Anti-inflammatory and Anti-oxidative Activities of Flavonoids Extracted from Dendranthema indicum Flowers in Jeju Island (제주 자생 감국 꽃 추출물 유래 flavonoid 화합물의 항산화 및 항염 활성)

  • Anti-inflammatory and anti-oxidative activities were examined on the extract of Dendranthema indicum (D. indicum) flowers. The flowers were extracted two times for 24 h each with 70% ethanol. Upon the biological activities screening, the ethanol extract exhibited potent free radical scavenging activities and inhibited the production of nitric oxide on LPS-induced RAW264.7 macrophages effectively without causing cell toxicity. Further purification by medium pressure liquid hromatography (MPLC) and identification of the isolates led to identification of cynaroside (1) and apigetrin (2). The chemical structures of the isolated compounds were elucidated based on spectroscopic data including nuclear magnetic resonance (NMR) spectra, as well as comparison of the data to the literature values. Also, the quantitative analysis of the compounds was perfromed by high-performance liquid chromatography (HPLC). The isolates 1 and 2 were determined to inhibite the nitric oxide (NO) production dose-dependently. Based on these results, it was suggested that D. indicum extract could be useful as anti-inflammatory agents in cosmetics applications.

Benthic Marine Algal Flora and Community Structure of Yongho-dong Area in Pusan, Korea (부산 용호동 일대의 저서 해조상 및 군집구조)

  • Benthic marine algal flora and community structure of Yongho-dong area in Pusan, Korea were investigated seasonally in intertidal and subtidal zones by a quadrat method along 5 transect lines from July 1996 to April 1997. In this area, a total of 99 species including 3 blue-green, 13 green, 26 brown and 57 red algae was found. Among them, 33 taxa, together with dominant Ulva pertusa and Corallina pilulifera, occurred throughout the year. Enteromorpha spp. and Ulva spp. vertically distributed in upper and middle zone, while Simphyocladia latiuscula, Chondria crassicaulis, Corallina pilulifera and Sargassum sup. in lower zones. Biomass per unit area exhibited a wide range of variation, from 1,241 g/$m^2$(spring) to 1,648 g/$m^2$(summer) in fresh weight. Maximum and minimum species diversity were recorded in spring and autumn, respectively. Results of the UPGAM cluster analysis suggest that the examined 5 sites can be divided into two groups, sites exposed to open sea (transects 1, 2, 3) and comparatively sheltered sites (transects 4, 5) faced with the Pusan Harbor. The former group is represented by large brown algae, Sargassum and Hizikia, while Ulva pertusa and Lomentaria species are mainly found in the latter group. A calcareous alga, Corallina pilulifera occurred dominantly in both groups throughout the year. In conclusion, number of species in this area was remarkably reduced as compared to the previous data. This result may suggest probably more change of algal vegetation in future, considering the physical and chemical pollutions loaded in the coastal marine environment of this area.

Whitening and Anti-inflammatory Constituents from the Extract of Citrullus lanatus Vines (수박 덩굴 추출물 유래 미백 및 항염 활성 성분)

  • In this study, we investigated whitening and anti-inflammatory constituents from a watermelon (Citrullus lanatus, C. lanatus) vines (leaves and stems). As anti-melanogenesis and anti-inflammatory activities were screened for the ethanol extract and solvent fractions, n-hexane (n-Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex and EtOAc fractions of C. lanatus; ${\alpha}-linolenic$ acid (1), sigmast-7-en-O-${\beta}$-D-glucopyranoside (2), 1-feruloyl-${\beta}$-D-glucopyrinoside (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Whitening and anti-inflammatory effects were studied for the isolated compounds. Upon the anti-melanogenesis tests using ${\alpha}-MSH$ stimulated B16F10 melanoma cells, the compounds 1 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. For the anti-inflammation tests using lipopolysaccharide (LPS)-induced RAW 264.7 cells, the isolates 1 and 3 were determined to decrease the production of nitric oxide (NO) and pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6). Based on these results, C. lanatus vines extract could be potentially applicable as whitening and anti-inflammatory ingredients in cosmetic formulations.

