Assessment of Environmental Flow Impacts for the Gosam Reservoir According to Climate Change (기후변화에 따른 고삼저수지의 환경유량 영향평가)

  • This study conducted a quantitative assessment on the environmental flows associated with climate change in the Gosam Reservoir, Korea. The application of RCP 8.5 climate change scenario has found that the peak value of High Flow Pulses has increased by 36.0 % on average compared to historical data (2001 ~ 2010), which is likely to cause disadvantage on flood control and management but the increase in peak value is expected to make a positive impact on resolving the issue of green algal blooms, promoting vegetation in surrounding areas and encouraging spawning and providing habitats for native species by releasing a larger amount of landslides as well as organic matters than the past. However, the decreasing pattern of the peak value of High Flow Pulses is quite apparent with the trend of delay on the occurrence time of peak value, necessitating a long-term impact analysis. The peak value of Large Floods shows a clear sign of decrease against climate change scenario, which is expected to lead to changes in fish species caused by degraded quality of water and decreasing habitats. A quicker occurrence of Small Floods is also expected to make an impact on the growth cycle of aquatic plants, and the reduction in occurrence frequency of Extreme Low Flows is to contribute to increasing the population of and raising the survival rate of native fish, greatly improving the aquatic ecosystem. The results of this study are expected to be useful to establish the water environment and ecological system in adapting or responding to climate change.

The Inhibitory Effects of GHJ on Allergic Inflammatory Response in Human Mast Cells (HMC-1) (고삼, 형개, 자초 혼합물(GHJ)의 인간비만세포에서의 항염증 효과)

  • Objectives : This study aimed to evaluate inhibitory effects of GHJ on allergic inflammatory response in human mast cells (HMC-1). Methods : To investigate the inhibitory effect of GHJ (62.5, 125, 250, 500, $1000{\mu}g/mL$), HMC-1 cells were stimulated with phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). Enzyme-linked immunosorbent assays (ELISAs), RT-PCR and Western blot analysis were investigated using GHJ extract. Results : GHJ inhibited levels of $TNF-{\alpha}$ and IL-6 of $1000{\mu}g/mL$ concentration in ELISA and mRNA expression. GHJ had inhibitory effects in level of MAPKs, $p-I{\kappa}B-{\alpha}$ and p-NF-kB also. GHJ attenuated Compound 48/80-stimulated histamine release. In addition, GHJ inhibited PCA reaction in vivo. Conclusion : This study indicated that GHJ extract can inhibit allergic responses in HMC-1 cell.

Screening of Insulin-like Substances from Traditional Herbs of Diabetes Prescription in Donguibogam (동의보감 당뇨 처방에 사용되는 한약재에서 인슐린성 물질(Insulin-like substances)의 탐색)

  • In order to search for insulin-like substances from the constituted herbs of Sogal prescriptions, we selected 19 traditional herbs, based on a review of the Donguibogam. The effects of the hot-water extract from the selected herbs on the proliferation and the differentiation of 3T3-L1 fibroblasts were tested. The various water-extracts from Pinellia ternata, Magnolia obovata, Rheum palmatum, Acanthopanax sessiliflorun, Atractylodes japonica and Strychnos ignatii inhibited the proliferation of 3T3-L1 fibroblasts, did not influence entirely in differentiation of 3T3-L1 fibroblasts. Treatment of 3T3-L1 fibroblasts with the extract from Ephedra sinica, Trichosanthes kirilowii, Scrophularia buergeriana and Sophora flavescens significantly increased the differentiation of the cells. In conclusion, these may contain such compounds that play a role of insulin-like action.

  • The winter cherry bug (Acanthocoris sordidus Thunberg, Coreidae, Hemiptera) is a insect pest that have a piercing sucking type mouthpart, and it is injured to solanaceous plants including pepper plant. For the control of the winter cherry bug, 20 organic synthetic insecticides and 7 environmental friendly agricultural materials (EFAMs) were selected with single formulation. Fenitrothion, fenthion and phenthoate were significantly higher mortality to adults and 3rd instar nymphs of winter cherry bug as over 95 and 100 % after 72 hours with treatment. Otherwise, in case of EFAMs, there is no significantly higher effective materials to adults and 3rd instar nymphs of winter cherry bug. Eighty percentage of Sophora flavescens was showed only around 50% mortality with the high variation after 72 hours with treatment against adult of the winter cherry bug.

