The Antioxidative Effects of Oregano (Origanum majorana L.) Extracts (오레가노(Origanum majorana L.) 추출물의 항산화 효과)

  • The objective of this study was to investigate the antioxidative capacity of oregano extracts. Amount of oregano extract at which DPPH radical scavenging activity was inhibited by 50% was $21.8{\mu}g$ as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against potent free radical reactions. Total antioxidant capacities of oregano extract at the amounts of 7.5 and $15{\mu}g$ were 15.1 and 31.4 nmol Trolox equivalents, respectively. Oxygen radical absorbance capacities of oregano extract at the amounts of 0.2 and $0.4{\mu}g$ were 1.4 and 2.4 nmol gallic acid equivalents, respectively. Total phenolic contents of oregano extract at the amounts of 30 and $75{\mu}g$ were 40.5 and 83.9 nmol gallic acid equivalents, respectively. The inhibitory effect of oregano extract on lipid peroxidation was examined using rat liver mitochondria induced by $FeSO_4$/ascorbic acid. Oregano extracts at the amounts of 20 and $50{\mu}g$ decreased TBARS level by 20 and 64%, respectively. Thus strong antioxidant effects of oregano extract seem to be due to, at least in part, the prevention against free radicals-induced oxidation, followed by inhibition of lipid peroxidation.

The Effect of Powdered Herb of Aster scaber Thunb. on Antioxidant System in Ethanol-Treated Rats (참취 분말이 에탄올을 투여한 흰쥐의 항산화계에 미치는 효과)

  • The present study was conducted to investigate effect of powdered herb of Aster scaber Thunb. (chamchwi) on antioxidant system in ethanol-administrated rats. Four week-old Sprague Dawley male rats which had initial body weights of 97.10$\pm$4.50 g were randomly divided into three groups: control (ethanol treated, vitamin E-deficient group); 5% chamchwi (ethanol-treated, 5% chamchwi powder-supplemented group): 10% chamchwi (ethanol-treated, 10% chamchwi powder-suplemented group). Three groups of rats were suplemented with three experimental diets for 4 weeks and orally administrated 10% ethanol (v/v) daily via drinking water in the last experimental week. Contents of TBARS (thiobarbituric acid reactive substance). glutathione in liver and kidney and serum albumin were determined. The activities of superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) in liver and kidney were also analyzed. Relative weight of liver and spleen to body in chamchwi groups was lower than that in control group (p<0.05). The most remarkable result was that liver TBARS contents in chamchwi groups (5% chamchwi group, 46 $\mu\textrm{g}$ in MDA value; 10% chamchwi group, 35 $\mu\textrm{g}$) were significantly lower (p<0.05) than that in control group (66 $\mu\textrm{g}$). The supplement of chamchwi powder lowered the activity of manganese- superoxide dismutase (Mn-SOD), catalase in liver and GSH-px in kidney. The levels of glutathione in liver and kidney and serum albumin were not significantly different in all experimental groups (p<0.05). These results indicate that powdered herb of Aster scaber decreases lipid peroxidation and acitvity of Mn-SOD increased by alcohol-induced oxidative stress in liver of rats.

Effects of Probiotics on Antioxidant Biochemical Parameters and Antioxidant Enzymesin the Blood, Intestinal Mucosal Tissues and Liver of Broiler Chicks under High Ambient Temperature Conditions (고온기 생균제 급여가 육계의 혈액, 소장 점막 및 간 조직에서 항산화 생화학 지표 및 항산화 효소에 미치는 영향)

  • Four-d-old broiler chicks were randomly assigned to 3 groups with 9 replicates (8 birds/cage) under high ambient temperature; birds fed a basal diet (CON), a basal diet supplemented with 0.25% of probiotic complex (LPB, 1 × 106 Lactobacillus plantarum, 1 × 106 Bacillus subtilis, and 1 × 106 Saccharomyces cerevisiae) and 0.5% probiotic complex (HPB). Immediately after 28-d feeding trial, 6 birds having average body weight per group were sacrificed for evaluating the effects of probiotics on antioxidant parameters in the serum, intestine, and liver of birds. As results, serum biochemical parameters of nitrogen components including total protein, albumin, urea nitrogen, and glutathione were unaffected by dietary probiotics. In addition, serum superoxide dismutase (SOD), glutathione peroxidase (GPX), and glutathione S-transferase (GST) activities, and lipid peroxidation (MDA) were not changed by dietary probiotic supplement in birds. In the intestinal mucosa, SOD activity in the HPB group significantly (P<0.05) increased compared with that in the CON and the LPB groups. Lipid peroxidation in the HPB group significantly (P<0.05) decreased compared with that in the CON group. However, there was no statistical difference in GPX, and GST activities in the intestinal mucosa among treatment groups. In the liver, the activities of SOD, GPX, and GST, and the level of MDA were unaffected by probiotic supplement. In conclusion, 0.5% of probiotics significantly increased SOD activity and decreased lipid peroxidation in the intestinal mucosa, suggesting that probiotic complex could be potential to improve the small intestinal antioxidant capacity of bird under high ambient temperature conditions.

