant in spring and summer season. 15 species of aloricate oligotrichs were identified representing 1 order, 5 families, 7 genera, and many belonged to genus Strombidium which was most abundant and most frequently occurred in many months. Abundance of aloricate oligotrichs ranged from 140 to 21,000 cells.1$^{-1}$ (mean 2,356 cells.1$^{-1}$ ). Species diversity and standing crops of tintinnids were quite different according to seasons, but few seasonal variations were detected in aloricate oligotrichs. In terms of water mass indication tintinnids represented obvious characteristics which were affected by marine environmental factors, but alor- icate oligotrichs had no such apparent indication as tintinnids. Ciliates were more abundant and more diverse in inshore than in offshore station. Total carbon biomass of ciliates ranged from 0.01 to 136.06 $\mu\textrm{g}$C.1$^{-1}$ (mean 5.01 $\mu\textrm{g}$C.1$^{-1}$ ). The carbon biomass, however, did not coincided with seasonal variations of abun- dance Vertical profiles of mean abundance of both tintinnids and aloricate oligotrichs were similar, and had same trend as those of mean chlorophyll o concentration. It suggests that phytoplankton and ciliates reflected the prey-predator relationship in the study area.

The VHR Dual-Specificity Protein Tyrosine Phosphatase (DS-PTPase) Inhibitor Isolated from Gastrodia elata (천마(Gastrodia elata)로부터 분리한 VHR DS-PTPase 저해 물질)

  • In the course of screening program for VHR DS-PTPase (dual-specificity protein tyrosine phosphatase) from natural sources, Gastrodia elata was selected. One compound showing potent inhibitory activity was isolated by the solvent extraction and column chromatography including silica gel, ODS RP-18, Sephades LH-20, and HPLC. This compound was identified as baicalein by several NMR techniques such as $^1H-NMR$, $^{13}C-NMR$, and DEPT. Baicalein showed selective inhibitory activity against VHR DS-PTPase with $IC_{50}=2.4\;{\mu}M$, and showed cytotoxicity against several human cancer cell lines with an $GI_{50}$ of $5.26{\sim}12.93\;{\mu}g/mL$ range, including, melanoma (LOX-IMVI), lung cancer (NCI H23 and A549), colon cancer (HCT 116 and SW 620), prostate cancer (PC-3), and leukemia (MOLT 4F).

Evaluation of biological activities of plasma-treated phloridzin (플라즈마 처리 phloridzin 반응물의 생리활성 평가)

  • Phloridzin is a predominant member of the chemical class of dihydrochalcones and mainly found in apple. The biological activity of phloridzin treated with dielectric barrier discharge (DBD) plasma was evaluated to investigate whether exposure to plasma can be used as a tools to enhance the biological activity of natural resources. DBD plasma treatment of phloridzin was carried out for three different exposure times. The antioxidant effects of degraded phloridzin for different reaction time were evaluated via radical scavenging assay using DPPH radical. In addition, the anti-diabetic and anti-obesity properties of the degraded phloridzin were measured based on ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities. Phloridzin treated for 60 min showed significantly higher radical scavenging, ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities compared to the tested positive controls. Degradation of phloridizin induced by DBD plasma might be responsible for enhancing the biological activity of phloridizin.
