Studies on the Optimal Culture Conditions for Itaconic Acid Production by Aspergilus Terreus (Aspergillus Terreus에 의한 이타콘산 생산을 위한 최적배양조건에 관한 연구)

  • The production of itaconic acid by Aspergillus terreus NRRL 1960 was studied. The optimal culture conditional such as pH, inoculum size and medium composition were established. Maximum production of itaconic acid, $19.18g/\ell$, was obtained when the cultivation was carried out at $37^{\circ}C$ and pH 2.5 for 7days, with medium containing 5%(w/v) glucose, 0.5%(w/v) NH4Cl, 0.2%(w/v) yeast extract 0.1%(w/v) CaC12, 0.1%(w/v) MgSO4 and 0.2%(w/v) NaCl. A proper medium for inoculum culture was found to be 2%(w/v) malt extract. The batch production of itaconic acid with free cells in a stirredtank reactor was not efficient compared to the shake-flask culture.

Optimization for Removal of Nitrogen Using Non-consumable Anode Electrodes (비소모성 Anode(산화전극)을 이용한 질소 제거 최적화)

  • Research was conducted to derive the optimal operation conditions and the optimal cathode for using a DSA electrode as an anode to minimize electrode consumption during the removal of nitrogen from wastewater by the electro-chemical method. Of the various electrodes tested as cathodes, brass was determined to be the optimal electrode. It had the highest NO3-N removal rate and the lowest concentration of residual NH3-N, a by-product when Cl is present in the solution. Investigating the effect of current density found that when the initial concentration of NO3-N was 50 mg L-1, the optimal current density was 15 mA cm-2. In addition, current densities above 15 mA cm-2 did not significantly affect the NO3-N removal rate. The effect of electrolytes on removing NO3-N and minimizing NH3-N was investigated by using Na2SO4 and NaCl as electrolytes and varying the reaction times. When Na2SO4 and NaCl are mixed at a ratio of 1.0 g L-1 to 0.5 g L-1 and reacted for 90 min at a current density of 15 mA cm-2 and an initial NO3-N concentration of 50 mg L-1, the removal rate of NO3-N was about 48% and there was no residual NH3-N. On the other hand, when using only 1.5 g L-1 of NaCl as an electrolyte, the removal rate of NO3-N was the highest at about 55% and there was no residual NH3-N.

  • The growth characteristics of Candida tropicalis KIST 359, isolated from soil samples collected at an oil depot in Korea, have been studied by cultivating batches under varying conditions. The conclusions of the study were: 1. The yeast easily assimilates hydrocarbons in a range of $C_{14}-C_{17}$, and the optimum cultivation temperature and pH are $30^{\circ}C$ and 5.5, respectively. 2. Using this strain of micro-organism, gas oil gives a higher cell yield than kerosine and with gas oil except urea all other nitrogen sources $(NH_4NO_3,\;NH_4Cl,\;(NH_4)_2SO_4\;and\;(NH_4)_2HPO_4)$ similarly support a satisfactory growth of the yeast. 3. The highest yield is obtained with a gas oil level of 10%(v/v), and concentrations of nitrogen source and $MgSO_4{\cdot}7H_2O$ of 0.5 and 0.05%(w/v), respectively. 4. The protein content of dried yeast cells is 59.8%. Its amino acid composition can be compared well with that of FAO provisional patterns, but with a low methionine and a high lysine content.

Characteristics of Ionic Components in Size-resolved Particulate Matters in Suwon Area (수원지역 분진의 입경별 이온성분 분포특성에 관한 연구)

