Comparative Effect of ${\gamma}$-Irradiation and Ozone Treatment for the Improvement of Hygienic Quality of Dried-Angelica Keiskei Koidz Powder (신선초 분말의 위생화를 위한 오존처리와 감미선 조사와의 비교 효과)

  • For the purpose of improving hygienic quality of dried-Angelica Keiskei Koidz powder, the effect of ozone treatment and gamma irradiation on the microbial decontamination and physicochemical properties were investigated. Gamma irradiation at 5 to 7.5 kGy resulted in sterilizing total aerobic bacteria, molds and coliforms below detective levels, while ozone treatment for 8 hours up to 18 ppm did not sufficiently eliminate the total aerobic bacteria of the sample. The physicochemical properties of the sample were not changed by gamma irradiation up to 7.5 kGy, whereas, ozone treatment caused remarkable changes in pH, TBA value, chlorophyll, carotenoid and fatty acid compositions. Therefore, this investigation demonstrated conclusively that gamma irradiation was more effective than ozone treatment for decontaminating and sterilizing the dried-Angelica Keiskei Koidz powder, with minimal effect on the physicochemical properties analyzed.

A Study on the Analysis of Five Artificial Sweetners in Beverages by HPLC/MS/MS (HPLC/MS/MS를 이용한 음료류 중 인공감미료 동시분석에 관한 연구)

  • A method for analysis of five artificial sweetners (sodium saccharin, aspartame, acesulfame-K, sucralose, cyclamate) in beverage samples was developed using high-performance liquid chromatography/triple quadrupole mass spectrometry (HPLC/MS/MS). The method uses a single-step dilution for sample preperation. Seperation was achieved on a $C_{18}$ column ($2.1{\times}150mm$, $3.5{\mu}m$) with A- 2% methanol (1 mM ammonium acetate), B-95% methanol (1 mM ammonium acetate) as mobile phase with gradient mode. The quantitation of target compounds was performed by external calibration in selected reaction monitorning (SRM) mode. The coefficient of determination of calibration curve for sodium saccharin, aspartame, acesulfame-K, sucralose and cyclamate were 0.9957, 0.9991, 0.9943, 0.9982 and 0.9948, respectively. The limits of detection (LODs) and limits of quantitation (LOQs) were in the range of 0.001~0.022 mg/L and 0.004~0.073 mg/L, repectively. Recoveries for beverage samples were in the range of 92.76~113.50% with RSD < 10.91%. The method has applied to the determination of the five sweetners in 102 beverage samples. Three artificial sweetners-aspartame, acesulfame-K, sucralose were detected from 42 samples. Sodium saccharin and cyclamate were not detected in all samples.

Monitoring of Sugar and Low-Calorie Sweetener Content in Alcoholic Beverages (유통 주류의 당류 함량 및 저칼로리 감미료 사용 실태 조사)

  • This study investigated the total sugar and low-calorie sweetener content in 72 alcoholic beverages. The samples included 10 takjus, 6 yakjus, 5 cheongjus, 5 beers, 12 fruit wines, 5 sojus, 5 general distilled alcoholic beverages, 9 liqueurs, and 15 other alcoholic beverages. Sugar and allulose content were analyzed using HPLC-RI, and the content of five sweeteners was analyzed using HPLC-UV and LC-MSMS. The average sugar content in the alcoholic beverages was 4.13±5.16 g/100 g. When categorized by type, the sugar content ranged from 0.00 to 8.92 g/100 g, 0.00 to 30.55 g/100 g, and 0.14 to 17.02 g/100 g in fermented (takju, yakju, cheongju, beer, and fruit wine), distilled (soju, general distilled alcoholic beverages, and liqueur), and other alcoholic beverages, respectively. Sugar content was the highest in liqueur, with the average content being 12.41±9.66 g/100 g. Among low-calorie sweeteners, acesulfame potassium, sodium saccharin, aspartame, and sucralose were detected in concentrations ranging from 28.6-121.5, 42.3, 34.1-141.5, and 23.3-88.1 mg/kg, respectively. Cyclamate and allulose were not detected in any of the alcoholic beverages. Sweeteners were detected in 24 out of the 72 alcoholic beverages, and their content complied with the standards stipulated for food additives.

Effect of Irradiation on Growth, Bulblet Formation, and Germination of Pollen and Seed of Several Lily Cultivars (나리 생장, 자구 발달 및 화분과 종자발아에 미치는 방사선의 영향)

  • To build up data for mutation breeding of lily, sensitivity to irradiation of bulb, seed and pollen was investigated. The bulbs of seven cultivars including 'New Wave' were very sensitive to gamma rays. Non-irradiated bulbs produced plants with bloom as normal. However, irradiated bulbs showed retarded growth and did not flower. Bulbs of 'New Wave' and 'Tiny Dino' irradiated with 125Gy gamma rays survived, but their growth rate dropped considerably. Part or whole leaves of the other cultivars except 'New Wave' and 'Tiny Dino' withered. Moisture content of individual scale was 72~78% depending on cultivar. Bulblet formation from untreated scales was 100%. Among all irradiated scales only 'Siberia' scales showed 8.5% bulblet formation when irradiated with 50Gy gamma rays. Pollen germination as affected by gamma ray irradiation had no consistent tendency. When dosage of $F_1$ 'Augusta' seeds reached 300Gy, their survival ratio was drastically decreased. Bulblet formation in vitro was observed only in non-irradiated seeds. Lethal dose ($LD_{50}$) of seed was less than 100Gy. Height and width of $F_1$ 'Augusta' bulblets in vitro irradiated with gamma rays decreased as dosage increased regardless of the culture method. The percentage shoot formation was 100% in non-irradiated bulblets and 15~60% in irradiated ones.

