평지방막에 융합된 골격근의 single ATP-sensitive K+ channel의 이온투과성에 대한 연구 (Permeability properties of skeletal muscle ATP-sensitive K+ channels reconstituted into planar lipid bilayer)

  • Properties of unitary ATP-sensitive $K^+$ channels were studied using planar lipid bilayer technique. Vesicles were prepared from bullfrog (Rana catesbeiana) skeletal muscle. ATP-sensitive $K^+$ (K (ATP)) channels were identified by their unitary conductance and sensitivity to ATP. In the symmetrical solution containing 200mM KCI, 10mM Hepes, 1mM EGTA and pH 7.2, single K (ATP) channels showed a linear current-voltage relations with slight inward rectification. Slope conductance at reversal potential was $60.1{\pm}0.43$ pS(n=3)). Micromolar ATP reversibly inhibited the channel activity when applied to the cytoplasmic side. In the range of -50~+50 mV, the channel activity was not voltage-dependent, but the channel gating within a burst was more frequent at negative voltage range. Varying the concentrations of external/internal KCl(mM) to 40/200, 200/200, 200/100 and 200/40 shifted reversal potentials to $-30.8{\pm}2.9$(n=3), $-1.1{\pm}2.7$(n=3), 10.5 and 30.6(mV), respecrivety. These reversal potentials were close to the expected values by the Nernst equation, indicating nearly ideal selectivity for $K^+$ over $Cl^-$. Under bi-ionic conditions of 200mM external test ions and 200mM internal $K^+$, the reversal potentials for each test ion/K pair were measured. The measured reversal potentials were used for the calculation of the releative permeability of alkali cations to $K^+$ ions using the Goldman-Hodgkin-Katz equation. The permeability sequence of 5 cations relative to $K^+$ was $K^+$(1), $Rb^+$(0.49), $Cs^+$(0.27), $Na^+$(0.027) and $Li^+$(0.021). This sequence was recognized as Eisenman's selectivity sequence IV. In addition, modelling the permeation of $K^+$ ion through ATP-sensitive $K^+$ channel revealed that a 3-barrier 2-site multiple occupancy model can reasonably predict the observed current-voltage relations.

Operators in L(X,Y) in which K(X,Y) is a semi M-ideal

  • Since Alfsen and Effors [1] introduced the notion of an M-ideal, many authors [3,6,9,12] have worked on the problem of finding those Banach spaces X and Y for which K(X,Y), the space of all compact linear operators from X to Y, is an M-ideal in L(X,Y), the space of all bounded linear operators from X to Y. The M-ideal property of K(X,Y) in L(X,Y) gives some informations on X,Y and K(X,Y). If K(X) (=K(X,X)) is an M-ideal in L(X) (=L(X,X)), then X has the metric compact approximation property [5] and X is an M-ideal in $X^{**}$ [10]. If X is reflexive and K(X) is an M-ideal in L(X), then K(X)$^{**}$ is isometrically isomorphic to L(X)[5]. A weaker notion is a semi M-ideal. Studies on Banach spaces X and Y for which K(X,Y) is a semi M-ideal in L(X,Y) were done by Lima [9, 10].

ON WEAKLY 2-ABSORBING PRIMARY SUBMODULES OF MODULES OVER COMMUTATIVE RINGS

  • Assume that M is an R-module where R is a commutative ring. A proper submodule N of M is called a weakly 2-absorbing primary submodule of M if $0{\neq}abm{\in}N$ for any $a,b{\in}R$ and $m{\in}M$, then $ab{\in}(N:M)$ or $am{\in}M-rad(N)$ or $bm{\in}M-rad(N)$. In this paper, we extended the concept of weakly 2-absorbing primary ideals of commutative rings to weakly 2-absorbing primary submodules of modules. Among many results, we show that if N is a weakly 2-absorbing primary submodule of M and it satisfies certain condition $0{\neq}I_1I_2K{\subseteq}N$ for some ideals $I_1$, $I_2$ of R and submodule K of M, then $I_1I_2{\subseteq}(N:M)$ or $I_1K{\subseteq}M-rad(N)$ or $I_2K{\subseteq}M-rad(N)$.