  • The main purpose of this study was to investigate air quality trends of ambient aerosol with obtaining size-fractionated information. The suspended particulate matters were continuously collected on membrane filters and glass fiber filters by an 8-stage cascade impactor for 2 years (Sep. 2005 $\sim$ Sep. 2007) in Kyung Hee University-Global Campus. 8 ionic species ($Na^+$, ${NH_4}^+$, $K^+$, $Mg^{2+}$, $Ca^{2+}$, $Cl^-$, ${NO_3}^-$, and ${SO_4}^{2-}$) were analyzed by an IC after performing proper pretreatments of each sample filter. The average concentration levels of each ion were $9.24{\mu}g/m^3$ of ${SO_4}^{2-}$, $7.35{\mu}g/m^3$ of ${NO_3}^-$, $2.81{\mu}g/m^3$ of ${NH_4}^+$, $2.11{\mu}g/m^3$ of $Ca^{2+}$, $1.65{\mu}g/m^3$ of $Cl^-$, $1.87{\mu}g/m^3$ of $Na^+$, $0.80{\mu}g/m^3$ of $Mg^{2+}$, and $0.54{\mu}g/m^3$ of $K^+$, respectively. The distribution pattern of $Na^+$, $Mg^{2+}$, $Ca^{2+}$, $Cl^-$, and ${NO_3}^-$ was bi-modal and two peaks appeared in the range of $0.4{\sim}0.7{\mu}m$ and $3.3{\sim}4.7{\mu}m$, respectively. On the other hand, ${SO_4}^{2-}$, ${NH_4}^+$, and $K^+$ showed patterns of uni-modal distribution, mostly abounded in the fine mode group.

Changes in Mineral Uptake and Hormone Concentrations in Rice Plants Treated with Silicon, Nitrogen and Calcium Independently or in Combination (규소, 질소, 칼슘 단독 및 혼합처리가 벼 식물체 내 무기성분 흡수 및 식물호르몬 함량 변화에 미치는 영향)

  • To elucidate the physiological responses of rice plants to the essential mineral silicon (Si), we assessed the effects of treatments with Si, nitrogen ($NH_4NO_3$; ammonium nitrate), and calcium ($CaCl_2$; calcium chloride), independently or in combination on mineral uptake rates and levels of the hormones abscisic acid (ABA), gibberellin ($GA_1$) and jasmonic acid (JA). We found that nitrogen and calcium uptake was inhibited by Si application. However, solo application of nitrogen or calcium did not affect Si uptake. Compared to the untreated plants, the application of Si, $NH_4NO_3$ or $CaCl_2$ increased the endogenous hormone levels in treated plants. In particular, the concentrations of $GA_1$ and JA increased significantly after the application of Si or $NH_4NO_3$. The level of $GA_1$ observed after a treatment (solo or combine) with Si, and $NH_4NO_3$ was higher than that of the control. By contrast, independent application of $CaCl_2$ or a combined treatment with Si and $CaCl_2$ did not alter $GA_1$ levels. The highest level of $GA_1$ was present in plants given a combination treatment of Si and $NH_4NO_3$. This effect was observed at all time points (6 h, 12 h and 24 h). Endogenous JA contents were higher in all treatments than the control. In particular, a combination treatment with Si and $NH_4NO_3$ significantly increased the JA levels in plants compared to other treatments at all time points. A small increase in JA levels was observed after 6 h in plants given the $CaCl_2$ treatment. However, JA levels did not differ between plants given a $CaCl_2$ treatment and controls after 12 h or 24 h of exposure. We conclude that treatment with $CaCl_2$ alone does not affect endogenous JA levels in the short term. Endogenous ABA contents did not show any differences among the various treatments.

  • Four component $Ag_2S$-PbS-$PbHAsO_4-Cu_2S$ and three component $Ag_2S$-PbS-$PbHAsO_4$ electrodes have been prepared and evaluated for the direct measurement of monohydrogen arsenate. The 3.0 : 0.5 : 1.0 : 0.25 (mole ratio, $Ag_2S$:PbS:$PbHAsO_4:Cu_2S$) composition is superior in terms of potentiometric response, stability, rapidity of response and reproducibility. Testing was done over the concentration range $10^{-1}$~$10^{-4}M\;HA_SO_4^{2-}$in 0.1F NH4Ac-NH4OH buffer solution at pH 8.50 with constant ionic strength. Interfering ions were $CN^-,\;I^-,\;S^{-2}$ and $Cl^-$.

  • The effects of salts which was used as a catalysis in formation of $\alpha$-calciumusulfate hemihydrate from dicalcium sulfate hydrate were investigated on the hydration of $\alpha$-calciumsulfate hemihydrate. The hydration of $\alpha$-calciumsulfate hemihydrate was studied by the measurements of crystalline water, heat evolution. Also the hydrates were analyzed by XRD, DSC and SEM. The promotive effect each salts on the hydration was as follows: NaCl>NH4Cl>NaNO3>NH4NO3, and the hydration rate was accelerated with concentration of salts. The effect of Al2(SO4)3 and potassium sodium tartrate on the hydration was slmilar to water, whereas sodium succinate and gelatin retarded the hydration in comparision with water. These salts affected the hydration time but total heat evoution was similar.

