Effect of Ge(Germanium) Treatment on Rice Quality (게르마늄 처리가 쌀 품질에 미치는 영향)

  • This study was conducted to investigate the effects of Ge(germanium) treatment on rice quality. Rice samples were divided into the following treatment groups: control(CON: cultivated without Ge), Ge-1(cultivated with 200 kg of rough stone powder containing 1.6 mg/kg germanium per 10 ha), and Ge-2(cultivated with 500 kg of rough stone powder containing 1.6 mg/kg germanium per 10 ha). The mean total Ge level in the Ge-2 sample was 20.47 ppb. The levels of Ca and Na in the Ge-2 rice increased by 65.12 and 110.28%, respectively, when compared to the control, whereas the Zn, Mn, Fe, Mg and K content decreased by 11.44~30.50%. No significant difference in the percentage weight of C and O was observed among samples. The order of the percentage weight of P, S, and Cl was Ge-2>Ge-1>CON. The free amino acids were higher in samples from the Ge-1 and Ge-2 groups than in samples from the control. The GABA($\gamma$-aminobutyric acid) amount in the Ge-2 products was significantly high compared to other groups. The micro structure of Ge-2 showed a firmer network than the control and had a macroporous structure. Conversely, the Ge-2 products had higher scores for stickiness, hardness and overall taste when compared to the other groups. These results suggest that rice treated with rough stone powder containing germanium can be used in the production of commercially-desired functional rice.

Occurrence and Chemical Composition of White Mica from Wallrock Alteration Zone of Janggun Pb-Zn Deposit (장군 연-아연 광상의 모암변질대에서 산출되는 백색운모의 산상 및 화학조성)

  • The Janggun Pb-Zn deposit has been known one of the four largest deposits (Yeonhwa, Shinyemi, Uljin) in South Korea. The geology of this deposit consists of Precambrian Weonnam formation, Yulri group, Paleozoic Jangsan formation, Dueumri formation, Janggum limestone formation, Dongsugok formation, Jaesan formation and Mesozoic Dongwhachi formation and Chungyang granite. This Pb-Zn deposit is hydrothermal replacement deposit in Paleozoic Janggum limestone formation. The wallrock alteration that is remarkably recognized with Pb-Zn mineralization at this deposit consists of mainly rhodochrositization and dolomitization with minor of pyritization, sericitization and chloritization. Wallrock alteration is divided into the five zones (Pb-Zn orebody -> rhodochrosite zone -> dolomite zone -> dolomitic limestone zone -> limestone or dolomitic marble) from orebody to wallrock. The white mica from wallrock alteration occurs as fine or medium aggregate associated with Ca-dolomite, Ferroan ankerite, sideroplesite, rutile, apatite, arsenopyrite, pyrite, sphalerite, galena, quartz, chlorite and calcite. The structural formular of white mica from wallrock alteration is (K0.77-0.62Na0.03-0.00Ca0.03-0.00Ba0.00Sr0.01)0.82-0.64(Al1.72-1.48Mg0.48-0.20Fe0.04-0.01Mn0.03-0.00Ti0.01-0.00Cr0.00As0.01-0.00Co0.03-0.00Zn0.03-0.00Pb0.05-0.00Ni0.01-0.00)2.07-1.92 (Si3.43-3.33Al0.67-0.57)4.00O10(OH1.94-1.80F0.20-0.06)2.00. It indicated that white mica from wallrock alteration has less K, Na and Ca, and more Si than theoretical dioctahedral micas. The white micas from wallrock alteration of Janggun Pb-Zn deposit, Yeonhwa 1 Pb-Zn deposit and Baekjeon Au-Ag deposit, and limestone of Gumoonso area correspond to muscovite and phengite and white mica from wallrock alteration of Dunjeon Au-Ag deposit corresponds to muscovite. Compositional variations in white mica from wallrock alteration of these deposits and limeston of Gumoonso area are caused by mainly phengitic or Tschermark substitution mechanism (Janggun Pb-Zn deposit), mainly phengitic or Tschermark substitution and partly illitic substitution mechanism (Yeonhwa 1 Pb-Zn deposit, Dunjeon Au-Ag deposit and Baekjeon Au-Ag deposit), and mainly phengitic or Tschermark substitution and partly illitic substitution or Na+ <-> K+ substitution mechanism (Gumoonso area).