  • Objective : This study was performed to investigate the effects of Sophorae Radix and Coptidis Rhizoma-iontophoresis in allergic contact dermatitis. Methods : Contact hypersensitivity assay, video microscope, melanin-erythema levels, pH levels, hydration levels, WBC count, RBC count, neutrophil ratio, lymphocyte ratio and total IgE levels were measured. Results : At contact hypersensitivity assay, the right ear swellings in Sophorae Radix and Coptidis Rhizoma-iontophoresis group were decreased compared with control group, but have no statistical significance. At observation of skin morphologic change, many papules were seen in control group and keratins were seen in Sophorae Radix-iontophoresis group. At melanin levels, Sophorae Radix and Coptidis Rhizoma-iontophoresis groups showed no significant difference compared with control group. At pH levels, Sophorae Radix-iontophoresis group showed significant decreased with control group. At hydration levels, Sophorae Radix and Coptidis Rhizoma-iontophoreses groups showed to significant difference compared with control group. WBC count, RBC count and neutrophil ratio were significantly increased in Sophorae Radix and Coptidis Rhizoma-iontophoresis group compared with control group. Lymphocyte ratio and total IgE levels were significantly decreased in Sophorae Radix and Coptidis Rhizoma-iontophoresis groups compared with control group. At morphology of skin, inflammation was decreased and the thickness of epidermis was well preserved in Sophorae Radix and Coptidis Rhizoma-iontophoresis group. Conclusion : Sophorae Radix and Coptidis Rhizoma-iontophoresis had some anti-allergy, anti-inflammatory effects on allergic contact dermatitis, but had no effects on melanin-erythema levels, pH levels and hydration levels of skin.

Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT. (고삼 수집종의 생육특성, 주요성분 및 항산화 활성)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.

Effects of Apoptosis of Sophorae Radix on Human Gastric Adenocarcinoma cells (인체 위암세포에서 고삼의 세포사멸효과)

  • Objective : The purpose of this study was to investigate the anti-cancer effects of Sophorae Radix and the effects of 5-Fluorouracil (5-FU) in human gastric adenocarcinoma cells (AGS). Method : We used human gastric adenocarcinoma cell line, AGS cells. We examined cell death by MTT assay and caspase 3 assay with Sophorae Radix. To examine the inhibitory effects of Sophorae Radix, cell cycle (sub G1) analysis was done the AGS cells after three days with Sophorae Radix. The reversibility of Sophorae Radix was examined on one day to five days treatment with 100 ${\mu}g/ml$ Sophorae Radix. Result : Sophorae Radix inhibited the growth of AGS cells in a dose-dependent fashion. Also we showed that Sophorae Radix induced apoptosis in AGS cells by MTT assay, caspase 3 assay and sub-G1 analysis. Sophorae Radix combined with 5-FU markedly inhibited the growth of AGS cells compared to Sophorae Radix or 5-FU alone. After 3 days treatment of AGS cells with Sophorae Radix, the fraction of cells in sub-G1 phase was much higher than that of the control group. Conclusion : Our findings provide insight into unraveling the effects of Sophorae Radix in human gastric adenocarcinoma cells and developing therapeutic agents against gastric cancer.

Effects of Sophorae Radix on Human Breast Adenocarcinoma Cells (고삼의 인체 유방암세포에 미치는 효과)

  • We investigated the inhibitory effect of an oral administration of a Sophora flavescens Aiton ethanol extract (SFE) on the development of atopic dermatitis (AD) by using NC/Nga model mice. The induction of atopic dermatitis-like lesion was conducted by the removal of the back hairs and topical application of a mite antigen (Dermatophagoides farinae, Df) on to the back skin twice a week for 8 weeks. SFE was orally administered at a different doses (100-400 mg/kg). Atopic dermatitis-like skin lesions were evaluated by dermatitis scores, skin histology and immunological parameters (serum levels of IgE, TARC/CCL17, MDC/CCL22, and CTACK/CCL27). Oral administration of SFE significantly inhibited the clinical sign of Df-induced atopic dermatitis, including dermatitis score and leukocyte infiltration. Moreover, SFE suppressed significantly the serum IgE and Th2 chemokine (TARC/CCL17, MDC/CCL22, and CTACK/CCL27) levels in a concentration dependent manner. These results suggest that oral administration of SFE could reduce significantly the clinical signs and Th2 chemokines in Df-induced atopic dermatitis model mice. Therefore, SFE may be effective substances for the management of AD in human.

Enhancement of Angiogenesis by Sophorae Radix in Full-thickness Skin Wound of Rats (흰쥐의 피부상처 동물모델에서 고삼(苦蔘)이 신생혈관형성에 미치는 영향)

  • Wound healing is the restoration in injured skin tissue and one of the most important therapeutic targets. The process consists of inflammation, proliferation, and remodeling. There have been reported multiple factors that accelerate and delay wound healing. In this study we tested the hypothesis that Sophorae Radix extract would improve the recovery of wound healing after full-thickness skin wound in rats. Twenty young male Sprague-Dawley rats were used for the studies. A full-thickness skin wound was made on the dorsal skin of the rats. Either Sophorae Radix water extract (SR) or saline (Control) was orally administrated every day. The wound area was measured and the percentages of wound contraction, wound healed and wound epithelization were evaluated. After 15 days, the skin tissues were excised and examined by histopathological and immunohistochemistrical method. In results, wound area in SR group was significantly decreased to compared with Control. SR group showed the significant enhancements in the percentages of wound contraction, wound healed and wound epithelization. Histopathological examination revealed that SR induces neo-vascularization potential in wound healing process. SR treatment in rats significantly accelerated cutaneous wound healing in the angiogenesis process by increasing VEGF and TGF-${\beta}1$ synthesis. These results suggest that Sophorae Radix enhance skin wound repair by increasing the angiogenic agents such as VEGF and TGF-${\beta}1$.
