Effects of controlled- and uncontrolled-pH operations on phenylalanine ammonia lyase (PAL) production by a recombinant Escherichia coli strain were investigated at uncontrolled-pH ($pH_{UC}$) and controlled-pH ($pH_C$) of 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, and 8.5 in bioreactor systems. The results showed that the recombinant PAL activity was improved significantly by controlled pH strategy. Among the $pH_C$ operations, the highest PAL activities were obtained under $pH_C$ 7.5 strategy where cell mass ($OD_{600\;nm}$) and PAL activity was 1.3 and 1.8 fold higher than those of $pH_{UC}$, respectively. The maximum PAL activity reached 123 U/g. The $pH_C$ 7.5 strategy made recombinant plasmid more stable and therefore allowed easier expression of PAL recombinant plasmid, which increased PAL production. It was indicated that the new approach (controlled-pH strategy) obtained in this work possessed a high potential for the industrial production of PAL, especially in the biosynthesis of L-phenylalanine.
A filamentous fungus, strain FM04 producing amylase was isolated from rotten yam peels and potatoes. The favorable conditions of cultivation factors such as, temperature, pH, and agitation speed of strain FM04 were 28∼30$^{\circ}C$, 5.0∼6.0, and 100 rpm, respectively. Starch was the best carbon source in the amylase production. Therefore, food wastes containing lots of starch were employed as the carbon source of the cultivation for the economical amylase production. 5.2 U/ml of amylase was obtained In the cultivation using 1 % (w/v) of food wastes. The amylase showed the highest activity at enzyme reaction conditions of 60$^{\circ}C$ and pH 4.5 and showed 90% of residual activity after the reaction at 50$^{\circ}C$ for 2 days. In the enzymatic hydrolysis reaction using 20% (w/v) of food wastes and 2.5 U/ml of amylase, 72.6 g/l of reducing sugar was obtained at the reaction condition of 50$^{\circ}C$, pH 4.5 for 2 days.
Batch sorption experiments were performed to remove the uranium (U) in groundwater by using the bamboo charcoal. For 2 kinds of commercialized bamboo charcoals in Korea, the U removal efficiency at various initial U concentrations in water were investigated and the optimal sorption conditions to apply the bamboo charcoal were determined by the batch experiments with replicate at different pH, temperature, and reaction time conditions. From results of adsorption batch experiments, the U removal efficiency of the bamboo charcoal ranged from 70 % to 97 % and the U removal efficiency for the genuine groundwater of which U concentration was 0.14 mg/L was 84 %. The high U removal efficiency of the bamboo charcoal maintained in a relatively wide range of temperatures ($10{\sim}20^{\circ}C$) and pHs (5 ~ 9), supporting that the usage of the bamboo charcoal is available for U contaminated groundwater without additional treatment process in field. Two typical sorption isotherms were plotted by using the experimental results and the bamboo charcoal for U complied with the Langmuir adsorption property. The maximum adsorption concentration ($q_m:mg/g$) of A type and C type bamboo charcoal in the Langmuir isotherm model were 200.0 mg/g and 16.9 mg/g, respectively. When 2 g of bamboo charcoal was added into 100 mL of U contaminated groundwater (0.04 ~ 10.8 mg/L of initial U concentration), the separation factor ($R_L$) and the surface coverage (${\theta}$) maintained lower than 1, suggesting that the U contaminated groundwater can be cleaned up with a small amount of the bamboo charcoal.