Protective Effects of Flavonoids from the Boehmeria quelpaertense against H2O2-Induced Cytotoxicity in H9c2 Cardiomyoblast Cells (H9c2 심근세포에서 제주모시풀(Boehmeria quelpaertense)로부터 분리된 flavonoids의 H2O2로 유도된 독성 보호 효과)

  • As a part of an infrastructure project on medicinal herb-based remedies, we conducted a phytohemical investigation of the 100% MeOH extract from the aerial part of Boehmeria quelpaertense; our findings resulted in the isolation of flavonoids (1-2), isoquercitrin (1) and hyperoside (2). The identification and structural elucidation of these compounds were based on $^1H$-, $^{13}C-NMR$, and LC ESI IT-TOF MS data. All the compounds isolated from this plant were reported for the first time. In this study, we examined the antioxidant activity of the 1 and 2 on the hydrogen peroxide ($H_2O_2$)-induced oxidative stress in a Rat Cardiomyoblast cell line (H9c2). The pretreatment of the flavonoids showed that it protects against $H_2O_2$-mediated cell death in the H9c2 cell line. Also, it decreases the intracellular reactive oxygen species (ROS) levels by the flavonoids in the $H_2O_2$-treated H9c2 cell line. These results showed that the 1 and 2 are a source of antioxidants. As a result, they might be helpful in preventing the progress of various oxidative stress mediated diseases, including myocardial infarction.

Antibiotics Resistance and Molecular Analysis of Enterococcus Isolated from the Han-river in Korea (한강에서 분리한 장구균의 항생제 내성과 분자생물학적 분석)

  • Identification was performed in March 2008 for the 76 Enterococcus strains isolated from the Han-river, which is used as water supply for Seoul citizens. The antibiotic susceptibility, antibiotic resistant structural analysis, trans-conjugation, pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST) were also carried out for the isolated strains. Among the isolated strains, 25 strains were E. casseliflavus, 4 strains were E. faecalis and 1 strain was E. hirae. Investigation of antibiotic susceptibility indicated that 15 strains demonstrated tolerance against vancomycin, and that 11 strains of E. faecium and 4 strains of E. casseliflavus were VRE. The vanA gene detection of the VRE strains revealed that 6 E. faecium strains were vancomycin-resistant Enterococcus faecium (VREF) possessing vanA. Analyses of transposon Tn1546 structure containing vanA demonstrated that Km36 and Km37 belonged to Tn1 type, Km20 and Km38 was Tn2 type, and Km39 and Km40 was Tn3 type. PFGE disclosed that among the 6 VREF strains, Km36 and Km37 exhibited equivalent subtype, while the rest 4 strains showed subtypes different to each other. MLST for the 6 VREF strains disclosed that 3 strains were ST78, while the rest 3 strains were ST18, ST192 and ST230, respectively. All these clonal complexes were derived from CC17 which has been isolated from clinical sources. 4 strains belonged to CC78, while the rest 2 strains were CC18 and CC192, respectively.

Neutron Capture Resonance Energy Identification of Indium by Time-of-Flight Method (중성자 비행시간법을 이용한 인듐의 공명에너지 동정에 관한 연구)

  • Prompt gamma ray from the natural Indium sample was measured by using an assembly of BGO($Bi_4Ge_3O_{12}$) scintillation detectors in the neutron energy region from 1 to 300 eV. The assembly was composed of pieces of BGO. The spectrometer was composed geometrically as total energy absorption detector. 46-MeV electron linear accelerator which is located at Research Reactor Institute, Kyoto University used for neutron sources from photonuclear reaction. The measurement of the neutron capture resonances was performed to below neutron energy 1 keV, because of strong X-ray effect from photonuclear reaction in Ta target and short distance from the target to an assembly of detector. The distance of neutron flight path is $12.7{\pm}0.02m$. The large neutron capture resonances were measured from 1 to 400 eV. The energy in the capture resonance was compared with the evaluated values. The large resonances were seen in the present measurement. General agreement can be seen between the present measurement and the previous evaluated data in relevant energy region. In the present study, we measured the continues resonance structure above 1 keV neutron energy region. 91.49 eV new neutron capture resonance was found in present measurement.

River Ecosystem and Floristic Characterization of Riparian Zones at the Youngjeong River, Sacheon-ci, Korea (사천시 용정천에서 하천 생태계와 하안단구 지역의 수변식물상)

  • This study is examined river naturality and vegetative composition of river riparian zones to identify their most important sources of variation. Information on plant species cover and on physical characteristics that occur at upper, medium, and low areas was collected for 30 riparian plots located throughout the Youngjeong River in Korea. The riparian areas of river banks are dominated by mixed sediment and the vegetation is composed of herbs, shrub, and trees. The floristic characterization of riparian at this river during 2015 season was identified with a total of 28 families, 72 genera, 75 species, 13 varieties, 23 associations. The vegetations of low water's edge and flood way at upper region were naturally formed various vegetation communities by natural erosion. Forty plant species were identified around the upper region, where the dominant growth form was mostly trees. The flood way vegetation at middle region was both of natural vegetation and artificial vegetation. Land uses in riparian zones river levee at low region were bush or grassland as natural floodplain. The values of cover-abundance at upper, middle, and low region were total 9.26, 7.24, and 7.56, respectively. Grasses and forbs at the Youngjeong River have similar cover-abundance values. Recent, many riparian areas of this river have been lost or degraded for commercial and industrial developments. Thus, monitoring for biological diversity of plant species of this river is necessary for an adaptive management approach and the successful implementation of ecosystem management.

