From the stem of Prunus davidiana, naringenin and its glucoside, kaempferol and its glucoside, dihydrokaempferol, kaempferide glucoside, hesperetin glucoside, quercetin glucoside, d-catechin and $\beta$-sitosterol glucoside were isolated.
강낭콩(Phaseolus vulgaris L.) 미숙종자로부터 신규 phytosteryl glucoside를 순수분리하였다. 이 화합물은 positive FAB-MS에서 m/z 613에 $[M+Na]^{+}$를, 696에 $[M+Matrix]^{+}$의 강한 ion peak를 나타내었고, negative FABMS에서 m/z 589에 $[M-1]^{-}$의 강한 ion peak를 나타내어 그 분자량은 590으로 확인되었다. 400 MHz $^1H-NMR$ 분석 결과 이 화합물은 24-hydroxy-24-vinyl-cholesterol(saringosterol)과 ${\beta}-D-glucopyranose$가 연결되어 있는 saringosteryl glucoside임을 알 수 있었고, 이 화합물의 acetyl화 화합물의 400 MHz $^1H-NMR$의 분석을 통하여 이 화합물은 saringosterol의 C-3 수산기와 ${\beta}-D-glucopyranose$의 C-1'의 수산기가 탈수축합되어 있는 $3-O-{\beta}-D-glucopyranosyl-24-hydroxy-24-vinyl-cholesterol$(saringosteryul glucoside)로 구조결정되었다. 이 saringosteryl glucoside는 천연으로부터 처음 동정된 신규 화합물이며, 이 화합물의 aglycone인 saringosterol도 고등 식물에서는 처음으로 발견된 것이다.
Isorhamnetin 3-O-glucoside는 플라보놀 그룹에 속하는 물질로서 염증이나 궤양에 효과가 있을 뿐만 아니라 신경장해, 신장병증, 망막증과 같은 당뇨합병증을 완화하는 것으로 보고되었다. Isorhamnetin 3-O-glucoside는 Tetraena aegyptia, Salsola oppositifolia, Salicornia herbacea, Sambucus ebulus와 같은 몇몇 식물에서 발견된다. 생물전환은 저렴한 화합물로부터 고부가가치 물질을 생산할 수 있는 유용한 방법이다. 본 연구에서 생물전환을 통해 quercetin으부터 isorhamnetin 3-O-glucoside를 생합성 하기 위해 두 개의 유전자(PGT E82L과, ROMT-9)를 각각의 대장균에 도입하였다. 대장균의 공조배양시스템을 이용하여 isorhamnetin 3-O-glucoside 생산 배양법의 최적화를 위해 생물전환배지, 배양온도, 세포의 혼합비율, 재조합 단백질 유도시간, 기질 공급 농도 등을 테스트하였다. 최적화된 생물전환 조건하에서 생물전환을 실시하였으며, 배양의 12시간 후 181.2 mg/L의 isorhamnetin 3-O-glucoside가 생합성 되었다. 이는 이전의 연구에서 보고된 isorhamnetin 3-O-glucosie (39.6 mg/L)의 생합성보다 4.7배 높았다.
