청미래덩굴 잎 및 뿌리 추출물의 항산화, ${\alpha}$-Glucosidase 억제 및 항염증 활성비교 (Comparison of antioxidant, ${\alpha}$-glucosidase inhibition and anti-inflammatory activities of the leaf and root extracts of Smilax china L.)

  • 본 연구에서 청미래덩굴 잎과 뿌리의 물추출물을 이용하여 총 폴리페놀과 플라보노이드의 함량을 측정한 결과 잎 추출물에서 더 많은 함량이 확인되었으며, DPPH 및 ABTS radical 소거능, 환원력 측정을 통한 항산화 활성을 비교한 결과도 폴리페놀과 플라보노이드함량 측정 결과와 마찬가지로 잎에서 높은 활성을 명확하게 확인할 수 있었다. 그리고 청미래덩굴 추출물의 항당뇨 효능에 대한 최초의 시도로서 ${\alpha}$-glucosidase 활성억제능을 측정한 결과 뿌리추출물에서는 효소의 억제력을 확인할 수 없었지만 잎 추출물에서는 농도 및 반응시간의 증가에 따라 억제력이 증가하여 물 추출물 상태에서 1 mg/mL 기준으로 acarbose의 ${\alpha}$-glucosidase 활성 억제능과 비교하여 71.8%의 활성을 관찰할 수 있었다. 또한 당뇨와 염증의 관련성이 대두되는 최근의 연구 상황에서 청미래덩굴 잎 추출물의 IL-$1{\beta}$, IL-6, iNOS, COX-2의 mRNA 발현 억제작용을 확인함으로써 염증반응의 개선 가능성도 제시할 수 있었다. 결과적으로 청미래덩굴 잎 추출물의 항산화능, ${\alpha}$-glucosidase 활성억제능 및 항염증효능을 확인함으로써 neutraceuticals 소재로서의 개발가능성을 증명하였고 추가 연구의 필요성을 제시하였다.

Antioxidant Properties of Water Extract from Acorn

  • Antioxidant and anti-diabetic activities of acorn were evaluated by its potential for scavenging stable DPPH free radical, inhibition of lipid peroxidation, reducing power, and inhibiton of ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water extract of acorn exhibited strong antioxidant and antidiabetic related activities in the tested model systems. Solvent fractionation of the water extract revealed that the water fraction and the EtOAc fraction had strong antioxidant activity, and inhibitory activity on ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water fraction exhibited higher DPPH radical scavenging activity ($EC_{50}=7.19{\mu}g/mL$) than that of ${\alpha}-tocopherol\;(EC_{50}=7.59{\mu}g/mL)$. It is considered that water extract of acorn has the potential for natural antioxidant and anti-diabetic products.

Fermentation of purple Jerusalem artichoke extract to improve the α-glucosidase inhibitory effect in vitro and ameliorate blood glucose in db/db mice

  • BACKGROUND/OBJECTIVES: Jerusalem artichoke has inhibitory activity against ${\alpha}$-glucosidase and decreases fasting serum glucose levels, which may be related to its fructan content. The biological activity of fructan can be influenced by the degree of polymerization. Thus, in this study, the inhibitory effects of original and fermented purple Jerusalem artichoke (PJA) on ${\alpha}$-glucosidase were compared in vitro. Additionally, the anti-diabetes effect of Lactobacillus plantarum-fermented PJA (LJA) was studied in a non-insulin-dependent diabetes mellitus animal model (C57BIKsJ db/db). MATERIALS/METHODS: The water extract of PJA was fermented by L. plantarum, and two strains of Bacillus subtilis to compare their anti-${\alpha}$-glucosidase activities in vitro by ${\alpha}$-glucosidase assays. The anti-diabetes effect of LJA was studied in a non-insulin-dependent diabetes mellitus animal model (C57BIKsJ db/db) for seven weeks. During the experiment, food intake, body weight, and fasting blood glucose were measured every week. At the end of the treatment period, several diabetic parameters and the intestinal ${\alpha}$-glucosidase activity were measured. RESULTS: The LJA showed the highest ${\alpha}$-glucosidase inhibitory activity in vitro. In the in vivo study, it resulted in a significantly lower blood glucose concentration than the control. Serum insulin and HDL cholesterol levels were significantly higher and the concentrations of triglycerides, non-esterified fatty acids, and total cholesterol were significant lower in mice treated with LJA after seven weeks. In addition, the intestinal ${\alpha}$-glucosidase activity was partially inhibited. CONCLUSIONS: These results suggested that LJA regulates blood glucose and has potential use as a dietary supplement.

