  • The Pichia pastoris expression system is widely used for e production of recombinant proteins. In this study, we investigated the characteristics of cell growth and PLC production of recombinant Pichia pastoris. Especially, severed carbon sources like glycerol, glucose, sucrose, mannose, dextrose, xylitol, lactic acid, and acetic acid etc. were used to fermentations. The PLC activity was measured photometically.

  • In order to maximize the cell growth and the polysaccharide production in Agaricus blazei, two kinds of fed-batch fermentation processes were performed with varying the feeding medium compositions and the feeding process. The relationship between dissolved oxygen and polysaccharide production in batch fermentation was applied to fed-batch fermentation. The biomasss concentration was 18.2 g/L and the polysaccharide production was 10.4 g/L.

  • The culture conditions of chinese cabbage medium for the growth of Bacillus lentimorbus G-74, having antifungal activities, were optimized. The optimal growth of B. lentimorbus G-74 was obtained when the concentration of chinese cabbage extract was 50%(v/v). The optimum temperature and pH for the cell growth of B. lentimorbus G-74 were $35^{\circ}C$ and 7.0, respectively.

  • Effect of Pluronic F-68, a nonionic surfactant, on the extracellular production of hGM-CSF in transgenic Nicotiαna tabacum cell suspension culture was investigated. The addition of 5 g/L Pluronic F-68 did not affect the cell growth but increased the extracellular production of hGM-CSF by two-fold. This may be due to the enhanced permeability of the cell membrane by the interaction between the Pluronic F-68 and the cell membrane.

  • In this work, effects of trehalose on the recovery of bioluminescence and viability of luxAB-containing recombinant bacteria were investigated. In case of a recombinant, UV2, only 2.5% of bioluminescence and 2.7% of cell viability were restored after 3.5hr of freeze-drying without trehalose, which implies that cells were heavily damaged during the dehydration. To improve these losses, trehalose was added before freeze-drying on different modes. Trehalose increased the bioluminescence and viability of freeze-dried UV2 in all conditions tested and it was observed that addition of trehalose into the broth(final concentraion, 0.08M) for 15min before the freeze-drying resulted in 45% of bioluminescence and 50% of cell viability. For the other luminescent recombinant, YH9, trehalose showed a similar efficacy.

  • Calcium carbonate $(CaCO_3)$ bead immobilized with alginate were developed as buffer system to enhance the cultivation efficiency of bifidobacteria. When Bifidobacteriuim longum KCTC 3128 and HLC 3742 were independently cultivated in 2.5-liter fermenter buffered the $CaCO_3$ bead, NaOH, $Na_2CO_3$, and $NH_4OH$. The proliferation of bifidobacteria and their storage stability were higher in culture broth buffered $CaCO_3$ beads than in culture broth buffered with NaOH, $Na_2CO_3$, and $NH_4OH$. Therefore, $CaCO_3$ bead may be useful as a buffer to enhance of the cultivation efficiency and viability of bifidobacteria.

  • Effect of immobilization on the production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) by Nicotiana tabacum cells was investigated using polyurethane foam as immobilization matrices. The cell activity and the hGM-CSF production were maintained for 16 days in spite of 3 times of media exchange. Under the same conditions of temperature and agitation rate, maximum concentrations of hGM-CSF in a 500-mL spinner flask and 100-mL Erleuneyer flasks were 17.3 ${\mu}g/L$ and 9.8 ${\mu}g/L$, respectively. Consequently high hGM-CSF production could be possible in spinner flask when the rate and amount of media exchange were optimized.

  • Effects of ultrasound on cell growth and the production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) were investigated using transgenic Nicotiana tabacum cell suspension cultures. The culture suffered a slight growth depression immediately after the sonication, but gradually recovered to normal growth within 2 days. When the cells were exposed to ultrasound, the level of secreted hGM-CSF was 2.14 times higher than that in normal condition. From the beginning to 6 days of culture, production of secreted hGM-CSF was higher than that of control and then decreased. At the end of culture, however, hGM-CSF was considerably increased up to 36.7%. In the case of intracellular hGM-CSF, the level was slightly higher than that obtained in normal condition. Total hGM-CSF production was 31.5% higher than that of control culture after 6 days. The highest amount of hGM-CSF was $34.9\;{\mu}g/L$.

  • In this study, we demonstrated that high cell density for Thiobacillus ferrooxidans could be obtained when optimal conditions for cell growth were maintained using electrochemical cultivation with sufficient aeration. The optimal pH for cultivation were determined to be $2.0{\pm}0.05$. When the current and potential were set to 0.15A, 4V, the Pt electrode reduced $Fe^{3+}$ to $Fe^{2+}$ with efficiency of 85%. Under these condition, cells at an initial density of 0.0025 g-dry cell/L grew for 8days until the cell density was 0.0576 g-dry cell/L. this was a 7-fold increase over conventional batch culture.

Optimum culture conditions of cell growth and polysaccharide production by Paecilomyces japonicain batch culture

  • To examine effects of agitation and aeration as well as adding of glucose and yeast extract on cell growth and polysaccharide production by Paecilomyces japonica, batch culture was carried out at 5L jar fermenter at $27^{\circ}C$ with the initial pH 7 for 7 days cultivation(innoculum size 2%, working volume 3L). Media compositions(g/L) were 30 glucose, 20 yeast extract, 0.5 $KH_2PO_4$, $0.1\;CuCl_2\;{\cdot}\;2H_2O$. Optimum culture conditions of agitation and aeration in batch culture were 400 rpm and 1.0 vvm, resulting in 23.1 g/L biomass and 2.5 g/L polysaccharide. Additional feeding of glucose and yeast extract with a pulse mode conferred an advantage on cell growth and polysaccharide production with showing the results of 29.2 g/L and 3.3 g/L, respectively.
