Termicidal Activity and Chemical Components of Wood Vinegar from Nipah Fruit against Coptotermes curvignathus

  • The termicidal activity and chemical components of wood vinegar from two sources of biomass, nipah fruit shells (NFS) and a mixture of shells and fiber (MSF), were evaluated against Coptotermes curvignathus. A no-choice test was carried out to evaluate their termicidal activity using filter paper samples treated with 2.5%, 5.0%, 7.5%, 10.0%, and 12.5% NFS or MSF vinegar. Both wood vinegars exhibited antitermitic activity against C. curvignathus. The results show that increased concentrations of NFS and MSF vinegar significantly increased termite mortality. In particular, the NFS vinegar caused complete mortality and the lowest filter paper mass losses at 2.18% when treated with 12.5% wood vinegar. The most abundant chemical compounds of NFS vinegar were cyclopropanecarbonyl chloride, 2,5-dichlorophenol, 2-propanone, acetic acid, propanoic acid, benzenesulfonic acid, 3,7-dimethyl-6-octenal, and trans-geraniol. Meanwhile, the main compounds in the MSF vinegar were 1,2-ethanediol, formic acid, acetic acid, ethanoic acid, 2-furancarboxaldehyde, phenol, 2-methoxy phenol, and 4-methyl phenol.

연속흐름반응기에서 바이오디젤 제조 특성 연구 (Biodiesel Production from Soybean Oil in Continuous Reactors)

  • 재생 가능한 자원인 동식물성 기름을 원료로 제조되는 수송용 연료 바이오디젤은 낮은 대기오염물질 배출과 $CO_2$ Neutral 특성으로 환경친화적인 연료로 인정을 받으며 전세계적으로 그 생산량이 급격히 증가하고 있다. 대부분의 상용화 공정은 염기촉매를 이용한 전이에스테르화 반응에 근거하고 있으며 높은 생산성을 위해 연속 공정을 채택하고 있다. 원료유 중의 유리지방산(free fatty acid, FFA)은 염기 촉매와 반응하여 지방산염(Soap)과 수분을 생성하며 반응촉매의 투입양을 증가시카고 반응 후에 글리세롤과 지방산 메틸에스테르와의 분리를 어렵게 만든다. 높은 수율과 후속공정의 부하를 줄이기 위해서는 식물성 원료유 중의 FFA는 고체 산촉매 하에서 메탄올과 에스테르화 반응시켜 전환 제거되어야 한다. 본 연구에서는 고체산 촉매인 Amberlyst-15을 충전한 4단 PBR(Packed Bed Reactor, 충전율 60%(v/v))에서 반응시간과 반응온도에 따른 대두원유의 전처리 효율을 조사하였으며 최적 전처리 조건을 도출하였다. 최적 전처리 조건에서 대두원유는 초기 산가 1.6에서 0.4-0.6으로 연속 전처리할 수 있었다. 본 연구에서는 연속 흐름 반응기인 PFR(Plug Flow Reactor)와 4단 CSTR(Continuous Stirred Tank Reactor)에서 균질계 촉매인 KOH 존재하에 대두유와 메탄올과의 전이에스테르화 반응 특성을 조사하였으며 각 연속 반응시스템에서 최적 운전 조건을 도출하였다. PFR 반응기에서 반응온도, 반응시간, 반응물 흐름방향, static mixer(SM) 개수에 따른 반응특성을 조사한 결과, PFR에서의 최적 반응조건은 하향류 흐름 방향과 3개의 SM를 설치한 조건에서 반응시간 5.8분, 반응온도 90$^{\cdot}C$, 메탄올:오일 몰비 9:1, KOH 농도 0.8%로 도출되었다. CSTR 반응기에서는 반응온도와 체류시간에 따른 반응특성을 조사하였으며 최적반응 조건으로 반응온도 80$^{\cdot}C$, 메탄올/오일 몰비 9:1, KOH 농도 0.8%, 체류시간 18.4분, 교반속도 250rpm로 조사되었다.

