  • In this study, we investigated the biological antioxidant and antibacterial activity of Chamaecyparis obtusa (C. obtuse) extracts by measuring DPPH radical scavenging and ABTS radical scavenging, and SOD-like activities. The DPPH and ABTS radical scavenging activities were increased in a dose-dependent manner, with maximum activities of 78% and 62% at an extract concentration of $50{\mu}l/ml$. The C. obtusa extracts also showed high SOD-like activity, with a maximum activity of 92.85% at a concentration of $50{\mu}l/ml$. The antibacterial activities of C. obtusa extracts were measured against six types of bacteria known to cause food poisoning and disease. Antibacterial activity was investigated against three gram-positive and three gram-negative bacteria using the paper disc agar diffusion method. The C. obtusa extracts showed antibacterial activities against B. cereus, E. coli, L. monocytogenes, S. aureus, S. typhi and V. parahaemolyticus, among which the activity against B. cereus was greatest. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of C. obtusa extracts were $30-40{\mu}l/ml$ for the 6 strains that showed an antimicrobial response by the paper disc agar diffusion method. These results suggest that C. obtusa extracts could serve as potential antibacterial agents to inhibit the growth of pathogens responsible for food poisoning and disease.

  • During the collection of boar semen, bacterial contamination usually occurs. The contamination has deleterious effects both on semen quality and on sow fertility. The majority of contaminants are gram-negative bacteria, especially Serratia marcescens. In this study, we developed a PCR assay for the identification of S. marcescens targeting the luxS gene (GenBank no. EF164926). S. marcescens yielded a specific 306 bp PCR product. However, no amplification was observed in the other strains tested. The detection limit of PCR was $50pg/{\mu}l$ of template DNA of S. marcescens. The antimicrobial susceptibility patterns of S. marcescens isolated from boar semen were tested using the disk diffusion method. Gentamicin, ceftiofur, florfenicol, and neomycin showed high sensitivity in this test. The minimum inhibitory concentration (MIC) was also determined by the broth microdilution method. The $MIC_{90}$ values of ceftiofur, enrofloxacin, gentamicin, and neomycin were 8, 8, 8, and $16{\mu}g/ml$, respectively. These results indicate that PCR amplification of the luxS gene is a reliable and effective method for the identification of S. marcescens and that ceftiofur, enrofloxacin, gentamicin, and neomycin are effective semen extenders for controlling S. marcescens.

  • The fruits of sweet cherry are highly appreciated by the consumer due to their precocity and quality, such as their sweetness, color and sourness. In this study, the hot-water extract and its sequential organic solvent fractions were prepared from domestic Napoleon sweet cherry (Prunus avium L.) to investigate antimicrobial and antioxidant activity. The hot-water extract contained about 40% sugars, and the solvent fraction yields for hexane, ethylacetate (EA), butanol, and water residue were 0.01%, 3.45%, 16.30%, and 80.24%, respectively. Contents of total polyphenol and total flavonoid of the fractions were 1.24~5.24%, and 0~3.76%, respectively. Among the fractions, EA fraction showed the highest total polyphenol and total flavonoid concentrations. Evaluation of antimicrobial activity of the extract and fractions revealed that EA fraction and butanol fraction contained strong antibacterial activity against Listeria monocytogenes, Staphylococcus aureus, and Salmonella typhimurium with minimal inhibitory concentration (MIC) of 0.5~1.0 mg/mL. But the extract and fractions tested were not active to Pseudomonas aeruginosa. In a while, only hexane fraction showed anti-Candida activity with 0.5~1.0 mg/mL of MIC. The fraction showed strong activity against different multi-antibiotics resistant strains such as C. albicans CCARM 14020. Antioxidative activity assay showed that EA fraction has a strong DPPH scavenging activity and a reducing power. The $IC_{50}s$ of vitamin E and EA fraction were 15.5 and $195.5\;{\mu}g/mL$, respectively. Our results suggest that the fruit of P. avium L. has high potentials with anti-Candida and antioxidative activity.

  • The antifungal activity of polygodial, which was isolated from Polygonum hydropiper used as a hot spice in Japan, was investigated through measuring its MICs and against food-contaminants including 4 strains of Saccharomyces cerevisiae, 3 strains of Zygosaccharomyces species, and 5 strains of Aspergillus species. All yeast-like fungi exhibited strong antifungal susceptivilities with MICs and MFCs below 3.13${\mu}g$/ml. Strains of Aspergillu species showed moderate with MIC of 25-50${\mu}g$/ml and MFC of 50-100${\mu}g$/ml. However these strains to sorbic acid, as already known, revealed very weak with MICs of 200-800${\mu}g$/ml and MFCs above 1600${\mu}g$/ml. In addition, effects of various pHs and temperatures on the antifungal activity of polygodial were tested against S. cerevisiae. At $4^{\circ}C$, the fungicidal activity of polygodial gradually increased with incubation time and reached the maximum(MFC of 3. 13${\mu}g$/ml) in 5 hours. At temperatures in the range of 30-$45^{\circ}C$, the activity of polygodial increased in proportion to temperature rise and in particular at $45^{\circ}C$ was possible with the concentration of 0.1${\mu}g$/ml. mOn the other hand, medium pH was also identified to affect the antifungal activity of polygodial. Namely, compared to neutral (pH7), acidic(pH 3) and basic(pH 9) medium synergized its activity (MIC and MFC) to 8- and 2- fold, respectively.