Isolation and Characterization of Dextrans Produced by Leuconostoc sp. strain JYY4 from Fermented Kimchi

  • Dextran is a generic term for a bacterial exopolysaccharide synthesized from sucrose and composed of chains of D-glucose units connected by ${\alpha}$-1,6-linkages by using dextransucrases. Dextran could be used as vicosifying, stabilizing, emulsifying, gelling, bulking, dietary fiber, prebiotics, and water holding agents. We isolated new strain capable of producing dextran from Korean traditional kimchi and identified as Leuconostoc sp. strain JYY4. Batch fermentation was conducted in bioreactor with a working volume of 3 L. The media was MMY and 15% (w/v) sucrose. Mineral medium consisted of $3.0g\;KH_2PO_4$, $0.01g\;FeSO_4$, $H_2O$, $0.01g\;MnSO_4$, $4H_2O$, $0.2g\;MgSO_4\;7H_2O$, 0.01 g NaCl, $0.05g\;CaCl_2$ per 1 liter deionized water. The pH of media was initially adjusted to 6.0. The inoculation rate was 1.0% (v/v) of the working volume. Temperature was maintained at $28^{\circ}C$. The agitation rate was 100 rpm. The production pattern of dextran was associated with the cell growth. After 24 hr dextran reached its highest concentration of 59.4 g/L. The sucrose was consumed completely after 40 hr. Growth reached stationery phase when sucrose became limiting, regardless of the presence of fructose or mannitol. When the specific growth rate was 0.54 hr-1, utilization averaged 5.8 g/L-hr. The yield and productivity of dextran were 80% and 2.0 g/L-hr, respectively. Dextrans produced by were separated to two different size by an alcohol fraction method. The size of high molecular weight dextran (45% alcohol, v/v), less soluble dextran, was between MW 500,000 and 2,000,000. Soluble dextran (55% alcohol, v/v) was between 70,000 and 150,000. The molecular weight average of total dextran (70% alcohol, v/v) was between 150,000 to 500,000. The enzymatic hydrolyzates of total dextran of ATCC 13146 showed branched dextrans by Penicillium dextranase contained of glucose, isomaltose, isomaltotriose, and isomaltooligosaccharides greater than DP4 (degree of polymerization) that had branch points. Compounds greater than DP4 were branched isomaltooligosaacharides. Hydrolysates by the Lipomyces dextranase produced the same composition of oligosaccharides as those by Penicillin dextranase.

  • Fungi which produce red pigment were isolated from tapioca chips, Korean Koji, air, and plant leaves. Among the Fungi, T-1 strain was selected as test strain. This strain was identified as a Monatcus sp. by the morphological propeties. Various culture conditions, and physical and physiological characteristics of red pigment were studied. According to the studies of culture conditions, optimum condition was found to be pH 6.5; 4 days of incubation; temperature, 32~33 c: 3.5% of Tapioca chips powder as carbon source, 0.2% of sodium nitrate as nitrogen source and 100 ml of medium in the 500 ml Erlenmyer flask at a rotary shaker (rpm180) as aeration condition. Also effective levels of vitamins, amino acids and inorganic compounds was found to be l$\mu\textrm{g}$/ml of folic acid and niacin; 0.3% of L-arginine, L-glutamic acid and L-proline; and 0.001% of manganese dioxide giving good results.

Physicochemical Characteristics of Dredged Soils in Reservoirs (저수지 준설대상 토양의 이화학적 특성)

  • This study was carried out to examine the physicochemical characteristics of dredged soils in reservoirs. Surveys and analyses of basic materials were made on 241 of 2,328 reservoirs in Chonbuk province through 2 years from December 1994 to November 1996. The results of this study are summarized as follows : 1. Soils were classified as 15 types according to physical properties, and some soils contain comparatively high percentage of sand and gravel. Considering only soil textures, useful and economical soils as aggregate are approximately 25% in all, and the other soils are arable for farm planting. 2. The results of chemical analyses of soils showed on the average 5.9 in pH, 1.lmmhos/cm in ECe, 14.6me/l00g in CEC, 460.0ppm in T-N, 119.0ppm in T-P, 264.9ppm in K, 134.2ppm in Na, l,335.0ppm in Ca, 575.9ppm in Mg, 486.Sppm in Fe, 57.7ppm in Mn, 3.3ppm in Cu, 21.9ppm in Zn, 0.49ppm in As, 0.34ppm in Cd, 0.O3ppm in Hg, 1.7% in OM, respectively. 3. General chemical components, heavy metals, organic matter contents were analyzed as similar to tlie mean values of common soils, therefore it was considered to be no significant effects on crop growth in the chemical properties. 4. Accodingly, the physicochemical characteristics of soils ought to be analyzed accurately before dredging for effective using of dredged soils. And it will be more effective, if the dredged soils are used with proper balance among each content of components with considering to the physicochemical properties of common soils.

