북방대합, Spisula sachalinensis의 산란유발 및 난발생 (Spawning Induction and Egg Development of Surf Clam, Spisula sachalinensis)

  • 한해성 패류인 북방대합을 대상으로 자극방법별 산란유발 및 수온별 난발생 과정을 조사하였다. 모패에 대한 산란유발 자극은 $NH_4OH$ 첨가와 serotonin 주사가 효과적이었으며, 수컷이 암컷보다 민감하게 반응하였다. 반응개시 시간에 있어서 $NH_4OH$ 첨가시 방란은 $3\~4$시간, 방정은 1시간 전후에 개시되어 암수간의 차이가 있었다. serotonin 주사는 암수 모두에서 5분 이내에 방란${\cdot}$방정을 유도하였다. 방출된 알의 수는 $NH_4OH$ 첨가에서 유의하게 많았다. 그러나 serotonin 주사에 의해 산란유발된 알의 GVBD율, 수정률 및 부화율은 $NH_4OH$ 첨가에 의한 알보다 높았다. 방출된 알은 평균 난경 $65.2{\pm}1.8\;{\mu}m$인 분리 침성란으로 GVBD 이전에도 수정이 가능하였으며, 적정 발생수온은 $15\~20^{\circ}C$였다. 초기 D상 유생까지는 $15^{\circ}C$에서 42시간, $20^{\circ}C$에서 27시간이 소요되었다.

산화적 스트레스에 대한 고온고압처리 인삼의 억제 효과 (Inhibitory Effects of Panax ginseng C. A. Mayer Treated with High Temperature and High Pressure on Oxidative Stress)

  • 체내에서 산화스트레스에 의해 생성되는 활성산소종(reacitve oxygen species, ROS)은 당뇨병, 비만 등과 같은 만성질환을 야기시키는 것으로 알려져 있다. 고려인삼(Panax ginseng)은 수천 년간 피로 회복 및 면역증강용 기능성식품으로 이용되어 왔고, 사포닌, 산성다당체, 페놀성 화합물과 같은 다양한 생리활성 물질을 함유하고 있다. 따라서 본 연구에는 고온, 고압처리하여 제조한 신규 인삼에 대한 산화적 스트레스 저감 효능을 평가하고자 하였다. C2C12 근육세포에 산화적 스트레스를 유도하기 위해 $H_2O_2$ 1 mM 처리하고, 전처리 조건을 달리한 인삼 시료를 처리하여 cell morphology 및 항산화 관련 유전자인 SOD, CAT 및 GPx를 살펴보았고, 3T3-L1 지방세포는 분화과정 중 ROS 생성 억제효과 및 CAT, GPx 및 Cu/Zn-SOD의 항산화효소 관련 유전자의 발현 정도를 조사하였다. 고온, 고압처리한 인삼은 산화적 스트레스가 유도된 C2C12 근육세포 및 3T3-L1 지방세포에서 유의적으로 산화적 스트레스를 저감하는 것으로 나타났다. 이상의 결과로 보아, 본 연구진에 의해 개발된 고온 및 고압 처리된 인삼은 항산화 및 항피로 효능이 기대되는 바이며, 본 연구는 동물세포 수준에서의 비교이며, 보다 정확한 작용기전의 구명을 위해 향후 추가적인 연구를 통한 비교 실험이 수행되어야 할 것으로 사료된다.

PTU(6-n-propyl-2-thiouracil)로 유발된 Rat 갑상선 기능저하증에 미치는 烏梅의 효과 (Effects of Mume Fructus on the Rat Hypothyroidism Induced by PTU(6-n-propyl-2-thiouracil))

  • Objectives : The aim of this study was to evaluate the effect of aqueous extracts of Mume Fructus(MF) on the 6-n-propyl-2-thiouracil(PTU)-induced rat hypothyroidism.Methods : Aqueous extracts of MF(yield = 19.38%) were administered, once per day for 42 days from 2 weeks before starting of PTU treatment as an oral dose of 300 and 150 ㎎/㎏(body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 ㎎/㎏ for 28 days. The changes in the body weight, thyroid gland weights, liver weight, serum levels of thyroid hormone-thyroid stimulating hormone(TSH), tri-iodothyronine(T3) and thyroxine(T4), total cholesterol, low density lipoprotein(LDL), high density lipoprotein(HDL) and triglyceride (TG), aspartate aminotransferase(AST), alanine aminotransferase(ALT), liver antioxidant defense system-lipid peroxidation, H2O2, superoxide dismutase(SOD) and catalase(CAT) were examined with histopathology of thyroid glands and liver.Results : Results were compared with LevoT40.5 ㎎/㎏ treated rats. MF extracts recovered from the decreases in the body weight, liver weight, T3and T4, TG, liver CAT activities as results of PTU treatment. And MF extracts recovered from the increases of thyroid gland weights, TSH, HDL contents, liver H2O2, AST as results of PTU treatment. In addition, these PTU-induced histopathological changes in thyroid glands and liver related to hypothyroidism were dramatically decreased by treatment of both different dosages of MF extract, respectively.Conclusions : According to the above results, it is suggested that MF extracts have advantageous effects on the thyroid hormone productions with beneficial effects on the hypothyroidism related liver injuries mediated by the modulation on the antioxidant system.

