Cortisol is present in high concentration in the ovary and its receptor is expressed in the ovarian cells. Moreover, cortisol is known to have a role in steroid synthesis and cell metabolism in human granulosa and lutein cells. However, little is known of the role of cortisol presenting in high concentration in the follicles after LH surge on the granulosa-lutein cells. Therefore, the this study we evaluated the apoptosis and the production of progesterone $(P_4)$ and estradiol $(E_2)$ in the granulosa-lutein cells that are obtained during oocyte-retrieval after treatment with 5, 50, and $500{\mu}g/m\ell$ cortisol and 1 IU/$m\ell$ FSH. Results of DNA fragment analysis and TUNEL assay demonstrated that DNA fragmentation and the rate of apoptotic cells were increased in a dose-dependent manner showing a significant increase in 50 and $500{\mu}g/m\ell$ cortisol treated cells. We found, however, that FSH did not suppress the apoptosis of the cells induced by cortisol. In the results of chemiluminescence assay for $P_4$ and $E_2$, $P_4$ production was decreased by cortisol treatment, whereas $E_2$ was not changed. We also demonstrated that FSH did not inhibit the suppressive effect of GnRH on $P_4$ production as the result of apoptosis. The present study suggests that cortisol of high concentration could cause the apoptosis of human granulosa-lutein cells by suppressing the production of $P_4$. However, we need more studies to elucidate the mechanism by which cortisol induces apoptosis in human granulosa-lutein cells in view of the fact that our results are inconsistent with previous reported data.
This study was undertaken to observe the histopathologic changes in submandibular glands of the white rats when exposed to megavoltage fractionated dose of CLINAC 2100 C-D 6 MV X-RAY irradiation and 42 female white rats, weighing approximately 100gm, were divided into control and 2 experimental groups. At sacrifice, submandibular glands were excised and examined microscopically and electromicroscopically. The results were as follows : 1. The acinar cells of submandibular gland showed damage varied with dose, 12 Gy resulted in very mild injury while 24 Gy caused extensive injury. 2. The acinar cells of sumandibular gland showed similar ultrastructual alterations, appeared as pleomorphic nucleus, decreased numbers and pleomorpgism of secretory granules, distention of rough endoplasmic reticulum, expansion and pallor appearance of mitochondria, and hypertrophy of Golgi complex. 3. A serous cells were the most sensitive components, displaying morphological alterations of radiation damage as early as 3 hours, followed by submandibular seromucinous cells and secretory tubular cells. 4. The mucous cells, as well as the whole ductal lining cells, displayed no significant alterations. 5. No evidence of microvascular injury through whole experimental groups indicated that microvascular impairment dose not contribute to early. salivary gland injury.
An immunohisochemical study on the cerebral ganglion of the African giant snail, Achatina fulica. was conducted by applying the AB/AY staining and the avidin-bovine-peroxidase complex staining methods. The followings are the results obtained throughout the study. The cerebral ganglion of Achatina fulica is an ellipsoidal body of 2 x 1 mm in size, which is connected by the cerebral commissure of 1 mm in diameter. The cross-section through the cerebral ganglion, shaped like a butterfly, is divided into the medio-dorsal parts, the latero-dorsal parts, the caudo-dorsal parts, and the lateral lobes. In the medio-dorsal and latero-dorsal parts, the LG cells and the DG cells are found mixed, although the LG cells are dominant. In lateral lobe, however, the Y cells are quite dominant, while the LG cells and the DG cells are seldom found. The LG cells are 20-70 $\mu\textrm{m}$ in sizes and circular or ellipsoidal in shapes. They are stained light green with the AB/AY. 1 - 3 nucleoli are found in karyolymph, where granular chromantins are evenly distributed. In cytoplasm, it is found that the secretory granules are evenly developed.

