대장균 트립토판 생성효소의 소단위체간 상호조절 (Intersubunit Communication of Escherichia coli Tryptophan Synthase)

  • 대장균 트립토판 생성효소는 ${\alpha}_2{\beta}_2$ 복합체로 구성되며, 트립토판 생합성에서 최종 2 단계의 반응에 관여한다. 두 개의 소단위체는 분자 터널로 연결되어 있어, 기질 채널링이 일어난다. 활성 부위간 상호 조절하는 정교한 조절 기작에는 ${\alpha}$-루프 L6(${\alpha}L6$), ${\alpha}L2$, ${\alpha}L3$이 관여한다. 본 연구에서는 이 자리의 잔기치환체를 써서 소단위체에 특이적으로 결합하는 리간드의 영향을 조사하여 소단위체간 상호 조절기작에 따른 구조 변화를 살펴보았다. ${\alpha}TS$의 활성부위에 결합하는 D,L-${\alpha}$-glycerophosphate(GP)는 모든 잔기치환체를 야생형 수준으로 회복시켰다. ${\beta}TS$의 기질인 L-Ser는 다양한 효과를 나타낸다. 야생형과 NS104에서는 속도가 감소한 반면, GD51과 PH53에서는 거의 영향이 없었고, PT53와 DG56은 증가하였다. 이는 반응 중간 화학종의 분포의 변화와 연관될 가능성을 제시한다. GP와 L-Ser를 동시에 처리했을 때는 특이하게도 PH53는 가장 안정한 잔기치환체였다. 이는 Pro53가 소단위체간의 조절기작에서 중요한 역할을 하는 것을 시사한다.

Tumor Necrosis Factor-alpha 저해제가 결핵 발생에 미치는 영향 (Effects of Tumor Necrosis Factor-alpha Inhibitors on the Incidence of Tuberculosis)

  • Objective: Tumor necrosis factor-alpha (TNF-alpha) inhibitors are used as a treatment in various immune-mediated inflammatory diseases (IMIDs). Tuberculosis (TB) risk is reported in several meta-analyses in patients treated with TNF-alpha inhibitors. The purpose of this study is to collect, review, and evaluate the TB risk in TNF-alpha inhibitors according to IMIDs indications and between soluble-receptor TNF-alpha inhibitor and monoclonal-antibody TNF-alpha inhibitors. Methods: A systematic literature search on systematic reviews and meta-analyses was performed in PubMed, MEDLINE, Cochrane library, and EMBASE. We identified meta-analyses that evaluated TB infection risk of TNF-alpha inhibitors in IMIDs patients. Results: Thirteen meta-analyses including 41 study results were included in this umbrella review. IMIDs patients treated with TNF-alpha inhibitors had an increased risk of TB than control group (placebo with or without standard therapy patients) (relative risk ratio (RR) 2.057, 95% confidence interval (CI) 1.697 to 2.495). Among them, RA patients with TNF-alpha inhibitors had a higher risk of TB than control group (RR 1.847, 95% CI 1.385 to 2.464), and non-RA patients with TNF-alpha inhibitors had an increased risk of TB (RR 2.236, 95% CI 1.284 to 3.894). In subgroup analysis on TB risk between soluble-receptor TNF-alpha inhibitor and monoclonal-antibody TNF-alpha inhibitors in RA patients, the analysis indicated that monoclonal-antibody TNF-alpha inhibitors had higher risk of TB than soluble-receptor TNF-alpha inhibitor (RR 2.880, 95% CI 1.730 to 4.792). Conclusion: This umbrella review confirms that the risk of TB is significantly increased in TNF-alpha inhibitor treated patients compared to control group.

