Transposition of IntAs into the Conserved Regions of IS3 Family Elements

  • Together with the previous reports, my computer survey revealed that several bacteria contain six copies of the type group II intron IntA. The sequence analysis of IntAs showed the high level of homology in the nucleotide sequence (91.9-99.8%). The consensus sequence, 2,270 base pair long, was derived from the nucleotide sequences of all IntA members. The size of the open reading frame intA was 502 amino acids long, that is homologous to reverse transcriptase-like proteins encoded within the group II introns. It was reported that EPEC.IntA and Sf.IntA were inserted into IS911 and IS629, respectively. The sequence of the flanking region IntA was analyzed here. The data show the insertion of EC.IntA into IS629, the insertion of EHEC.IntA into IS3, the insertion of Yp.IntA into IS904-like sequence, and the insertion of EK12.IntA into IS911. Interestingly, these IS elements nested by IntAs were the members of IS3 family elements. The sequences of the IS3 members correspond to the OrfB with the DDE motif conserved in retroviral integrases. Alignment of the flanking sequences of IntAs revealed that the flanking regions -25 to + 10 of insertion sites, that are generally believed to be required for the retrohoming, were not strongly conserved. The data presented here suggests that the retrohoming pathway of IntA seems to differ from those of other group II introns.

Occurrence and Control System of Aneilema japonica in Dry Seeded Rice (벼 건답직파답(乾畓直播畓)에서 사마귀풀 발생양상(發生樣相)과 방제체계(方劑體系))

  • The experiment was conducted to obtain basic information on occurrence ecology system of Aneilema japonica and to establish its effective control systems in dry seeded rice at National Yeongnam Agricultural Experiment Station in 1995. Total amount of weed in dry seeded rice in the following year('95) as affected by control of barnyardgrass at 70 days after seeding(DAS) in previous year('94) was reduced 47.7% at 30 DAS and 7.8% at 60DAS and also exhibited different weed occurrence as compared with no control plot. Especially in control plot at 60 DAS, occurrence of Echinochloa crus-galli was reduced by 46.9%, But the growth of Aneilema japonica which is one of the problems in weed species was drastically increased by 231 % and Persicaria hydropiper by 11.8%, respectively. Suppressed tillering of rice plant as affected by occurring of A. japonica(300-750 plants/$m^2$) occurred from about 40 DAS and rice grain reduced 74-93% compared with hand weeding due to reduced panicle number. Soil applied Butachlor(EC) at 5 DAS, foliar applied Propanil -Butachlor(EC) at 13 DAS and Lgc 40863+Pendimethalin at 20 DAS were very effective for Aneilema japonica. As considering both weed suppression of Aneilema japonica and yield capacity, middle late foliar application of Lgc 40863+Pendimethalin and systematic application of Butachlor at 5 DAS followed by Propanil/Molinate at 25 DAS were the most effective control systems.

Optimal HPLC Condition for Simultaneous Determination of Catechins and Caffeine in Green Tea Extracts (녹차 함유 카테친 및 카페인 동시분석을 위한 최적 HPLC 분석 조건)

  • The health benefits associated with tea consumption have resulted in the wide inclusion of green tea extracts in botanical dietary supplements, which are widely consumed as adjuvants for complementary and alternative medicines. Tea contains polyphenols such as catechins or flavan-3-ols including (-)-epicatechin (EC), (-)-epigallocatechin (EGC), (-)-epicatechin gallate (ECG), and (-)-epigallocatechin gallate (EGCG), as well as the alkaloid, caffeine. The contents of catechins and caffeine in green tea are considered as a standard of quality evaluation of green tea. Therefor, the purpose of this study was to investigate the most suitable HPLC condition for simultaneous determination of catechins and caffeine in green tea extracts. The efficient HPLC analytical condition of catechins and caffeine contained green tea extracts was developed. The gradient elution employed a $250\;mm\;{\times}\;4.6\;mm$ i.d. YMC-pak ODS-AM 303 column. The gradient system was used two mobile phases. A gradient elution was performed with mobile phase A, consisting of 0.1% aqueous phosphoric acid, and mobile phase B, comprising 100% MeOH, and delivered at a flow rate of 1 mL/min as follows: $0{\sim}25\;min$, 80% A; $26{\sim}50\;min$, $80{\sim}70%$ A; 51 min, 80% A. $51{\sim}55\;min$, 80% A. The UV detection wavelength was set at 280 nm. The limit of detection (LOD) for catechins and caffeine standards were under 50 ng/mL.