옻나무 추출물의 침지가 가스조절포장으로 저장한 한우고기 스테이크의 총환원력, 단백질산화 및 육색소산화에 미치는 영향 (Effect of Immersion in Rhus verniciflua Stokes Extract on the Total Reducing Ability, Protein Oxidation and Myoglobin Oxidation of Hanwoo Beef Steak Stored with Modified-atmosphere Packaging)

  • 본 연구는 옻나무 추출물(Rhus verniciflua Stokes extract, RVSE)의 침지가 가스조절포장(modified-atmosphere packaging, MAP)으로 저장한 한우고기 스테이크의 총환원력, 단백질산화 및 육색소산화에 미치는 영향을 구명하고자 실시하였다. 시료(Hanwoo longissimus muscle)는 옻나무 추출물의 침지 여부에 따라 처리한 후 high oxygen-MAP(HOMAP; 75% $O_2$+20% $CO_2$+5% $N_2$)와 low oxygen-MAP (LOMAP; 0% $O_2$+20% $CO_2$+80% $N_2$)으로 포장하여 $4^{\circ}C$에서 9일 동안 저장하였다. 총환원력은 RVSE의 침지에 의해 증가되었으며(p<0.05), LOMAP에 의해 저장기간 동안 높게 유지되었다. Carbonyl 함량은 RVSE의 침지와 LOMAP에 의해 억제되었다. Metmyoglobin의 축적은 RVSE의 침지에 의해 증가되었으나, HOMAP에 의해 낮게 유지되었다. 이와 반대로 oxymyoglobin에 의한 적색 강도의 지표인 R630-R580은 RVSE의 침지에 의해 감소되었던 반면, HOMAP에 의해 높게 유지되었다. 명도($L^*$), 적색도($a^*$) 및 chroma($C^*$)는 RVSE의 침지에 의해 감소되었으나, HOMAP에 의해 높게 유지되었다. 반면에 hue-angle($H^{\circ}$)은 RVSE의 침지에 의해 증가되었으나, HOMAP에 의해 낮게 유지되었다. 따라서 RVSE의 침지는 한우고기 스테이크의 총환원력과 단백질산화 안정성을 증가시켰으나, 육색소산화 안정성 감소시켰다. 또한 HOMAP는 총환원력과 단백질산화 안정성을 감소시켰으나, 육색소산화 안정성은 증가시켰다.

홍미삼 에탄올 추출분획의 항산화 활성 (Antioxidative Activity of Ethanol Extraction Fraction from the Korean Red Tail Ginseng)

  • 본 연구의 목적은 홍미삼의 기능성 연구의 일환으로 총 페놀성 화합물의 추출방법과 DPPH에 대한 수소공여능, linoleic acid의 산화방지활성 및 LDL에 의한 항산화활성 등을 중심으로 연구한 결과이다. 홍미삼에 60% 에탄올를 첨가하여 추출회수별 총 페놀성 화합물을 조사한 결과 3회까지 추출했을 때 대부분이 추출되었기 때문에 추출횟수는 3회가 적당한 것으로 판단되었다. 60% 에탄올 추출액의 수율은 3회까지 추출했을 때 약 37.35%가 추출되었다. 추출 용매에 따른 총 페놀성 화합물의 추출 효율은 물 추출구를 100%로 했을 때, 60% 에탄올 용액으로 추출한 뒤 농축하여 물로 부피를 재조정한 시험구는 122%로 조사되었다. 항산화활성 조사에서 DPPH에 대한 수소공여능에서 시간이 경과함에 따라 항산화 활성이 약간씩 증가하나 활성은 약한 편이었다. Linoliec acid 산화에 대한 산화억제 효과는 1,500 ppm 농도에서 약 72.23%의 저해율을 나타났으며, LDL에 대한 산화방지 효과는 250 ppm 농도에서 그 저해율이 약 22.52%로 나타났다.