  • $\text\tiny{D}$-Psicose 3-Epimerase (DPE)는 $\text\tiny{D}$-Fructose의 C3 Epimerase로써 $\text\tiny{D}$-Fructose를 $\text\tiny{D}$-Psicose로 전환해 주는 효소이다. $\text\tiny{D}$-Psicose는 설탕 대신 사용하는 감미료로 몸에 흡수되지 않아 칼로리가 없다고 알려져 있고 자연에서는 오로지 DPE에 의해서만 생산되는 희귀당이다. 이에 따라 DPE를 통한 $\text\tiny{D}$-Psicose 대량생산의 필요성이 대두되고 있는 등 이 분야에 대한 관심이 뜨거운 실정이다. 본 연구팀은 이 당과 관련된 작용기작 연구를 수행하기 위하여 아직 단백질 3차구조가 알려지지 않은 Clostridium hylemonae DPE (chDPE) 단백질의 3차 구조예측 연구를 수행 하였다. 우리는 HHsearch를 이용하여 agrobacterium tumefaciens의 DPE 외 2개의 구조를 호몰로지 모델링 연구를 위한 주형으로 선정하였다. 다음으로 PROMALS3D를 이용하여 주형들과 chDPE의 multiple sequence alignment를 수행하였고 이를 바탕으로 3차구조 예측 연구를 수행 하였다. 예측된 구조를 검증하기 위하여 ProSA와 Ramachandran plot분석을 이용하였고 Ramachandran plot에서 단백질의 94.8%에 해당하는 잔기들이 favoured regions에 위치하였다. ProSA에서는 Z-score값이 -9.3으로 X-선 결정학이나 핵자기 공명법으로 밝혀진 구조들에서 관측되는 범위 내에 위치하였다. 나아가 예측된 구조에 $\text\tiny{D}$-Psicose와 $\text\tiny{D}$-Fructose의 결합모드를 규명하기 위하여 도킹을 시도하였다. 이번 연구를 통하여 chDPE의 구조를 예측 할 수 있었고 이를 바탕으로 이 단백질의 기능을 이해하는데 도움을 줄 것으로 기대된다.

Effect of Heat Treatment on Mechanical Properties of Cross-Linked Ultra-High Molecular Weight Polyethylene Used for Artificial Joint Liner (인공관절 라이너용 가교된 초고분자량폴리에틸렌(UHMWPE)의 열처리 조건에 따른 기계적 특성 변화)

  • The mechanical characteristics of gamma-ray irradiated UHMWPE specimens were investigated under various heat treatment conditions. The heat treatment was performed in the range of annealing and remelting temperatures. The annealing treatment below the temperature of $130^{\circ}C$ hardly induced changes in the tensile strength, the strain at the failure and the hardness. However the remelting treatment above $140^{\circ}C$ deteriorated those mechanical properties. It was shown in an FTIR analysis that the annealing treatment caused some oxidation of free radicals created by the pretreatment of the irradiation. These quantitative data represented by the behavior of mechanical properties might be used as basic informations for the design and analysis of various artificial joints.

Study on the Proper D-Xylose Concentration in Sugar Mixture to Reduce Glycemic Index (GI) Value in the Human Clinical Model (설탕에 대한 Glycemic Index(GI) 저감효과가 있는 D-Xylose의 적정 농도에 관한 연구)

  • The objective of this study was to investigate the proper concentration of D-xylose which is expected to reduce the GI (Glycemic index) value of sucrose in the human body. When subjects took a sucrose mixture containing 5% and 10% D-xylose, the blood glucose levels were lowered by approximately 27.5% and 25.9%, respectively, compared to those of sucrose. The GI values of sucrose mixtures containing 5% and 10% D-xylose were 49.3 and 50.4, respectively. The reduction in GI value was not dependent on the D-xylose concentration, as the GI value of sucrose mixture containing 5% D-xylose (XyloSugar) was similar to that of sucrose mixture containing 10% D-xylose (XyloSugar10). D-xylose is not only more expensive but also less sweet than sucrose. So, low concentration of D-xylose has the advantage in the price and taste. It was determined that the proper concentration of D-xylose expected to reduce GI value of sucrose was 5% (w/w).

Optimization of Production Yield for Neohesperidin by Response Surface Methodology (반응표면 분석법을 이용한 neohesperidin 생산 수율의 최적화)

  • Neohesperidin is a natural new nutrition sweetener, widely existing in plants of dry citrus peel, which can be derived from extraction. Since the sweetness is 1,300-1,500 times greater than that of sugar, neohesperidin are widely used in fruit juices, wines, beverages, bakeries and pharmaceutical formulations, and are particularly suitable for consumption by diabetic patients. However, the yield of extraction from citrus peel waste is very low. In this study optimal yield conditions were determinedusing response surface methodology (RSM) in order to increase the neohesperidin extraction yield. The critical factors for maximum extraction yield were selected extraction pressure ($x_1$), extraction time ($x_2$), and concentration of ethanol ($x_3$). As a result, the extraction yield was improved when the extracting pressure increased. The extraction yield also increased in a time-dependent manner. When adding ethanol as an assistance solvent to the supercritical carbon dioxide, extraction yield was increased as more ethanol concentration was added. Finally, the extraction yield of neohesperidin was improved to about 162.22% compared to ethanol extraction as a conventional method.