Concentration of Heavy Metals in Natural Soils of Jeju Island, Korea (제주도 자연토양에서 중금속의 농도)

  • For 63 soil series distributed in Jeju Island, natural uncultivated soils in each soil series were collected, and their physicochemical properties and their concentrations of 19 heavy metals including 8 heavy metals which are regulated by Korean Soil Environment Conservation Law, were analyzed. Moreover, the correlations between physicochemical properties and heavy metal concentrations, and between heavy metal concentrations were analyzed. The heavy metals distributed in the higher concentrations and the lower concentrations with arithmetric mean value, were Mn(730 mg/kg) and Ba(493 mg/kg), and Hg(0.146 mg/kg) and Tl(0.096 mg/kg), respectively. The correlations between pH($H_2O$) and heavy metals(Hg, Ni, Co, Se), between pH(NaF) and heavy metals(Hg, Ba, Se, Tl), and between organic matter content and heavy metals(Hg, Tl) were significant at the 0.01 level. From the correlations between heavy metal concentrations, there were 22 where there were significant at the 0.01 level and they showed positive correlation. Among those, the heavy metals showing the correlation higher than r=0.5, were Sb-V(0.878), Mo-Sn(0.867), Co-V(0.654), Co-Sb(0.648), Be-Sn(0.546), and Sn-Tl(0.528).

  • Quartz veins from the Jukwangri area of Hwawon-myeon are epithermal quartz veins that are filling the NW or NE-trending faults within Precambrian metasedimentary rocks. Based on their prolongation and ore grades, No. 1 quartz vein can be traced for about 200 m and varies 0.1 to 3 m in thickness. Mineralization of No. 1 quartz vein can be divided into hypogene and supergene stages. Hypogene stage is associated with hydrothermal alteration minerals(phyllic and argillic zones) such as illite, sericite and sulfides such as pyrite, arsenopyrite, sphalerite. chalcopyrite, galena, argentian tetrahedrite. Supergene stage is composed of Fe-Mn oxide, Zn-Fe oxide and Pb oxide. Fluid inclusion data indicate that homogenization temperature and salinity of hypogene stage range from 187 to $306^{\circ}C$ and ken 0.0 to 6.2 wt.% eq. NaCl, respectively. They suggest that ore forming fluids were progressively cooled and diluted from mixing with meteoric water. Oxygen($-4.1{\sim}4.1%o$) and hydrogen($-107{\sim}-88%o$) isotope com-positions indicated that hydrothermal fluids were derived from meteoric and evolved by progressive mixing with meteoric water during mineralization.

Optimization of Submerged Culture Conditions for Mycelial Growth and Exopolysaccharides Production by Agaricus blazei

  • The influences of inoculum size, pH, and medium composition on mycelial growth and exopolysaccharides (EPS) production were investigated in shake flasks and in a bioreactor. The optimum inoculum size for both mycelial growth and EPS production was identified to be 10% (v/v) in shake flask cultures. The optimal initial pH for mycelial growth and EPS production in shake flask cultures were found to be 5.0 and 7.0, respectively. However, the optimal pH was 5.0 for both mycelial growth and EPS production in bioreactor cultures where the pH was regulated. The optimal mass ratio of the two major carbon sources, glucose to dextrin, was 1:4. The optimal mass ratio of the two major nitrogen sources, yeast extract to soy tone peptone, was 2:1. When 500 mg $1^{-1}$ of $MnSO_4-5H_2O$ was added to the bioreactor culture, both mycelial growth and EPS production were enhanced by approximately 10%. Under the optimized conditions, a mycelial biomass of 9.85 g $1^{-1}$ and an EPS concentration of 4.92 g $1^{-1}$ were obtained in 4 days.