The activity of ${\alpha}$-amylase and ${\alpha}$-glucosidase as anti-diabetic function

  • BACKGROUND/OBJECTIVES: We investigated total 26 ingredients of Saengshik which will be commercially produced as an anti-diabetic dietary supplement. SUBJECTS/METHODS: Thirteen vegetables, nine cereals, three legumes and one seed were extracted with aqueous ethanol for 2 h at $60^{\circ}C$, and evaluated for their inhibitory effects against ${\alpha}$-amylase and ${\alpha}$-glucosidase and for total phenolic and flavonoid contents. RESULTS: All ingredients inhibited ${\alpha}$-amylase activity except cabbage. Strong inhibitory activity of ${\alpha}$-amylase was observed in leek, black rice, angelica and barley compared with acarbose as a positive control. Stronger inhibition of ${\alpha}$-glucosidase activity was found in small water dropwort, radish leaves, sorghum and cabbage than acarbose. All Saengshik ingredients suppressed ${\alpha}$-glucosidase activity in the range of 0.3-60.5%. Most ingredients contained total phenols which were in the range of 1.2-229.4 mg gallic acid equivalent/g dried extract. But, total phenolic contents were not observed in carrot, pumpkin and radish. All ingredients contained flavonoid in the range of 11.6-380.7 mg catechin equivalent/g dried extract. CONCLUSIONS: Our results demonstrate that Saengshik containing these ingredients would be an effective dietary supplement for diabetes.

약용식물 물 추출물이 Goto-Kakizaki 흰쥐의 간 세포액에서 당대사 관련효소 활성에 미치는 영향 (Effect of Medicinal Plant Water Extracts on Glucose-regulating Enzyme Activities in Goto-Kakizaki Rat Liver Cytosol)

  • 본 연구에서는 유전적으로 제2형 당뇨를 가진 Goto-Kakizaki 흰쥐의 간 세포액에 함유된 당 이용 대사 관련 주요 효소인 glucokinase(GK)와 acetyl CoA carboxylase (ACC)에 약용 식물 물 추출물들이 어떤 영향을 미치는지 알아보았다. $\alpha$-Glucosidase 저해제는 제2형 당뇨 치료와 예방을 위해 사용되어져 오고 있다. 따라서 약용식물 물 추출물들의 $\alpha$-glucosidase 저해 활성을 알아보았다. Goto-Kakizaki 흰쥐의 간 세포액 분획물을 GK과 ACC 활성을 알아보기 위해 사용하였다. 본 실험에 사용된 약용 식물들은 Lycium chinense(JGP), Discorea japonica Thunb.(SY), Pyrus pyrifolia(YSB), Cornus officinalis(SSY), Paeonia suffruticosa ANDR.(MDP), Cordyceps militaris(DCH) 그리고 Acanthopanax senticosus(GSO)였다. 지골피(JGP), 산약(SY), 야생배(YSB) 그리고 산수유(SSY) 물 추출물들은 GK 활성을 증가시켰고, 실험에 사용된 모든 약용식물 물추출물들은 간 ACC 활성을 증가시켰다. 야생배(YSB), 산수유(SSY), 목단피(MDP) 그리고 가시오가피(GSO) 물 추출물들은 대조군보다 현저히 높은 $\alpha$-glucosidase 활성 억제를 보여주었다. $\alpha$-Glucosidase 활성을 가장 억제시킨 것은 GSO 물 추출물이었고, 그 억제율은 $\alpha$-glucosidase 치료제인 acarbose보다 높았다. 따라서 제2형 당뇨에서 지골피 (JGP), 산약(SY), 야생배(YSB) 그리고 산수유(SSY) 물 추출물들은 글루코스 대사과정을 활성화시켜 항당뇨 효과를 나타내는 천연 소재일 것으로 생각되고 야생배(YBS), 목단피(MDP) 그리고 가시오가피(GSO)는 천연 $\alpha$-glucosidase 저해제로써 사용될 수 있을 것이다. 약용식물 물 추출물에 의한 증가된 ACC 활성은 더해진 항당뇨 효과를 나타낼 것이다.

당뇨 처방에 근거한 생약재의 α-Glucosidase 활성 저해 효과 및 이를 활용한 미백 소재 평가법 (Effect of Medicinal Herb Prepared through Traditional Antidiabetic Prescription on α-Glucosidase Activity and Evaluation Method for Anti-Melanogenesis Agents Using α-Glucosidase Activity)

  • 본 연구에서는 선정한 생약재 및 복합처방단의 ${\alpha}$-glucosidase 저해 활성을 알아보았으며, 이 방법이 미백 소재 스크리닝을 위한 유용한 평가법인지를 알아보았다. 한의학과 민간에서 당뇨의 개선 및 치료 효과가 우수하다고 알려진 생약재 및 처방 중 죽력, 귀전우, 적양, 연자육, 마인, 청심연자음의 ${\alpha}$-glucosidase 활성 저해 효과는 식후 혈당조절제인 acarbose와 비교하여 볼 때 우수한 효과를 나타내었다. 미백 효과가 알려진 연자육을 함유한 청심연자음 hydrolyzed EtOAc layer는 $100{\mu}g/mL$ 농도에서 약 50% 멜라닌 생성 저해 효과를 보였다. 또한 청심연자음 hydrolyzed EtOAc layer는 ${\alpha}$-glucosidase 활성 저해 효과가 우수하였으나 mushroom tyrosinase 활성 저해 효과는 나타나지 않았다. 이로써 청심연자음 hydrolyzed EtOAc layer는 ${\alpha}$-glucosidase 활성을 저해시켜 tyrosinase의 glycosylation을 저해함으로써 멜라닌 생성 억제 효과가 나타나는 것으로 생각된다. 이상의 결과로 볼 때 ${\alpha}$-glucosidase 활성 억제 효과가 있으면서 당뇨병에 효과가 있는 생약재들은 N-linked glycoprotein인 tyrosinase의 glycosylation을 저해하여 tyrosinase의 세포 내 이동이나 활성을 억제함으로써 멜라닌 생성을 억제할 것으로 사료되며, 본 연구에서 선정된 생약재들은 당뇨병 치료를 위한 목적뿐만 아니라 화장품에서 새로운 미백 소재로서의 활용가치가 있을 것으로 판단된다. 또한 미백에 효과가 있는 소재 스크리닝을 위해 현재 널리 사용되고 있는 mushroom tyrosinase 활성 저해 효과와 다른 접근 방법으로써 ${\alpha}$-glucosidase 활성 측정 방법도 하나의 평가법으로 유용할 것으로 생각된다.