Environment-Friendly Control of Beet Armyworm, Spodoptera exigua (Noctuidae: Lepidoptera) to Reduce Insecticide Use (농약 사용 저감화를 위한 환경 친화적인 파밤나방(Spodoptera exigua)의 방제)

  • For the environment-friendly control of beet armyworm, Spodoptera exigua, in spring onion fields, we have examined an alternative application method. Twenty-five insecticides registered for spring onion were tested for control effect against the beet armyworm in the laboratory, then the best 9 chemical and a single biological insecticides were selected and compared with 2 new isolates of Bacillus thuringiensis in a polyvinyl house. After that, 4 chemical and 3 B. thuringiensis treatments were used in the field for the control of beet armyworm in the spring onion. Two application methods are used: one is triple treatments with the same chemical and the other is alternative application with different chemicals and B. thuringiensis for 7 days intervals. Indoxacarb WP - chlorfluazuron EC - B.t. var. kurstaki CAB141 and indoxacarb WP - methoxyfenozide + spinosad SC - Chlorfluazuron EC - B.t. var. aizawai CAB109, B.t. var. kurstaki CAB141 showed greater than 78% mortality of beet armyworm larvae and greater than 43% damage decrease in spring onions infested by beet armyworm. These results showed that alternative applications had higher control effect than any other applications. It was suggested that alternative applications with microbial biological agents such as B. thuringiensis might minimize the development of insecticide resistance and might be used as the environment-friendly control of the beet armyworm.

Efficacy of Tomato Powder as Antioxidant in Cooked Pork Patties

  • This study was carried out to determine the effects of tomato powder (TP) on cooked pork patties during storage at $10{\pm}1^{\circ}C$ in the dark. The total phenolic and flavonoid contents of TP extract were 26.22 mg gallic acid/100 g and 3.52 mg quercetin/100 g, respectively. The extract of TP showed a potential antioxidant activity in the DPPH radical-scavenging assay ($EC_{50}=16.76{\mu}g/mL$). Pork patties were manufactured with 0.25% (T1), 0.5% (T2), 0.75% (T3) and 1.0% (T4) TP in a basic formula (C). The pH and volatile basic nitrogen (VBN) values of T2, T3 and T4 patties were lower (p<0.05) than the C patties during storage. Increased concentration of TP in meat patties decreased (p<0.05) the 2-thiobarbituric acid reactive substances (TBARS) and total plate count (TPC) values at d 7 of storage. Tomato treated-patties had lower (p<0.05) values for lightness ($L^*$), but higher (p<0.05) values for redness ($a^*$) and yellowness ($b^*$) at d 3 and 7 of storage compared with the C. In the case of sensory evaluation, the scores of colour, flavour and overall acceptability of T3 and T4 patties were higher (p<0.05) than those of the C patty after 3 or 7 days of storage.

Germination Dependency of Antioxidative Activities in Brown Rice (현미의 발아정도에 의한 항산화활성의 변화)

  • The change of antioxidative character by germination of brown rice was evaluated. From the total methanolic extract of brown rice, 2.5 mm-germinated brown rice, and 5 mm-germinated brown rice, SOD-like activity and nitrite scavenging ability were identified as antioxidative character. SOD-like activities and nitrite scavenging abilities of all samples were changed dose-dependently and germination-dependently. After successive partitioning with hexane, ethyl acetate (EtOAc) and water, each fraction was tested for these activities. SOD-like activities of all fractions were increased by germination, and especially hexane fraction and EtOAc fraction of 5 mm-germinated brown rice had more strong activities than 50 ppm vitamin C. The $EC_{50}$ values of SOD-like activity showed a gradual decrease by germination and that of EtOAc fraction of 5 mm-germinated brown rice was 17 ppm, which was lower concentration than that of 50 ppm vitamin C. The $IC_{50}$ values of nitrite scavenging ability at PH 1.5 also underwent a great decrease by germination and germinated brown rice had the nitrite scavenging ability at lower concentration than brown rice. The results suggest that SOD-like activity and nitrite scavenging ability are thought to be enhanced by the germination effect.