Effects of Salvia plebeia R. Br. on Antioxidative Enzyme Activities and Oxidative Damage in Rats Fed High-Fat and High-Cholesterol Diets (곰보배추섭취가 고지방과 고콜레스테롤 식이 랫드에서의 항산화 효소활성 및 산화적 손상에 미치는 영향)

  • The purpose of the present study was to investigate the effect of Salvia Plebia R. Br. (SP) powder on the antioxidative defense system and oxidative stress in rats which were fed a high fat high cholesterol diet. Accordingly, the rats were divided into four experimental groups which were composed of a high fat high cholesterol diet group (HF), HF diet with 5% SP powder supplemented group (PA), a HF diet with 10% SP powder supplemented group (PB), and a normal group (N). Consequently, the hepatic catalase activity of the HF group was decreased compared to the normal group (N), but it is recorded that of the PA and PB groups were significantly increased. With this in mind, the PA and PB groups resulted in the case of significantly increased activities of hepatic GSH-px and SOD. The hepatic superoxide radical and hydrogen peroxide contents of the PA and PB groups were significantly decreased, as compared to the HF group. The GOT and GPT activities of the PB group were also significantly decreased when thus compared to the HF group. Notably, the carbonyl values contents of the PA and PB groups were significantly reduced compared to the HF group. The hepatic TBARS values in the liver were significantly reduced as measured in the PA and PB groups. These results suggest that the SP powder may reduce the incidence of oxidative damage, by the activation of an antioxidative enzyme in rats fed with high fat high cholesterol diets.

Antioxidant Activities of the Mycelium and Culture Broth of Phellinus igniarius and Agrocybe cylindracea (말똥진흙버섯 및 버들송이버섯의 균사체 및 배양액의 항산화활성)

  • The fractions obtained from the mycelia and their culture broth of Agrocybe cylindracea and Phellinus igniarius were tested for the antioxidation activities. In electron donating ability test, strong activities more than 70% were observed in ethyl acetate and butanol fractions of A. cylindracea mycelium, butanol fraction of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether and ethyl acetate fractions of P. igniarius culture broth. Lipid peroxidation inhibition was observed in diethyl ether, butanol and water fractions of A. cylindracea mycelium, and water fraction of A. cylindracea induced the decrease in uric acid production. Activities of $50{\sim}60\;%$ of nitrite scavenging ability were observed in diethyl ether fraction of A. cylindracea mycelium, ethyl acetate and water fractions of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether, chloroform, butanol and water fractions of P. igniarius culture broth. In melanin biosynthesis inhibition activity measurement, water fraction of A. cylindracea mycelium and butanol fraction of P. igniarius mycelium showed twice higher activities than control. From these results, the fungi of P. igniarius and A. cylindracea had strong antioxidant activities.

Effects of Duchesnea chrysantha on Regulation of Antioxidative defense System in Rats Fed a High-fat·High-cholesterol Diet (뱀딸기풀의 항산화 활성 및 고지방·고콜레스테롤 식이 흰쥐의 항산화 방어계 조절에 미치는 영향)

  • This study was attempted to investigate the effects of Duchesnea chrysantha (DC) on antioxidative activities by in vivo. Rats were divided into four experimental groups which are composed of normal diet group (N group), high fat high cholesterol diet group (HF group), high fat high cholesterol diet with 5% DC powder supplemented group (DA group) and high fat high cholesterol diet with 10% DC powder supplemented group (DB group). Supplementation of DC powder groups resulted in increased activities of hepatic glutathione peroxidase and catalase. The microsomal superoxide radical contents of the DA and DB groups were significantly reduced compared to the high fat high cholesterol diet group. The mitochondrial superoxide radical contents of the DB group were significantly reduced compared to the high fat high cholesterol diet group. Hepatic hydrogen peroxide contents in cytosol were significantly reduced 5% and 10% DC powder supplemented group. The carbonyl values contents in mitochondria and microsome of the DA and DB groups were significantly reduced compared to the HF group. Thiobarbituric acid reaction substance (TBARS) values in liver were reduced in 10% DC powder supplemented group compared to the HF group. These results suggest that DC powder may have a strong regulatory effect in the activation of the antioxidative defense system.

Effect of Korean Traditional Tea Materials Water Extract on Hepatic Oxygen Free Radical Generating and Scavenging Enzyme Activities in Lead Administered Rats (한국전통차 재료의 열수추출물이 납투여 흰주의 간조직 중 유해 활성산소 생성과 제거효소 활성에 미치는 영향)