Protective Activity against Ionizing Radiation of Antioxidative Plants Indigenous to Korea

  • We have screened the cytoprotective effect on ${\gamma}-ray$ radiation induced oxidative stress from forty one Korean plant extracts. Carpinus laxiflora (caulis), Quercus salicina (caulis), and Castanopsis cuspidata (caulis) were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of three plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by ${\gamma}-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Carpinus laxiflora, Quercus salicina, and Castanopsis cuspidata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

  • A lean alloy is defined as a low alloy steel with a minimum amount of the alloying element that maintains the characteristics of the sintered alloy. It is well known that the addition of elements such as Cr, P, Si, or Mn improves the mechanical characteristics of the alloy, but decreases the sinterability. The mother alloy is used to avoid an oxidation reaction with the alloying elements of Cr, P, Si or Mn. The purpose of this study is to determine the change in the mechanical properties of Fe-P-Mo and Fe-P-Mn alloys as a result of the addition of Si. In this article, the Fe-P-Mo and Fe-P-Mn alloys to which Si is added are compacted at $7.0g/cm^3$ and then sintered in $H_2-N_2$ at $1120^{\circ}C$. The P around the macropores and large grains reduces due to the formation of $SiO_2$ as the Si content increases. This is caused by the increase in strength owing to reducing intergranular fracture by suppressing the reaction with oxygen.

Antioxidant and Hepatoprotective Effects of Tomato Extracts

  • The objective of present study was to investigate the anti oxidative and hepatoprotective effects of tomato extracts. Total antioxidant capacity and total antioxidant response were 5.5 and $19.8{\mu}g$ Trolox equivalent per mg of tomato extract, respectively. DPPH radical scavenging activity of tomato extracts ($10mg\;ml^{-1}$) was 70% as compared to 100% by pyrogallol solution as a reference. The effect of the tomato extracts on lipid peroxidation was examined using rat liver mitochondria induced by iron/ascorbate. Tomato extracts at the concentration of $0.5mg\;ml^{-1}$ significantly decreased TBARS concentration. Tomato extracts prevented lipid peroxidation in a dose-dependent manner. The effect of the tomato extracts on reactive oxygen species (ROS) generation was examined using cell-free system induced by $H_2O_2/FeSO_4$. Addition of $1mg\;ml^{-1}$ of tomato extracts significantly reduced dichlorofluorescein (DCF) fluorescence. Tomato extracts caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that tomato extracts significantly prevented ROS generation in vitro. The effect of tomato extracts on cell viability and proliferation was examined using hepatocyte culture. Primary cultures of rat hepatocytes were incubated with 1mM tert-butyl hydroperoxide (t-BHP) for 90 min in the presence or absence of tomato extracts. MTT values by addition of tomato extracts at the concentration of 2, 10, and $20mg\;ml^{-1}$ in the presence of t-BHP were 13, 33 and 48%, respectively, compared to 100% as control. Tomato extracts increased cell viability in a dose-dependent manner. These results demonstrate that tomato extracts suppressed lipid peroxidation and t-BHP-induced hepatotoxicity and scavenged ROS generation. Thus antioxidant and hepatoprotective effects of tomato extracts seem to be due to, at least in part, the prevention from free radicals-induced oxidation, followed by inhibition of lipid peroxidation.

Methanol 자화세균에 의한 아미노산의 생성에 관한 연구 (Studies on the Production of Amino Acids by Methanol-Utilizing Bacteria)

  • About 600 strains of bacteria which can utilize methanol as the sole source of carbon were isolated from natrual sources, and their ability to produce amino acids on the medium containing methanol as the sole source of carbon was tested by paper chromatography, by which 3 strains were screened by virtue of their relatively shperior amino acid production. These were tentatively identified as follows ; one strain belonged to the genus methylomonas, while the remaining two were members of the genus Hyphomicrobium. Further studies on their growth characteristics were carried out with relation to amino acid production. According to these experiments, the following results were obtained. 1) No vitamin added was effective for the growth enhancement. 2) Optimum initial concnetration of methanol was proved to be one per cent (v/v), and no conspicuous effect of feeding methanol on the growth of these bacteria ould be observed, provide that the initial methanol concentration was controlled to 1 per cent. 3) Among the nitrogen sources tested, 0.1 per cent of ${(NH_4)}_2SO_4$ enabled the best growth. 4) Addition of 0.001 per cent $MgSO_4.$ $7H_2O$ largely increased the growth of all three strains. Other metal ions tested were either not effective or strongly inhibitory for the growth. 5) Addition of 0.1 per cent each of yeast extract, corn steep liquor, beef extract, and peptone also increased the growth markedly. 6) About $75{\mu}$g/ml of alanine could be obtained finally after 64 hr culture of Hypm. A on the medium lcontaining 0.1 per cent yeast extract and 1 percent methanol. Approximately $40.{\mu}$g/ml of glutamic acid by Hypm. GA and $43.{\mu}$g/ml of alanine by Mtlm. A were produced at the same culture conditions.