Effect of Feeding Ca-salts of Fatty Acids from Soybean Oil and Linseed Oil on c9,t11-CLA Production in Ruminal Fluid and Milk of Holstein Dairy Cows

  • The objective of this study was to investigate the effect of dietary supplementation with calcium salts of soybean oil fatty acids (CaSO) and linseed oil fatty acids (CaLO) on c9,t11-CLA production in ruminal fluid and milk fat from Holstein dairy cows. Rumen fermentation, lactational performances and fatty acid profiles in ruminal fluid and milk fat were also investigated. Twenty multiparous Holstein dairy cows were allotted randomly into two groups consisting of ten cows in each group according to calving date and average milk yield. The first group of cows was fed a control (without calcium salts) diet and a treatment as 1.0% of CaSO (on DM basis) for 30 days in each period. In the second group, cows were fed the same control diet and 1.0% of CaLO as a treatment in the same manner. The forage: concentrate ratio was 52:48, and diets were formulated to contain 17% crude protein (DM basis) for both groups. Ruminal pH, protozoal numbers and the concentration of total volatile fatty acids were unchanged, however, the ruminal ammonia-N decreased by feeding CaSO or CaLO treatment compared to the control diet. The vaccenic acid (trans-11 C18:1; VA) in rumen fluid increased (p<0.01) by 169% and 153%, and the c9,t11-CLA content of rumen fluid increased (p<0.01) by 214% and 210% in the CaSO and CaLO treatments, respectively, compared to the control diet. In milk fatty acids, the VA content increased by 130% and 132% in the evening and morning milking times, respectively, and the c9,t11-CLA content increased by 125% in both milking times for the CaSO supplementation than that of control diet. In the case of CaLO supplementation, the VA increased by 117% and 114%, and the c9,t11-CLA increased by 96% and 94% in the evening and morning milking times, respectively, compared to the control diet. The contents of VA and c9,t11-CLA of milk fatty acids were numerically higher in the evening milking time compared to the morning milking time for control and both treatments. Finally, these results indicated that the supplementation of CaSO or CaLO treatment increased the VA and the c9,t11-CLA in both ruminal fluid and milk fat of Holstein dairy cows.

The Effect of Body Energy Reserve Mobilization on the Fatty Acid Profile of Milk in High-yielding Cows

  • We investigated the effect of the amount of body condition loss in the dry period and early lactation in 42 high-yielding Holstein-Friesian cows on milk yield and the share of fatty acids in milk fat. Energy reserves were estimated based on the body condition scoring (BCS) and backfat thickness (BFT). Milk yield and milk composition were determined over 305-d lactation. From d 6 to 60 of lactation, the concentrations of 43 fatty acids in milk fat were determined by gas chromatography. Cows were categorized based on body condition loss from the beginning of the dry period to the lowest point of the BCS curve in early lactation into three groups: low condition loss group (L) ${\leq}0.5$ points (n = 14); moderate condition loss group (M) 0.75 to 1.0 points (n = 16) and high condition loss group (H) >1.0 points (n = 12). Cows whose body energy reserves were mobilized at 0.8 BCS and 11 mm BFT, produced 12,987 kg ECM over 305-d lactation, i.e. 1,429 kg ECM more than cows whose BCS and BFT decreased by 0.3 and 5 mm, respectively. In group H, milk yield reached 12,818 kg ECM at body fat reserve mobilization of 1.3 BCS and 17 mm BFT. High mobilization of body fat reserves led to a significant (approx. 5%) increase in the concentrations of monounsaturated fatty acids-MUFA (mostly $C_{18:1}$ cis-9, followed by $C_{18:1}$ trans-11), a significant decrease in the levels of fatty acids adversely affecting human health, and a drop in the content of linoleic acid, arachidonic acid and docosahexaenoic acid in milk fat. In successive weeks of lactation, an improved energy balance contributed to a decrease in the concentrations of unsaturated fatty acids (UFA) and an increase in the conjugated linoleic acid (CLA) content of milk fat.