Role of α-Actinin 2 in Cytoadherence and Cytotoxicity of Trichomonas vaginalis

  • Trichomonas vaginalis is a pathogen that triggers severe immune responses in hosts. T. vaginalis ${\alpha}$-actinin 2 ($Tv{\alpha}$-actinin 2) has been used to diagnose trichomoniasis. $Tv{\alpha}$-actinin 2 was dissected into three parts; the N-terminal, central, and C-terminal portions of the protein (#1, #2, and #3, respectively). Western blot of these $Tv{\alpha}$-actinin 2 proteins with pooled patients' sera indicated that #2 and #3, but not #1, reacted with those sera. Immunofluorescence assays of two different forms of T. vaginalis (trophozoites and amoeboid forms), using anti-$Tv{\alpha}$- actinin 2 antibodies, showed localization of $Tv{\alpha}$-actinin 2 close to the plasma membranes of the amoeboid form. Fractionation experiments indicated the presence of $Tv{\alpha}$-actinin 2 in cytoplasmic, membrane, and secreted proteins of T. vaginalis. Binding of fluorescence-labeled Trichomonas to vaginal epithelial cells and prostate cells was decreased in the antibody blocking experiment using anti-$Tv{\alpha}$-actinin 2 antibodies. Pretreatment of T. vaginalis with anti-$rTv{\alpha}$-actinin 2 antibodies also resulted in reduction in its cytotoxicity. Flow cytometry, ligand-binding immunoblotting assay, and observation by fluorescence microscopy were used to detect the binding of recombinant $Tv{\alpha}$-actinin 2 to human epithelial cell lines. Specifically, the truncated N-terminal portion of $Tv{\alpha}$-actinin 2, $Tv{\alpha}$-actinin 2 #1, was shown to bind directly to vaginal epithelial cells. These data suggest that ${\alpha}$-actinin 2 is one of the virulence factors responsible for the pathogenesis of T. vaginalis by serving as an adhesin to the host cells.

  • The recombinant soluble human tumor necrosis factor-alpha (hTNF-$\alpha$) was expressed in a yeast Saccharomyces cerevisiae and its cytotoxicity was evaluated. A cDNA encoding hTNF-$\alpha$ was placed under the control of two different promoters: a glyceraldehyde-3-phosphate dehydrogenase (GPD) promoter and a yeast hybrid ADH2-GPD promoter, consisting of alcohol dehydrogenase II (ADH2) and the GPD promoter. A Northern blot analysis revealed that, although variation in the expression level of hTNF-$\alpha$ existed among transformants, the higher expression was obtained with the GPD promoter. Expressed hTNF-$\alpha$ protein (rhTNF-$\alpha$) was successfully secreted into the culture medium, producing 2.5 mg per liter of culture filtrate, with no changes in cell growth. The bioassay for observing the cytotoxicity to the murine L929 fibroblast cell line, with serial dilution of rhTNF-$\alpha$, indicated that the secreted rhTNF-$\alpha$ was bioactive and its dose-response was improved eight to ten times over that of the E. coli-derived rhTNF-$\alpha$.

전분으로부터 α-amylase에 의한 배당체의 합성 (Synthesis of Glycosides by Transglycosylation of α-Amylase from Starch)

  • 수계에서 전분 가수분해 효소의 transglycosylation 반응을 이용하여 배당체(glycoside)를 합성하였다. Glycosyl donor인 가용성전분과 glycosyl acceptor인 benzylalcohol을 기질로 하여 ${\alpha}$-amylase에 의해 합성되는 배당체는 glucose의 1번 OH기에 benzylalcohol이 ${\alpha}$형태로 결합한 benzylalcohol-${\alpha}$-glucoside(BG)와 benzylalcohol-${\alpha}$-maltoside(BM)이었다. pH 5.0의 반응에서는 주로 BG가, pH 8.0의 반응에서는 BM만이 합성되는 특이한 반응양상을 보였다. Transglycosylation 반응의 최적조건은 가용성전분 50 mg/ml, benzylalcohol 50 mg/ml, 온도 $30-35^{\circ}C$, 효소량 10 unit/ml이었으며, 합성된 BG는 ${\alpha}$-glucodisase에 의해 glucose와 benzylalcohol로 가수분해되었으며 BM의 경우는 pH 5.0에서는 glucose와 BG로 가수분해되었으나 pH 8.0이상에서는 전혀 분해되지 않았다. BM과 구조적으로 유사한 maltotriose는 pH 5.0에서 glucose와 maltose로 가수분해 되었으나 transglycosylation반응은 거의 일어나지 않았으며 pH 8.0에서는 가수분해도 transglycosylation반응도 일어나지 않았다.