  • This study was designed to test the effect of Korean traditional tea materials on oxygen-free radical metabolism in lead (Pb) -administered rats. Male rats were divided into normal, Pb-control (Pb-Con) and Pb-water extract of green tea (Camellia sinensis; GT) , persimmon leaf (Diospyros kaki; PL) , safflower seed (Carhamus tinctorius: SS) , Du-Zhong (Eucommia ulmoides; EU) groups, respectively. Pb intoxication was induced by administration of lead acetate (25 mg/kg. B.W., oral) weekly. The extract was administered based on 1.26 g of raw material/kg B.W./day for 4 weeks. When the GT, PL, SS and EU were supplemented to the Pb-administered rats, hepatic lipid peroxide levels were significantly lower compared to the Pb-Con group. Hepatic cytochrom P-450 content and aminopyrine N-demethylase activity was lower in the Pb-Con group than in the normal group, whereas xanthine oxidase activity was significantly elevated in Pb-administered rats. The water extract of GT, PL, SS and EU supplementation attenuated changes in enzyme activities generating reactive oxygen species in the liver. Hepatic superoxide dismutase, catalase and glucose 6-phosphate dehydrogenase activities were significantly higher in the Pb-Con group than in the normal group, while monoamine oxidase activity also tended to increase in the Pb-administered rats. However, glutathione peroxidase and glutathione S-transferase activities, and glutathione content significantly decreased through Pb intoxication. The supplementation of GT, PL, SS and EU induced alleviation changes of hepatic antioxidant enzyme activity.

Free Radical Scavenging Activities and Inhibitory Effect on Xanthine Oxidase by Acetone Extract from Buckwheat (수원 5호 메밀 채소 추출물의 유리라디칼 소거 및 Xanthine Oxidase 활성 저해)

  • To examine the characteristics of the antioxidative property of Buckwheat components, acetone extracts from a buckwheat, Suwon 5, was fractionated using five solvents. Hexane, ethylacetate, ether, butanol and water fractions were obtained. Butanol fraction showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. It also showed the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. Spectrophotogram of butanol fraction was similar to that of rutin. Superoxide radical scavenging activity was related to the contents of rutin. Inhibitory effect of each fraction on xanthine oxidase was also measured. Butanol fraction had the strongest inhibitory effect on xanthine oxidase and $IC_{50}\;was\;3.1\;{\mu}g$. The inhibition type of butanol fraction on xanthine oxidase turned out to be a mixture of the uncompetitive and non-competitive modes.

The Oxidative Stress Induction and Response of Antioxidative Enzymes in the Large Patch-Infected Zoysiagrass I. Oxidative stress induction (라지 팻치에 감염된 잔디의 산화적 스트레스 발현과 항산화효소의 활력의 변화 I. 산화적 스트레스 발현)

  • To investigate the effect of large patch infection on oxidative stress induction, growth, $H_2O_2$ concentration and lipid peroxidation were compared between pathogen-infected and healthy (control) zoysiagrass. The sampling for leaves and roots were carried out every 2 days for a period of 6 days. Pathogen-infection increased root mortality by 30% compared to control. Dry mass was not significantly affected by pathogen-infection until day 4, but significant decreases in both leaves (-14%) and roots (-20%) were observed at day 6. The $H_2O_2$ concentration in pathogen-infected leaves rapidly increased within the first 2 days(+28%) and then slightly decreased. The increase of $H_2O_2$ in pathogen-infected roots was distinct, showing 1.7-fold higher level than control at day 6. The extent of lipid peroxidation caused by pathogen-infection continuously increased for the first 4 days. This was then stagnated until day 6. In roots, the significant increase of lipid peroxidation was observed only at day 2. These results indicate that large patch-infection induces oxidative stress, and that the oxidative stress responsive pattern was plant organ specific.

Effect of Monascus purpureus-Fermented Korean Red Ginseng Powder on the Serum Lipid Levels and Antioxidative Activity in Rats (홍국발효홍삼 분말 급여에 의한 흰쥐의 혈청 지질 농도 및 항산화 활성에 미치는 영향)

  • Monascus-fermented Korean red ginseng (MFRG), fermented by Monascus purpureus KCCM 12002, may be an ideal candidate for the hepatoprotectic, hypolipidemic and antioxidative activities. Effect of MFRG powder on these parameters in rats was investigated. Body weight gain, food intake, and water consumption were not significantly different among the groups. Total and relative weights of liver were significantly higher in MFRG group than that in other groups. The activities of AST and $\gamma$-GTP were highly lowered in MFRG group compared to control group. Contents of serum total lipid and triglyceride were significantly lowered in silymarin group and were significantly increased in MFRG group compared to control group, but tended to be lowered in RG group. Serum content of total cholesterol tended to be lowered in silymarin, RG, and MFRG groups compared to control group. HDL-cholesterol contents was only significantly increased in MFRG group compared to control group. At the same time, atherogenic index (AI) was also significantly lowered in silymarin, RG and MFRG groups compared to control group, and this effect was more pronounced in MFRG group. Content of thiobarbituric acid reactive substances (TBARS) in the liver was significantly lowered in MFRG group and tended to lowered in silymarin and RG groups compared to control group. The hepatic glutathione concentration was significantly higher in silymarin and MFRG groups. Hepatic morphology in all experimental groups revealed clear-cut hepatic lobules with the uniform pattern of the polyhedral hepatocytes radiating towards the periphery from the central vein. These results suggested that MFRG may have anti-atherogenic index (AI) and antioxidative activity in normal dietary feeding rats.