  • Copper-dimethylglyoxime complex (CuDMG) modified Copper electrode (Cu/CuDMG) showed a catalytic activity towards cyclohexanol oxidation in NaOH solution. The modified electrode prepared by the dimethylglyoxime anodic deposition on Cu electrode in the solution contained 0.20 M $NH_4Cl\;+\;NH_4OH\;(pH\;9.50)\;and\;1\;{\times}\;10^{-4}$ M dimethylglyoxime. The modified electrode conditioned by potential recycling in a potential range of -900${\sim}$900 mV vs. Ag/AgCl by cyclic voltammetry in alkaline medium (1 M NaOH). The results show that the CuDMG film on the electrode behaves as an efficient catalyst for the electro-oxidation of cyclohexanol in alkaline medium via Cu (III) species formed on the electrode.

Characterization of $NH_4\;^+$ and $Zn^{2+}$ Adsorption by Korean Natural Zeolites (한국산(韓國産) 천연(天然) 제오라이트의 암모늄과 아연이온의 흡착(吸着)특성)

  • The adsorption of $NH_4\;^+$ and $Zn^{2+}$ by four Korean zeolites, the major species of which are clinoptilolite, clinoptilolite with mordenite, mordenite with clinoptilolite, and mordenite was measured in different concentrations of solutions of $NH_4\;^+$ and $Zn^{2+}$, and their mixtures. The adsorption of $NH_4\;^+$ was greater than that $Zn^{2+}$ far all samples at the concentrations of the added solutions from $1\;to\;7{\times}10^{-3}N$ and this difference was greater at the higher concentrations. Also, $Zn^{2+}$ adsorption by samples was decreased by the presence of $NH_4\;^+$, but that of $NH_4\;^+$ by the presence of $Zn^{2+}$ was not. The extent of $NH_4\;^+$ selectivity among samples was increased in order of clinoptilolite with mordenite$NH_4\;^+$ adsorbed by six successive equilibrations with the solution containing both $NH_4\;^+$ and $Zn^{2+}$ each at a concentration of $3{\times}10^{-3}N$ were in range from 43.7 to 50.4 me/100g, whereas those amounts of $Zn^{2+}$ were in the range from 6.6 to 17.0 me/100g. It was suggested from these results that mordenite and clinoptilolite, particularly the former, can be used for removal of $NH_4\;^+$ from municipal wastewater and those zeolites treated with wastewater can be applied to agricultural land.

Influences of Culture Medium Components on the Production Poly (γ-Glutamic Acid) by Bacillus subtilis GS-2 Isolated Chungkookjang (청국장에서 분리한 Bacillus subtilis GS-2에 의한 Poly(γ-Glutamic Acid) 생산의 최적 배양조건)

  • A bacterium strain GS-2 isolated from the Korean traditional seasoning food, Chungkookjang and was determined to produce large amounts of ${\gamma}$-PGA with high productivity when provided with simple nutrients (L-glutamic acid 2.0%, glucose 1.0%, $NH_4Cl$ 0.5%, $KH_2PO_4$ 0.05%, $MgSO_4{\cdot}7H_2O$ 0.01%, pH 7.0). In this study, the culture medium for this strain was optimized for the production of ${\gamma}$-PGA. The Bacillus subtilis GS-2 required supplementation with L-glutamic acid and other nutrients for maximal production of ${\gamma}$-PGA. The optimal culture conditions for ${\gamma}$-PGA production were a 48 hr culture time, a temperature of $33^{\circ}C$ and initial pH of 6.5 by rotary shaking (220 rpm). A maximum ${\gamma}$-PGA production of 31.0 $g/{\ell}$ was obtained with L-glutamic acid (30 $g/{\ell}$), sucrose (the main carbon source, 30 $g/{\ell}$), $NH_4Cl$ (the main nitrogen source, 2.5 $g/{\ell}$), $KH_2PO_4$ (1.5 $g/{\ell}$) and $MgSO_4{\cdot}7H_2O$ (0.15 $g/{\ell}$) in the culture medium.