The Structural and Functional Role of p53 as a Cancer Therapeutic Target (암 치료 표적으로서 p53의 구조적 및 기능적 역할)

  • The p53 gene plays a critical role in the transcriptional regulation of cellular response to stress, DNA damage, hypoxia, and tumor development. Keeping in mind the recently discovered manifold physiological functions of p53, its involvement in the regulation of cancer is not surprising. In about 50% of all human cancers, inactivation of p53's protein function occurs either through mutations in the gene itself or defects in the mechanisms that activate it. This disorder plays a crucial role in tumor evolution by allowing the evasion of a p53-dependent response. Many recent studies have focused on directly targeting p53 mutants by identifying selective, small molecular compounds to deplete them or to restore their tumor-suppressive function. These small molecules should effectively regulate various interactions while maintaining good drug-like properties. Among them, the discovery of the key p53-negative regulator, MDM2, has led to the design of new small molecule inhibitors that block the interaction between p53 and MDM2. Some of these small molecule compounds have now moved from proof-of-concept studies into clinical trials, with prospects for further, more personalized anti-carcinogenic medicines. Here, we review the structural and functional consequences of wild type and mutant p53 as well as the development of therapeutic agents that directly target this gene, and compounds that inhibit the interaction between it and MDM2.

Manufacturing of Meat Flavor Extract used for Browning Reaction (Browning Reaction을 이용한 Meat Flavor Extract의 개발)

  • Separation-concentration of sulfur containing heterocyclic compounds(SCHC) from many reaction meat flavors manufactured with Maillard reaction was carried out. Profile of SCHC was identified and analyzed by GC and GC-MSD. The results were as follows. 1. Profile of SCHC could be identified 7 kinds thiazole and 11 kinds thiophene, the major produced compounds were thiophene, thiazolidine, 4-methyl-5-thiazole ethanol. 2. In the case of SCHC, relationship between changes of reaction conditions and the kind of produced components were as same, but produced amounts appeared the difference. Producing amount of complexed SCHC and caramellike note as well as oxygen containing heterocyclic compounds were high level more than high reaction temperature and long time reaction period. 3. Producing ratio of comparative simple structural SCHC were the highest level at reaction conditions of moisture content 50%, reaction temperature 100$^{\circ}C$ and reaction time 2 hours. Reaction conditions for the revelation of reaction meat flavor were below 110$^{\circ}C$ and less than 2 hours. 4. Relationship between moisture content and reaction temperature as well as reaction time had very relative relation. From the change of moisture content and reaction conditions could be obtained the simultaneously profile. Signal presentation for production of reaction meat flavor could be from extraction-separation-concentration of SCHC through simplification of raw-materials in the flavor and seasoning food industry.

Benthic Marine Algal Flora and Community Structure of Eocheongdo in Western Coast of Korea (한국 서해 어청도 해조상 및 군집구조)

  • This study was carried out to investigate the community structure and biomass of benthic marine algae at the intertidal and subtidal zones of Eocheongdo on the western coast of Korea seasonally from July 2008 to April 2009. Total 101 species including 12 of green algae, 23 of brown algae and 66 of red algae were collected and identified. Among these species, 19 species were found throughout the year. The dominant species which contributed significantly to the total biomass were Corallina pilulifera, Gelidium elegans, Sargassum thunbergii, Sargassum fusiforme, Saccharina japonica, Sargassum honeri and Sargassum filicinum. Annual seaweed biomass in dry weight was $237.5gm^{-2}$ and maximum biomass was recorded seasonally in winter ($272.1gm^{-2}$), while minimum was recorded in summer ($204.5gm^{-2}$). The number of species and biomass of the subtidal zone were greater than those of the intertidal zone. The number of species and biomass in Eocheongdo seem to be higher than those of other islands in the western coast of Korea, due to the environmental conditions characterized by the increased transparency, and the well established rocky shore.