A Study on the Safety of Mycotoxins in Grains and Commonly Consumed Foods (곡류 등 다소비 식품 중 곰팡이독소 안전성 조사 연구)

  • The purpose of this study was to investigate and evaluate the safety of the grains, nut products, beans and oilseeds being sold in Gyeonggi province by analyzing mycotoxins. A multi-mycotoxins analysis method based on LC-MS/MS was validated and applied for the determination of eight mycotoxins, including aflatoxins ($B_1$, $B_2$, $G_1$ and $G_2$), fumonisins ($B_1$, $B_2$), zearalenone and ochratoxcin A in 134 samples. The limit of detection (LOD) and limit of quantitation (LOQ) for the eight mycotoxins ranged from 0.14 to $8.25{\mu}g/kg$ and from 1.08 to $7.21{\mu}g/kg$, respectively. Recovery rates of mycotoxins were determined in the range of 61.1 to 97.5% with RSD of 1.0~14.5% (n=3). Fumonisin $B_1$, $B_2$, zearalenone, and ochratoxin A were detected in 22 samples, indicating that 27% of grains, 12.5% of beans and 11.8% of oilseeds were contaminated. Fumonisin and zearalenone were detected simultaneously in 2 adlays and 3 sorghums. Fumonisin $B_1$ and $B_2$ were detected simultaneously in most samples whereas fumonisin $B_1$ was detected in 1 adlay, 1 millet and 1 sesame sample. The average detected amount of fumonisin was $49.3{\mu}g/kg$ and $10.1{\mu}g/kg$ for grains and oilseeds, respectively. The average detected amount of zearalenone was $1.9{\mu}g/kg$ and $1.5{\mu}g/kg$ for grains and beans, respectively. In addition, the average amount of ochratoxin A was $0.08{\mu}g/kg$ for grains. The calculated exposure amounts of fumonisin, zeralenone and ochratoxin A for grains, beans and oilseeds were below the PMTDI/PTWI.

Effect of Microorganism Mixture Application on the Microflora and the Chemical Properties of Soil and the Growth of Vegetables in Greenhouse (미생물혼합제제 처리가 토양의 미생물상과 화학적 특성 및 시설 채소 생육에 미치는 영향)

  • BACKGROUND: The urgency of feeding the world's growing population while combating soil pollution, salinization and desertification requires suitable biotechnology not only to improve crop productivity but also to improve soil health through interactions of soil nutrient and soil microorganism. Interest in the utilization of microbial fertilizer has increased. A principle of nature farming is to produce abundant and healthy crops without using chemical fertilizer and pesticides, and without interrupting the natural ecosystem. Beneficial microorganisms may provide supplemental nutrients in the soil, promote crop growth, and enhance plant resistance against pathogenic microorganisms. We mixed beneficial microorganisms such as Bacillus sp. Han-5 with anti-fungal activities, Trichoderma harziaum, Trichoderma longibrachiatum with organic material degrading activity, Actinomycetes bovis with antibiotic production and Pseudomonas sp. with nitrogen fixation. This study was carried out to investigate the mixtures on the soil microflora and soil chemical properties and the effect on the growth of lettuce and cucumber under greenhouse conditions. METHODS AND RESULTS: The microbial mixtures were used with each of organic fertilizer, swine manure and organic+swine manure and compared in regard to changes in soil chemical properties, soil microflora properties and crop growth. At 50 days after the treatment of microorganism mixtures, the pH improved from 5.8 to 6.3, and the EC, $NO_3$-Na and K decreased by 52.4%, 60.5% and 29.3%, respectively. The available $P_2O_5$ and $SiO_2$ increased by 25.9% and 21.2%, respectively. Otherwise, the population density of fluorescent Pseudomonas sp. was accelerated and the growth of vegetables increased. Moreover, the population density of E. coli and Fusarium sp., decreased remarkably. The ratio of bacteria to fungi (B/F) and the ratio of Actinomycetes bovis to fungi (A/F) increased 2.3 (from 272.2 to 624.4) and 1.7 times (from 38.3 to 64), respectively. Furthermore, the growth and yield of cucumber and lettuce significantly increased by the treatment of microorganism mixtures. CONCLUSION(S): These results suggest that the treatment of microorganism mixtures improved the chemical properties and the microflora of soil and the crop growth. Therefore, it is concluded that the microorganism mixtures could be good alternative soil amendments to restore soil nutrients and soil microflora.