  • There studies were carried for evaluation of the efficiency of freezing of hamster oocytes for use in a human sperm penetration assay. The hamster oocytes fully equilibrated in various cryoprotectant agents and inseminated with human sperm. After insemination with hamster oocytes, there was no difference in penetrated rates. Cumulus free oocytes equilibrated in 1.5M various cryoprotective agents and slowely cooled to temperature $-30^{\circ}C$ before rapid cooling and storage in nitrozen tank. After rapid thawing, survival rates of frozen oocytes according to cryo-protective agents were examined and the human sperm penetration assay with zona free hamster oocytes was conducted. 1. Survival rates of oocytes after cryoprotectants exposure have no significant difference (range 88-91%) and peneration rate was 51.1%. 2. Recovery and survival rate of frozen-thawed oocytes were 85.1 and 66.8%. There was no significant difference on cryoprotective agents. 3. Penetration rates of the frozen-thawed and intact oocytes were 69.0 and 77.0%, respectively. 4. Hamster oocytes cryopreservation provides a convenient way of supplying and trans-porting hamster oocytes for the assessment of the fertilizing potential of human spermatozoa.

Fatty Acid Composition of Grain- and Grass-Fed Beef and Their Nutritional Value and Health Implication

  • Beef contains functional fatty acids such as conjugated linoleic acid and longchain fatty acids. This review summarizes results from studies comparing the fatty acid composition of beef from cattle fed either grass or grain-based feed. Since functional lipid components are contributed through dietary consumption of beef, the fatty acid composition is reported on mg/100 g of meat basis rather than on a percentage of total fat basis. Beef from grass-fed contains lesser total fat than that from grain-fed in all breeds of cattle. Reduced total fat content also influences the fatty acid composition of beef. A 100 g beef meat from grass-fed cattle contained 2,773 mg less total saturated fatty acids (SFA) than that from the same amount of grain-fed. Grass-fed also showed a more favorable SFA lipid profile containing less cholesterol-raising fatty acids (C12:0 to C16:0) but contained a lesser amount of cholesterol-lowering C18:0 than grain-fed beef. In terms of essential fatty acids, grass-fed beef showed greater levels of trans-vaccenic acid and long-chain n-3 polyunsaturated fatty acids (PUFA; EPA, DPA, DHA) than grain-fed beef. Grass-fed beef also contains an increased level of total n-3 PUFA which reduced the n-6 to n-3 ratio thus can offer more health benefits than grain-fed. The findings signify that grass-fed beef could exert protective effects against a number of diseases ranging from cancer to cardiovascular disease (CVD) as evidenced by the increased functional omega-3 PUFA and decreased undesirable SFA. Although grain-fed beef showed lesser EPA, DPA, and DHA, consumers should be aware that greater portions of grain-fed beef could also achieve a similar dietary intake of long-chain omega-3 fatty acids. Noteworthy, grain-fed beef contained higher total monounsaturated fatty acid that have beneficial roles in the amelioration of CVD risks than grass-fed beef. In Hanwoo beef, grain-fed showed higher EPA and DHA than grass-fed beef.

Comparison of miR-106b, miR-191, and miR-30d expression dynamics in milk with regard to its composition in Holstein and Ayrshire cows