Geochemistry and Mineralogy of Mine Drainage Water Precipitate and Evaporite Minerals in the Hwasoon Area (화순 폐탄광지역 광산배수와 침전 및 증발잔류광물에 대한 지구화학적 및 광물학적 연구)

  • This study investigated the geochemical characteristics of mine drainage discharged from an abandoned coal mine in the Hwasoon area. Surface water samples were collected from 23 locations along the Hancheon creek. The concentration of Zn and Cu in stream waters was highest at low pH (3.53), whereas the content of TDS and TDI was highest at high pH (7.78) due to the concentration of Ca, $HCO_3$ and $SO_4$. At the upstream site, the Ba, Fe, Mn, Zn, and $SO_4$ contents were relatively high but decreased significantly with the distance from the coal mine. On the contrary, the Na and $NO_3$ contents were low at the upstream site but increased downstream. Yellow precipitate material collected in the Hancheon consisted mainly of iron and LOI. This yellow precipitate was heated from 100 to $900^{\circ}C$ for 1 hour. With increasing temperature, the intensity of hematite peaks were sharply produced in X-ray pattern and the absorption band Fe-O of hematite increased in IR due to dehydration and melting. The yellow to brown precipitate and evaporite materials were collected by a air-dry from the acid mine water at the laboratory. After drying, the concentration of ions in the acid water samples increased progressively in oversaturation with respect to either gypsum, ferrohexahydrite or quenstedetite. The X-ray powder diffraction studies identified that the precipitated and evaporated materials after drying were well crystallized gypsum, ferrohexahydrite and quenstedetite. Diagnostic peaks used for identification of gypsum were the 7.65, 4.28, 3.03, 2.87 and 2.48$\AA$ peaks and those for ferrohexahydrite were the 5.46, 5.12, 4.89, 4.44, 4.05, 3.62, 3.46, 3.40, 3.20, 3.03, 2.94, 2.53, 2.28, 2.07, 1.88 and 1.86${\AA} peaks. The IR spectra with OH-stretching, deformation of $H_2O$and ${SO_4}^{2-}$stretching vibration include the existence of gypsum, ferrohexahydrite and quenstedetite in the precipitated and evaporite materials. In the SEM and EDS analysis for the evaporite material, gypsum with well-crystallized, acicular, and columnar form was distinctly observed.

Estimation of Characteristics and Methane Production Rate of Food Waste (음식물류 폐기물 특성 및 메탄 발생가능량 평가)

  • This research was performed to evaluate the characteristics of food waste from 5 areas in Gangwon Province, Korea and to predict the $CH_4$ gas production rate. Food wastes were sampled in July and September, 2017. The amount of methane gas generation was evaluated through the biochemical methane potential (BMP) test and a calculation method using chemical composition. Average bulk density and pH of the food wastes were in the range of $0.758{\sim}0.850g\;cm^{-3}$ and 4.29 ~ 4.75, respectively. By physical composition, vegetables were the highest with 56.43 ~ 72.81% with fruits recording 5.31 ~ 8.95%, cereals 1.60 ~ 18.73%, fish and meat 4.47 ~ 12.11%, and filtrate 1.76 ~ 3.64%. The average water content was 69.30 ~ 75.87%, and VS and ash content were 22.50 ~ 27.98% and 1.63 ~ 2.48%, respectively. In addition, $BOD_5$, $COD_{Cr}$, and $COD_{Mn}$ were in the ranges of $17,690.3{\sim}33,154.9mg\;L^{-1}$, $106,212.3{\sim}128,695.5mg\;L^{-1}$, and $51,266.1{\sim}63,426.3mg\;L^{-1}$, respectively. The NaCl content ranged from 0.81 to 1.17%. The results of elemental analysis showed that the contents of C, H, O, N, and S were 44.87 ~ 48.1%, 7.12 ~ 7.57%, 40.13 ~ 43.78%, 3.22 ~ 4.14%, and 0.00 ~ 0.02%, respectively. In a comparison of the methane production yield per VS mass of food waste, there was no significant difference between the cumulative amount (${0.303{\sim}0.354m_{CH4}}^3\;{kg_{VS}}^{-1}$) by the BMP test and the theoretical amount (${0.294{\sim}0.352m_{CH4}}^3\;{kg_{VS}}^{-1}$) calculated by chemical composition.