The mucosal folds of the stomach are regular, and the muscularis consists of longitudinal, oblique and circular muscle layer. The chief cell of the gastric gland have a tubular mitochondria, endoplasmic reticula and numerous secretory granules in electron-dense. However, parietal cell contains small mitochondria, endoplasmic reticula and vacuoles in low electron density. Mucosal epithelium of the pyloric caeca and intestine composed of columnar epithelium, goblet cell, rodlet cell and dark cell. Columnar absorptive cell in the pyloric caeca and intestine contains well developed mitochondria, endoplasmic reticula, vesiculated granules in high electron density, pinocytotic vesicles and multivesicular body. Rodlet cell have a well developed cytoplasmic capsule and the endoplasmic reticula in the cytoplasm. Dark cell showing a high electron density in the cytoplasm and contains well developed mitochondria. Columnar epithelium of the intestine have a well developed intercellular junction and the microvilli which contains actin filament originated from the cytoplasm. Mucosal epithelium of the intestine have a longer microvilli and more abundant goblet cells than in the pyloric caeca.
This study was carried out to determine whether dermal mast cell responses to Parasoninn westemoni in an abnormal host, the mouse, were dependent on the site of metacercarial inoculation. In mice during subcutaneous infection, the number of der- mal mast cells were increased significantly (p<0.05) at the first week ($38.3/\textrm{mm}^2$) and then persisted at a high level until the sixth week ($45.2/\textrm{mm}^2$) of infection compared with PBS- injected (control) mice (range: $19.4-25.1/\textrm{mm}^2$). In mice during oral infection, the number of dermal mast cells were increased significantly (p<0.05) at two weeks ($33.5/\textrm{mm}^2$) after infection and remained at these levels thereafter compared loth non-infected (control) mice (range: $17.4-22.3/\textrm{mm}^2$). In mice both during subcutaneous and oral infection, the recruited dermal mast cells showed extensive degranulation at the second week (68.4%) and 60.7%, respectivelyl, reached a peak at the third week (81.4%, and 92.1%, respectively) and then declined slightly thereafter. By contrast, in both control mice, about 10% of dermal mast cells were degranulated. In conclusion, this study suggests that dermal mast cell responses to p. westemcni in mice are dependent on cutaneous sensitization by larval excretory-secretory antigens, irrespective of infection route.
Ethanol extracts from Rhododendrom brachycarpum and Abies koreana were investigated for their anti-allergic activities using RBL-2H3 cell line. After treatment with ethanol extracts of various concentrations on the immune response induced mast cell by concanavalin A (Con A), the expressions of cytokine interleukin-4 (IL-4), interleukin-13 (IL-13) were determined by using RT-PCR and the degranulation of mast cells was determined by measuring ${\beta}$-hexosaminidase release. Expression level of IL-4 was decreased by the extract from Rhododendrom brachycarpum in $10^{-7}$, $10^{-5}$ and $10^{-3}%$ concentrations. Expression level of IL-13 was also decreased by both extracts. ${\beta}$-Hexosaminidase release by RBL-2H3 cells was inhibited at the $10^{-5}$ and $10^{-3}%$ concentration of extracts from Rhododendrom brachycarpum and Abies koreana, respectively. These results demonstrate that ethanol extracts of Rhododendron brachycarpum and Abies koreana exert anti-allergic effects by regulating the reduction of IL-4 and IL-13 genes expression and also the secretion of ${\beta}$-hexosaminidase.
Cardiovascular disease (CVD) is increasingly increasing as the main cause of death worldwide, and activation of platelet in vascular damage is one of the important causes of CVD. In recent, there is a growing interest in anti-thrombotic materials through platelet suppression, and efforts are being made to reduce side effects by using natural bioactive compounds. Known as one of the Flavonoids, hydroxygenkwanin (HGK) is a purified substance in Daphne Genkwa, which is known to have antibacterial, anti-inflammatory and anti-cancer effects, and has been reported to serve as an inhibitor of tissue factor that prevents thrombosis, but its anti-platelet effects and the action mechanisms is not known. In this study, we confirmed that the effects of HGK on the collagen-induced human platelets activation. HGK suppressed phosphorylation of PI3K/AKT and mitogen-activated protein kinases during platelet signaling, and reduced granule secretion in platelets such as ATP and serotonin. In addition, HGK inhibited the phosphorylation of cPLA2 and strongly undermined the production of TXA2, which is a powerful aggregation amplifier. As a result, the platelet aggregation derived by Collagen, a cohesive induced substance, was strongly suppressed by HGK to an IC50 of 86.36 µM. Therefore, HGK might be worth the antithrombotic substance that inhibits the activation and aggregation of human platelets that occur through blood vessel damage.

비만세포의 한 종류인 rat basophilic leukemia (RBL-2H3) 세포를 이용하여 교계사물탕-가감의 항알레르기 효과를 확인하고자 하였다. Phorbol 12-myristate 13-acetate (PMA)와 calcium ionophore A23187을 이용하여 RBL-2H3 세포를 자극한 후 세포의 탈과립 정도를 $\beta$-hexosaminidase assay로 확인한 결과, 전 처리한 교계사물탕-가감의 농도 의존적으로 탈과립을 억제하였다. Pro-inflammatory cytokines인 tumor necrosis factor (TNF)-alpha와 interleukin(IL)-4의 분비량을 enzyme-linked immunosorbent assay (ELISA)로 확인한 결과 교계사물탕-가감의 농도 의존적으로 감소하였으며, 이들 cytokines와 염증 반응에 주요한 인자인 cyclooxygenase (COX)-2 의 mRNA 발현 정도 역시 교계사물탕-가감에 의해 감소함을 확인할 수 있었다. 이러한 실험 결과로 보아 교계사물탕-가감은 알레르기 관련 질환의 치료에 응용될 수 있을 것으로 사료된다.
Three fertilization envelopes (FE) have been observed after the artificial insemination of U. unicinctus oocytes. The substances of the first fertilization envelope, which is an effective barrier against excessive sperm, come mainly from the surface coat of the oocyte. The secretions of the cortical granules take part in formation of the 2nd fertilization envelope. Histologically, the 3rd fertilization envelope is not amorphous as seen under light microscope, but contains numerous panicles under electron microscope, which would be contributed to harden the envelope by 60 min after the fertilization. With the substantial similarity between the 1st fertilization envelope and the surface coat of the oocyte, and the coincidence of retraction of microvilli and the formation of the 1st fertilization envelope, it is suggested that the microvilli contain the sperm receptors in U. unicinctus. Some granular substances from the distal part of the acrosome diffuse on the surface coat of the oocyte while the acrosomal tubules penetrate into the surface coat. The acrosomal tubules arise from the proximal part of the acrosome and pass through the acrosomal lumen.

This study has been carried out to understand histochemical and ultrastructural changes by acupuncture stimulation at the acupoint of the Zusanli (St. 36) in the rat. A lot of mast cells are observed in the peripheral of the hole with a sparrow-pecking and twistin -needle manipulation to the acupoint Zusanli for induction of 'Qi'. These cells contained a lot of granules with varied electron density and exocytosis granules were observed near the mast cell. H-bands of the muscle fiber that situated near the hole were shortened. It is assumed that these muscles are contracted by acupuncture stimulation. These results imply that functional relationship between mast cells in the dermis and Qi-sensation induced by acupuncture plays an important role on the specific receptor response to the mechanical stimulation.