  • Objective: Milk composition varies considerably and depends on paratypical, genetic, and epigenetic factors. MiRNAs belong to the class of small non-coding RNAs; they are one of the key tools of epigenetic control because of their ability to regulate gene expression at the post-transcriptional level. We compared the relative expression levels of miR-106b, miR-191, and miR-30d in milk to demonstrate the relationship between the content of these miRNAs with protein and fat components of milk in Holstein and Ayrshire cattle. Methods: Milk fat, protein, and casein contents were determined in the obtained samples, as well as the content of the main fatty acids (g/100 g milk), including: saturated acids, such as myristic (C14:0), palmitic (C16:0), and stearic (C18:0) acids; monounsaturated acids, including oleic (C18:1) acid; as well as long-, medium- and short-chain, polyunsaturated, and trans fatty acids. Real-time stem-loop one-tube reverse transcription polymerase chain reaction with TaqMan probes was used to measure the miRNA expression levels. Results: The miRNA expression levels in milk samples were found to be decreased in the first two months in Holstein breed, and in the first four months in Ayrshire breed. Correlation analysis did not reveal any dependence between changes in the expression level of miRNA and milk fat content, but showed a multidirectional relationship with individual milk fatty acids. Positive associations between the expression levels of miR-106b and miR-30d and protein and casein content were found in the Ayrshire breed. Receiver operating characteristic curve analysis showed that miR-106b and miR-30d expression levels can cause changes in fatty acid and protein composition of milk in Ayrshire cows, whereas miR-106b expression level determines the fatty acid composition in Holsteins. Conclusion: The data obtained in this study showed that miR-106b, miR-191, and miR-30d expression levels in milk samples have peculiarities associated with breed affiliation and the lactation period.

에탄올이 신경아세포종 B103세포의 Protein Kinase C Isozyme 활성에 미치는 영향 (Effect of Ethanol on the PKC Isozyme Activities in B103 Neuroblastoma Cells)

  • 에탄올이 지속적으로 뇌 신경세포에 미치는 영향을 조사하기 위하여 흰쥐의 신경세포로부터 유래 된 B103 neuroblastoma cell을 사용하여 세포독성이 나타나지 않는 에탄을 농도(0, 50, 100, 200 mM)에서의 1, 2, 8, 18, 24시간 경과에 따라 유도되는 PKC $\alpha$, ${\gamma}$, $\varepsilon$, ζ isozyme들의 양을 세포질 분획과 세포막 분획으로 나누어 Western blot으로 각각 분석하였다. 100 mM의 에탄올 농도에서 분석된 PKC isozyme들 중 PKC-$\varepsilon$는 18시간대의 세포질에서 그리고 PKC-$\varepsilon$은 8∼18시간대의 세포막분획에서 각각 현저한 유도현상을 보였다 PKC-$\alpha$는 200 mM의 에탄을 첨가 후 18시간과 24시간에 세포질과 세포막 분획에서 모두 대조군의 150%까지 현저한 증가를 나타낸 반면 PKC-ζ는 100, 200 mM 에탄올농도에서 배양(18, 24시간 동안)한 세포의 세포막분획에서만 유도되었다. 그리고 50, 100, 200 mM의 에탄올 농도에서 24시간동안 배양한 세포질 분획에서 PKC-${\gamma}$는 농도 의존적으로 감소하여 200 mM의 에탄올 농도에서는 대조군의 47%까지 현저한 감소를 나타내었으며, 세포내에 세포독성을 나타내지 않는 농도 특히 100∼200mM농도범위의 에탄올을 첨가하여 24시간 동안 지속적으로 배양할 때 PKC-${\gamma}$$\varepsilon$이 관련된 신호전달체계가 억제됨을 보였다. 이는 에탄올이 PKC isozyme들의 상호간 조절을 통해 신호전달계 또는 신경전달 물질들의 변화에 영향을 줄 수 있음을 시사하며 에탄올의 중추신경계에 미치는 지속적 영향으로 나타나는 행동장애 및 뇌 기능의 손상 또는 보호과정 에 PKC-isozyme들이 관여할 수 있음을 시사한다.