Characteristics of Manure and Estimation of Nutrient and Pollutant of Holstein Dairy Cattle (홀스타인 젖소 분뇨의 특성과 비료성분 및 오염물질 부하량 추정)

  • This study was conducted to determine fertilizer nutrient and pollutant production of Holstein dairy cattle by estimating manure characteristics. The moisture content of feces was 83.9% and 95.1% for urine. The pH of feces and urine were in the ranges of 7.0~7.4 and 7.5~7.8, respectively. The average BOD5, COD, SS, T-N, T-P concentrations of the dairy feces were 18,294, 52,765, 102,889, 2,575, and 457mg/ℓ, respectively. Dairy urine showed lower levels of BOD5(5,455mg/ℓ), COD(8,089mg/ℓ), SS(593mg/ℓ), T-N(3,401mg/l), and T-P(13mg/ℓ) than feces. The total daily produced pollutant amounts of a dairy cow were 924.1g(Milking cow), 538.8g(Dry cow), 284.4g(Heifer) of BOD5, 2,336.5g (Milking cow), 1,651.8g(Dry cow), 734.1g(Heifer) of COD and 4,210.1g(Milking cow), 2,417.1g(Dry cow), 1,629.1g(Heifer) of SS and 194.8g(Milking cow), 96.4g(Dry cow), 58.3g(Heifer) of T-N and 24.0g(Milking cow), 10.2g(Dry cow), 6.1g(Heifer) of T-P. The calculated amount of pollutants produced by a 450kg dairy cow for one year were 181.3kg of BOD5, 492.5kg of COD, 899.9kg of SS, 36.0kg of T-N and 4.1kg of T-P. The total yearly estimated pollutant production from all head(497,261) of dairy cattle in Korea is 90,149 tons of BOD5, 244,890 tons of COD, 447,491 tons of SS, 17,898 tons of T-N and 2,008 tons of T-P. The fertilizer nutrient concentrations of dairy feces was 0.26% N, 0.1% P2O5 and 0.14% K2O. Urine was found to contain 0.34% N, 0.003% of P2O5 and 0.31% K2O. The total daily fertilizer nutrients produced by dairy cattle were 197.4g (Milking cow), 97.4g(Dry cow), and 57.9g(Heifer) of Nitrogen, 54.2g(Milking cow), 22.2g(Dry cow), and 14.2g(Heifer) of P2O5 and 110.8g(Milking cow), 80.4g (Dry cow), and 39.5g(Heifer) of K2O. The total yearly estimated fertilizer nutrient produced by a 450kg dairy animal is 36.2kg of N, 8.8kg of P2O5, 24.6kg of K2O. The estimated yearly fertilizer nutrient production from all dairy cattle in Korea is 18,000 tons of N, 4,397 tons of P2O5, 12,206 tons of K2O. Dairy manure contains useful trace minerals for crops, such as CaO and MgO, which are contained in similar levels to commercial compost being sold in the domestic market. Concentrations of harmful trace minerals, such as As, Cd, Hg, Pb, Cr, Cu, Ni, Zn, met the Korea compost standard regulations, with some of these minerals being in undetected amounts.