본 실험은 포도주스의 가공 공정중 열처리 조건이 anthocyanin색소 각각에 미치는 영향을 검토함으로서 anthocyanin 색소의 파괴가 최소화 되는 열처리 조건을 결정하는 기초자료를얻고자 실시하였다 Anthocyanin 색소 동정을 위한 분석결과 딸기의 주요 적색소는 pelargonidin-3-glucoside이며, 미국산 포도의 과피 및 주스의 주요 적색소는 delphinidin-3-glucoside와 cyanidin-3-glucoside이며, 유색미의 주요 적색소는 cyanidin-3-glucoside로 나타났다. 총 anthocyanin 함량 변화는 30초간 열처리한 처리구가 비열처리구에 비하여 100$^{\circ}C$까지 변화가 없었으며, 1분간 열처리한 처리구는 비열처리구에 비하여90$^{\circ}C$ 까지는 변화가 없었으며, l00$^{\circ}C$, 110$^{\circ}C$에서 각각 3.5$\%$, 9.3$\%$의 감소를 나타냈다. 열처리에 따른 anthocyanin 색소의 변화를 실험한 결과 1분간 열처리 하였을 경우 delphinidin-3-glucoside와 cyanidin-3-glucoside의 경우 90$^{\circ}C$까지 변화가 없었으며, 100$^{\circ}C$부터 급격히 감소하였다. Petunidin-3-glucoside와 malvidin-3-glucoside의 경우 100$^{\circ}C$까지 변화가 없었으며, 110$^{\circ}C$에서 3.5$\%$, 2.7$\%$로 비교적 열에 안정한 것으로 나타났다. 2분간 열처리시 delphinidin-3-glucoside와 cyanidin-3-glucoside의 경우 90$^{\circ}C$까지 3.2$\%$, 5.3$\%$와 1.9$\%$, 4.3$\%$로 감소하였으나, 100$^{\circ}C$부터 12.4$\%$, 13.2$\%$와 9.1$\%$, 11.8$\%$로 급격히 감소하였다. Petunidin-3-glucoside와 malvidin-3-glucoside의 경우 110$^{\circ}C$ 까지 4.8$\%$, 5.2$\%$의 감소를 나타내며 다른 anthocyanin 색소에 비하여 비교적 열에 안정한 것으로 나타났다. 따라서 포도주스 제조시 열처리 조건을 90$^{\circ}C$에서 1분간 살균하는 것이 anthocyanin 파괴를 최소화하여 고품질 포도 주스를 생산하는데 최적의 가공조건인 것으로 사료된다.

Flavonoids have an ability to suppress various ion channels. We determined whether one of flavonoids, cyanidin-3-glucoside, affects adenosine 5'-triphosphate (ATP)-induced calcium signaling using digital imaging methods for intracellular free $Ca^{2+}$ concentration ([$Ca^{2+}$]i), reactive oxygen species (ROS) and mitochondrial membrane potential in PC12 cells. Treatment with ATP ($100{\mu}M$) for 90 sec induced [$Ca^{2+}$]i increases in PC12 cells. Pretreatment with cyanidin-3-glucoside ($1{\mu}g/ml$ to $100{\mu}g/ml$) for 30 min inhibited the ATP-induced [$Ca^{2+}$]i increases in a concentration-dependent manner ($IC_{50}=15.3{\mu}g/ml$). Pretreatment with cyanidin-3-glucoside ($15{\mu}g/ml$) for 30 min significantly inhibited the ATP-induced [$Ca^{2+}$]i responses following removal of extracellular $Ca^{2+}$ or depletion of intracellular [$Ca^{2+}$]i stores. Cyanidin-3-glucoside also significantly inhibited the relatively specific P2X2 receptor agonist 2-MeSATP-induced [$Ca^{2+}$]i responses. Cyanidin-3-glucoside significantly inhibited the thapsigargin or ATP-induced store-operated calcium entry. Cyanidin-3-glucoside significantly inhibited the ATP-induced [$Ca^{2+}$]i responses in the presence of nimodipine and ${\omega}$-conotoxin. Cyanidin-3-glucoside also significantly inhibited KCl (50 mM)-induced [$Ca^{2+}$]i increases. Cyanidin-3-glucoside significantly inhibited ATP-induced mitochondrial depolarization. The intracellular $Ca^{2+}$ chelator BAPTA-AM or the mitochondrial $Ca^{2+}$ uniporter inhibitor RU360 blocked the ATP-induced mitochondrial depolarization in the presence of cyanidin-3-glucoside. Cyanidin-3-glucoside blocked ATP-induced formation of ROS. BAPTA-AM further decreased the formation of ROS in the presence of cyanidin-3-glucoside. All these results suggest that cyanidin-3-glucoside inhibits ATP-induced calcium signaling in PC12 cells by inhibiting multiple pathways which are the influx of extracellular $Ca^{2+}$ through the nimodipine and ${\omega}$-conotoxin-sensitive and -insensitive pathways and the release of $Ca^{2+}$ from intracellular stores. In addition, cyanidin-3-glucoside inhibits ATP-induced formation of ROS by inhibiting $Ca^{2+}$-induced mitochondrial depolarization.