The sorption and desorption properties of U(VI), Th(IV), Zr(IV), Cu(II), Pb(II), Ni(II), Zn(II), Cd(II) and Mn(II) ions on XAD-16-[2-(2-thiazolylazo)-p-cresol](TAC) chelating resin were studied by elution method for selective separation, concentration and recovery of trace metal ions in sea water. The optimum conditions for the sorption of metal ions were examined with respect to flow rate, pH and concentration of buffer solution. The overall capacities of some metal ions on this chelating resin were 0.41mmol U(VI)/g resin, 0.55mmol Th(IV)/g resin, 0.43mmol Cu(II)/g resin, and 0.32mmol Zr(IV)/g resin, respectively. The elution order of metal ions obtained from breakthrough capacity and overall capacity at pH 5.0 was found as Th(IV)>Cu(II)>U(VI)>Zr(IV)>Pb(II)>Ni(II)>Zn(II)>Cd(II)>Mn(II). Desorption of characteristics for metal ions were investigated with desorption agents such as $HNO_3$, HCl, $HClO_4$, $H_2SO_4$, and $Na_2CO_3$. It was found that most of metal ions except Zr(IV) showed high desorption efficiency with 2M $HNO_3$. But, desorption and recovery of Zr(IV) ion were successfully performed with 1M $H_2SO_4$. The resin was applied for separation and preconcentration of trace amount of U(VI) ion from artificial sea water and the recovery of U(IV) was over 96%.

A bacterium producing the extracellular cellulase was isolated from cattle feces and screened as cellulase activity was excellent upon congo red straining method and activity measurements. Isolate was identified as Bacillus subtilis CH-10 on the basis of morphological and biochemical properties as well as cellular fatty acids composition. The enzyme which the isolate secretes had the optimum initial pH and temperature for its induction was 7.5 and 50${\circ}C$, respectively. The maximum CMCase activity in crude enzyme solution was observed at pH 7.5 and 75${\circ}C$ and was stable for pH 7.5 to 9.0 to maintain 70% activity. When the isolate was cultured in CMC media at 37${\circ}C$ for 24 hrs, CMCase and FPase activity was 1.13 U/㎖and 0.16U/㎖, respectively whereas Avicelase and ${\beta}$-glucosidase activity was not detected. When crude supernatant was used for zymogram, three major bands, cel 1, cel 2 and cel 3, were detected approximately 39, 41 and 57 KDa, respectively on CMC-SDS-PAGE.
The objective of the study was to determine the effects of animal related factors on bruising in slaughter cattle, creatine kinase (CK) and beef quality. Three hundred and twenty one cattle from three breeds (108 Bonsmara, 130 Beefmaster and 83 Brahman) were used in this study. The animals were grouped as follows: Group 1 (16 months old), Group 2 (18 months old) and Group 3 (24 months old). At exsanguinations, blood samples for CK determination were collected using disposable vacutainer tubes. Muscularis longissimus thoracis et lumborum (LTL) was collected 24 h after slaughter to determine the colour ($L^*$, $a^*$, and $b^*$) and ultimate pH ($pH_u$) of beef. Breed, sex and age had significant effects (p<0.05) on bruising score, CK levels and beef quality. Bonsmara breed had the highest (80%) bruising score percentage, CK ($705.3{\pm}80.57U/L$) and $pH_u$ ($6.3{\pm}0.05$) values while the Bonsmara had the highest $L^*$ ($24.8{\pm}0.78$) $a^*$ ($17.5{\pm}0.53$) and $b^*$ ($12.8{\pm}0.53$) values. Higher CK levels were also observed in winter compared to summer, spring and autumn respectively. Therefore, animal factors (sex, breed and animal age at slaughter) contribute to the development of bruises and have an effect on the levels of CK and meat quality. It was also concluded that there is no significant relationship between meat parameters (L,* $a^*$, and $b^*$) and CK levels.

In this study, the physical factors for amylase production by Arthrobacter sp. were optimized using response surface methodology(RSM). Antarctic microorganism Arthrobacter sp. PAMC 27388 was obtained from the Polar and Alpine Microbial Collection(PAMC) at the Korea Polar Research Institute. This microorganism was confirmed for the excretion of amylase with Lugol's solution. The amylase activity was after flask culture was as low as 1.66 mU/L before optimization. The physical factors including the inoculum volume, the initial culture pH, and the medium volume were chosen to be optimized for the enhanced amylase production. The calculated results using RSM indicate that the optimal physical factors were 2.49 mL inoculum volume, 6.85 pH and 42.87 mL medium volume with a predicted amylase production of 2.84 mU/L. The experimentally obtained amylase activity was 2.50 mU/L, which was a 150% increase compared to the level before optimization.