  • The antibacterial activity of ethanol (99.9%) extracts from marine micro-algae, namely, Mixed A (Pavlova sp., Thalassiosira weissflogii, Tetraselmis suecica and Isochrysis galbana were mixed with 1:1:1:1 ratio), Chlorella vulgaris, Nannochloropsis oculata and Chaetoceros calcitrans were estimated against food-borne bacteria, namely, Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus and Bacillus cereus. The extracts from these marine micro-algae showed potent antibacterial activity against all tested bacteria by the paper disk method. The extracts from C. vulgaris showed the strongest antibacterial activity against E. coli with minimum inhibitory concentration (MIC) of 0.62 mg/mL, and minimum bactericidal concentration (MBC) of 2.50 mg/mL. The extract from C. vulgaris contained 2 active compounds, 38.8% linoelaidic acid and 30.0% phytol. These results indicated that the ethanol extract from C. vulgaris may be a putative natural antibacterial agent against food-borne bacteria.

  • To develop the safe and natural antimicrobial agents, the 68 ethanol extracts from the 61 different kinds of oriental herbal medicine were prepared and their antimicrobial activities were evaluated. The herbal medicine used were from China (46 kinds), South Korea (14 kinds), North Korea (5 kinds) and Vietnam (3 kinds), respectively, and the root (27 species) was popular part in this study. The average water content and extraction ratio for ethanol were 7.10% and 6.75%, respectively. Determination of antimicrobial activity by disc-diffusion assay at 0.5 mg/disc concentration showed that the extract of Angelica tenuissima Nakai (china), Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum has strong antimicrobial activities against different food spoilage and pathogenic bacteria and fungi. Determination of MIC and MBC/MFC further showed that the extract of Syzygium aromaticum has MIC of 1.25 mg/mL and MBC/MFC of 1.25~5.00 mg/mL against Listeria monocytogenes, Bacillus subtilis, Staphylococcus epidermidis, Staphylococcus aureus, Salmonella typhimurium, Proteus vulgaris, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Saccharomyces cerevisiae. And, the extract of Junci medulla, Rhus javanica L. and Salvia miltiorrhiza Bunge showed strong antibacterial activities with MIC of 0.08~0.63 mg/mL and MBC/MFC of 0.08~2.50 mg/mL against the tested bacteria except E. coli and P. aeruginosa. In a while, the results of hemolytic activity of 68 different herbal extracts against human red blood cells showed that the extract of Angelica tenuissima Nakai has hemolytic activity at 0.5 mg/mL concentration. Therefore, Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum were finally selected for natural antimicrobial resources. Further research on active substances and the mode of action of the selected herbal medicine is necessary.

  • This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalis L. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH ($3.22\;{\mu}g/ml$) and ABTS ($5.05\;{\mu}g/ml$) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of $5.9{\pm}0.3\;{\mu}M/{\mu}g$, and buthanol fraction and rosmarinic acid exhibited values of $4.8{\pm}0.2\;{\mu}M/{\mu}g$ and $5.1{\pm}0.1\;{\mu}M/{\mu}M$, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureus and Escherichia coli to form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 mm. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.

  • To select the effective fungicides for the control of leaf spot disease of jujube tree (Zizyphus jujuba) caused by Phoma sp., inhibitory effects of 26 fungicides for mycelial growth were investigated at $250{\mu}g\;a.i./m{\ell}$. In the test, eight fungicides were selected and minimum inhibitory concentration (MIC) for mycelial growth and an inhibitory effect for spore germination were investigated. Among the fungicides, myclobutanil, hexaconazole, and triflumizole were excluded in control effect tests because of their relatively high MICs. MICs were ranged $10-50{\mu}g\;a.i./m{\ell}$ for benomyl, carbendazim + kasugamycin (CK), and thiophanate-methyl. triflumizole (TT), and $50-250{\mu}g\;a.i./m{\ell}$ for iprodione + propineb (IT) and iminoctadine-triacelate (IT). However, benomyl and IP showed very low inhibitory effect on conidial germination. When the fungicides were sprayed on the seedlings before the leaves were inoculated with conidial suspension of Phoma sp., the protective values of CK and TT were around 70% at 1,000 ppm and around 90% at 2,000 ppm. The protective values were around 70% at 2,000 ppm (benomyl), 4,000 ppm (IP), and 8,000 ppm (IT). When the fungicides were sprayed after inoculation, benomyl showed the highest curative values of over 90% at 1,000 ppm and the values of CK and TT ranged $70{\sim}80%$ at 1,000 ppm. However, IP and IT had little or no effect on therapy of the disease. IT caused necrotic phytotoxicity on the leaves of jujube seedlings. As results, the best fungicides for the protection of jujube trees from leaf spot disease were CK (2,000 ppm) and TT (2,000 ppm) and for the remedy of the tree, benomyl (1,000 ppm) was the best. Therefore, alternate application of benomyl and CK or TT will be effective in the disease control.