잣버섯(Neolentinus lepideus) 에탄올 추출물의 혈당상승 억제 효과 (Antihyperglycemic α-Glucosidase Inhibitory Activity of Ethanol Extract from Neolentinus lepideus)

  • 버섯을 이용하여 새로운 혈당상승 억제 물질을 개발하여 건강 기능성 식품 소재 산업에 응용하고자 느타리버섯, 노랑느타리버섯, 분홍느타리버섯, 새송이버섯과 잣버섯들의 ${\alpha}$-glucosidase 저해활성을 측정한 결과 잣버섯 자실체의 에탄올 추출물이 가장 높은 ${\alpha}$-glucosidase 저해활성을 보였다. 잣버섯 자실체 분말을 95% 에탄올로 $30^{\circ}C$에서 48시간 추출하였을 때 ${\alpha}$-glucosidase 저해물질이 가장 많이 추출되었고 이 ${\alpha}$-glucosidase 저해물질을 함유한 잣버섯 에탄올 추출물은 일반 쥐와 streptozotocin으로 당뇨를 유발시킨 당뇨 쥐에서 모두 농도 의존적으로 식후 혈당상승 억제 효과를 보였다.

추출방법에 따른 소목 심재의 항산화 및 항당뇨 활성 평가 (Evaluation of Antioxidant and Anti-diabetic Effects of Sappan Lignum by Extraction Method)

  • Objectives : The heartwood of Sappan Lignum has been used since ancient times as an ingredient in folk medicines against anti-bacterial and anti-anemia purposes. Many bioactive constituents have been derived from this biomass such as chalcones and homoisoflavonoids. In the current investigation, the antioxidant and anti-diabetic properties using DPPH and $ABTS^+$ radicals scavenging, ${\alpha}-glucosidase$, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Sappan Lignum. Methods : In our continuing investigation for bioactive natural ingredients, the antioxidant and ${\alpha}-glucosidase$ inhibitory properties of Sappan Lignum extracts were prepared from different extraction methods and the biological efficacies were investigated in vitro. The antioxidant properties were evaluated employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effects of Sappan Lignum extracts were tested via ${\alpha}-glucosidase$ and AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested samples showed dose-dependent radical scavenging and ${\alpha}-glucosidase$ inhibitory activities. Among the tested extracts, the 80% methanolic extract of Sappan Lignum was showed the most potent activity with an $IC_{50}$ value of $82.3{\pm}1.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. While, $ABTS^+$ radical scavenging activity of 80% methanolic extract was higher than those of other extracts. Also, ${\alpha}-glucosidase$ inhibitory and AGEs formation effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Sappan Lignum can be considered as a new effective source of natural antioxidant and anti-diabetic materials.

산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성 (Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

In vitro biological evaluation of 100 selected methanol extracts from the traditional medicinal plants of Asia

  • BACKGROUND/OBJECTIVES: In Asia, various medicinal plants have been used as the primary sources in the health care regimen for thousands of years. In recent decades, various studies have investigated the biological activity and potential medicinal value of the medicinal plants. In this study, 100 methanol extracts from 98 plant species were evaluated for their biological activities. MATERIALS/METHODS: The research properties, including 1,1-diphenyl-2-pic-rylhydrazyl (DPPH) radical scavenging activity, ${\alpha}$-glucosidase and ${\alpha}$-tyrosinase inhibitory effects, anti-inflammatory activity, and anticancer activity were evaluated for the selected extracts. RESULTS: Fifteen of the extracts scavenged more than 90% of the DPPH radical. Among the extracts, approximately 20 extracts showed a strong inhibitory effect on ${\alpha}$-glucosidase, while most had no effect on ${\alpha}$-tyrosinase. In addition, 52% of the extracts showed low toxicity to normal cells, and parts of the extracts exhibited high anti-inflammatory and anticancer activities on the murine macrophage cell (RAW 264.7) and human colon cancer cell (HT-29) lines, respectively. CONCLUSIONS: Our findings may contribute to further nutrition and pharmacological studies. Detailed investigations of the outstanding samples are currently underway.