GENERAL TYPES OF (α,β)-FUZZY IDEALS OF HEMIRINGS

  • W. A. Dudek, M. Shabir and M. Irfan Ali discussed the properties of (${\alpha},{\beta}$)-fuzzy ideals of hemirings in [9]. In this paper, we discuss the generalization of their results on (${\alpha},{\beta}$)-fuzzy ideals of hemirings. As a generalization of the notions of $({\alpha},\;\in{\vee}q)$-fuzzy left (right) ideals, $({\alpha},\;\in{\vee}q)$-fuzzy h-ideals and $({\alpha},\;\in{\vee}q)$-fuzzy k-ideals, the concepts of $({\alpha},\;\in{\vee}q_m)$-fuzzy left (right) ideals, $({\alpha},\;\in{\vee}q_m)$-fuzzy h-ideals and $({\alpha},\;\in{\vee}q_m)$-fuzzy k-ideals are defined, and their characterizations are considered. Using a left (right) ideal (resp. h-ideal, k-ideal), we construct an $({\alpha},\;\in{\vee}q_m)$-fuzzy left (right) ideal (resp. $({\alpha},\;\in{\vee}q_m)$-fuzzy h-ideal, $({\alpha},\;\in{\vee}q_m)$-fuzzy k-ideal). The implication-based fuzzy h-ideals (k-ideals) of a hemiring are considered.

Roles of mTOR and p-mTOR in Gastrointestinal Stromal Tumors

  • Objective: This study aimed to examine the relationship between expression of mammal target of rapamycin (mTOR) and phosphorylation of mTOR (p-mTOR) protein in the PI3K/Akt/mTOR signaling pathways in gastrointestinal stromal tumors and relatiuonships with clinical factors. Methods: Immunohistochemistry was used to detect the expression of the associated proteins mTOR, p-mTOR, and phosphorylation of the tumor suppressor genes PTEN, P27, VEGF, and EGFR in 40 cases of gastrointestinal stromal tumors, with division into a very low and low risk group as well as a moderate and high risk group. Results: The positive rate of mTOR and p-mTOR was significantly increased in the moderate and high risk group compared with the very low and low risk group. The difference was statistically significant (P<0.05). When grouped according to size, the positive mTOR expression rate exhibited a statistical difference (P<0.05), which was significantly increased in the group of tumors larger than 5 cm. The difference in the positive mTOR and p-mTOR expression rate exhibit no statistical significance among the PTEN, P27, VEGF, and EGFR expression subgroups (P>0.05). Conclusion: The different expressions of mTOR and p-mTOR in the signal transduction pathway of gastrointestinal stromal tumor in the different degree-of-risk groups suggested that the mTOR and p-mTOR of the signal transduction pathway serve an important function in the occurrence and development of gastrointestinal stromal tumors.

Toxoplasma gondii: Ultrastructural localization of specific antigens and inhibition of intracellular multiplication by monoclonal antibodies

  • This experiment was focused on the characterization of anti- Toxoplasma monoclonal antibodies (mAbs) and the effect of mAbs on the parasite invasion of mouse peritoneal macrophages. Twenty eight mAbs including M110, M556, R7A6 and M62l were characterized by Ab titer, immunoglobulin isotyping and western blot pattern. Antibody titer (optical density) of 4 mAbs. Ml 10. M556. R7A6 and M62l. were 0.53,0.67, 0.45 and 0.39 (normal mouse serum; 0.19) with the same IgGl isotypes shown by Enzyme-linked immunosorbent assay (ELISA). Western blot analysis showed that Ml 10. M556. R7A6 and M62l reacted with the 33 kDa (p30),31 kDa (p28),43 kDa and 36 kDa protein. Immuno-gold labelling of mAbs M110, M556, R7A6 and M621 reacted with the surface membrane, dense granules and parasitophorous vacuolar membrane (PVM) , rhoptries and cytoplasm of tachyzoite, respectively. For in vitro assay, preincubation of tachyzoties with four mAbs, Ml 10, M556, R7A6 and M62l resulted in the decrease of the number of infected macrophages (p < 0.05) and the suppression of parasite multiplication at 18 h post-infection. Four monoclonal antibodies including Ml 10 (SAGI) were found to have an important role in the inhibition of macrophage invasion and T. gondii multiplication in vitro, and these mAbs may be suitable for vaccine candidates, diagnostic kit and for chemotherapy.