사료의 $\alpha-cellulose$ 함량이 조피볼락 Sebastes schlegeli의 성장, 사료 효율 및 체성분에 미치는 영향 (Effects of Dietary $\alpha-cellulose$ Levels on the Growth, Feed Efficiency and Body Composition in Korean Rockfish, Sebastes schlegeli)

  • 사료의 $\alpha-cellulose$ 함량이 조피볼락의 성장, 사료 효율, 영양소 이용 효율 및 체조성에 미치는 영향을 조사하기 위해 209 전후의 조피볼락 치어를 대상으로 사료의 $\alpha-cellulose$ 함량이 $0\~20\%$가 되도록 총 5종의 실험 사료를 제조하여 10주간 사육 실험한 결과는 다음과 같다. $\alpha-cellulose$ 함량 $10\%$ 첨가구까지는 대조구와 유의차없이 성장 및 사료 효율이 양호하였고, $\alpha-cellulose$ $15\%$이상 첨가 실험구들은 유의하게 낮은 값을 보였다(P<0.05). 일간 먹이 섭취량은 $\alpha-cellulose$ $15\%$ 이상 첨가구에서 유의하게 높은 결과를 보였다(P<0.05). 영양소 축적효율 중 단백질 및 에너지 축적 효율은 $\alpha-cellulose$ $10\%$ 첨가구까지 대조구와 유의차 없이 좋은 결과를 보인 반면, 지질 축적 효율은 a-cellulose 함량이 증가함에 따라 계속 감소하였다 (P< 0.05). 전어체의 단백질 함량은 실험 구간별로 차이가 없었지만, 지질 함량은$\alpha-cellulose$ $10\%$ 첨가구에서 유의하게 높은 값을 보였다(P<0.05).

${\alpha}$-Mangostin Reduced ER Stress-mediated Tumor Growth through Autophagy Activation

  • ${\alpha}$-Mangostin is a xanthon derivative contained in the fruit hull of mangosteen (Garcinia mangostana L.), and the administration of ${\alpha}$-Mangostin inhibited the growth of transplanted colon cancer, Her/CT26 cells which expressed Her-2/neu as tumor antigen. Although ${\alpha}$-Mangostin was reported to have inhibitory activity against sarco/endoplasmic reticulum $Ca^{2+}$ ATPase like thapsigargin, it showed different activity for autophagy regulation. In the current study, we found that ${\alpha}$-Mangostin induced autophagy activation in mouse intestinal epithelial cells, as GFP-LC3 transgenic mice were orally administered with 20 mg/kg of ${\alpha}$-Mangostin daily for three days. However, the activation of autophagy by ${\alpha}$-Mangostin did not significantly increase OVA-specific T cell proliferation. As we assessed ER stress by using XBP-1 reporter system and phosphorylation of $eIF2{\alpha}$, thapsigargin-induced ER stress was significantly reduced by ${\alpha}$-Mangostin. However, coadministration of thapsigargin with ${\alpha}$-Mangostin completely blocked the antitumor activity of ${\alpha}$-Mangostin, suggesting ER stress with autophagy blockade accelerated tumor growth in mouse colon cancer model. Thus the antitumor activity of ${\alpha}$-Mangostin can be ascribable to the autophagy activation rather than ER stress induction.

  • Lf는 cytokines의 생성 및 면역반응 등의 생체 방어적 기능을 하는 물질로 알려져 있다. 본 연구에서는 Lf과 Lf-h에 의해 macrophage에서 TNF-$\alpha$와 NO 생성에 미치는 효과를 조사하였다. K-Lf 및 K-Lf-h는 단독으로 TNF-$\alpha$의 생성을 증가시켰으며, Lf의 농도에 따라 그 생성량이 증가하였다. LPS와 함께 작용시켰을 경우에는 큰 효과가 없는 것으로 나타났으나, 세포 성장률은 증가시켰다. 그러나 B-Lf, H-Lf, 그리고 이들의 가수분해물들은 단독으로는, RAW264.7 cell을 자극하여 TNF-$\alpha$나 NO의 생성을 증가시키지 못하였다. 또한, K-Lf는 그 자체만으로 TNF-$\alpha$에서 보여준 것처럼 NO생성에 영향을 미치며 농도가 높아짐에 따라 NO 생성을 증가시키는 경향을 보여주었다.