최근 방제 및 예찰과 같은 농작업에 단일 UAV(Unmanned Aerial Vehicle)시스템이 적용되고 있지만, 가반하중과 체공시간 등 기존시스템의 문제가 점차 대두되면서 작업 시간을 보다 단축시키고 작업 효율을 극대화 할 수 있는 농업용 멀티 UAV시스템의 필요성이 증대되고 있다. 본 논문에서는 작업자가 다수의 농업용 UAV를 효과적으로 제어할 수 있는 분산군집제어 알고리즘을 제안하며 알고리즘 검증 및 평가를 위한 시뮬레이터를 소개한다. 분산군집제어는 UAV 제어 계층, VP(Virtual Point) 제어 계층, 원격제어 계층으로 이루어진 3계층 제어구조를 가진다. UAV 제어 계층에서 각 UAV는 point mass로 모델링 되는 VP의 이상적인 경로를 추종하도록 제어한다. VP 제어 계층에서 각 VP는 입력 $p_i(t)=u^c_i+u^o_i+u^{co}_i+u^h_i$-(1)을 받아 제어되는데 여기서, $u^c_i{\in}{\mathbb{R}}^3$는 VP 사이의 충돌방지제어, $u^o_i{\in}{\mathbb{R}}^3$는 장애물과의 충돌방지제어, $u^{co}_i{\in}{\mathbb{R}}^3$는 UAV 상호간의 협조제어, $u^h_i{\in}{\mathbb{R}}^3$는 작업자로부터의 원격제어명령이다. (1)의 제어입력에서 충돌방지제어는 각 $u^i_c:=-{\sum\limits_{j{\in}{\eta}_i}}{\frac {{\partial}{\phi}_{ij}^c({\parallel}p_i-p_j{\parallel})^T}{{\partial}p_i}}$-(2), $u^o_c:=-{\sum\limits_{r{\in}O_i}}{\frac {{\partial}{\phi}_{ir}^o({\parallel}p_i-p^o_r{\parallel})^T}{{\partial}p_i}}$-(3)로 정의되면 ${\phi}^c_{ij}$와 ${\phi}^o_{ir}$는 포텐셜 함수를 나타낸다. 원격제어 계층에서 작업자는 햅틱 인터페이스를 통해 VP의 속도를 제어하게 된다. 이때 스케일변수 ${\lambda}$에 대하여 VP의 원격제어명령은 $u^t_i(t)={\lambda}q(t)$로 정의한다. UAV 시뮬레이터는 리눅스 환경에서 ROS(Robot Operating Systems)를 기반한 3차원 시뮬레이터인 Gazebo상에 구축하였으며, 마스터와 슬레이브 간의 제어 명령은 TCPROS를 통해 서로 주고받는다. UAV는 PX4 기반의 3DR Solo 모델을 사용하였으며 MAVROS를 통해 MAVLink 통신 프로토콜에 접속하여 UAV의 고도, 속도 및 가속도 등의 상태정보를 받을 수 있다. 현재 멀티 드론 시스템을 Gazebo 환경에 구축하였으며, 추후 시뮬레이터 상에 분산군집제어 알고리즘을 구현하여 검증 및 평가를 진행하고자 한다.

Dissolved hexavalent uranium can exist in the form of several different chemical species. Furthermore, species distributions depend on the pH value of the aqueous solution. Representatively, UO22+, UO2OH+, (UO2)2(OH)22+, and (UO2)3(OH)5+ species coexist in solutions at acidic and circumneutral pH values. When amorphous silica particles are suspended in an aqueous solution, the dissolved chemical species are easily adsorbed onto silica surfaces. In this study, it was examined whether the species distribution of the adsorbed U(VI) on a silica surface followed that of the dissolved U(VI) in an aqueous solution. Time-resolved luminescence spectra of three different dissolved species (UO22+, UO2OH+, and (UO2)3(OH)5+) and two different adsorbed species (≡SiO2UO2, ≡SiO2(UO2)OH-, or ≡SiO2(UO2)3(OH)5-) were measured in the pH range 3.5-7.5. The spectral shapes of these chemical species were compared by changing the pH value; consequently, it was confirmed that the species distribution of the adsorbed U(VI) species was different from that of the dissolved U(VI) species.