Comparison of Various Single Chemical Extraction Methods for Predicting the Bioavailability of Arsenic in Paddy Soils

  • The Codex Committee of Contaminants in Food (CCCF) has been discussing a new standard for arsenic (As) in rice since 2010 and a code of practice for the prevention and reduction of As contamination in rice since 2013. Therefore, our current studies focus on setting a maximum level of As in rice and paddy soil by considering bioavailability in the remediation of As contaminated soils. This study aimed to select an appropriate single chemical extractant for evaluating the mobility of As in paddy soil and the bioavailability of As to rice. Nine different extractants, such as deionized water, 0.01 M $Ca(NO_3)_2$, 0.1 M HCl, 0.2 M $C_6H_8O_7$, 0.43 M $HNO_3$, 0.43 M $CH_3COOH$, 0.5 M $KH_2PO_4$, 1 M HCl, and 1 M $NH_4NO_3$ were used in this study. Total As content in soil was also determined after aqua regia digestion. The As extractability of the was in the order of: Aqua regia > 1 M HCl > 0.5 M $KH_2PO_4$ > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.1 M HCl > 0.43 M $CH_3COOH$ > deionized water > 1 M $NH_4NO_3$ > 0.01 M $Ca(NO_3)_2$. Correlation between soil extractants and As content in rice was in the order of : deionized water > 0.01 M $Ca(NO_3)_2$ > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.5 M $KH_2PO_4$ > 1 M $NH_4NO_3$ > 0.2 M $C_6H_8O_7$ > 0.43 M $HNO_3$ > 1M HCl > Aqua regia. BCF (bioconcentration factor) according to extractants was in the order of : 0.01M $Ca(NO_3)_2$ > 1 M $NH_4NO_3$ > deionized water > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.5 M $KH_2PO_4$ > 1 M HCl > Aqua regia. Therefore, 0.01 M $Ca(NO_3)_2$ ($r=0.78^{**}$) was proven to have the greatest potential for predicting As bioavailability in soil with higher correlation between As in rice and the extractant.

Moderate Elevation of Extracellular $K^+$ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries: Role of Na Pump and Ba-Sensitive Process

  • Cerebral blood vessels relax when extracellular $K^+$ concentrations $([K^+])_e$ are elevated moderately $(2{\sim}15$ mM, $K^+-induced$ vasorelaxation). We have therefore studied the underlying mechanism for this $K^+-induced$ vasorelaxation in the isolated middle cerebral arteries (MCAs). The effects of ouabain and $Ba^{2+}\;on\;K^+-induced$ vasorelaxation were examined to determine the role of sodium pump and/or Ba-sensitive process (possibly, inward rectifier K current) in the mechanism. Mulvany myograph was used to study 24 rats, 18 rabbits, and 10 humans MCAs $(216{\pm}3\;{\mu}m,\;347{\pm}7\;{\mu}m,\;and\;597{\pm}39\;{\mu}m$ in diameter when stretched to a tension equivalent to 55 mmHg). High $K^+$ (125 mM) and $PGF_{2{\alpha}}\;(1{\sim}10\;{\mu}M)$ induced concentration-dependent contractions in all 3 species, while histamine $(10{\sim}50\;{\mu}M)$ evoked contraction only in the rabbits and induced relaxation in the rats and humans. Addition of $K^+\;(2{\sim}10\;{\mu}M)$ to the control solution induced vasorelaxations. These effects were inhibited by the pretreatment with both ouabain $(10\;{\mu}M)$ and $Ba^{2+}\;(0.1{\sim}0.3\;mM)$ in the rat, but only with ouabain $(10\;{\mu}M)$ in the rabbit and human. These results suggest that $K^+-induced$ vasorelaxation occurs via the stimulation of electrogenic Na pump in the rabbit and human MCAs, while in the rat MCAs via the activation of both Na pump and Ba-sensitive process.

  • Let $\mathcal{K}^{\prime}_M$ be the generalized tempered distributions of $e^{M(t)}$-growth, where the function M(t) grows faster than any linear functions as ${\mid}t{\mid}{\rightarrow}{\infty}$, and let $K^{\prime}_M$ be the Fourier transform spaces of $\mathcal{K}^{\prime}_M$. We obtain the relationship between certain classes of analytic functions in tubes, $\mathcal{K}^{\prime}_M$ and $K^{\prime}_M$.

  • For an irreducible binomial polynomial $f(x)=x^n-b{\in}K[x]$ with a field K, we ask when does the mth iteration $f_m$ is irreducible but $m+1th\;f_{m+1}$ is reducible over K. Let S(n, m) be the set of b's such that $f_m$ is irreducible but $f_{m+1}$ is reducible over K. We investigate the set S(n, m) by taking K as the rational number field.