Alkyl-glucoside의 생산을 위하여 상용화 cellulase인 Celluclast의 당전이 반응을 사용하였다. 5가지 종류의 알코올을 acceptor molecule로 하여 반응을 살펴본 결과 methyl alcohol, ethyl alcohol, isopropanol 그리고 butanol에서 당전이 반응이 일어남을 확인하였다. 반응 수율이 높았던, methyl alcohol과 ethyl alcohol의 반응산물을 MALDI-TOF MS와 효소적인 방법을 사용하여 각각의 산물이 methyl ${\beta}$-D-glucopyranoside와 ethyl ${\beta}$-D-glucopyranoside임을 확인하였다. 시간대별 methyl-glucoside와 ethyl-glucoside의 생산량을 비교하여 본 결과 9시간에서 최대 생산 수율 65% (mol/mol)와 59%(mol/mol)를 각각 보였으며, 이후 반응은 진행되지 않았다. Cellulose의 당전이 반응으로 생성된 부산물인 glucose를 제거하기 위하여 고정화 효모 system을 도입하였고, 그 결과 glucose를 모두 제거할 수 있었다. 이상의 결과에서 Celluclast를 이용한 alkyl-glucoside의 생산을 성공적으로 수행하였고, 고정화 효모 system을 도입하여 친환경적으로 부산물을 제거하여 고순도의 ethyl-glucoside를 생산하였다.
From the 70% EtOH extract of the roots of Astragalus membranaceus (Leguminosae), eleven flavonoid derivatives and a lignan, were isolated and identified as liquiritigenin (1), daidzein (2), formononetin (3), sophorophenolone (4), calycosin (5), methylnissolin (6), isomucronulatol (7), isomucronulatol 7-O-glucoside (8), methylnissolin 3-O-glucoside (9), calycosin 7-O-glucoside (10), (+)-syringaresinol O-${\beta}$-D-glucoside (11), and isomucronulatol 7,2'-di-O-glucoside (12), by spectroscopic methods. This is the second report of the isoflavonoid derivatives sophorophenolone (4) and isomucronulatol 7,2'-di-O-glucoside (12) from a natural source, as well as the first report of compounds liquiritigenin (1), daidzein (2) and (+)-syringaresinol O-${\beta}$-D-glucoside (11) from the species A. membranaceus.
Cyclodextrin glucanotransferase (CGTase) from Bacillus stearothermophilus synthesized vanillin and ethyl vanillin monoglucoside, with a series of its maltooligoglucosides by transglycosylation with dextrin as a donor, and vanillin or ethyl vanillin as an acceptor. The monoglucoside formed from reaction mixture of vanillin or ethyl vanillin by the successive actions of CGTase and Rhizopus glucoamylase was isolated by extraction with n-butanol saturated with water and silica gel column chromatography. The structure of the isolated monoglucoside was identified as vanillin- $\alpha$ -D-glucoside and ethyl vanillin- $\alpha$ -D-glucoside, respectively, by FAB-MS, UV, IR, 1H-NMR, 13C-NMR spectra and products by hydrolysis with acid, $\alpha$ - and $\beta$ -glucosidases.

The monoglucoside formed from reaction mixture of maltol or ethyl maltol by the successive actions of Bacillus stearothemophilus cyclodextrin glucanotransferase and Rhizopus glucoamylase was isolated by Diaion HP-20 column and silica gel column chromatography. The structure of the isolated monoglucoside was identified as maltol-$\alpha$-D-glucoside and ethyl maltol-$\alpha$-D-glucoside, respectively, by FAB-MS, UV, $^1$H-NMR, $^{13}$ C-NMR spectra and products by hydrolysis with acid, $\alpha$ - and $\beta$ -glucosidases.