한우의 생식세포 보존에 관한 연구 I. 한우 정액의 일반성상 및 동결후 생존성에 미치는 영향 (Studies on Preservation of Germ Cells in Hanwoo I. Effects of Frozen Thawed Viability and Characteristics in Bovine Semen)

  • 한우의 유전자원을 보존하기 위하여 정액동결 시험을 수행하였으며 고급육계통은 육질에 대한 육종가 상위 10% 이내의 자손에서 선발하였고 다 유계통은 어미의 이유시 체중에 대한 육종가 상위 10%이내의 자손에서 선발하였으며 2개 계통에서 종모우 총 13두를 선발하여 공시하였다. 정액채취는 인공질법으로 실시하였고 의빈대에 수소를 계류하고 채정대상우를 승가시켰으며, 3회 가승가후에 채정하였다. 채정후 10분 이내에 실험실로 옮겨와 검사항목을 조사하고 37$^{\circ}C$에서 1차 희석을 하고 5$^{\circ}C$까지 하강한 후 2차 희석을 하였으며, 액체 질소 5cm 위에서 5분간 평형후 침지하여 동결하였다. 이들 종모우 정액의 일반적 성상에 있어서 채정된 정액량은 1차와 2차를 합하였을 때 평균 채정량이 5.7 ml였고 정자농도는 975${\times}$$10^{6}$개였으며 정액의 색상은 유백색이었고 pH는 6.8이었다. 채취시 생존성은 90.2% 그리고 동결 융해후에는 65.7 %가 생존하였으며 총 6,870스트로우의 동결 정액을 생산하여 보존하였다.

서울지역 소매업체 제빵류의 트랜스지방 및 콜레스테롤 조사 (Monitoring of Trans Fatty Acid and Cholesterol of Bakery Products Sold at Retail in Seoul Area)

  • 서울시내 제과점중 소매점을 중심으로 제빵류 54건에 대하여 조지방함량, 지방산조성, 포화지방산과 불포화지방산조성, 트랜스지방 함량과 콜레스테롤 함량을 분석하였다. 조지방 [mean (minimum-maximum), %]은 식빵에서 6.46% (3.51~8.69%)로 나타나서 페이스츄리 16.23% (3.55~25.56%)와 생크림케이크 16.26% (8.61~31.58%) 보다 낮았다. 식빵, 페이스츄리, 생크림케이크의 지방산 중 palmitic acid가 각각 29.98% (19.84~34.64%)와 35.06% (27.73~40.91 %), 그리고 21.33% (13.43~33.02%)로서 가장 높은 지방산 함량을 보여주었다. 생크림케이크의 경우 lauric acid가 22.17% (4.36~30.82%)로 식빵이나 페이스츄리에 비해 높은 함량을 보여 주었다. 포화지방산은 식빵 54.04%, 페이스츄리 53.41%, 불포화지방산은46.05%, 46.63%이다. 생크림케이크 경우 포화지방산이 70.09${\pm}$8.64%인 반면에 불포화지방산은 29.06${\pm}$8.41%로 나타났다. 트랜스지방산 이성질체는 C18:1t 만이 검출되었다. 트랜스지방의 함량은 생크림케이크가 0.14 g/100 g food으로 가장 높았다. 빵류의 l 회 제공량인 70 g을 기준으로 트랜스지방이 0.2 g을 초과하는 시료의 시료를 판정한 결과 생크림케이크에서만 17건 중 4건의 시료가 0.2 g을 초과하였다. 콜레스테롤 함량은 생크림케이크 30.55%(0.0~132.99%), 페이스츄리 8.11 % (0.0~42.80%), 식빵 2.36%(0.0~12.86%) 순이었다. 빵류의 1회제공량인 70 g을 기준으로 콜레스테롤이 2mg을 초과하는 시료의 수는 식빵에 비해 페이스츄리와 생크림케이크에서 다소 검출건수가 높은 것으로 나타났다. 바쁜 현대인들의 식습관에 따라 점점 빵의 소비가 증가하고 있지만 대량으로 시판되는 빵에 비해 소매 업체 빵은 표기가 부족하므로 불신을 먼저 갖기 보다는 올바른 정보제공으로 빵을 선택할 때 소비자들에 안심을 주도록해야 할 것이다.
